▶ 중간엽 줄기세포로부터 연골, 골 및 지방세포로의 최적 분화 조건을 확립하였으며, Troglitazone을 이용한 지방세포로 분화 촉진법을 개발하였고, 프로테오믹스를 이용하여 연골세포 분화시 변화 단백질 약 18개와 골아세포 분화시 약 21개의 단백질을 동정하였음. 특히 연골세포 분화에서 증가하는 것으로 밝혀진 MMP13은 연골분화 후기에 나타나는 단백질로서, in vitro 분화기간을 결정하는 지표로서 제시할 수 있었음. 또한 3차 배양 및 정수압과 전단력을 가할 수 있는 bioreactor를 고안하였으며, 정적 배양법에 비하여 3차원 배양조건에서의 생역학적 자극 (정수압 +2기압, 전단력 30 rpm)에 의해 골아세포의 분화가 최적으로 촉진되는 것으로 밝혔음. 또한 기존에 알려지지 않은 본 연구 분화 모델에서의 생역학적 자극에 의해 촉진 되는 세포신호전달 기전을 밝힘. (4) 임상적용을 위한 동물 모델 확립 ▶ 임상에서 세포 주입에 용이하도록 고안된 온도 민감성 주사형 폴리머를 개발 및 보완하여, 질병치료 모델로서, 요로 결석 및 역류성 식도염과 관절염 치료모델을 제시할 수 있었음. 또한, 세포 이식 후 동반될 수 있는 세포의 독성 테스트를 통해 토끼 모델에서 관찰한 결과, 근육에 이식된 자가 중간엽 줄기세포가 신장으로 이동되는 것을 관찰 할 수 있었으며, 이는 줄기세포의 타 조직으로 이동에 대한 문제 해결방안의 필요성을 제시하고 있음.

Mesenchymal stem cells are a kind of adult stem cells and mainly existed in the bone marrow. Recently, mesenchymal stem cells are known to be separated from cord blood and adipose tissue and so on. They are known to multipotent cells which can differentiate into bone, cartilage, muscle, and fat. Because they can be easily separated from bone marrow and applied as a cell therapy for regeneration of injured tissue through relatively simple manipulation without any morbidity, several researches have been performed. The purposes of this particular project are to develop the cell therapy and apply this concept from bench to bed through establishment of mesenchymal stem cell line and induction of differentiation from multipotent cells into several tissues. In order to realize these final goals, we will try to find out the mechanism of differentiation and transdifferentiation into several tissues from mesenchymal stem cells, interpretate the function and control mechanism of genes and proteoms related differentiation, establish the system of biomechanical stimuli for enhancement of differentiation, and construct the cutting edge technologies to produce the artificial tissues through the tissue engineering concepts. Our research team will be composed of medical doctors, cell biologists, and specialists for biomaterials, biomechanics and tissue engineering. For effective progress and accomplishment of our purposes, we will focus on four target diseases such as vesicoureteral reflux, urinary incontinence, osteoporosis, and articular cartilage defects for early clinical application of cell therapy concept. Specifically, we will try to develop the minimally invasive cell therapy technology through combination of mesenchymal stem cells and injectable biomaterals. The main strong points of this project from others are early clinical application using noble injectable biomaterials, the development of proliferation and differentiation of mesenchymal stem cell through biomechanical stimuli, and its originality to observe the changes of proteoms related to biomechanical stimuli in the process of differentiation.
