인간 뇌에는 1000억 개에 달하는 신경세포가 있으며 세포 하나하나는 주위의 뉴런과 1만개 정도의 시냅스를 갖고 있다. 인간이 정상적인 생활을 영위하기 위해서 우리 뇌안에 있는 신경세포간의 긴밀한 정보교환과정 (information processing)이 필수적이며, 그 역할을 수행하는 물질이 바로 신경전달물질 (neurotransmitter)이다. 이러한 신경 전달 물질은 크게 small molecules에 biogenic amines, amino acids, nucleotides, NO등과 neuropeptides에 opioid
인간 뇌에는 1000억 개에 달하는 신경세포가 있으며 세포 하나하나는 주위의 뉴런과 1만개 정도의 시냅스를 갖고 있다. 인간이 정상적인 생활을 영위하기 위해서 우리 뇌안에 있는 신경세포간의 긴밀한 정보교환과정 (information processing)이 필수적이며, 그 역할을 수행하는 물질이 바로 신경전달물질 (neurotransmitter)이다. 이러한 신경 전달 물질은 크게 small molecules에 biogenic amines, amino acids, nucleotides, NO등과 neuropeptides에 opioid peptides, posterior pituitary peptides, tachykinins, glucagon-related peptides, pancreatic polypeptide-related peptides, somatostatin 등으로 구분할 수 있다. 여기서 흥분성 아미노산(excitatory amino acids)은 small molecules에 속하는 신경전달 물질이다. 뇌세포에 있어서 흥분성 아미노산 (excitatory amino acids: kainic acid, NMDA. quisqulate AMPA, glutamate 등)은 흥분 독성을 일으켜 신경세포 죽음을 일으키고 뇌 기능의 장애를 유발하여 여러 가지 뇌 질환(경련, 뇌졸중, 알츠하이머병, 파킨슨 병, 척수 손상 등)을 일으키는 중요한 인자이다. 지금까지 많은 과학자들이 흥분성 아미노산을 이용한 흥분 독성 모델을 통해서 뇌 세포 사멸에 대한 연구를 수행해 왔다. 그러나 아직 glutamate, NMDA, kainic acid(KA)와 같은 흥분성 아미노산을 이용한 뇌세포 죽음에 대한 기전 연구에 있어서 다른 신경 전달물질에 대한 연관성에 대해서는 연구가 미흡하다. 본 연구는 이에 관심을 두고 mouse에서 KA를 뇌실내로 투여한 모델에 있어서 여러 신경전달물질의 길항제 등을 전처치 하여 흥분성 아미노산 (excitatory amino acid)에 의한 세포사멸 (cell death)과 seizure activity, 메모리 손상, 고용량 주입시 lethal toxicity에 관련된 여러 신경 전달물질의 연관성을 알아보고자 하였다.
Kainic acid(KA)는 잘 알려진 흥분성 신경독성물질이다. Mouse에서 뇌실내로 투여한 KA는 mouse 해마의 조직학적 손상을 야기시키는데 특히 CA3 피라미달 신경세포가 현저하게 죽는 것을 관찰 할 수 있었다. 또한, 뇌실내로 투여한 KA는 seizure activity를 나타내며 기억력 손상과 고용량 주입시 lethal toxicity를 나타낸다. 먼저 KA가 유도하는 mouse 해마 CA3 피라미달 신경세포 죽음에 있어서 5-HT, adrenergic, A1 adenosine, $GABA_{A}$, muscarinic acetylcholine (mACh) 수용체 길항체인 methysergide, yohimbine, PACPX, SR95531, scopolamine 등은 영향을 주지 못했고, A2 adenosine, $GABA_{B}$, nicotinic acetylcholien (nACh) 수용체 길항제인 DMPX, 5-Aminovaleric acid, methyllycaconitine 등이 KA에 의한 마우스 해마 cell death를 억제하였다.
첫 번째로 뇌실 내로 투여한 KA(0.1 ㎍)가 유도하는 다양한 약리학적 신경독성 반응에서 cycloheximide(CHX)의 효과에 대해서 연구하였다. 기억력테스트 중의 하나인 passive avoidance test에서 복강으로 20분 전처치한 CHX(200 ㎎/㎏)는 KA가 유도하는 메모리 손상을 유의성 있게 억제하였다. 또한, CHX는 KA가 유도하는 해마 CA3 region의 cell death를 효과적으로 억제하였다. 면역조직화학염색법으로 조사하였을 때 KA가 유도하는 p-ERK, p-JNK1, p-CaMK Ⅱ, c-Fos, c-Jun의 현저한 발현이 mouse 해마에서 관찰되었다. CHX를 전처치 했을 경우 이렇게 mouse 해마에서 KA가 유도하는 p-ERK, p-JNK1, p-CaMK Ⅱ, c-Fos, c-Jun의 발현이 뚜렷하게 줄어드는 것을 알 수 있었다. Microglia의 활성화를 보기 위해서 OX-42의 면역염색반응을 보았는데, KA가 유도하는 mouse 해마 cell death region에서 OX-42의 면역반응이 증가하였고 이를 CHX가 억제하는 것을 확인하였다.
