○세포의 발생과 분화 및 노화 과정에서 많은 세포들이 apoptosis라는 자연적인 세포사멸과정에 의해 제거된다.
○ 그러나 세포의 분화 및 발생과정에서 뿐만 아니라 질병이나 방사선 조사 등에 의해서도 apoptosis의 특징적인 현상들이 나타나는 것으로 밝혀져 apoptosis의 원인 및 발생기전 나아가 apoptosis를 방지할 수 있는 약물의 개발 등에 관한 연구가 활발히 이루어지고 있다.
○ 세포의 사멸과 연관된 세포내 신호전달과정이 많이 있으나 근래 $Ca^{2+}\;homeostasis$의

○ 세포의 사멸과 연관된 세포내 신호전달과정이 많이 있으나 근래 $Ca^{2+}\;homeostasis$의 상실도 세포의 사멸을 유발하는 요인으로 생각되고 있다.
- 그러나 세포내 칼슘증가가 어떤 기전에 의해 일어나는지 아직 밝혀져 있지 않음.
○ 따라서 본 연구에서는 대표적인 세포사멸 유발물질인 staurosporine을 사용하여 세포내 칼슘의 동원 부위 및 칼슘증가가 세포사멸에 미치는 영향을 밝히고자 하였다.
○ 타액선세포에서 staurosporine에 의해 세포사멸이 유도될 때 세포내 칼슘이 증가하는데 이때 칼슘을 동원하는 부위가 어디인지 확인.
▶ 타액선세포를 carbachol로 자극하고 세포외부의 칼슘을 제거한 후 staurosporine을 투여하면 세포내 칼슘농도가 증가하는데 이는 staurosporine에 의한 세포질내 칼슘증가가 세포외부나 소포체 (endoplasmic reticulum) 이외의 다른 세포내 소기관에서 칼슘이 유리되어 일어남을 보여주는 결과이다.
▶ 타액선세포를 carbachol로 자극한 후 세포외 칼슘을 제거하여 소포체의 칼슘을 고갈시키고, 미토콘드리아의 막전위차를 없애주어 미토콘드리아내 칼슘을 세포질로 유리시키는 carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP)를 투여하였으나 칼슘농도의 증가는 거의 없었으며 칼슘 ionophore인 ionomycin을 투여하여도 역시 칼슘증가가 매우 작은 것으로 보아 타액선세포에서는 미토콘드리아에 많은 양의 칼슘이 저장되어 있지 않으며, 따라서 staurosporine에 의한 칼슘증가에 미토콘드리아는 관여하지 않음을 확인하였다.
▶ Carbachol로 타액선세포를 자극한 후 세포외 칼슘을 제거하고 ionomycin과 monensin (세포 소기관내 pH를 외부와 같게 해주는 $Na^{+}/H^{+}\;exchanger$)을 투여하면 세포내 칼슘이 즉각 증가하는데, 이는 분비과립과 같은 세포내 산성 소기관에 고농도의 칼슘이 들어있음을 의미한다. 또한 staurosporine을 처리하여 세포내 칼슘농도를 증가시킨 후 ionomycin과 monensin을 투여하면 세포내 칼슘 농도가 전혀 증가하지 않아, staurosporine에 의한 칼슘증가와 ionomycin과 monensin에 의한 칼슘증가는 동일한 세포내 칼슘 저장고로부터 나온다는 것을 알 수 있었다.
▶ 나아가 분비과립이 staurosporine에 의해 칼슘을 유리하는 소기관이라는 것을 좀 더 정확하게 증명하기 위하여 isoproterenol을 흰쥐 복강내 투여하여 분비과립이 고갈된 상태의 세포를 이용하여 같은 실험을 시행한 결과, 분비과립을 유출시키지 않은 세포에 비해 칼슘증가가 크게 줄어들었음을 확인하여 분비과립이 고갈된 세포에서는 staurosporine에 의한 칼슘 증가가 거의 나타나지 않았다. 이와 같은 실험 결과는 staurosporine이 타액선세포의 분비과립으로부터 칼슘을 유리하여 세포질내 칼슘농도를 증가시킴을 보여 준다.
○ Staurosporine에 의한 칼슘증가가 protein kinase C의 억제 때문인지 확인
▶ Staurosporine은 강력한 비특이적 protein kinase C 억제제이다. 따라서 staurosporine에 의한 세포내 칼슘증가 효과가 protein kinase C를 억제하여 나타나는 현상인지를 확인하기 위하여 protein kinase C의 catalytic domain을 봉쇄하는 K252a와 chelerythrine 그리고 regulatory domain을 봉쇄하는 H-7과 calphostin C를 투여하였을 때 이들 모두 세포내 칼슘을 증가시켜, 세포내 칼슘을 증가시키는 staurosporine의 효과가 protein kinase C를 억제하기 때문일 것으로 생각된다.
