(SmartRodTM 상표등록:상 2003-0046684, 특허등록:KR0486028). 최적화된 제형 제조 조건에 따라 3 batch를 생산하여 약물 함량 분석과 체외 방출 실험을 통하여 제조 공정의 재현성을 확인하였으며, 제조된 인터페론 제형을 이용하여 전임상 시험을 진행하였다. 위탁기관인 안전성 평가연구소에서는 토끼를 이용한 국소독성시험 (피부자극 시험), 약동력학 및 약물역학 (쥐, 원숭이), 단회투여 독성시험 (쥐, 원숭이), 반복투여 독성시험 (쥐, 원숭이)과 시험물질의 항체 및 역가측정 실험 등의 전임상 시험을 진행하였다. 3. 연구결과 제조된 최종 제형은 RP-HPLC와 SDS-PAGE를 통하여 약물의 안정성과 순도, 약물 방출 경향 등을 확인하였고, 단회 및 반복투여독성시험 결과 임상 예정 투여 용량의 1000배에서도 독성을 나타내지 않는 안전성을 확인하였다. 서방성 인터페론 로드 제형의 기준을 설정하고, 기준 및 시험법에 따라 제형의 안정성을 평가하였다. 4°C에서 12개월 보존 후에 인터페론의 함량 및 체외 방출 시험 분석 결과 4°C에서는 12개월 이상 안정함이 확인되었다. 약물의 서방성은 원숭이 독성동태시험결과 인터페론이 10일까지 방출됨을 확인하였으며, 이것으로 본 제형이 인터페론의 1회/2주 제형으로 적합함이 판명되었다. 4. 결론 이상의 결과로 2주 1회 주사되는 서방출성 인터페론 로드 제형 제조 기술을 확립하였고, 이에 대한 안정성, 안전성 자료를 확보함에 따라 향후 국내외 제약 회사에 라이센싱을 진행할 예정이다.

1. Research background and aim Interferon-alpha is being used for the treatment of chronic hepatitis C as a three times a week injectable. In order to reduce inconvenience for the patients, we have developed a twice a month injectable formulation of interferon-alpha using our SmartRodTM technology (Trademark registration No. 2003-0046684), an innovative drug delivery system for protein and peptide drugs. In this study, we aim to obtain basic data of preclinical studies sufficient to partner with a global pharmaceutical company in developing it further and proceed to the development of a new drug. 2. Contents and methods of research Stabilized interferon-alpha microparticles were prepared by bubble drying of aqueous interferon solution containing excipients such as human serum albumin and polyethylene glycol. Interferon-rods were manufactured by extrusion of mixed powders of stabilized interferon microparticles and triglyceride powders through a ram extruder (Patent No. KR0486028). After optimization of manufacturing parameters, three batches of interferon-rod were prepared and confirmed reproducibility of manufacturing process by examining drug content and in vitro release. Preclinical studies such as local toxicity test (skin irritation test, rabbit models), pharmacokinetic and pharmacodynamic (rats, monkeys) studies, single injection toxicity test (rats, monkeys), weekly four repeating injections toxicity test (rats, monkeys), antibody formation and antiviral activity assays were performed at the Korea Institute of Toxicology. 3. Results The formulation was evaluated in terms of stability and purity of interferon-alpha in the formulation by RP-HPLC and SDS-PAGE and in vitro release of the protein. Based on the results of single and repeating injection toxicity tests, the formulation was found to be safe even at 1,000 fold dose of expecting human clinical dose. Storage stability of the formulation was evaluated by quality control methods after setting up the standards and methods of quality control. The formulation was found to be stable for 12 months at 4°C determined by interferon content analysis and in vitro release test. The formulation seemed to be very promising as twice a month release injectable based on the data of sustained release for 10 days obtained from toxicokinetic studies in monkeys. 4. Conclusion Manufacturing process for twice a month release injectable interferon formulation has been established. With safety data obtained from animal toxicity studies and long-term storage stability data, we are currently searching for a global partner to license this technology and develop it into a drug approved by the FDA.