두 번째로 adenosine receptor가 KA가 유도하는 mouse 해마 CA3 region의 cell death에 어떤 관련이 있는지를 조사하였다. A2 adenosine receptor 길항제인 DMPX (20 ㎍)를 뇌실내로 10분 전처치 하였을 때 KA가 유도하는 mouse 해마 CA3 region의 cell death가 억제 되었다. 뇌실내로 투여한 KA는 mouse 해마에서 p-ERK, c-Jun, c-Fos, Fra-1, Fra-2의 면역염색반응이 증가하게 하였고 DMPX는 KA가 유도하는 p-ERK, c-Jun, Fra-1, Fra-2의 면역염색반응을 억제시켰다.DMPX는 KA가 유도하는 mouse 해마 CA3 region의 cell death 부분에서의 OX-42의 면역반응을 억제하였다, 그러나 A1 adenosine receptor 길항제인 PACPX (20 ㎍)는 KA가 유도하는 cell death나 면역 반응을 억제하지 못하였다. 다만 KA가 유도하는 c-Fos의 면역반응을 DMPX가 아닌 PACPX가 억제하는 현상을 보였다. 이러한 결과들을 통해서 무엇보다도 KA가 유도하는 mouse 해마 cell death에 관련해서 A1 adenosine 수용체가 깊이 연관되어 있음을 알 수 있고 특별히 p-ERK나 c-Jun 단백질들이 이러한 cell death에 연관될 것이라고 생각할 수 있겠다.
세 번째로 뇌실내로 투여한 KA가 유도하는 해마 cell death에 있어서 $GABA_{B}$ receptor의 가능한 역할을 조사하였다. $GABA_{B}$ receptor 길항제인 5-Aminovaleric acid (5-AV; 20 ㎍)를 mouse 뇌실내로 10분 전처치 하였을 때 KA가 유도하는 CA3 피라미달 cell death를 억제하였다. 뇌실내로 투여한 KA는 KA 투여 후 30분에서 p-ERK와 p-CaMK Ⅱ가 분포가 다르지만 해마에서 증가시켰다. KA는 투여 후 2시간후에는 분포가 다르지만 c-Fos, c-Jun 면역반응이 mouse 해마에서 증가되었다. KA 투여 하루 후에는 GFAP와 OX-42의 면역반응이 해매 CA3 region에서 증가하였다. 5-AV는 KA가 유도하는 p-CaMK Ⅱ, GFAP, OX-42의 면역반응을 현저히 감소 시켰다. 이상의 결과는 $GABA_{B}$ 수용체가 KA가 유도하는 해마 cell death에 깊이 연관되어 있다고 판단할 수 있게 해주고 동시에 p-CaMK Ⅱ가 KA가 유도하는 해마 cell death에 있어서 $GABA_{B}$ 수용체와 관련하여 중요한 역학을 할 것이라고 추측할 수 있게 해준다.
네 번째로 뇌실내로 투여한 KA가 유도하는 해마 cell death에 있어서 nicotinic acetylcholine receptor(nAChR)의 가능한 역할을 조사하였다. nAChR의 길항제인 methyllycaconitine (MC; 20 ㎍)를 mouse 뇌실내로 10분 전처치 하였을 때 KA가 유도하는 CA3 피라미달 cell death를 억제하였다. 뇌실내로 투여한 KA는 KA 투여 후 30분에서 p-ERK와 p-CaMK Ⅱ가 분포가 다르지만 해마에서 증가시켰다. KA 투여 후 2시간 후에는 분포가 다르지만 c-Fos, c-Jun 면역반응이 mouse 해마에서 증가되었다. KA 투여 하루후에는 GFAP와 OX-42의 면역반응이 해마 CA3 region에서 증가하였다. MC는 KA가 유도하는 p-CaMK Ⅱ, GFAP, OX-42의 면역반응을 현저히 감소 시켰다. 이상의 결과는 nACh 수용체가 KA가 유도하는 해마 cell death에 깊이 연관되??체와 관련하여 중요한 역할을 할 것이라고 추측할 수 있게 해준다.
