본 연구는 대표적 식중독균에 대해서 항균작용을 갖는 천연 식물소재를 발굴하여 실용화할 수 있는 기반을 제공하고 아울러 다양한 종류의 유산균으로부터 유용한 박테리오신을 생산하여 항균제로 활용할 수 있는 기술을 개발하며, 살모넬라와 패혈증 비브리오균을 모델로 하여 병원성 유전자의 발현조절 기전에 대한 연구를 통하여 세균성 식중독을 제어할 수 있는 새로운 차원의 방법을 제시함과 동시에, 주요 세균성 식중독의 위해도 평가방법을 확립하여 식품의 안전성을 제고할 것을 목표로 수행되었다.
제1세부과제에서는 800여종의 식물소재로부터 항균소
본 연구는 대표적 식중독균에 대해서 항균작용을 갖는 천연 식물소재를 발굴하여 실용화할 수 있는 기반을 제공하고 아울러 다양한 종류의 유산균으로부터 유용한 박테리오신을 생산하여 항균제로 활용할 수 있는 기술을 개발하며, 살모넬라와 패혈증 비브리오균을 모델로 하여 병원성 유전자의 발현조절 기전에 대한 연구를 통하여 세균성 식중독을 제어할 수 있는 새로운 차원의 방법을 제시함과 동시에, 주요 세균성 식중독의 위해도 평가방법을 확립하여 식품의 안전성을 제고할 것을 목표로 수행되었다.
제1세부과제에서는 800여종의 식물소재로부터 항균소재를 선발한 결과 동백나무잎과 은행나무 잎의 추출물이 소재자원의 채취활용성과 안전성면에서 유용한 소재로 선정되었고, 특히 은행나무잎의 부탄올추출물에서 주요식중독균에 대해 항균성이 높게 나타났으며 이 항균성분은 6-hydroxykynurenic acid(6-HKA)로 밝혀졌다. 6-HKA를 함유한 은행잎추출물들은 실제 식품시스템에 첨가하거나 주방용세제와 함께 사용할 때 효율적으로 유해세균들을 제어할 수 있음이 밝혀졌다. 한편 Lactobacillus 등 실험에 사용한 다양한 유산균들은 식중독균들에 대해서 항균성을 나타냈으며 이들로부터 유효성분인 Pediocin K1과 reuterin을 분리, 동정하였다. 이중 주요식품유해균에 대해서 폭넓은 항균특성을 가진 reuterin을 효율적으로 생산하기 위한 연속공정을 개발하였다. 또한 항균소재들의 Salmonella균의 독성유전자의 발현에 미치는 영향을 밝혀 그 활용가능성을 높게 하였다.
제 2세부과제에서는 살모넬라에서 독성에 관여하는 많은 유전자들이 모여 있는 pathogenicity island (SPI)가 coding하는 type III secretion system (TTSS)이 살모넬라의 발병에 중요한 역할을 하는 SPI의 발현 및 조절 기작을 분자 유전학적 수준에서 연구하여 살모넬라균의 발병 기작에 대한 이해를 증진시키고 이를 바탕으로 살모넬라에 의한 식중독을 방지할 수 있는 새로운 방법을 개발하고자 하였다. 숙주 세포 내로 감염 후 살모넬라의 증식과 관련된 기능을 수행하는 SPI-2 TTSS의 구성인자인 ssaG 유전자의 발현 조절 기작을 in vitro 혹은 in vivo 단계에서 분석하여 SPI-2 유전자의 발현 양상에 대한 이해를 넓힐 수 있었다. 또한 살모넬라가 숙주세포 내로 침투할 때 중요한 기능을 수행하는 것으로 알려진 SPI-1 유전자들의 발현을 조절하는 InvF의 발현 양상과 조절 기작을 분석하였다. 이들 일련의 결과들을 바탕으로 살모넬라의 병원성을 조절하는 환경 인자들과 살모넬라 내 다양한 조절 인자들의 영향을 밝힐 수 있었다. 또한 살모넬라의 병원성을 모니터링 할 수 있는 시스템을 개발하여 제시하였다.
