연구의 목표는 동맥경화증과 혈관재협착의 발생에 중요한 혈관평활근세포의 기능을 제어할 수 있는 새로운 기술을 개발하는 것임. 이를 위하여 혈관평활근세포 증식이 증가된 대사성 증후군환자에서 1)혈관평활근세포의 증식증가에 미치는 인자, 2)혈관평활근세포의 신생내막억제 기전, 3)혈관평활근세포의 석회화의 발생기전을 규명함. 나아가 각각의 과정을 성공적으로 제어할 수 있는 약물표적과 이를 조절하는 선도물질을 발굴하고 동맥경화증 등의 혈관질환에서 혈관평활근세포의 기능제어를 통한 새로운 치료법과 동시에 대사성질환의 혈관합병증의 치료 및 대사성질
연구의 목표는 동맥경화증과 혈관재협착의 발생에 중요한 혈관평활근세포의 기능을 제어할 수 있는 새로운 기술을 개발하는 것임. 이를 위하여 혈관평활근세포 증식이 증가된 대사성 증후군환자에서 1)혈관평활근세포의 증식증가에 미치는 인자, 2)혈관평활근세포의 신생내막억제 기전, 3)혈관평활근세포의 석회화의 발생기전을 규명함. 나아가 각각의 과정을 성공적으로 제어할 수 있는 약물표적과 이를 조절하는 선도물질을 발굴하고 동맥경화증 등의 혈관질환에서 혈관평활근세포의 기능제어를 통한 새로운 치료법과 동시에 대사성질환의 혈관합병증의 치료 및 대사성질환 자체의 치료 및 예방의 기반기술을 확보하고자 함.
※ 본 연구는 지난 연구기간 동안 연구계획서에 제시된 바와 같이 성실히 대사성질환과 혈관질환의 상호관계와 이들의 치료시스템에 대해 연구를 진행한 결과 다음과 같은 연구결과를 도출하게 됨.
1) 대사성질환에 의한 혈관세포의 기능이상에서 신규 표적분자 발굴 및 분자적 기전규명: 미토콘드리아와 ER의 기능에 관여하는 표적분자 PGC-1, PDK4, ATF6, TRB3, SHP, clusterin, Nrf2, NQO1등을 발굴하였고, 이 들의 분자적 기능을 혈관세포와 타 대사성 질환의 조직에서 명백히 하였음.
2) 발굴된 표적분자의 기능연구를 통해 대사성질환과 동맥경화증의 연관성 규명: 대사성 질환와 동맥경화증의 표적분자가 서로 공유함을 관찰하고, 표적분자 중 Clusterin, SREBP-1c, SHP, LXR등을 이용하여, 동맥경화증 즉 혈관세포의 기능이상이 대사성질환과 밀접한 관계임을 확인함.
3) 표적분자를 이용한 혈관평활근세포의 기능제어 시스템 발굴: 표적분자를 이용하여 혈관평활근세포의 기능을 제어할 수 있는 선도물질 ALA, β-Lapachone, AG110, DCA, DMF등을 발굴하였으며, 특히 ALA는 혈관재협착의 치료에 있어서 DES에 적용 가능성이 높고, 또한 β-Lapachone의 경우 혈관재협착의 치료제로써의 가능성을 인정받아 기술이전 하였음.
4) 동맥경화증에서 발굴된 표적분자와 제어시스템을 대사성질환에 적응증확대: 발굴된 표적분자와 제어시스템을 타 조직의 대사성질환에 적응/응용연구를 통해 표적분자와 제어시스템의 응용범위를 넓힘. PDK4: 혈관석회화, insulin resistant. SHP: 혈관섬유화, 신장섬유화, insulin 생성. ALA: 혈관재협착/석회화, 지방간.
5) 동맥경화증의 진단/예측 신규표식자 개발: 대사성질환 (당뇨병) 환자에서 미토콘드리아 활성에 관여하는 PPARg, PSMB5, Kir6.2의 유전자의 SNP를 발굴하였으며, 제 2형 당뇨병환자에서 혈관석회화와 관계있는 바이오마커 ENPP-1, MMP3를 발굴하였음.
본 연구에서 발굴된 연구결과는 타 연구자와 기초지식를 공유하여 기초연구 발전에 기여할 것이며, 응용연구를 통하여 치료제개발에 활용가능성이 높다고 사료됨. 연구기술적면에서 개발되는 각종 in vitro (세포)실험구축과 in vivo (동물모델)실험구축은 대사성 질환 및 혈관질환을 연구하는 타 연구자에게 기술지도를 통해 대사성질환 연구분야에서 국가 경쟁력을 향상시킬 것으로 사료됨.

The purpose of this project is to develop the strategies that can control the function of the vascular smooth muscle cells (VSMCs), which play a crucial role in the pathogenesis of atherosclerosis and restenosis. To achieve this goal, we will 1)identify the factors regulating VSM C proliferation, 2)
The purpose of this project is to develop the strategies that can control the function of the vascular smooth muscle cells (VSMCs), which play a crucial role in the pathogenesis of atherosclerosis and restenosis. To achieve this goal, we will 1)identify the factors regulating VSM C proliferation, 2) elucidate the molecular mechanism of inhibition of VSM C proliferation and 3) elucidate the mechanism of vascular calcification in the patients with the metabolic syndrome (M S). Furthermore, we will discover new drug targets and novel compounds which can modulate their activity. On the basis of these achievements, we will establish the novel therapeutic strategies to treat metabolic syndrome as well as its related complications including atherosclerosis.
1) Identification and elucidation of molecular mechanism of action of target genes involved in vascular cell dysfunction
- Identified target gens that are associated with mitochondria and ER function such as PGC-1, PDK4, ATF6, TRB3, SHP, Clusterin, Nrf2 and NQO-1
- Defined the functions and roles of identified genes in vascular cells and other MS-related tissues.
2) Verification of the corelation between MS and atherosclerosis through the study of identified genes
- found that MS-related genes are closely related to vascular cell dysfunction
- suggested that these identified genes are common targets for the treatment of MS and atherosclerosis.
3) Development of control system of VSMC function by modulating target gene expression and function
- identified lead and natural compounds that can control VSMC function such as ALA, β-Lapachone, AG110, DCA and DMF.
4) Application of regulators and modulators of identified target genes to other MS-associated complications
- ALA: liver steatosis, vascular restenosis and calcification
5) Development of new prognostic or diagnostic markers
- identified SNPs of the genes (PPARg, PSMB5, and Kir6.2) which are associated with mitochondria function in diabetic patients
- identified polymorphisms of the genes (ENPP-1 and MMP-3) which are associated with vascular calcification in diabetic patients
Successful accomplishments of this project will open new windows for the discovery of new molecular targets for the prevention and treatment for vascular diseases such as vascular restenosis and calcification as well as other MS-associated complications. Moreover, results achieved through this project will provide the fundamental knowledge for the advancement of other basic researches. The in vitro model of cell experiments and in vivo models of animal experiments established in this research project will be shared and transferred to other researchers who are not able to apply these techniques to their researches.
