심해생물 자원을 대상으로 유전적 다양성을 확보하고 기능성 소재로의 유용성 평가 기법을 이용하여 유용생체기능 소재를 개발하고 이를 통한 심해 생물자원의 생명공학소재로서 지속 가능한 이용 및 산업화 기반을 구축한다.
바다는 지구 표면의 70% 이상을 차지하고 그 중 60%는 수심이 2,000m가 넘으며, 특히 심해는 생물학적 사막으로 간주되었으나 지금은 미생물의 다양성을 포함하는 열대 다우림과 같은 지역으로 밝혀졌다. 해양생물 다양성은 종의 수가 1,000 만 종을 상회하는 것으로 추정되며 그 중 60%는 새로운 종으로 여겨지고 이러한 생물체는 대부분이 신규의 유기적 조직체로 활용 가능한 자원으로 여겨지고 있다. 육상 유래의 생물체들은 놀라운 종 다양성을 보여준 반면, 해양유래의 생물들은 거대한 계통학적인 생물 다양성(진핵생물류, 세균규, 원시세균류 의 도메인수준)을 보여 주고 있어, 지구상의 식물, 동물, 그리고 미생물의 주요한 분류군들이 우세하게 혹은 절대적으로 해양에서 나타나며, 따라서 해양생물은 새로운 유전학적인 정보와 기능을 이용할 수 있는 유용생물소재의 발견 확률이 가장 높다고 할 수 있다.
심해 생물자원은 다음과 같이 생물공학적 활용 가능성이 다양하다.

국제적인 지구생태환경에 대한 보호 및 생물다양성 협약에 대처하기 위하여 해양생물자원의 지속적 이용 및 적극적 활용에 대한 국가적인 전략이 요구되며, 생물자원의 지속적 확보방안은 선진국의 생물자원 패권주의에 대응하는 수단으로 인식되어야 한다. 우리나라는 협소한 국토를 가지고 있으나 삼면이 바다로 둘러싸여 있어 해양자원의 활용을 극대화해야 할 필요성이 있으며, 심해생물자원은 마지막 남은 지구자원으로 지속이용 가능한 자원으로 활용되어야 하며, 생명공학의 소재 개발이라는 측면에서 높은 가치가 있다.
III. 연구개발의 내용 및 범위

IV. 연구개발결과
1. 심해생물연구 시료확보 및 연구기반확립
가. 일본 JAMSTEC 프론티어 사업단 팀 리더인 C. Kato 박사를 초청하여 세미나개최 및 공동연구협의를 추진하였다. 이후 심해생물 배양장치 공동 활용 및 공동승선연구 및 시료공유를 추진하고 MOU 체결을 준비하고 있으며, 호압성 심해미생물 배양에 필요한 고압펌프 및 고압배양장치를 확보하였다.
2. 동해 및 일본연안 태평양 심해저 퇴적물시료 확보 및 미생물 분리
가. 일본 해양과학기술센터 방문 및 'YK01-06 Cruise' 공동 승선 연구(2001. 6. - 2001. 8.)를 실시하였다.
나. 일본해구, 쿠릴해구, 동해 (일본홋가이도 연안)지역의 심해 퇴적물 및 미생물막 시료를 확보하였다.
다. JAMSTEC 유인잠수정 (신카이 6500)으로부터 획득한 시료에서 중온성 및 저온성 미생물 균주와 유전자원을 확보하였다.
라. 채취된 심해퇴적물로부터 중온성, facultative anaerobes, 저온성 세균을 분리하고 16S rDNA 염기서열 분석에 의한 분자분류를 실시하였다.
마. 중온성 세균; 20 균주를 분리하여 RFLP/(RsaI+MspI) typing 결과 14 형태가 얻어졌으며, 14 형태에 대한 각 균주의 16S rDNA 염기서열 결정하여 분류학적 위치를 결정하였다.
바. 중온성 심해고유 균주 RT-17, 18의 최적 성장온도는 약 30℃이었다.
사. 통성 혐기성 중온세균 21주를 분리하였다.
아. 저온성 세균 20 주를 분리하여 16S rDNA를 증폭하고 6 가지의 서로다른 RFLP 형태를 얻어 염기서열을 결정하고 데이터베이스와 비교한 결과 96.6 - 99.6%의 염기서열 상동성을 보이며 다양한 속으로 나타났다.
자. 심해퇴적물로부터 추출된 핵산으로부터 세균류 및 고세균류의 16S rDNA 클론 분리하여 다양성을 분석한 결과 19개의 세균류 클론에 18개의 서로다른 RFLP 형태가 관찰되었고 16S rDNA 염기서열 분석 결과 72.8 - 98.8% 의 범위에서 유사도를 보였고, 21개의 고세균류 클론을 얻어 RFLP typing 결과 5개의 서로 다른 형태가 관찰되어 각 형태에 대한 클론의 16S rDNA 염기서열 결정 및 분류학적 위치 결정결과 93.4 - 99.5%의 유사도를 보였다.
3. 서태평양 심해저 퇴적물시료 확보 및 미생물 분리
가. 온누리 'Cruise 2001 Ayu Trough'로부터 태평양 수심 약 4000 m의 심해퇴적물을 확보하고 저온성, 중온성, 고온성(55℃) 균주를 분리하였다.