제 3세부과제에서는 패혈증 비브리오균의 gsr (Genes in Survival Regulon) 유전자의 발현에 관여하는 global regulator로 보고되고 있는 Lrp (leucine-responsive protein)와 cycle AMP receptor protein(CRP)이 결여된 V. vulnificus lrp, 또는 crp, isogenic mutant를 개발하였으며, 이변이주들의 각 환경에 대한 생존 정도를 정량적으로 비교 분석함으로써 이들 global regulator 들이 패혈증비브리오균의 생존에 주는 영향을 결정하였다. CRP와 Lrp protein은 V. vulnificus가 다양한 스트레스 조건 하에서 생존을 하는데 중요한 역할을 하는 global regulator임이 규명되었으며, CRP에 의하여 조절 받는 고 삼투압 내성유전자로서 putAPR operon을 동정하였다. 이put operon은 putA (proline dehydrogenase), putP (proline permease), 그리고 putR (Lrp likr transcription regulator) 등 3개의 operon으로 구성되어 있었다. Proline dehydrogenase 활성은 대수기에서 최고조에 이른 후 점차 감소하였으며 이러한 양상은 전사수준에서 조절되고 있었다. 또한 putAP 유전자는 하나의 전사단위로 이루어져 있으면서, putAP 전사체와 putA 전사체라는 두 개의 서로 다른 위치에 존재하는 PputA 및 PputAP promoter로부터 시작됨을 확인하였다.
Allelic exchange에 의해서 제조된 crp 돌연변이주에서 proline dehydrogenase의 생성과 전사 정도가 감소하였기 때문에, CRP가 전사수준에서 putAP의 positive regulator로 작용함을 알 수 있었다.
제4세부과제는 패혈증비브리오균의 항원성인자의 탐색 및 성격규명으로 패혈증비브리오균의 표면단백질으로 FlgE와 NlpD를 알아내고 패혈증비브리오균의 병원성에 있어서의 그 기능을 알아내는 연구를 진행하였다. FlgE가 구성하는 편모는 패혈증비브리오균이 숙주세포에 부착하는데 중요하고 그 결과 숙주세포독성능 및 mouse 독성능에 주요 역할을 한다고 밝혀졌다. 또 다른 인자인 NlpD는 흥미롭게도 숙주세포의 부착에는 중요한 역할을 하나 세포독성능에는 관여하지 않는 것을 알 수 있었고 인체세포로부터 cytokine을 생성시키는 면역단백질임을 알 수 있었다.
제 5세부과제에서는 전국 4개 대도시(서울, 부산, 대전, 광주)를 중심으로 시중에서 유통되는 식육 중 고위해 식중독균인 Salmonella 와 Campylobacter의 오염실태를 파악하고, 이들 오염도 자료를 토대로 Food MicroModelⓇ과 같은 Gompertz Model을 활용하여 환경 조건에 따른 균의 성장을 예측하여 식품의 보존 기간 산정 시 필요한 자료로 제공하였다. 국내 유통 중인 닭고기시료에서 Campylobacter 가 55.8% 검출되었으며 지역별 검출율은 서울(90건/139건, 64.7%), 광주(10건/20건, 50%), 대전(6건/20건, 30%), 부산(5건/20건, 25%)의 순으로 확인되었다
Campylobacter의 오염도는 겨울보다는 여름철이, 판매 장소 별로 시장(69건/102건, 67.6%), 슈퍼 (26건/59건, 44.1%), 백화점(16건/38건, 42.1%)의 순으로 높았다. 오염도 자료를 토대로 환경적 인자인 Food matrix, pH, Aw, 온도에 변화를 주어 균주의 성장 및 사멸 시간을 예측하였다.

The risk of meats in markets contaminated by Salmonella and Campylobater was also monitored and evaluated in this study.
