Ⅰ. 제 목 탄수화물 전이효소의 반응과 조절연구 Ⅱ. 연구개발의 목적 및 필요성 가. 연구개발의 경제․사회․기술적 필요성 ◦기술적 측면 현재 화학반응에 의한 탄수화물의 합성범위는 한정되어 있어서 단순한 화합물을 합성하는 수준에 머무르고 있다. 반면, 생체 내에서는 복잡한 탄수화물은 물론 고분자화합물까지 일사불란하게 합성되고 있다. 이는 생체의 효소와 생체 합성계가 합성정보에 따라 생체탄수화물과 그 관련물질을 합성할 수 있음을 알 수 있다. 이러한 견지에서 탄수화물의 합성과 이용에 관련된 효소반응의 중요성이 인식되고 있고있다. 그러나, 최근까지 생물이 분비하는 가수분해효소(加水分解酵素)와 식물씨앗의 발아시 생성되는 탄수화물 가수분해효소 등 일부의 효소활용에 국한되어 왔다. 그 이유는 대부분의 효소가 생체 내에 극미량 존재하므로 실제 활용하기에 충분한 양을 확보할 수 없었다. 최근에 와서 분자생물학의 발달로 신기능 효소의 탐색이 용이해졌고, 효소의 유전자정보만 알면 어떠한 효소도 대량 제조할 수 있는 유전공학기술(遺傳工學技術)이 정착되었다. 이는 효소를 이용한 생물산업의 커다란 전환기가 마련된 셈이다. 이러한 흐름에 편승하여 최근에는 미국, 일본, 유럽 등에서 효소와 생체반응을 이용하는 탄수화물공학(炭水化物工學)이라는 대규모 연구사업으로 이어지고 있다. 더욱이 생체의 세포 내에서는 복합 탄수화물의 생합성이 탄수화물 전이효소에 의하여 이루어진다. 그러나 현재 생명공학기술측면에서 복합 탄수화물을 대량 제조할 수 있는 합성방법이 정립되지 못한 실정이다. 이러한 측면에서 탄수화물전이효소의 반응연구는 기능성 생체탄수화물의 합성뿐만 아니라, 효소저해제 등을 선별하여 의약품개발에까지 이어질 수 있어서 시급히 정립되어야할 연구분야이다. Helicobactor pylori의 세포표면은 fucose가 부착되므로서 인체 위벽세포를 인식하고 위궤양 등의 만성질환을 유발하는 것으로 알려졌다. 본 연구에서는 한국인 유래 Helicobactor pylori의 fucosyltransferase를 유전공학적으로 생산하고, 이 효소를 이용하여 복합기능당의 합성기술을 개발하는 것이며 fucosyltransferase의 효소저해제를 유기화학적으로 합성하여 그 저해제의 성능을 검증하는 연구를 수행할 계획이다. ◦경제. 산업적 측면 생명과학의 추진과 발달은 유전공학, 단백질공학, 효소공학 등의 새로운 기술이 등장하면서 탄수화물활용의 잠재력과 실상이 현실로 나타나기 시작하였다. 또한, 최근에 생명과학의 발달로 탄수화물이 단순한 생체구조물질과 에너지원으로 뿐 아니라 생체기능의 신호전달과 인식 등의 신호전달물질로서의 중요한 역할이 알려지고 있다. 이러한 복합 탄수화물의 생합성과 그 생체기능 이해는 기능성 생물소재의 산업적인 활용 뿐 아니라 암종양과 같은 질병의 억제제 및 치료제로서 새로이 부상하고있는 신약연구영역이다. 더욱 중요한 재인식은 생체가 필요로 하는 생체화합물과 생체고분자들의 초기 및 중간체가 탄수화물에서부터 효소화학반응(酵素化學反應)에 의하여 합성되어가고 있다는 사실이다. 이러한 측면에서 탄수화물은 생체내의 기능 측면뿐 아니라 생물산업과 화학산업의 가장 중요한 출발물질이 될 수 있고 의약품과 같은 고도의 정밀화합물의 잠재력을 내포하고 있다고 말할 수 있다. 특히, 본 연구에서 개발목표로 하는 복합기능성 탄수화물의 효소합성기술과 fucosyltransferase 저해제 탐색은 신약개발로 이어져서, 동양인 다수가 감염되어 있는 Helicobactor pyrori의 퇴치에 큰 역할을 할 것으로 본다. ◦사회. 문화적 측면 1980년대에 들어 유전공학이 등장하여 생명과학의 핵심적인 연구수단을 제공하였고, 이어서 등장한 탄수화물공학은 새로운 생명공학의 연구방향을 제시하고 있다. Ⅲ. 연구개발의 내용 및 범위 본 연구과제 수행에서는 1차년도, 2차년도 및 3차년도에 걸쳐서??장균에서 효소를 정제하고 효소의 생체외에서의 특성을 확립하는 것이다. 또한, 효소에 대한 반응조건을 확립하여 인간위벽세포에 존재하는 Helicobacter pyroli의 생체내 다양한 활성중 이 효소로부터 GDP-fucose에서 glycoconjugate의 합성 최종단계에 fucose가 부가됨으로서 암의 발생이나 전이에 영향을 준다는 최근의 보고를 토대로 위암의 진단이나 치료와 연관하여 전이효소의 억제제를 유기합성을 통해 개발하고 생물학적인 활성을 연구하는 것이다. 화학 합성으로 새롭게 효소 억제제를 이용하여 fucosyltransferase 전이 활성을 효소 반응속도론적인 측면에서 이를 검증하였다. Ⅳ. 연구개발결과 1. 한국형 Helicobacter pylori에서 유래한 두 종류의 fucosyltransferase의 클로닝과 발현 두 종류의 fucosyltransferase 유전자를 한국형 Helicobacter pylori (strain 51과 137)에서 클로닝하였다. 이 두 효소의 유전자는 사람, 쥐, 닭에서 유래한 fucosyltransferase (FucT)의 상보 DNA와 비교하여 21개의 염기를 각각 strain 57에서 6번, strain 137에서 5번 반복하는 특이한 구조를 지니고 있었다. 원핵생물에서 라이보좀 (ribosome)의 결합 부위인 Shine-Dalgarno (SD) 서열이 전사 (translation) 개시의 앞쪽에 위치하였으며 일반적으로 알려진 -10 과 -35의 위치도 확인하였다. Strain 51에서 유래한 유전자는 H. pylori 26695 균주에서 이미 알려진 cytochrome C biogenesis 단백질의 앞쪽으로 (5‘ up-stream) 위치하였다. 유추된 아미노산의 서열은 signal peptide나 transmembrane domain을 지니고 있지 않았다. 반복된 서열은 단백질의 c 말단의 42 아미노산에 위치하였다. 아미노산인 leucine이 진핵생물에서 잘 알려진 leucine zipper motif와 유사한 7개의 아미노산의 간격으로 나타나고 있다. 이 두 전이효소는 사람이나 소에서 유래한 효소와 비교해 볼 때 유사성이 없었으나 활성에서 중요하다고 예상되는 보존된 위치에서 유사성을 발견할 수 있었다. 한편, 이미 보고된 H. pylori α-1,3-fucosyltransferase와이 두 유전자 공히 70% 이상의 유사성을 지니고 있었으며, 이는 이 효소들이 α-1,3-fucosyltransferase와 동일한 활성을 지닐 것으로 예측되고 있다. 이 두 유전자를 발현벡타 pKK223-3에 각각 클로닝하고 IPTG 의한 유도를 통해 발현시켰다. 2. 재조합 효소의 정제와 활성 측정 유전자 클로닝을 통하여 확보한 α-1,3-fucosyltransferase에 대한 생물학적 정보를 알아보기 위하여 일반적인 칼럼크로마토그래피를 이용하여 정제하였다. 