다섯 번째로 위에서 언급한 결과를 토대로 기억과 연관이 있는 해마에서의 cell death가 기억력에도 관련이 있지 않는가 생각하는 가운데 cell death가 일어나는 시점인 KA(0.02 ㎍) 투여 하루후에 여러 길항제들과 관련하여 기억력테스트를 수행해 보았다. 또한 lethality를 나타내는 고용량의 KA(0.5 ㎍)를 투여하기 전에 여러 길항제들과 cycloheximide (CHX)를 전처치하여 lethality에 미치는 신경전달 수용체들과 on-going protein synthesis의 관련을 조사하였다. 먼저 KA(0.005-0.01 ㎍)는 메모리 테스트중의 하나인 passive avoidance test에서 거의 용량의존적으로 메모리 손상을 보였다, 현저하게 메모리 손상을 보이는 가장 낮은 KA의 농도인 0.02 ㎍을 선택하여 여러 길항제들을 전처치하여 실험한 결과 PACPX, DMPX, MC, SR, 5-AV, MS, YB 모두 KA가 유도하는 메모리 손상을 억제하지 못하였다. Lethality test에서는 ACPX, DMPX, MC, SR, 5-AV, MS, YB, CHX 모두가 KA가 유도하는 lethaality를 어느정도 억제시켰지만 그중에서도 유의성 있게 억제시킨 것은 PACPX, DMPX, MS, YB, CHX 이었다. 이러한 결과를 토대로 생각해 볼 때 KA가 유도하는 메모리 손상은 cell death가 일어나지 않는 저농도의 KA(0.02 ㎍)에서도 발생하며 여러 신경전달물질의 수용체와 관련이 부족함을 생각할 수 있게 해준다. 또한 KA가 유도하는 lethal toxicity에 있어서는 A1, A2 adenosine 수용체와 5-HT. adrenergic 수용체 그리고 on-going protein synthesis가 중요하게 알 수 있었다.
이상 모든 연구를 정리한다면, 뇌실내로 투여한 KA가 유도하는 신경 흥분 독성 반응에서 나타나는 다양한 세포내 메카니즘과 기능들을 단편적이지만 관찰할 수 있었다. 뇌실내로 투여한 KA는 다양한 세포내 물질들, p-ERK, p-JNK, CaMK Ⅱ, p-CaMK Ⅱ, c-Fos, c-Jun, Fra등의 변화를 mouse 해마에서 보인다. KA는 astrocyte와 microglia의 활성화를 마우스 해마에서의 cell death와 연관하여 일으키고 이것은 신경독성을 나타내주는 좋은 지표가 되었다. 또한 다양한 신경수용체와 on-going protein synthesis가 KA가 유도하는 해마 cell death와 메모리 손상, lethality에 관련되어 있음을 조사하였다. 이러한 연구들은 다양한 신경독성 또는 신경질환 연구에 기초 자료가 되며 중추신경계에 있어서 KA나 흥분성 아미노산 연구에 도움이 되는 정보를 줄 것이다.

Kainic acid (KA) is a well-known excitatory, neurotoxic substance. In mice, morphological damage of hippocampus induced by KA administered intracerebroventricularly (i.c.v,) was markedly concentrated on the CA3 pyramidal neurons.
In the first study, we investigated the effect of cycloheximide (CHX) on various pharmacological responses induced by KA administered i.c.v. in mice. In a passive avoidance test, a 20 min CHX (200 ㎎/㎏, i.p.) pretreatment prevented the memory impairment induced by KA. The morphological damage induced by KA (0.1 ㎎) in the hippocampus was markedly concentrated in the CA3 pyramidal neurons and CHX effectively prevented the KA -induced pyramidal cell death in CA3 hippocampal region. In immunohistochemical study, KA dramatically increased the phosphorylation of extracellular signal-regulated protein kinase (p-ERK), c-Jun N-terminal kinase 1 (p- JNK1) and calcium/calmodulin-dependent protein kinase Ⅱ (p-CaMK Ⅱ). CHX attenuated the increased p-ERK, p- JNK1 and p-CaMK Ⅱ levels induced by KA. Furthermore, CHX inhibited the increased c-Fos and c-Jun protein expression levels induced by KA m the hippocampus. The activation of microglia was detected in KA-induced CA3 cell death region by immunostaining with a monoclonal antibody against the OX-42. CHX inhibited KA-induced increase of OX-42 immunoreactivity. Our results suggest that the increased expression of the c-Fos, c-Jun and phosphorylation of ERK, JNK1 and CaMK Ⅱ proteins may play important roles in the memory impairment and the cell death in CA3 region of the hippocampus induced by i.c.v. KA administration in mice. Furthermore, the activated microglia may be related to phagocytosis of degenerated neuronal elements induced by KA.
