- 연구의 목적

Glycosidase나 glycose transferase의 기질은 당쇄 (carbohydrate)이므로 이들의 저해제들을 탄수화물 구조를 모방한 구조가 될 수 있다. 또한, 암세포 및 HIV 세로의 표면에는 비정상적으로 과도한 당쇄 및 당 단백질이 존재하며, 이러한 복합 당쇄 구조를 형성하기 위해서는 많은 양의 glycosidase나 glycose transferase가 요구된다. 따라서, 특정한 glycosidase나 glycose transferase의 과다한 검출은 특정한 암의 발병 및 AIDS를 진단하는 marker로 이용될 수도 있다.
기존에 임상에서 사용되고 있는 의약품의 단점들은 이들과 작용기전이 다른 탄수화물의 구조를 모방한 의약품에 의하여 개선될 여지가 많이 있다. 당쇄를 이용한 의약품들은 세포의 표현형질에 작용하므로 다른 작용기전에 의하여 작용하는 의약품에 비하여 독성이나 면역반응 둥이 낮을 수 있으며, 약제에 대한 내성이 적을 것으로 기대되므로 glycosidase 저해제를 이용한 새로운 질병 치료제의 개발을 본 연구의 목적으로 하였다.
- 연구방법
최근 자연계에 손재하는 azasugar류나 또는 합성되어진 유사 화합물들이 glycosidase 억제제의 기능이 있다는 것이 알려진 이후로 이들 화합물의 유도체들에 관한 연구가 활발히 진행되고 있다. 이들 화합물들은 공통적으로 sugar와 같이 polyhydroxyl기를 함유하고 sugar ring의 산소 대신에 질소를 함유하는 azasugar의 일반적인 구조를 갖고 있다. 이들 azasugar 화합물을 구조에 따라 대표적인 화합물로 구분하여보면, 5원환 구조의 polyhydroxypyrrolidine 화합물, 6원환의 가지는 polyhydroxypiperidine 화합물, bicyclic 형태의 polyhydroxyindolizidine 또는 polyhydroxypyrrolizidine 화합물과 드물게 7원환 구조의 polyhydroxyazepane 화합물로 구분할 수 있다.본 연구에서는 7원환 형태의 azasugar 화합물의 제조를 목적으로 하였고, 목적으로 하는 화합물인 신규의 azasugar 화합물을 손쉽게 합성하기 위한 방법으로 하나의 주요중간체로부터 aminoalcohol 및 diamaino 화합물과 같은 여러 amine류와 반응하여 합성하기로 하였다. Glucose를 출발물질로 하여 주요 합성중간체인 epoxyaldehyde를 합성하였고, 이 화합물을 이용하여 bicycloazasugar 화합물 유도체들을 합성하였다. 또한 합성되어진 신규의 bicyclioazasugar 화합물로부터 개환반응을 통하여 monocyclic 형태의 azasugar 화합물을 합성하였다.
- 연구결과
본 연구에서 주요중간체인 epoxyaldehyde와 여러종류의 amine 화합물과 반응을 하여 최종 10종의 bicyclic azasugar 화합물과 6종의 monocyclic azasugar 화합물을 합성하였다. 합성되어진 bicyclic 형태의 azasugar 화합물의 β-glucosidase에 대한 활성시험 결과, 대조물질로 사용된 천연에서 분리된 강력한 당분해효소 억제제인 deoxynojirimycin에 비하여 우수한 억제 활성을 보여주었다.

Every living organism ranged from microorganism to higher organism has a complex and variety of carbohydrates in cell surface. Carbohydrates and their conjugates such as glycoproteins and glycolipids are recognized to play a wide kind of metabolic roles, both as free-standing chemical mediators and as cell surface receptors and markers. Such carbohydrates mediate an enormous repertoire of biologically important events, including cell-cell recognition, cell growth, cell development, cell-cell adhesion, inflammation, and metastasis. Carbohydrates in cell surface are matured by two families of enzymes, glycosidases and glycosyl transferases. The two enzymes, glycosidases and glycosyl transferases, are vital to the growth of cell. For this reason, the inhibition of glycosidases or glycosyl transferases could interfere the maturation of cell surface carbohydrates leading to promising therapeutic applications in the area of diabetes, antiviral, anticancer, and anti-AIDS.
Most of commercially available medicines have somewhat weak point such as side effect and toxicity etc. However, carbohydrate drugs could avoid immune response or toxicity. A major goal of this research has been to design and synthesize new glycosidase inhibitors which can control effectively over glycoside hydrolysis.
- Method of Study
After the naturally-occurring and synthetic azasugars are known to inhibit the glycosidases there is continuing interest in the synthesis and mechanism study of this class of compounds. Structurally azasugar derivatives possess polyhydroxyl group like sugar and the ring oxygen atom has been replaced with a nitrogen atom. Among them are polyhydroxypyrrolidines with five-membered ring, polyhydroxypiperidines with six-membered ring, polyhydroxyindolizidines or polyhydroxypyrrolizidines with bicyclic azasugar, and polyhydoxyazepane with seven-membered ring.A major goal of this study is the preparation of seven-membered azasugar which can be synthesized by the condensation reaction with aminoalcohol or diamino compounds and epoxyaldehyde from glucose. Using this condensation reaction, we could make bicyclic seven-membered azasugar compounds and monocyclic seven-membered azasugar derivatives via ring opening reaction of bicyclic compounds.
- Results and Discussions
We made 10 bicyclic azasugars and 6 monocyclic azasugars via the condensation reaction with aminoalcohol or diamino compounds and epoxyaldehyde. The synthesized bicyclic azasugars were evaluated as inhibitor against β-glucosidase. Most of them showed better inhibition activity than deoxynojirimycin which was a naturally-occurring potent glycosidase inhibitor.
