I. 연구개발의 목적 및 필요성 1) 루푸스 발병이 T 세포의 peripheral tolerance 기전의 결함에 미치는 영향에 대해서 분석된 바가 없음 2) 자연발생 루푸스 마우스에서 자가항체를 생성하는 종자중심 (germianl center, GC)의 특징을 형태학적 및 발생빈도 측면에서 분석 3) 자가항체 형성 GC의 형성이 T 세포 (Tfh, Treg)의 분포와 기능적인 차이에서 비롯된 것인지에 대한 연관성을 규명 Ⅱ. 연구개발의 내용 및 범위 1) Lupus-prone 마우스를 이용하여 자발적으로 형성된 germinal centers (GCs)의 형태학적 및 기능적인 특성 규명 - B6와 B6.Sle 마우스간의 GCs의 phenotype 분석 - B6와 B6.Sle 마우스간의 자가항체 형성분석 - GC B cells과 Post GC B cells의 특성 분석 2) Pathogenic GCs를 유도하는데 있어서 GC CD4+ T cells 역할을 규명 - B6와 B6.Sle 마우스간의 GC/follicular helper CD4+ T cells의 형태 분석 - B6와 B6.Sle 마우스간의 GC regulatory CD4+/CD8+ T cells의 형태 분석 Ⅲ. 연구개발결과 1) 자가항체를 생성하는 마우스와 생성하지 않는 마우스를 비교하여 자발적으로 형성된 germinal center와 자가항체 생성과의 상관관계를 분석하고, 루푸스 감수성 마우스에서 germinal center (GC)의 수가 증가하는 것을 밝힘 2) 루푸가 감수성 유전자위가 GC B cell 및 post GC B cells의 과도한 분화를 유도함을 밝힘 3) B cells의 분화를 조절하는 GC내에 존재하는 종자중심 CD4+ T 세포인 follicular helper T (Tfh), Foxp3+ CD4+ Treg, 및 GC CD8+ Treg의 빈도 및 기능에 미치는 분자의 발현 차이를 루푸수 감수성 마우스에서 밝힘 4) 즉, 루푸스 감수성 마우스 GC내의 CD8+ Treg 세포가 증가한 반면, CD4+Treg는 감소하였고, GC형성을 조절할 것으로 기대되는 CXCR5- splenic Treg의 수가 늘어났지만, 이의 기능과 중요하게 관련된 CD62L+ regulatory T 세포의 수가, GITR의 발현이 증가하여 GC에서 peripheral tolerance 기전이 결함되어 루푸스가 발병하는 것으로 사료됨 5) 특히 GITR+ Foxp3+CD4+ Treg은 suppressive function에 이상이 있는 것으로 보고됨 Ⅳ. 연구개발결과의 활용계획 1) Foxp3 regulatory T cell의 기능 분석을 위해서 Foxp3-GFP 마우스와 backcross를 완료하고, 2) GC regulatory CD4+ T 세포와 CXCR5- Regulatory CD4+ T 세포의 기능과 관련된 marker의 이상과 실제 기능의 이상이 연관성이 있는지 검증하고자 함 3) GC C8+ Treg이 기능을 갖는지에 대해서 B6와 B6.Sle 마우간에 비교 4) 이로부터 루푸스에서 regulatory T cell이 peripheral tolerance에 미치는 영향을 밝히게 되면, 내성기전의 조절을 가능하게 하는 표적치료제의 개발에 근거를 제공할 것으로 기대함

I. Objective It has not been known which cellular factor(s) may affect the pathogenesis of lupus erythematosus by breaking a peripheral tolerance mechanism(s). Therefore, we firstly analyzed the frequency and size of germinal center in the B follicle of mice, which have germinal center naturally, and control mice. Secondly, the cellular characteristics and the frequency of regulatory T cells including foxp3+ CD4+ Tregs, germinal center CD8+ Tregs were analyzed by flow cytometry and confocal microscopy. In addition, we measured the frequency of B cells including plasma cells, plasmablasts, centrocytes, and centroblasts, germinal center CD4+ T cells as well as CXCR5-splenic CD4+ T cells. Ⅱ. Scope of research 1) Phenotypical characterization of spontaneously developed germinal center in lupus-prone mice and B6 mice in terms of numbers and cellularity in the germinal center •Analysis of germinal center between B6 and B6.Sle mice • Measurement of anti-dsDNA antibodies by pathogenic germinal center in lupus-prone mice • Analysis of pre-GC B cells and Post-GC B cells between B6 and B6.Sle mice 2) Analysis of the role of T cells in pathogenic germinal center by using cellular markers related to function • Analysis of germinal center/follicular helper CD4+ T cells between B6 and B6.Sle mice in terms of the frequency and the markers related to function • Analysis of regulatory T cells including CD4+ and CD8+ Treg cells between B6 and B6.Sle mice in terms of the frequency and the markers related to function • SLAM, activation marker, functional marker for regulatory T cells we re used to characterize the T cells Ⅲ. Results 1) Increased number of germinal center in lupus-prone B6.Sle1 congenic mice compared to B6 mice and non-lupus prone B6.Sle3 congenic mice, resulting in correlation between increased GCs and titers of anti-ds DNA Abs. 2) Increased number of centroblasts in B6.Sle1 mice compared to B6 and B6.Sle3 mice, but decreased centrocytes in B6.Sle1 mice 3) Increased number of foxp3+ CD4+ Treg with high levels of GITR in CXCR5- CD4+ T cells of B6.Sle1 mice, which is defective marker for Treg, suggesting that these cells may defect in suppressive functions. 4) Moreover, the expression levels of CD62L were significantly decreased in regulatory T cells in both germinal center and splenic T zone 5) The levels of OX40 and CTLA-4 expression were comparable between B6 and B6.Sle congenic mice. 6) Increased germinal center CD8+ Tregs and decreased in foxp3+ CD4+ Treg in B6.Sle1 mice compared to B6 and B6.Sle3 mice Ⅳ. Future directions Our study will be further investigated as follows, 1) Completely change the background of Foxp3-GFP mice with B6.Sle1 2) To address the question that Treg cells are defect in lupus-prone, we are going to analyze the suppressive function using effector CD4+ T cells and B cells 3) Differentiate the function of GC regulatory T cells from CXCR5- splenic regulatory CD4+ T cells This study is innovative and novel since little is known about such abnormalities in regulatory T cell that is critically involved in pathogenic germinal center formation. Clinically, this study will support a potential application of regulatory T cells in suppressing the autoimmune response in developed autoimmune disease.
