본 연구 과제의 목표는 마이크로어레이 기술을 이용하여 유전독성/비유전독성 발암성을 시스템 독성학적 측면에서 예측하고자 하는 데에 있다. 이에 따라 구체적인 연구 방법으로서 유전독성 발암물질로 알려진 2-acetamidofluorene, 3’ methyl dimethylaminoazobenzene, N-nitroso-diethylamine의 마이크로어레이 데이터와 이의 비교를 위한 비 유전독성 발암물질 clofibrate, ethionine, dioxane의 마이크로어레이 데이터를 식약청으로부터 전달받아 분석하여 다양한 각도의 예측 모델을 확보하고자 하였으며 유전독성/비유전독성 여부를 알지 못하는 식약청 데이터(DCP, methyleugenol, urethane, sodium nitrate)와 GEO를 통해 얻은 국외 데이터를 이용하여 이 모델을 검증하고자 하였다. 예측 모델은 크게 유전체학 데이터의 분석을 위해 표준화된 상용프로그램인 GeneSpring과 다양한 데이터의 처리가 가능한 오픈 소스인 R 프로그램을 이용하여 확보하였으며, 방법 또한 기본적인 통계적 분석 방법인 t-test, ANOVA에서부터 보다 전문화된 판별분석(classification) 모델링까지 여러 분석방법을 경우에 따라 선택, 적용하여 결과적으로 본 연구가 다음 연구자들이 상황에 맞는 분석법을 선택하고자 할 때에 도움이 되는 지침서가 되도록 하고자 노력하였다. 또한 기존의 연구들에서 예측 모델 확보 자체에 의미를 두고 이의 정확성을 근거로 제공하는 단계에서 연구를 종료하는 것에 비하여 본 연구에서는 확보한 예측 모델의 생물학적작용기전에서의 위치를 규명하고 pathway를 밝힘으로서 예측모델에 시스템 독성학적인 의미를 추가하는 단계를 진행하였다. 이 과정에는 ResNet과 Chemeffect의 상용 데이커베이스를 기반으로 하는 프로그램 PathwayStudio와 기존에 밝혀진 pathway와의 연관성을 검색할 수 있는 데이터베이스 인 DAVID가 이용 되었으며 그 외의 자세한 유전자의 정보는 NCBI의 Entrez gene을 참고하였다. 이 결과로 예측모델로서의 유전자 목록은 독성학적 의미를 가진 바이오마커로서 기능하게 되었으며 이의 발현 패턴을 통해 발암물질의 특성을 예측하는데 도움을 줄 뿐 아니라 유전독성과 비 유전독성 발암물질 각각의 투여 시에 시스템 독성학적 차원에서의 변화가 무엇인지까지도 예측할 수 있게 해주었다. 따라서 이의 모든 과정을 통해 ‘시스템 독성학적 유전독성 발암성 예측 연구’라는 본 연구의 주제를 만족하였으며 이의 결과를 포스터와 논문 등을 통해 대외적 성과로서 제시하였다

The aim of this research project is to apply the microarray technology to predict genotoxicity and carcinogenicity in the aspect of systems toxicology. For a concrete research method, the microarray data on 2-acetamidofluorene, 3’ methyl dimethylaminoazobenzene and N-nitroso-diethylamine known as genotoxic carcinogen and the microarray data on clofibrate, ethionine and dioxane known as non-genotoxic carcinogen were provided by KFDA (the Korean Food and Drug Administration) for their comparison and analysis in order to obtain a predictive model approached at various angles. For the verification of this model, the following data were introduced : the KFDA data (DCP, methyleugenol, urethane, sodium nitrate) indicating nothing about the presence of genotoxicity and non-genotoxicity, as well as the foreign data acquired through GEO. The predictive model was acquired largely by GeneSpring, a commercial program standardized for the analysis of genomics data, and a open-source R program offering various data processing techniques. Multiple analysis methods were adopted and used selectively for suitable applications, ranging from fundamental methods of statistical analysis such as t-test or ANOVA to more specialized classification modeling. As a result, this aimed to present a helpful guideline for next researchers who would intend to choose an appropriate analysis for a situation. Besides, unlike the existing researches putting great importance on obtaining the predictive model itself and ending the researches at the stage of providing necessary support data based on the accuracy of the predictive model, this research provided insights into the position of the acquired predictive model in biological mechanisms and into its pathway, to the stage of adding the systems toxicological significance to the predictive model. During the process, applied was DAVID, a search database for connections between PathwayStudio based on the commercial database including ResNet and chemeffect and the pathway revealed previously. Other detailed gene information for reference was from NCBI Entrez gene. Based on this result, the gene list as a predictive model could serve as a biomarker of toxicological significance, and its expression pattern facilitates predicting not only the carcinogen characteristics but also even changes in the systems toxicology when applying genotoxical and non-genotoxical carcinogens respectively. Throughout the process, therefore, the research subject 'Study on Predictions over Genotoxicity and Carcinogenicity of Systems Toxicology' was satisfied and the research findings were presented as the outside performance through posters and papers.