나. 31 균주의 저온성 미생물을 선택하여 16S rDNA RFLP/HaeIII 분석 결과, 10개의 서로 다른 형태를 얻었고, 대표 균주의 염기서열 분석결과 92.4 - 99.7%의 유사도를 보이며 다양한 속의 세균종이 분리되었다.
V. 연구개발결과의 활용계획
본 연구로부터 확보된 미생물 및 유전자원은 보존체계를 마련하고 향후 유용성 검색을 실시하여 유용한 생물소재 개발 연구에 활용할 것이다.

II. Objective and Significance
The aims of this study are the establishment of collection system for microorganisms from deep sea environment and the development of technologies for utilization of the bioresources. For this, we will assess and access the microbial diversity in deep sea environments and develop screening system for useful bioactive and biocatalytic properties of marine microorganisms.
The oceans constitute more then 70% of the earth's surface, of which about 60% is covered by water more than 2,000 m deep. In the estimation of macrofaunal species richness of the deep sea, the diversity could exceed 10 million species. The deep sea encompasses the extremes of most environmental conditions found on earth. Studies of the deep ocean environments have led to significant new discoveries of unusual microbial diversity, metabolic activity, and natural products of interest to biotechnology. Most of the metabolic and biochemical diversity of life resides in microorganisms including the domains Bacteria and Archaea, and unicellular members of the Eukarya. Marine microbial diversity is extremely high, which is a direct consequence of the extraordinary variability of the marine biosphere. This biosphere encompasses a huge thermal range (-1.5℃ in Antartic waters to temperatures in excess of 100℃ in shallow and 350℃ in deep hydrothermal vent), pressures ranging from 1 to over 1000 atmosphere, nutrient variations ranging from eutrophic to oligotrophic, and extensive photic and non-photic zones. There are much potential of marine biodiversity for commercial development and exploitation in every area of biotechnology. The biotechnological implications of exploring deep-sea environments are as follow:
Novel and improved biocatalysts and chemicals
Cold-active biocatalysts; surfactants; bioantifreeze
High-affinity catalsysts and ligands
Thermostable and solvent-stable biocatalysts; biohydrometallurgy
Novel bioactive chemicals; sensing, signaling, and defense chemicals; consortia for enhanced turnover rates
Halotolerant biocatalysts; novel metabolites
Biotransformation; Bioremediation
Deep-sea environments are usually considered as extreme environments. The microbial communities living there are enormously diverse. The mechanisms involved in adaptation to these conditions represent a variety of research topics. The properties of extreme conditions may represent novel sources of valuable molecules or bioprocesses. But the access to these deep-sea is difficult, expensive, and limited to a small number of investigators. Much efforts in the research, accompanied by improved accessibility of microorganism collections in both academic institutions and industry, are the key in the future.

IV. Results
1. Acquisition of deep sea sediment and establishment for research basis
During this study, we have invited Dr. Kato, a team leader of DEEPSTAR frontier program of JAMSTEC, and discussed about molecular mechanism of adaptation in piezophilc bacteria isolated from deep sea, and about co-work in the future. Also, we prepared the high pressure pump and culture vessel for studying piezophilic microorganisms.
2. Visit to JAMSTEC and participate in 'YK01-06 cruise' (2001. 6. - 8.)
In the YK01-06 cruise, we obtained sediment samples and microbial films from Japan Trench, Kuril Trench, and East sea (Japan sea) using manned submarine, Shinkai 6500.
3. Isolation of microorganisms from deep sea sediments
We have isolated heterotrophic mesophiles, facultative anaerobes, and psychrophiles from the samples, and have determined taxonomical position using 16S rDNA sequencing. Twenty heterotrophic mesophiles which showing different colony morphology were isolated. The RFLP/(RsaI+MspI) analysis of the strains have shown 14 different types of which representative 16S rDNA sequences were determined. Among them, strain RT-17 and RT-18 were similar to genus Shewanella and psychrophilic Peusoalteromonas, respectively. The optimal growth temperature of both strains was about 30℃. Mesophilic facultative anaerobes (21 strains) were isolated and stored at -70℃. 20 strains which were grown at 4℃ were isolated and their RFLP/HaeIII types were classified into 6 types. By comparison of 16S rDNA sequences with database, they resembled diverse microorganisms in genus level including Pseudoalteromonas, Roseobacter, Alteromonas, Cytophaga, Pseudomonas, and Vibrio with similarity range of 96.6 - 99.6%.
4. Microbial diversity of deep sea samples
The analysis of microbial diversity using 16S rDNA cloning and sequencing has been conducted. Bacterial 16S rDNA clones (No. 19) were classified into 18 RFLP types and were in the range of 72.3 - 98.8% similarity with database. Among 21 archaeal 16S rDNA clones, 5 different RFLP types were obtained. They showed higher level of similarity (93.4 - 99.5%) with known archaeal sequences.
5. Analysis of deep sea samples from Ayu Trough
We obtained deep sea (about 4,000 m) sediment samples from Ayu Trough cruise. 168 strains including mesophiles, psychrophiles and thermophiles were isolated from the sediments using different media. Of the 31 psychrophiles, 10 RFLP types were obtained and sequenced. Analysis of 16S rDNA sequences have shown that there were diverse psychrophiles with similarity 92.4 - 99.7%.
V. Application of this Research
We will develop the storage system for obtained deep sea samples and microorganisms during this study, and screening system for bioactive properties from isolated strains. Microbiological resources will be investigated continually for the comprehensive analysis of biotechnological potential in the following area:

Genomics and proteomics of useful microorganisms
Metagenomics for the analysis of multiple genomes of uncultured microorganisms