배양된 대장균은 초음파 파쇄기를 통하여 strain 57과 137에서 유래한 대장균을 파쇄하고 (NH4)2SO4 분별침전한 후 투석된 시료를 DEAE-Sephacel 크로마토그래피를 이용하여 정제하였다. 여기서 얻은 효소가 N-acetyllactosamine의 3'-OH 잔기에 fucose를 전이하는 활성을 보여 주었다. 3. α-1,3-fucosyltransferase 억제제의 화학 합성 생체내의 다양한 활성중 fucose가 전이됨으로서 암의 발생이나 전이에 영향을 준다는 여러 보고가 있어왔다. 본 연구는 위암의 발생에 원인균이라 알려진 H. pylori에서 fucosyltransferase의 유전자를 클로닝하였으며, 위암의 진단이나 치료와 연관하여 이 전이효소의 억제제를 개발하는 방향으로 연구에 목적을 두고 있다. 이러한 억제제는 국제 공동으로 중국의 상해 유기화학 연구소에서 화학합성하고 일부는 본 연구실에서 합성하였다. 4. α-1,3-fucosyltransferase 억제제의 저해 합성된 저해제 중 6가지를 선별하여 α-1,3-fucosyltransferase 활성 억제메카니즘을 효소속도론적인 측면에서 확인하였다. 효소반응의 acceptor인 N-acetyllactosamine을 고정시킨 후 donor인??모델을 분석한 결과, 6가지 저해제 모두 경쟁적인 저해반응 모델에 적합하였고, 결과적으로 donor인 GDP-fucose를 기질로 모사한 저해제가 α-1,3-fucosyltransferase의 반응모델인 ordered, sequencial Bi-Bi 메카니즘에서 초기 효소와 GDP-fucose가 유사상태 전이과정인 <효소-GDP-fucose>로 이동해가는 과정을 경쟁적으로 저해하여 <효소-저해제>가 생기도록 함으로써 효소반응의 초기단계를 억제하였다. Ⅴ. 연구개발결과의 활용계획 본 연구 결과, 한국형 Helicobacter pylori에서 분리한 α-1,3-fucosyltransferase를 클로닝하고 발현시킴으로서 새로운 암전이 억제를 차단할 수 있는 의약품 개발에 활용할 수 있으며, 다양한 억제제에 대한 분석을 통해, 새로운 억제제의 개발에 대한 정보를 충분히 제공해줄 것으로 생각된다.

Initially, it was assumed that the oligosaccharides serve as energy stock and structural materials. With the development of molecular biology, it is well known that oligosaccharides are mediated in every stage of biological phenomena. The glycoconjugates found on cell surface membranes, in which the oligosaccharide residues are exposed, play important roles in intercellular recognition and interaction. Over the past decade, a large number of evidence has been accumulated, indicating that the glycoconjugates become altered during abnormal cellular development. Carbohydrate structures of glycoproteins and glycolipids as differentiation antigens, tumor-associated antigens, and components of receptor systems, compared with normal one. Among the abnormal alterations, their characteristic shifted toward the synthesis and expression of large, high relative molecular mass, asparagine-linked oligosaccharides following neoplastic transformation. Transfection by human oncogenes. Concomitant induction of tumorigenicity and tumor-associated membrane alteration. One of the typical changes includes the occurrence of poly-fucosylated, repeated N-acetyllactosamine chain Novel fucolipids accumulating in human adenocarcinoma. These aberrant glycosylation processes result in consistent changes in cell surface carbohydrate chains and are known to accompany the development of human melanoma, neuroblastosma, and gastric, pancreatic carcinom. Aberrant glycosylation in cancer cell membranes as focused on glycolipids. Fucosyltransferases, that are responsible for transferring L-fucose from GDP-fucose to variety of oligosaccharide substrates, are involved in the last steps of biosynthesis of ABH and Lewis oligosaccharide as cell-surface antigen. Fucose is a component of glycoprotein, glycolipids and cellular oligosaccharides, and is commonly glycosidically linked to galactose (Gal), glucose (Glc) and N-acetylglucosamine (GlcNAc). Fucose is linked to these sugars via their 2, 3, 4 or 6 hydroxyl groups of substrates to produce a multitude of structures. For example, attachment of fucose to the 4-hydroxyl group of GlcNAc of type 1 core structure produces Lea blood group antigen (i.e. Galb1-3 (Fuca1-4)GlcNAc), whereas attachment of fucose to 2-hydroxyl of Gal, or to 3-hydroxyl group of GlcNAc of a Type 2 core structure produces the H or Lewis x blood group antigen, respectively. In addition to their roles in defining blood group antigens, fucosylated structures are expressed in a highly regulated fashion during embryonic development and tumorigenesis Stage-specific embryonic antigen involves α-1,3 fucosylated Type 2 blood group chains. The new structures that appear at the cell surface during these processes are referred to as tumor-associated antigens. Consequently, the majority of tumor-associated antigens are glycoconjugates in which carbohydrate moieties are altered. The functional significance, if any, of these structures is still unclear, but these aberrant oligosaccharide structures are already attracting a great deal of interest as potential tumor markers. Thus, in the last ten years, a great deals of studies focused on fucosyltransferases themselves that are responsible for synthesizing these structures and on discovering inhibitors of FucT that may prevent the aberrant glycosylation of glycoconjugates that accompanies cancers. This report will cover three major topics. In the first part, gene cloning and expression of α-1,3-L-fucosyltransferase (FucT) from H. pylori Korean-type strain 137 and 51 will be discussed. In the second part, biochemical characteristics of recombinant fucosyltransferase were examined. Last part describes candidates of FucT inhibitors, covering their design and synthesis, and their inhibitor assay. III. Result and Discussion 1. Molecular cloning of α-1,3-fucosyltransferase gene from H. pylori strain 51 and 137 In this study, two of the H. pylori α-1,3-fucosyltransferase genes have been cloned and expressed successfully. The two genes were consisted of 1371 base pair and 1329 base pair, respectively. Sequences analysis and expression of these two genes in E. coli indicated that they coded for proteins of ~51 kDa and ~53kDa, respectively. α-1,3-fucosyltransferase activity of them was assayed by in vivo biochemical characterization, which demonstrated the two of enzymes may use Type II core structure oligosaccharides, for example N-acetyllactosamine, as acceptors. The fact presented in this study strongly supports that the two enzymes represent for two novel a-1,3-fucosyltransferases from H. pylori Korean-type Strain 137 and 51. Although they shared little homology with FucT genes from human sources (less than 30%) in amino acid and in topology, they shared similarities to the genes from H. pylori. Unlike mammalian FucTs, the two FucTs do not contain a N-terminal transmembrane segment, a typical feature for mammalian FucTs, primarily consisting of hydrophobic residues. And to be similar to FucTs from H. pylori, the two FucTs contain a C-terminal region including 5, 6 direct repeats, respectively. The numbers of the repeats region of all H. pylori FucT genes vary from 2 to 10. The homology search of the region indicated that it might be leucine zipper motif. 