목적 1. 구강점막 biofilm인 잔존타액의 조성을 규명 2. 인공타액 성분인 동물성 mucin이 타액 peroxidase의 활성에 미치는 영향 내용 및 범위 1. 생화학적 방법 2. Hydroxyapatite beads, disc, bovine tooth, microtiter plate를 solid-phase로 활용 3. 구강건조증 환자를 대상으로 한 임상연구 결과 1. 구강점막 biofilm내 total protein, slgA, lactoferrin, total carbohydrate, 및 sialic acid의 양은 전타액에 비해 유의하게 높음. 2. 구강점막 biofilm은 탄수화물 및 lactoferrin의 양이 상대적으로 많음. 3. 돼지 위장 mucin은 용액상태에서 peroxidase 활성을 증가시킴. 4. 돼지 위장 mucin이 부착된 HA는 추가적인 peroxidase 활성을 증가시키며, 그 정도는 HA 표면의 종류에 따라 다름. 5. 구강건조증 관련 증상 및 행동의 정도는 자극성 타액분비율과 더욱 뚜렷한 상관관계를 보임. 기존의 인공타액은 환자의 불편감을 일부 개선 시킴. 이상의 결과는 점막방어 기전을 이해하고 효과적인 타액대체제 개발에 활용될 것임

I. Purpose of Study 1. Understanding of composition of residual saliva, oral mucosal biofilm 2. Influence of animal mucins on salivary antimicrobial components in solution and on hydroxyapatite surfaces 3. Clinical studies in patients with dry mouth for the development of effective artificial saliva II. Contents of Study 1. Composition of residual saliva, oral mucosal biofilm (1) Comparative study between residual saliva and whole saliva: 1) Biochemical analysis on important salivary antimicrobials 2) Total protein, total carbohydrate, slgA, lysozyme, lactoferrin, peroxidase, sialic acid, amylase 2. The residual saliva (RS) showed much higher concentrations of total protein, slgA, lactoferrin, total carbohydrate, and sialic acid than unstimulated (UWS) and stimulated whole saliva (SWS). In terms of the relative proportion or specific enzymatic activity of each components, the RS had a higher proportion of carbohydrate and lactoferrin and lower specific activity of lysozyme compared with the UWS and SWS. 2. Porcine gastric mucin (PGM) enhanced bovine lactoperoxidase (bLPO) activity in solution. HA-adsorbed PGM increased subsequent bLPO adsorption in all three HA phases. The effects of the animal mucins were dependent on the type of HA surface. 3. The intraclass correlation coefficients of 6 questions to evaluate the subjective oral dryness were as significantly high as 0.563 - 0.767. Dry mouth-related symptoms and behaviors were significantly associated with whole salivary flow rate. The severity of dry mouth-related symptoms was mere closely correlated with the flow rate of SWS, compared with UWS. Among the dry mouth-related symptoms, there were statistically significant differences in Dry-PM (oral dryness at night or on awakening), Dry-day (oral dryness at other times of the day), and Eff-life (effect of oral dryness on daily life) after use of CMC-based artificial saliva.