○ Staurosporine에 의한 세포사멸과정에 caspase-3 활성화 여부 확인
▶ Staurosporine을 처리하면 농도와 시간에 비례하여 세포사가 증가하는데 이 때 PARP와 Ac-DEVD-pNA의 절단도 함께 나타남을 확인하여 staurosporine에 의한 세포사는 caspase-3의 활성화에 의한 것임을 확인하였다. 그러나 caspase-3의 mRNA 양은 변화가 없음을 확인하여 staurosporine에 의한 세포사멸은 caspase-3의 양적 변화 보다는 caspase-3의 활성화 때문임을 알 수 있었다.
○ Staurosporine에 의한 세포사멸에서 Bcl-xL의 역할
▶ Bcl-xL을 과발현시킨 세포에서 staurosporine에 의한 세포사멸이 현저히 억제되었으며 caspase-3의 활성화도 억제되어 Bcl-xL이 staurosporine에 의한 세포사멸 과정에 관여하는 것을 확인하였다.
○ Staurosporine에 의한 세포사멸과정에 세포내 칼슘증가의 역할 확인
▶ 세포내 칼슘을 제거하는 chelating agent인 EGTA/AM을 전처치하고 staurosproine을 처리하면 세포사멸이 통계적으로 의의있게 억제되는 것을 확인하여 staurosporine 처리에 의해 증가되는 세포내 칼슘은 세포사멸을 유발하는 요인임을 확인하였다. 이와 같은 실험 결과는 staurosporine에 의한 세포사멸 과정에서 세포내 칼슘증가, bcl family, caspase-3가 서로 밀접한 관계가 있음을 보여주는 것으로 생각된다.

However, apoptotic pathways are also known to be activated by various diseases, chemicals and radiations. Although the mechanisms by which such insults induce apoptosis remain unknown, they may serve as therapeutic targets for the diseases.
It is now believed that the disruption of intracellular $Ca^{2 }$ homeostasis is a critical event in apoptosis. The $Ca^{2 }$ -ionophore, such as A23187 and ionomycin, and the selective inhibitor of endoplasmic reticular $Ca^{2 }$-ATPase thapsigargin elevate cytosolic $Ca^{2 }$ concentration ([$Ca^{2 }$]c) and cause apoptosis. In addition, increase in [$Ca^{2 }$]c has been reported to contribute to apoptosis in response to various pathological conditions.
A variety of compounds and experimental conditions, including staurosporine, oxidative stress and withdrawal of trophic factors, induce apoptosis in various cells. Staurosporine, a potent inhibitor of protein kinases, has been widely used as a useful tool in understanding the biochemical chain of events involved in apoptosis. The exact mechanism by which staurosporine induces apoptosis has not been established yet.
In the present study, we aimed to elucidate the source and the role of $Ca^{2 }$ in staurosporine-induced apoptosis in salivary acinar cells. Staurosporine increased [$Ca^{2 }$]c after inositol 1,4,5-trisphosphate (IP3)-sensitive $Ca^{2 }$ store was depleted. Ionomycin caused only small increases in [$Ca^{2 }$]c after the depletion of $IP_{3}-sensitive$ $Ca^{2 }$ store, whereas ionomycin monensin caused large increases. However, ionomycin monensin did not increase [$Ca^{2 }$]c when added after [$Ca^{2 }$]c was increased by staurosporine, indicating that acidic $Ca^{2 }$ store was the main source of $Ca^{2 }$. The acidic $Ca^{2 }$ store appeared to be associated with secretory granules, since ionomycin monensin- and staurosporine-induced [$Ca^{2 }$]c increases were significantly reduced when the acinar cells were degranulated using isoproterenol. The effect of staurosporine on [$Ca^{2 }$]c was mimicked by other protein kinase C inhibitors. These data indicate that staurosporine mobilizes $Ca^{2 }$ from secretory granules probably through the inhibition of protein kinase C in rat ubmandibular acinar cells.
Staurosproine-induced cell death was accompanied by DNA fragmentation and PARP cleavage, suggesting that the cell death was due to apoptosis. In addition, Ac-DEVD-pNA, a specific caspase-3 substrate, was cleaved although total amount of caspase-3 mRNA was not affected, indicating that activation of caspase-3 was responsible for the staurosporine-induced apoptosis. Pretreatment of the cells with EGTA/AM, an intracellular $Ca^{2 }$ chelator, significantly attenuated the staurosporine-induced apoptosis. The over expression of Bcl-xL prevented PARP cleavage and cell death. In conclusion, staurosporine mobilizes $Ca^{2 }$ from secretory granules probably by the inhibition of protein kinase C and the staurosporine-induced cell death is attributed to the $Ca^{2 }$ increases