2. Characterization of α-1,3-fucosyltransferase Metal ion manganese was known to be important for FucT to possess ability of transferring fucose to oligosaccharide. For different FucTs reported, the concentration of manganese ion varies a lot. So the factor to the activity of FucT from H. pylori strain 137 was studies. The concentration of manganese needed to achieve maximal activity was determined to be 2mM at fixed concentrations of both GDP-Fuc (1mM) and LacNAc (1mM). In Tris-HCl buffer, the maximal enzyme activity was obtained when pH of the reaction was 7.0. Moreover, in the pH range of 7.0-8.0, the Tris-HCl buffer could help the enzyme to achieve the highest activity. So the optimal pH for FucT standard assay is 7.0 while the Tris-HCl buffer is applied. The substrate specificity of the FucT was also determined. Apparently, the most preferred substrate for the enzyme is LacNAc, which was followed by Lacto-N-biose. The factor demonstrated that the FucT had abilities to transfer fucose both to Type I oligosaccharides to form 1-4 linkage, and to Type II oligosaccharide to form 1-3 linkage. Even the novel FucT can use lactose as substrate, which is similar to FucT III from human, although the activity is tiny. But when the substituted lactose was used, the activity increased. The factor that activity of entry 6 was zero suggested that the FucT can not produce Sialyl-Lewis x tertrasaccharide in vivo. In contract, the FucT used another sialylated oligosaccharide as substrate, although it is possible that fucose was transferred to the glucose residue, not the glucosamine. The possibility was indirectly proved by the similarity of the activity among substrates. The same pattern of the substrate specificity of FucT from strain 51 was discovered. In conclusion, the substrate specificity of the two enzymes was determined, which showed that they possessed same pattern of the substrate specificity. Of special notice, they could bind both Type I and Type II core structure oligosaccharide to form Lea and Lex, respectively. So the two FucTs could be called b-galactoside a-1-3/4-fucosyltransferase, which is for the first time that the enzyme possessing the ability to form both 1-3 and 1-4 linkage, had been discovered in bacterium. 3. Design and synthesis of α-1,3-fucosyltransferase inhibitors 2-amino-6-chloro-purine severed as the starting material that reacted with bromo-acetic acid to give the compound 50. The chloro group of compound 50 was replaced by benzoxyl group for protection 6-position of purine, in 45% yield. Fucose was ammoniated at 1-position at the presence of ammonia and ammonium sulfate. The unstable intermediate was ligated with N-protection amino acid to give substituted amide. Because of difficulty to be purified, the amide was further reacted to protect the hydroxyl group of fucosyl unit. The purified compound 53 was deprotected by hydrogenation to be ligated with another N-protected amino acid. The ligated product was deprotected and ligated with compound 51 to give the protected target molecule 55. After hydrogenation of 55, the base N2H4 was used to remove the acetyl group to produce compound 56. 4. Kinetics of α-1,3-fucosyltransferase on inhibitors It was fact that the 6 types of inhibitors competitively inhibited the enzyme on GDP-fucose by ordered, sequencial Bi-Bi mechanism, after activity of α-1,3-fucosyltransferase on enzyme inhibitor syntehsized from chemical synthesis was tested. the first step of enzyme mechanism, transition state analog was inhibited by formation of .
