Objectives: to evaluate anti-obesity activity of 8 natural candidate plants, including Artemisia iwayomogi, Juniperus chinensis, Allium cepa, Astragail Radix, Coix lachryma-jobi var. ma-yuen, Cassiatora, Rehmanniae Radix Preparata, and Pterocarpus santalinus, in a rodent model with high-fat diet (HFD)-induced obesity.
Results: SD rats were fed HFD, or 8 different test diets including hot-water extract of Artemisia iwayomogi, Juniperus chinensis, Allium cepa, Astragail Radix, Coix lachryma-jobi var. ma-yuen, Cassiatora, Rehmanniae Radix Preparata, and Pterocarpus santalinu for 11 weeks, and measured for diverse obesity related biomarkers. Among eight plant extracts, Juniperus chinensis and Artemisia iwagomogi exhibited the most prominent anti-obesity effects based on body weight gain, visceral fat-pad weights, blood and hepatic lipid profiles and other obesity-related biomarkers, such as blood levels of glucose, insulin and leptin.
<2nd year>
Objectives: to evaluate the protective effect of Juniperus chinensis ethanol extract (JC) and Artesimia iwayomogi ethanol extract (Al) against high-fat-diet (HFD)-induced obesity and their molecular mechanisms in the visceral adipose tissue of mice.
Results: JC or Al ethanol extract added at 0.5%, 1%, and 2% levels to the HFD significantly lowered body weight gain, visceral fat-pad weights, blood lipid levels, and blood insulin and leptin levels of mice. Anti-obesity activities of JC or Al ethanol extracts were most significant at a 0.5% level added to the HFD than at higher dietary levels. Feeding with JC or Al ethanol extract significantly reversed the HFD-induced down-regulation of the epididymal adipose tissue genes implicated in adipogenesis, such as the peroxisome proliferator-activated receptors $\gamma$ 2 (PPAR$\gamma$ 2), adipocyte protein aP2 (aPE), sterol regulatory element binding protein 1c (SRESP1c), fatty acid synthase (FAS), and HMG-CoA reductase (HMGR), as well as those involved in uncoupled respiration, such as the uncoupling protein 2 (UCP2) and uncoupling protein 3 (UCP3). Dietary supplementation with JC also reversed the HFD-induced decreases in the AMP-activated protein kinase (AMPK) and the acetyl-CoA carboxylase 2 (ACC2) expressions at both the mRNA and protein levels and restored the HFD-induced inhibitions in the AMPH and ACC2 phosphorylation, which are related to fatty acid $\beta$-oxidation, in the edipidymal adipose tissue. This study reports that the JC or Al can have an anti-obesity effect in a rodent model with HFD-induced obesity through an enhanced gene transcription of the uncoupling protein as well as an elevated AMPK protein expression and phosphorylation in the visceral adipose tissue.
