Hep G2 세포를 이용하여 염증유발과 관련 있는 유전자들의 발현 양상을 조사한 결과 전통주 추출액과 10 ㎍/㎖의 LPS를 혼합처리 했을 경우, LPS를 단독처리한 군에 비해 추출액 농도가 증가할수록 이 유전자들의 발현이 감소하며 특히 10 ㎎/㎖의 시료를 처리하였을 때 이들 유전자 발현의 억제가 현저하였다. 따라서 기능성 전통주가 LPS에 의해 유도된 염증을 완화 시켜줄것이라고 판단된다.
4. 실험동물모델을 통한 기능성 전통주에 대한 효능 검정
상황버섯발효 전통주 알코올 농도 8% 5 ㎖을 흰쥐에 1일 2회 나누어 14일간 경구 투여하였다. 대조군으로 동일한 농도의 ethanol을 2주간 투여 하여 간의 염증관련 유전자 및 단백질의 발현에 대한 결과, 정상군에 비해 ethanol투여에 의한 발현의 변화는 관찰되지 않으나 전통주투여군의 COX-2, iNOS, TNF-α의 유전자 및 단백질 발현이 감소하였다. 상황버섯 발효 전통주가 간 염증에 효과적일 것이라고 판단된다.

1). Analysis of biomass produced from alcohol fermentation
230 ton of materials such as Koji, rice, mycelia, and water produces 150 kg of biomass, which is 15% of total input, after alcohol fermentation.
Biomass contains 75% starch and mycelia which can be reusable. Since amount of mycelia is 15% among them it is conceivable that biomass can be reusable to make functional traditional wine.
2). Optimization of biomass with pretreatment for alcohol fermentation
It was investigated whether mycelia in the biomass contain ability to fermentation under the condition of 7 days incubation at 25C. Alcohol amount was 8.0%, and acidity was 17.8%.
Therefore, it was confirmed that mycelia in the biomass was alcohol-fermentable. However, fermentation process was slower possibly by the biomass without glucose hydrolysis. Glucose or steamed rice was additionally added.
Alcohol fermentation after addition of glucose or steamed rice for 6 days at 25℃ produced 8.8% and 9.1% of final alcohol concentration, respectively, which is approximately 1% greater than biomass only. In contrast, pH from glucose- or steamed rice-added fermentation product was decreased to 6.0 and 6.1, respectively.
The results indicated that it is possible for alcohol fermentation with the biomass and mycelia in the biomass has weak fermenting ability and starch hydrolysis and is not enough for alcohol fermentation.
3) Optimization for mycelia-fermented alcohol beverage in small scale.
It was necessary to add Koji as starch hydrolysis and fresh-cultured mycelia other than mycelia in the biomass from the previous fermentation. With them alcohol fermentation for 6 days at 25℃ produced 12.4% of final alcohol concentration, pH4.0, suggesting that production condition is established.
4). Making commercial product of functional alcoholic beverage
Scale-up was carried out based on the condition for small scale. Briefly, rice was absorbed in water, drained, and steamed. Aspergillus kawachii was innoculated and incubated for one day at 35 ~ 40℃ to produce Koji. The first fermentation of alcohol was done with Koji (1,700kg), newly cultured mycelia (65kg), and water (2,600kg). After 24 hr, rice (1,240kg) and water (3,000ℓ) were added. The second addition of the same amounts of rice and water was done at 24 hr post-fermentation. Pressure filtration was necessary to make the final product, 8% alcohol beverage.
2. Analysis of Nutritional Compounds and the physiological activity in Alcohol Product
1). Analysis of Nutritional Compounds
Amounts of organic acids and amino acids are influent to taste and flavor of alcohol beverage. Therefore, amounts of organic acids and amino acids of mycelia-fermented alcohol beverage were analyzed using HPLC. we identified 5 different organic acids (oxalic acid, malonic acid, lactic acid, acetic acid, citric acid). Among these amounts of lactic acid and citric acid were relatively greater than others.
Also, vitamin B1 was detected as a concentration of 3ug/ml of the three-days old product. Storage temperature and period caused change of each compounds. Amounts of lactic acid and citric acid increased to 1.9 fold and 3.7 fold, respectively, when stored at 25℃. Other organic acids showed a similar pattern. Increased amounts of organic acid result in change of taste and flavor of alcohol beverage. Amounts of most amino acids increased when stored at 25℃. In contrast, aspartic aicd and arginine were detected only when stored at 4℃.
2). Analysis of functional activity at cellular level
Alcohol metabolism occurs in the liver tissue. Therefore, a cell line from liver tissue was used for analysis. The expression of inflammation-related proteins in HepG2 cells such as iNOS, COX-2, and TNF-α was examined. The results showed that the treatment of extract of traditional wine inhibited the expression of these proteins even in the presence of LPS.
3). Analysis of the physiological activity with animal model
8 week old male Sprague-Dawley rats were purchased from Samtaco company and fed orally with the mycelia-fermented alcohol beverage for 14 days with twice of 5 ml everyday.
Control group were fed the same protocol except alcohol instead of the mycelia-fermented alcohol beverage. Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in blood were analyzed with automatic analysis system (COBAS MIRA, Roche, Germany).
Normal group AST without any feeding was 107.8±3.9 IU/L, while control group was 129.8±9.1 IU/L (P<0.05). Experimental group AST was 115.8±5.8 IU/L, which was regarded as nonsignificant. Normal group ALT was 38.4±1.1 IU/L, while control group was 42.5±2.0(IU/L). Experimental group ALT was 39.8±2.7 IU/L and showed statistically nonsignificant. The expression of inflammatory proteins such as cyclooxygenase (COX)-2, inducible nitric oxide synthase (iNOS), tumor necrosis factor (TNF)-α were decreased in the mycelia-fermented product-treated rat compared with ethanol-treated ones.
