연구의 목적 및 내용
- 연구목적 : Selenium, EGCG, 머루 anthocyanin의 조절능력을 상실한 세포자연사멸 과정이나 암세포 전이 상태를 원상복귀과정에서 핵심적인 주요 신호전달체계 확립과 이 과정 중AMPK 및 새로 발굴한 목표물질들과의 상호 연관적인 기전을 규명하고자 함
- 연구내용 : Selenium, EGCG, anthocyanin을 암세포에 처리하였을 때 in vitro 및 in vivo에서 암세포의 증식억제 효과 및 apptosis 유도 확인

연구의 목적 및 내용
- 연구목적 : Selenium, EGCG, 머루 anthocyanin의 조절능력을 상실한 세포자연사멸 과정이나 암세포 전이 상태를 원상복귀과정에서 핵심적인 주요 신호전달체계 확립과 이 과정 중AMPK 및 새로 발굴한 목표물질들과의 상호 연관적인 기전을 규명하고자 함
- 연구내용 : Selenium, EGCG, anthocyanin을 암세포에 처리하였을 때 in vitro 및 in vivo에서 암세포의 증식억제 효과 및 apptosis 유도 확인
Selenium, EGCG, anthocyanin이 조절하는 AMPK의 활성을 통한 Akt/mTOR 신호경로 및 Wnt 신호경로의 조절 관계 확립
연구결과
1) 세포자연사멸 및 전이조절능력 복구 현상 확인
- 암세포에서 과다하게 활성화되어 있는 Wnt, PI3K/Akt pathway가 암세포의 비정상적 증식에 관여하는지 알아보기 위하여 IGF-1을 처리하여 세포성장조절 효과를 확인하였음
- Selenium, EGCG 및 anthocyanin의 세포증식억제효과 및 apoptosis 유도효과를 확인하고, 이들 효과가 Wnt, PI3K/Akt pathway의 표적분자들을 조절함에 따라 일어나는 것임을 확인하였음
2) 세포 자연사멸 및 전이조절 능력 복구기전 확립
- Selenium이 HT-29 대장암세포를 이용한 xenograft model에서 종양의 성장을 억제하고 AMPK-Akt- mTOR 신호경로를 조절함으로서 일어남을 확인하였음
- Hep3B 간암세포에서 selenium 및 anthocyanin이 조절하는 AMPK/Wnt 신호경로의 조절에 있어서 AMPK가 GSK3β의 조절자로서 작용함을 확인하였음
- 혈관신생에 관여하는 인자인 VASP와 VEGF 및 전이인자인 MMPs의 발현을 확인하였으며, EGCG와 anthocyanin의 전이억제를 확인해 보기 위하여 wound healing assay를 실시하여 전이억제 효과를 확인하였음
3) in vivo에서 자연사멸 및 전이조절 능력 복구 기전의 역할 탐색
- in vivo xenograft model에서 Akt, mTOR 및 AMPK 신호분자들의 tumor 성장 억제효과 확인하였음
- EGCG의 new target인 CSK와 AMPK/COX-2 신호경로의 조절기전을 확립하였음
- Hep3B 간암세포주를 이용한 in vivo xenograft model에서 Wnt/AMPK 신호경로의 조절기전을 확인하였음
- DEN과 2-AAF를 이용한 mouse 간암유도모델 제작 및 selenium의 초기발암억제 효과 확인하였음

- Purpose : The mechanism of selenium, EGCG, and anthocyanin in their protective effect of malignant transformation by balancing survival and apoptosis through the regulation of major survival pathways such as PI3-kinase/Akt and Wnt is explored. The possible links between AMPK and newly discovered target protein by these agents, and major survival pathways are investigated.

Relationship of Wnt and Akt/mTOR signaling pathway through AMPK activation which is regulated by selenium, EGCG and anthocyanin
Result
1) Confirmation of apoptosis and angiogenesis-regulation
-Verification of cell growth by treating IGF-1 to confirm that if Wnt, PI3K/Akt pathway which is over-activated in cancer cells is related to abnormal proliferation of cancer cells.
-Verification of that anti-proliferatory effects and apoptosis of Selenium, EGCG and anthocyanin and these effects are consequent upon the regulation of the target molecules of Wnt, PI3K/Akt pathway
2) Establishment of apoptosis and angiogenesis-regulation
-Selenium inhibit the tumor growth in xenograft model with using HT-29, colon cancer and it is consequent on the regulation of signalling pathway of AMPK-Akt-mTOR by selenium.
-In regulation of AMPK/Wnt signalling pathway which is regulated by selenium and anthocyatnin in Hep3B, liver cancer, AMPK acts on GSK3β as a regulator.
-Expressions of VASP, VEGF and MMPs were verified.
3) Research to define the role of apoptotic effects and anti-angiogenic effects in vivo
- Inhibitory effects of tumor growth by regulation of Akt, mTOR and AMPK signal molecule in vivo xenograft model
- Establishment the regulatory mechanism of CSK, new target of EGCG, with AMPK and COX-2
- Verification of the regulatory mechanism of Wnt/AMPK signaling pathway with using Hep3B hepato-carcinoma xenograft model
- Production of liver cancer mouse model with using DEN and 2-AAF and verification of early anti-carcinogenic effects by selenium
Expected Contribution
This study can prove very valuable insights into the molecular basis of the chemopreventive agents such as selenium, EGCG and anthocyanins with the emphasis on their ability to control intracellular signaling cascades of AMPK, mTOR, Akt, Wnt the restoration fo dyregulated cancer cell survival, apoptosis and metastasis. Discovering new targets and resolution of complex interrelationships among important signals can provide the detailed mechanism of selenium, EGCG and anthocyanin for their potentails as anti-tumor agents. Further, this work is to contribute to the economic value or efficacy of resources of selenium-containing constituents, green tea or wild berry. The advances in this research can further strengthen the ultimate application of chemopreventive agents to control
cancer and for the training of anti-cancer expertises. It also can provide the sound basis for utilization of chemopreventive agents to maximize industrial outcome.
