Forty eight field isolates of IBV were recovered from commercial chicken flocks in Korea since 20036 and characterized phylogenetically by nucleotide sequence analysis of the IBV S1 gene hyper-variable region. Our phylogenetic analysis reveals that recent field isolates of IBV form at least three distinct phylogenetic types including K-I, K-II and K-III types. K-I type IBV consisted of indigenous 13 IBV isolates which evolved from the Kr-EJ/95 strain and then separated into lineages, type K-Ia and type K-Ib. K-II type IBV isolates (19 isolates) were closely related to nephropathogenic IBV variants from China and Japan. K-III type isolate (Kr/D064/05), first identified by this study, was closely related to enteric IBV variants from Chinese strains which cause proventriculitis. Sequence comparisons showed amino acid differences of at least >27.5% between IBV types. Next, pathogenic and antigenic characteristics of field strains of IBV in Korea were assessed. Specific pathogen free chickens of 5 day-old after K-IIb type IBV isolate challenge showed severe respiratory signs (dyspnea) and nephropathogenic lesions (swollen kidney with nephritis and urate deposition). The pathogeny was similar to K-Ia IBV strain. However, K-IIb field strain and K-IIa M91 strain calculated by virus neutralization index (NI) was not antigenically related, indicating that K-IIb type isolate is a new variant different antigenically from KM91 strain. This is the first report that demonstrates new antigenic variant of nephropathogenic IBV in chicken from Korea. Due to variations in serotypes among different strains of avian infectious bronchitis viruses (IBV), vaccination of chicks with imported vaccines fails to protect them from IBV infections in Korea. Therefore, we develop attenuated vaccines from local strains in Korea. A Korean Group I (KI) strain was passaged 80 times or more through SPF chicken embryonated eggs, and then tested in SPF chickens. Vaccinated birds with attenuated IBV had efficacy of 90% for a Korean Group II (KII) field strain as well as a Korean Group K1 strain. Additionally, the KIIb type isolate attenuated vaccine strain had no reversion to virulence after back passages six times in chicks. In conclusion, vaccinated birds with K-IIb type isolate were also protected from new variant nephropathogenic IBV isolated since 2003. These vaccine strains have potential for controlling local Korean IBV infections in chickens.