In the second study, the possible role of adenosine receptors in hippocampal cell death induced by KA (0.1 ㎍) administered i.c.v. was examined. 3,7-Dimethyl-1-propargylxanthine (DMPX; A2 adenosine receptors antagonist, 20 ㎍) reduced KA-induced CA3 pyramidal cell death. KA dramatically increased the p-ERK immunoreactivities (IR) in dentate gyrus (DG) and mossy fibers. In addition, c-Jun, c-Fos, Fra-l and Fra-2 IR were increased in hippocampal area in KA -injected mice. DMPX attenuated KA -induced p- ERK, c-Jun, Fra-l and Fra-2 IR. However, 1,3-dipropyl-8-(2-amino-4-chlorophenyl)-xanthine (PACPX; Al adenosine receptor antagonist, 20 ㎍) did not affect KA-induced p-ERK, c-Jun, Fra-l and Fra-2 IR. KA also increased OX -42 IR in CA3 region of hippocampus. DMPX, but not PACPX, blocked KA -induced OX -42 IR. Our results suggest that p-ERK and c-Jun, may playas important regulators responsible for the hippocampal cell death induced by KA administered i.c.v. in mice. Furthermore. it is implicated that A2. but not AI. adenosine receptor appear to be involved in hippocampal CA3 pyramidal cell death induced by KA administered i.c.v. in mice.

KA increased the p-ERK, p-CaMK Ⅱ immunoreactivities (IR) at 30 min after KA treatment, and c-Fos, c-Jun IR at 2 hr, and GFAP, OX-42 IR at 1day in hippocampal area in KA-injected mice. 5-AV attenuated KA-induced p-CaMK Ⅱ, GFAP and OX-42 IR in the hippocampal CA3 region. Our results suggest that p-CaMK Ⅱ may playas an important regulator responsible for the hippocampal cell death induced by KA administered i.c.v. in mice. Methyllycaconitine (MC; nicotinic acetylcholine receptors antagonist, 20 ㎍) reduced KA-induced CA3 pyramidal cell death. KA increased the p-ERK, p-CaMK Ⅱ immunoreactivities (IR) at 30 min after KA treatment, and c-Fos, c-Jun IR at 2 hr, and GFAP, OX -42 IR at 1day in hippocampal area in KA -injected mice. MC attenuated selectively KA-induced p-CaMK Ⅱ, GFAP and OX-42 IR in the hippocampal CA3 region. Our results suggest that p-CaMK Ⅱ may playas important regulators responsible for the hippocampal cell death induced by KA administered i.c.v. in mice. Furthermore, it is implicated that niconitic receptors appear to be involved in hippocampal CA3 pyramidal cell death induced by KA administered i.c.v. in mice.
In the fifth study, we examined the effect of various antagonists and CHX on memory loss and lethality induced by KA administered i.c.v. in mice. KA arose memory loss in passive avoidance test by a dose-dependent manner. KA (0.02 ㎎, i.c.v.)-induced memory loss did not affected by pretreatment with PACPX, DMPX, MC, SR, 5-AV, MS and YB. In lethality test, KA (0.5 ㎎, i.c.v.)-induced lethal toxicity reversed by pretreatment with (PACPX, DMPX, MC, SR, 5-AV, MS, YB and CHX. Among these data, unequivocal reversions were that pretreatment of PACPX, DMPX, MS, YB and CHX against the KA-induced lethal toxicity. Thus, our results suggest that KA (0.02 ㎎, i.c.v.)-induced memory loss may not be related to A1 and A2 adenosine, $GABA_{A and B}$, nACh, 5-HT and adrenergic receptors in mice. KA (0.5 ㎎, i.c.v.)-induced lethal toxicity may be related to A1 and A2 adenosine, 5-HT, adrenergic receptors and on-going protein synthesis.
In conclusion, we studied various intracellular mechanisms and functions of neurotransmitters involved in KA-induced neurotoxicity. I.c.v. administered KA gave rise to various changes of intracellular molecules, those were p-ERK, p-JNK, CaMK Ⅱ, p-CaMK Ⅱ, c-Fos, c-Jun and Fra in mouse hippocampus. Thus, we studied that KA induces activation of astrocytes and microglia in mice hippocampus. We investigated that various neuronal and neurotransmitter receptors, and on-going protein synthesis involved in KA -induced hippocampal cell death, memory loss and lethality. We think that these studies are important to various neurotoxicity or neurodisease researches, and KA or excitatory amino acids researches in the central nervous system.
