Reporter enzyme으로 이용된 CMCase의 아미노 말단부위에 library를 구축하여 anchoring motif를 선별할 경우 매개 단백질의 카르복시 말단을 repoter enzyme 융합에 이용함으로써 기존의 motif와는 다른 새로운 motif를 선별할 수 있는 가능성이 높았으며, signal sequence를 포함한 새로운 promoter의 cloning도 가능하였다.4) 표면 발현계 구축과 비교 분석 새로이 cloning된 세포벽 부착 단백질 유전자와, 기존에 보고된 유전자 중, 매개 단백질로 이용될 가능성이 있는 다양한 유전자에 대하여 표면 발현계를 구축하고 그 효율을 비교 분석하였다. 또한 다양한 reporter enzyme을 이용하여 비교 검토하고 최적의 표면 발현 매개 단백질을 선정하였다.

To develope a new surface display system in methylotrophic yeast Hansenula polymorpha, several putative cell wall proteins were cloned and tested. In Saccharomyces, immobilization of heterologous protein on the cell surface is possible by making use of the C-terminal GPI anchor domains of glucanase extractable cell wall proteins. At first, we constructed two glycosylation mutant strains, mnn9 and pmtl, to inhibit the hyperglycosylation of the cell wall protein that hinders the purification of cell wall proteins. These two mutant strains, mnn9 and pmtl, which have defects in the N- and O-linked glycosylation, respectively, showed the supersecretion ability and some changes in cell wall protein profiles. This approach, however, could not render cell wall proteins purifiable. Therefore, we cloned potential GPI anchored cell wall protein gene homologues, Tiplp, Sedlp, Gaslp, and Cwplp from H. polymorpha. These were tested as an anchoring motif for the surface expression of carboxymethyl cellulase (CMCase). The CMCase expression vector was constructed using the secretion signal sequence of killer toxin, CMCase from Bacillus sp., and either the entire mature part or the C-terminal 40 amino acids of the putative cell wall proteins. Most of the fusion proteins were incorporated into the cell wall and showed CMC hydrolyzing activity in the whole cell fraction. CMCase immobilized by the Cwplp, Gaslp, and 40 amino acids of Tiplp was most effective in the hydrolysis of high molecular weight substrate, CMC, showing higher CMCase activity than anchored by the S. cerevisiae Cwp2p known as one of the most efficient anchor motifs in yeast. These results indicate that all four genes cloned from H. polymorpha have efficient surface targeting signals and, thus, show a great potential for a new cell surface anchoring system. In addition, Pir2p, another NaOH extractable cell wall protein, Wsclp derived from H. polymorpha, and the glucoamylase serine/threonine-rich domain from S. diastaticus were also demonstrated to be possible surface anchors. The cell suface expression system of the present study is expected to provide an immobilization effect as biocatalysts by adhereing a desired protein to the cell surface, and a means of altering target protein characteristics such as binding affinity and stability for library screening.