In the first subproject, about 800 plant extracts were screened for the antibacterial activities against E.coli, Salmonella thphymurium, Staphylococcus and Listeria monocytogenes. Methanol extracts of Camellia japonica and Gingko biloba leaves were selected and an active antimicrobial compound, 6-hydroxykynurenic acid was isloated and identified from the butanol fraction of G.biloba extract. We also confirmed and characterized the production of bacteriocins and anti-microbial compounds from various strains of Lactobacillus, Bifidobacterium, Streptococcus, Lactococcus. The anti-microbial spectrums against various strains of food-poisoning or pathogenic bacteria were differential and specific depending on the target organisms. Evaluation on the various environmental factors and continuous processes were employed to optimize the production of bacteriocins and anti-microbial compounds from lactic acid bacteria. Purified Pediocin K1 and reuterin were more extensively assessed on their anti-microbial characteristics. The feasibility study of the application of the anti microbial compounds from lactic acid bacteria and natural biomaterials were successfully performed in the real food models such as milk, soy-milk and Kimchi. A novel anti-microbial evaluation system measuring the inhibition of the expression Salmonella genes such as hilA와. invF will be valuable.
Salmonella harbors a special gene compartment of SPI (Salmonella pathogenicity island) which is implicated in the virulence of Salmonella and composed of dozens of virulence genes encoding components of TTSS (type III secretion system), effector proteins, chaperones, regulator, and so on. In the second subproject, we investigated the regulation mechanism of SPI-1 and SPI-2 genes in order to expand our knowledge about salmonellosis and tried to develop methods controlling the pathogenicity of Salmonella. Gene ssaG, a component of SPI-2 which is essential for the survival and proliferation of Salmonella within host cells, was analyzed in vitro and in vivo. The study of ssaG helped to understand the regulation mechanism of SPI-2. We also examined the regulation of invF encoding InvF regulator of SPI-1 which is required for the invasion of Salmonella into host cells. Based on the experimental results, we could evaluate the influences of various environmental conditions and Salmonella regulators on the virulence of Salmonella. Furthermore, we developed the system capable of monitoring Salmonella virulence and explored novel materials controlling the virulence.
In the third subproject, to better understand the strategies of Vibrio vulnificus to survive osmotic stress, a mutant that was more sensitive to high osmolarity was screened from a library of mutants constructed by a random transposon mutagenesis. By a transposon-tagging method, putAP genes encoding a proline dehydrogenase and a proline permease were identified and cloned from V. vulnificus. Functions of putAP genes were assessed by the construction of mutants, whose putAP genes were inactivated by allelic exchanges. When compared to the wild type, the putA mutant exhibitedreduced survival under hyperosmotic stress. Proline dehydrogenase activity and the level of putA transcript were determined to reach a maximum in exponential phase and were then repressed when growth slowed down. Northern blot and primer extension analyses revealed that transcription of putAP genes results in two different transcripts, transcript A (putA transcript) and transcript AP (putAP transcript). Expression of putAP genes was directed by two promoters. Transcription of putAP is under the positive control of CRP (cyclic AMP receptor protein).
In the fourth subproject, libraries of Vibrio vulnificus causing septicemia were screening for its surface proteins, resulted in identification of two important clones. The FlgE compring the hook of a flagellum played an important role in adherence of V. vulnificus as well as in cytotoxicity to the host cells including mice. The other molecule identified in this process was NplD, a putative lipoprotein. A nlpD knockout mutation suggests that this protein involves in adherence, but not in cytotoxicity.
The prevention of infectious disease from contaminated foods is very important in public health. Quantitative microbial risk assessment has been used in advance countries to achieve the safety of public health against hazardous microbial causing contaminated foods. The fifth subproject was conducted to estimate risk level based on monitoring data of Salmonella and Campylobacter from porks and chickens selling at different domestic stores and regions in summer and winter. The detection rates by sampling season were higher in summer than in winter and were 67.6% in market , 44.1% in supermaket. Campylobacter was detected in 64.7% of selling chickens in Seoul , and 50% in Gwangju. Based on monitoring data, we tried to predict microbial growth according to the environmental factor such as temperature, pH, aw(water activity) using FoodMicroModelⓇ. and to propose conservation time of food.
