Purpose Hsp70 acts as an anti-apoptotic factor to protect cells from apoptosis induced by stressful stimuli, such as anticancer agents, tumor necrosis factor (TNF) and UV irradiation. In response to a variety of cellular stresses, the expression levels of Hsp70 are elevated and block the accumulation of proteins denatured by stresses. In many cancers, the expression levels of Hsp70 are remarkably increased, which leads to tumor cell survival via multiple anti-apoptotic processes. It has been suggested that the increased expression of Hsp70 promotes cancerous properties of cells and deteriorates the efficacy of anti-cancer treatments. Thus, Hsp70 could be an effective target for anti-cancer drug development. In this study, we aimed to synthesize a new apoptosis-inducing small molecule that binds to Hsp70 and Hsc70, apoptozole and to extend our studies on the potential role and mechanism of apoptozole in apoptosis-related signaling pathways. We also deduced the effect of apoptozole in various cancer cell lines and in nude mouse models xenografted with various cancer cells to examine the potential of apoptozole being used as an antitumor agent either alone or in combination with other anticancer agents.

Results In this study, we have uncovered evidence that shows that apoptozole(Az) inhibits Hsp70 activity by binding to its ATPase domain and blocks the interaction of Hsp70 with Apaf-1, thereby inducing apoptosis. We also show that Az neither suppresses the interaction of Hsp70 with the apoptosis inducing factor (AIF) nor does it influence the expression levels of Hsp70 and other apoptosis related proteins such as Apaf-1 and AIF. These results suggest that apoptosis elicited by Az takes place via a caspase activation process. We also synthesized 96 apoptozole derivatives and examined their cell death-inducing activities, demonstrating that the phenyl group and CF3 of phenyl group attached to the N of imidazole play critical roles for their activity. In addition, the results of studies using model mice xenografted with tumor cells show that Az markedly reduces the tumor size and increases the number of apoptotic cells, indicating that this substance effectively inhibits tumor growth in tumor-bearing mice. Interestingly, treatment of cancer cells and tumor-xenografted mice with a combination of Az and doxorubicin enhances apoptosis in comparison with a single treatment with either doxorubicin or Az. These findings suggest that Az has the potential of being used alone or in combination with other substances as an anticancer agent.
