본 연구는 N. fowleri의 감염 시 병인을 규명하고 진단 시료를 개발하는 일환으로 다양한 분자생물학적 및 면역학적 접근 방법으로 N. fowleri의 병원성과 관련이 있는 항원의 유전자를 클론닝하고 그 정보를 이용하여 재조합 항원을 제조하고자 하였으며 또한, 얻어지는 다양한 유전자를 in vivo 체계에서 발현할 수 있는 모델을 개발하며 더 나가서는 여기서 얻어진 재조합 항원들의 진단적 가치를 평가하고자 실험을 계획하였다. 본 연구의 내용은 다양한 분자생물학적 및 면역학적 접근 방법으로 N. fowleri의 병원성과 관련이 있는 다양한 유전자를 탐색하고자 immunoscreening 방법으로 cDNA expression library로부터 클론닝하고, 얻어진 유전자의 염기서열을 밝히었고, 또한 재조합 단백질을 생산하여 제특성을 검정하였다. 이들 중 병원성과 관련이 있을 유전자를 택하여 in vivo에서 유전자를 발현시킬 수 있는 system으로 N.fowleri로부터 새로이 클론닝된 nfa1 유전자를 pEGFP-C2 벡타에 클론닝하여 비병원성인 N. fowleri에 transfection 시켰으며 그결과를 확인하였다. 그다음은 아메바의 항원성 및 병원성과 관련이 있는 유전자들을 선택하여 생화학적 제특성 및 발현정도를 관찰하고 재조합 항원을 제조하여 진단적 가치를 평가하였다. 병원성 N. fowleri의 cDNA library를 구축하여 N. fowleri의 감염혈청 및 면역혈청으로 immunoscreening 과정을 통해 얻어진 양성 clone들 중에서 DNA sequencing을 통하여 360 bp의 DNA 염기서열 (120개의 아미노산)을 가진 유전자 (nfa1 유전자) 및 654 bp (190개의 아미노산)을 가진 nfCBP 유전자를 클론닝하여 GeneBank에 등록하였으며 상동성을 비교하였다. 한편으로, 본 실험에서 새로이 클로닝된 nfa1 유전자가 병원성인 N. gruberi에서는 발현되지 않음이 관찰되어 Nfa1은 아메바의 병원성과 연관 있을 것으로 주목하고 in vivo transfection 실험을 진행하였는데, nfa1 유전자가 N. gruberi에 잘 transfection되어 Nfa1 단백질을 지속적으로 분비됨이 관찰되었다. 또한, nfa1 유전자로부터 발현된 재조합 단백질 (Nfa1)을 정기영동한 뒤 13.1 kDa의 분자량이 확인되었으며 감염 및 면역 혈청으로 western blotting을 수행한 결과 13.1 kDa에서 반응대가 잘 관찰되었다. 항-Nfa1항체를 생산하여 immunocytochemistry를 수행한 결과 아메바의 위족에 강하게 분포하였다. 또한, N. fowleri에 감염된 마우스 조직 내에서도 N. fowleri에만 특이적으로 반응을 보여 진단적인 이용가치가 큰 항원으로 관찰되었다. 한편, 아메바의 위족 활동과 연관이 있고 병원성 아메바에서 왕성하게 발현되는 것으로 보아 병원성과의 연관이 있을 것으로 사료되어, 기존에 알려진 fibronetin을 이용한 병원성 N. fowleri의 세포독성에 대한 실험을 진행하였다. 본 실험에서 클론닝된 nfa1 유전자도 위족 활동과 연관이 있었으므로 아메바의 병원성과 연관이 있을 것으로 사료되어, 병원성 N. fowleri의 배양에 있어서 항-Nfa1 항체를 투여할 경우 어떠한 영향이 있는지를 관찰하고자 하였는데, 본 실험 결과 N. fowleri의 증식률과 CHO 세포들에 대해 세포독성이 항-Nfa1 항체의 투여에 의해 현저히 감소함이 관찰되었으며 그런 현상은 항체의 희석농도 즉, 투여량에 의존적이었다. 결론적으로 본 연구에서 다양한 유전자를 새로이 클론닝할 수 있었으며, 그중 nfa1 유전자는 아메바의 병원성과 연관이 있는 유전자로 관찰되어 추후의 연구에 많은 정보와 재료를 제공할 것으로 생각되며, 본 연구에서 개발된 in vivo system은 새로이 클론닝될 많은 아메바 관련 유전자들의 기능을 연구하는데 중요한 모델이 될 것이며, 본 연구에서 처음 클론닝된 nfa1 유전자는 아메바의 진단에 유용하게 사용될 것이다.

In order to elucidate the pathogenicity for the infection of Naegleria fowleri and production of diagnostic agents, this study was tried to produce the pools of pathogen-related genes by various immunological and molecular biological approaches (first year), and then an in vivo model that a pathogenic-related gene (nfa1) was transfected into nonpathogenic amoeba was established (second year), and the evaluation of recombinant antigen that produced those genes was evaluated (third year). 1) genes cloning from N. fowleri. A nfa1 (360 bp) and nfCBP (564 bp) gene were cloned from pathogenic N. fowleri. An α-actinin gene cloned partly from N. fowleri was full sequenced using Genome-working PCR system. At addition, the promoter region of nfa1 gene was sequenced in order to use on transfection vector. Otherwise, the first collaborator observed the response between the fibronectin (FN) and N. fowleri, as concerning with pathogenicity of N. fowleri. 2) In vivo transfection of a newly cloned nfa1 gene into nonpathogenic amoeba Using a pEGFP-c2 vector including nfa1ORF DNA, the pathogenic-related genes, nfa1, was transfected into nonpathogenic N. gruberi. The recombinant Nfa1 protein was identified from transgenic N. gruberi. 3) In vitro cytotoxicity of N. fowleri In order to observe the relationship of the gene with amoeba pathogenicity, cytotoxicity N. fowleri was examined on the monolayered CHO cells with/without an anti-Nfa1 polyclonal antibody. An anti-Nfa1 antibody showed the decreasing effect on the cytotoxicity N. fowleri. 4) Immunocytochemistry of Nfa1 protein An anti-Nfa1 obtained by immunizing with a recombinant 13.1 kDa protein was not cross-reactive with non-pathogenic N. gruberi, and lysates of some Acanthamoeba species. By immunohistochemistry of the Nfa1 protein in vivo, trophozoites were well immunostained on imflamatory and necrotic region of mouse brain infected with amoeba. This staining signal may be the result of antigen presentation. For the immunolocalization study of the Nfa1 protein, it was located on the cell surface of pseudopodia, it may be used as an useful antigen. Various gene pools established in this study may be used in studying for the pathogenicity, antigenicity, physiology, and host-relationship of free-living amoebae. Especially, the nfa1 gene may be used in studying for the pathogenicity and antigenicity of pathogenic N. fowleri. And then, it may be useful on the immunodiagnosis of amoebic infection. In addition, in vivo eukaryotic transfection system using nonpathogenic N. gruberi is established in this study, it may be useful for identifying the function of genes cloned newly from various organisms.
