암(Tumor)은 유전적 변이로 인하여 세포주기 (Cell cycle)가 조절이 되지 않는 질병으로, 특히 세포 분열(Mitosis)에 직접적으로 관여하는 Mitotic checkpoint tumor suppressor (항암유전자)의 기능 연구는 암 발생의 원인과 암의 주된 특징인 세포 분열과 정에서의 Chromosomal Instability (CI)생성과 Aneuploidy 형성을 생리, 생화학적으로 이해함으로써 암 발생 기전의 확립 및 치료에 기여할 수 있다.

암(Tumor)은 유전적 변이로 인하여 세포주기 (Cell cycle)가 조절이 되지 않는 질병으로, 특히 세포 분열(Mitosis)에 직접적으로 관여하는 Mitotic checkpoint tumor suppressor (항암유전자)의 기능 연구는 암 발생의 원인과 암의 주된 특징인 세포 분열과 정에서의 Chromosomal Instability (CI)생성과 Aneuploidy 형성을 생리, 생화학적으로 이해함으로써 암 발생 기전의 확립 및 치료에 기여할 수 있다.
최근 본 연구자는 유전자 손상 신호 전달 회로에 관하여 연구하는 과정에서, 세포 분열 과정에 관여하는 새로운 단백질 network를 구축하였으며, 그 중에서 암의 발병 원인 유전자의 기능연구를 수행하여 발병 원인을 규명할 수 있는 중요한 단서를 얻을 수 있었다. 이들 중 RASSFIA, RABPI, RAN GTPase, Aurora A kinase등은 Mitotic checkpoint에 관여하여 세포분열 과정을 조절함으로써 정확한 염색체 분열을 유도하였다. 또한 이들을 정상세포에서 그 기능을 제어하는 system을 구축하고, Chromosomal Instability, 세포사멸 (Apoptosis), 암 발생 기전 및 대표적인 항암 단백질인 p53과의 유전적 관계를 규명하고자하였다.
본 과제의 연구결과를 종합해 보면, (1) 정상세포에서 세포 분열 제어에 관여하는 Cdc20, Mad2, Bub1, BubR1, Emil, RASSF1A 단백질들의 기능을 억제함으로써 Chromosomal Instability 및 Mitotic Exit를 유도하는 system을 확보하였으며, (2) 비정상적 mitosis에 의하여 유도되는 Chromosomal Instability(CI)와 p53 단백질에 의한 G1 세포 주기억제 및 사멸과의 유전적 연과성을 조사하였으며, (3) 정상세포에서 Mitotic checkpoint 와 p53 checkpoint의 비정상적 조절이 암 세포로의 형질전환의 원인일 수 있다는 가능성을 조사하였으며 (4) Mitosis기간 동안에 항암 단백질 RASSF1A과 결합하는 RABP1, RAN GTPase, Aurora A kinase 단백질의 기능적 연구를 수행하였다.
마지막으로 본 연구결과에 근거하여 Mitosis의 이상으로 인한 G1 aneuploidy arrest 암 세포의 초기 단계에서 염색체 이상이 암 발생의 initiator로 암을 유발 또는 발전시킬 것이라는 가설을 제시하였으며 또한 일부를 확인/증명하였다. 이러한 연구 결과는 암 발생 기전의 이해와 암 치료를 목적으로 mitosis에 관여하는 새로운 molecular therapeutic targets의 발굴기초를 확립할 수 있다.

Tumor can be formed due to a cell cycle deregulation with genetic changes of either oncogene or tumor suppressor. Especially, the functional study of mitotic checkpoint tumor suppressor, which is essential for mitosis, contributes to development of anti-cancer treatment through understanding physiological and biochemical mechanism(s) of tumorigenesis, chromosomal instability (CI) and aneuploidy.
Since we have studied on the DNA damage response pathway, we have defined new protein network related to the cell division and have obtained the clues for cancer development. We found that the tumor suppressor RASSF1A, RABP1, RAN GTPase and Aurora A kinase regulate the mitotic checkpoint during cell division, which are faithful to correctly segregate two daughter chromosomes. Additionally, we established the systems which control their functions in normal cells and investigated the genetic correlation between their biological roles and chromosomal instability, apoptosis, tumorigenesis and the tumor suppressor p53.
In this research project, (1) we established the systems which induce chromosomal instability and mitotic exit by inhibiting the function of Cdc20, Mad2, Bub1, BubR1, Emi1, RASSF1A proteins in normal cells, (2) we examined the effects of chromosomal instability (CI) resulting from abnormal mitosis on the G1 arrest and apoptosis by p53, (3) we also examined the effects of deregulation of both mitotic checkpoint and p53 checkpoint on the tumor formation even in normal cells, (4) we finally determined the functions of RABP1, RAN GTPase and Aurora A kinase which interact with tumor suppressor RASSF1A.
Based on our results, we had provide evidences of the correlation between G1 aneuploidy arrest induced by mitotic defects and p53-Rb proteins. And we also proposed chromosomal instability as a initiator of tumorigenesis in normal cells. These results may be helpful to identify the mechanism of tumorigenesis and mitosis and to develop new molecular-targets in cancer therapy.
