Purpose(500자내외) Cell death is one of the major factors maintaining body homeostasis. Abnormality in cell death-controlling mechanism is associated with development of cancer, ischemic and neurodegenerative diseases. The aim of this research center is to understand molecular pathogenesis of diseases related to cell death, and to find candidate targets for early diagnosis and new therapeutic agents through integrated work of basic medical scientists and clinical practitioners. The main purposes of this project are as follows. ① To establish basic techniques for understanding cell death mechanism and the integrated analysis. ② To establish basic techniques for biotechnology and secure necessary resources. ③ To develop tolerance-inducing substances for cell death. ④ To understand cell injury model and cell death mechanism occurring in tumor. ⑤ To find substances inducing or inhibiting cell death. ⑥ To apply candidate targets to develop new early diagnostic technique and therapy for the general improvement in public health. ⑦ To overcome human diseases through understanding cell death mechanisms. ⑧ To enhance national competitiveness by rearing young basic medical researchers. Contents (1000자내외) Basic medical professors specializing in anatomy, physiology, biochemistry, pathology and pharmacology, and clinical professors specializing in internal medicine, surgery, and psychiatry participated to study diverse approach to cell death and methods to put research to practical use. The research has been carried out under the following five major categories. Chapter 3: The applicational research of oxidative injury Section 1: The induction of tolerance for cell death in oxidative injury model We have carried out research on cell death in normal cytogenesis or pathological process based on kidney development and disease model. We have developed 4 different models to study the importance of cell death in pathological process: 1) Stage-specific changes in various carriers/transporters, such as EGF, LYVE-1, AQP2, UT-A and TonEBP, related to concentrating urine in the process of kidney development. 2) Differentiationof intercalated cell, which plays an important role in maintaining body acid-basis balance. 3) Ischemic damage. 4) Kidney toxicity caused by an immunosuppressant, cyclosporine. Section 2: Development of therapeutic target proteins for diseases caused by oxidative cell death We have identified target gene by screening expression level of genes, which are induced by increased cellular metabolism and oxidative substances inevitably resulting from environment. We then examined molecular functions and working mechanisms of the target gene to provide the basis of developing new therapeutic techniques for diseased related to oxidative cell damage. Chapter 4: Practical study of neuronal disorders Section 1: Neuronal ion channelopathies Channelopathy refers to diseases induced by abnormal activation of cells. This abnormal activation results from from functional defect of ion channels by either genetic or environmental factors. We carried out electrophysiological research on channelopathy. Based on this research, we studied cell death and many other neurodegenerative diseases. Section 2: Neurodegenerative diseases in the brain - Based on models for cerebral infarction, ischemic stroke and reiterated/repetitive epilepsy, we searched for mechanism and regulatory factors (drug, gene, environment etc.) of neuronal death and production. Chapter 5: Cell death oncology for translational research Section 1: Cell death oncology for translational research We have collected more than 200 samples each for gastric, liver, colorectal and breast cancer, to establish cancer tissue bank withclinical information. We selected candidate gene based on cDNA, miRNA chip and array CGH research results. We examined possible gain or loss of function due to different expression level, mutation, genetic loss, methylation and transposition of the candidate gene. Finally, we studied the prospect of putting these results to practical use Performance 174 SCI researchpublication, 14 patent application, 611 presentation in symposiums, 18 symposiums (including 6 international symposiums) Fostering basic medical scientists We have nurtured 32 individuals with master’s degree, 19 individuals with Ph.D. degree and 8 basic medical physicians. Out of 17 physicians, 6 have been appointed as the medical school professors. Together, we hope that translational study in cell death research will provide excellent target molecules for early diagnosis and treatment, and improve the patients’ prognosis and quality of life.
