연구목표 미토콘드리아에서의 에너지 대사 억제로 유도된 세포노화와 세포고사의 기전을 에너지 대사와 신호전달 측면에서 그 기전을 밝힘으로서, 1) 노화와 암과 같은 질병 유도에 있어서의 에너지 대사의 중요성과 역할을 밝히고, 2) 두개의 다른 세포내 현상으로 이끄는 주요 인자가 무엇인지를 찾아 내려고 한다. 연구내용 철이온 흡착제인 Deferoxamine로 유도된 간장세포의 세포노화에서 미토콘드리아에서의 대사 및 기능 이상 연구 1) 세포노화 조건 설정 및 확인 Iron chelator인 Deferoxamine을 이용하여, 세포 노화성 성장억제를 유도시키는 조건을 deferoxamine의 여러가지 다른 농도와 처리시간을 사용하여 확립. 세포노화성 성장억제의 기준은 세포노화성 beta-galactosidase 활성의 발현과 세포형태의 변화, 그리고 성장억제를 growth curve를 통해 관찰. 노화성 세포성장 조절인자로 TGF-b1 및 p27단백질이 관여함을 확인 2) 미토콘드리아에서의 에너지 대사 기능을 ATP level, membrane potential, complexes의 발현도와 활성으로 확인 i)미토콘드리아의 membrane potential의 변화를 JC-1 및 TMRM staining을 통해 확인. ii) oxidative phosphorylation에 관련된 단백질의 발현도를 western blot으로 관찰. iii) 미토콘드리아에서의 호흡율을 O2 소비량 측정을 통해 확인 3) 세포노화의 주요 매개체를 ATP회복과 ROS 제거법을 적용하여 확인 2)번을 통해 세포노화 유도의 주요원인이 ATP나 ROS level임이 확인된 경우 STP를 회복시켜 주거나, ROS생성을 억제시켜 줌으로서 세포노화가 억제되는지를 확인. 연구성과 본 연구에서 2001년 초기에는 간장세포에서 complex I 저해를 통한 세포고사 및 철이온 흡착을 통한 세포노화에서 미토콘드리아에서의 대사 및 기능 이상 연구를 통해 미토콘드리아의 중요성을 연구하고자 하였으나, 좀 더 깊이 있는 연구를 위해 연구기간 2년동안 세포노화만를 중심으로 연구하게 되었슴 1) 철이온 흡착제인 deferoxamine을 간장세포에 처리하여 세포가 세포주기의 G1 phase에 축적됨과 동시에 세포노화가 유도되는 것을 확인하고, 이 과정에서 TGF- $\beta$1과 p27 단백질이 매개된 신호체계를 통해 매개되고 있슴을 확인하여 논문으로 보고하였슴 -- Biochemical Journal (2002) 366: 613-621. 2) 철이온 흡착제로 유도된 간장세포의 노화유도과정에서 미토콘드리아의 대사이 상과 10배 이상으로 길어진 형태학적이상을 관찰하고, 이 과정은 활성 산소 생성에 의한 산화적 스트레스와 미토콘드리아 DNA의 감소가 연관되어 있슴을 밝힘으로서 철이온 흡착을 통한 세포노화과정에서도 미토콘드리아의 기능이상에 주요하게 관여 하고 있슴을 보고 -- Annals of New York Acad. (2003) in press 3) 철이온 흡착제로 유도된 간장세포의 노화유도과정에서 미토콘드리아의 대사 이상은 complex II의 두번째 subunit인 iron sulfur protein의 발현 억제에 의한 것이며, 이로 인해 세포 내 에너지 수준이 저하되고, complex II의 활성도가 초기에 특이적으로 저해됨으로서 세포주기가 지연됨을 확인하여 J. Purpose of Research Recently, we developed the cellular senescence model in Chang liver cells by applying a iron chelator, deferoxamine, which abruptly decrease ATP synthesis in mitochondria. On the other hand, apoptosis in Chang cells was induced via inhibition of complex I activity by Rotenone.
Purpose of Research Recently, we developed the cellular senescence model in Chang liver cells by applying a iron chelator, deferoxamine, which abruptly decrease ATP synthesis in mitochondria. On the other hand, apoptosis in Chang cells was induced via inhibition of complex I activity by Rotenone. From those two systems, we will analyze the changes of energy metabolism and signal transduction pathway in the process of cellular senescence and apoptosis. The specific aim of this study is to elucidate the mechanism of cellular senescence in the view of energy metabolism and signal transduction and finally understand their relationship. Contents of Research Elucidation of the mechanism of cellular senescence induced by deferoxamine 1) Conditioning and confirmation of cellular senescence in Chang cells - examination of cellular growth rate, cell cycle pattern, cellular morphology, and senescence-associated $\beta$-galactosidase - examination of G1 arrest regulators 2) Mitochondrial energy metabolism: - ATP level, membrane potential, - activities and expression of complexes - mitochondrial respiration 3) Importance of mitochondrial function: - Effect of ATP supply using high glucose media - ROS and damage of mitochondrial DNA - mitochondrial morphological changes Effectiveness of Research 1) we observed that sub-cytotoxic concentration of deferoxamine mesylate (DFO), an iron chelator, specifically inhibited transition of a hepatocyte cell-line, Chang cell, from G1 to S phase, accompanied with appearance of senescent biomarkers. DFO induced senescence-like G1 arrest in hepatocyte cell lines and the arrest was associated by induction of p27Kip1 through TGF-b1, independent of p53. (published on Biochemical Journal (2002) 366: 613-621.) 2) Under the treatment of 1M DFO, total cellular ATP level was irreversibly decreased with concurrent disruption of mitochondrial membrane potential (rYm) implying that it might be one of the crucial factors involved in the arrest. DFO did not directly inhibit the mitochondrial respiratory activities in vitro. Among the respiratory activities, complex II activity was specifically inhibited through down regulation of expression of its iron-sulfur subunit. We also observed that mitochondrial morphology was dramatically changed to highly elongated form. (Annals of New York Acad. Sci. (2003) in press) 3) We found that the decrease of ATP level in DFO-induced senescence-like G1 arrest was most likely due to loss of SQR activity through down regulation of iron-sulfur protein expression by iron-chelation. From these results, complex II could be considered as one of critical factors to regulate mitochondrial function by responding to the iron level, and thus influencing mitochondrial function and finally cellular growth by delaying cell cycle.
