연구의 목적 및 내용
우리나라를 포함하여 전 세계적으로 비만인구와 암으로 인한 사망률은 급격히 증가하고 있음. 본 연구의 목적은 비만에 의한 암전이 촉진 기전을 종양미소환경내의 변화를 중심으로 규명하고, 이를 효과적으로 저해하는 기능성 식품성분의 효능을 평가하는 것임. 본 연구의 세부 목표는 다음과 같음. (1) 다양한 mouse tumor model을 확립하여 고지방식이가 고형암의 성장과 전이, 종양조직내의 신생혈관생성과 신생림프관생성에 미치는 영향 규명. (2) M2-대식세포가 암의 성장과 전이에 미치는 영향 규명. (3)

연구결과
다양한 mouse tumor model과 in vitro co-culture model를 확립하여 얻은 결과들의 요약.
(1) 고지방식이는 종양조직에서 HIF-1α, VEGF-A, VEGF-C, VEGF-D의 발현 증가와 다양한 cytokines/chemokines의 분비 증가를 통해 신생혈관생성과 신생림프관생성을 증가시키고, 그로인해 고형암의 성장과 암전이가 증가되어 마우스의 생존률이 감소함.
(2) 고지방식이에 의해 증가된 종양조직내의 M2-대식세포와 종양세포의 상호작용은 다양한 cytokines (MCP-1, M-CSF 등)의 분비 증가를 통해 종양조직으로 대식세포의 유입을 증가시킴. 또한 암세포와 M2-대식세포의 상호작용은 신생혈관생성과 신생림프관생성 증가를 통해 고형암의 증식과 암전이를 촉진함.
(3) In vitro co-culture mode을 이용한 실험에서도 암세포와 M2-대식세포는 서로에게 영향을 주어 MCP-1과 M-CSF를 비롯한 cytokines의 발현을 변화시킴. 또한 고지방식이를 공급한 마우스의 지방조직 배양액에는 다앙햔 cytokines의 수준이 증가되어 암세포의 이동과 신생혈관생성을 증가시키며, 이는 M2-대식세포 혹은 암세포와의 co-culture에 의해 촉진됨.
(4) Short hairpin RNA를 이용하여 암세포에서의 MCP-1과 M-CSF 발현을 억제시키면 종양조직에서 대식세포의 유입과 분화가 억제되고, VEGF-A, VEGF-C의 발현 감소를 통해 신생혈관생성과 신생림프관생성이 억제되어 고형암의 성장과 암전이가 감소됨.
(5) In vitro 실험을 통해 항염증, 항비만, 항전이 활성을 나타내는 식품유래 성분을 탐색하고, 선별된 BITC와 piceatannol은 mouse tumor model에서 다양한 cytokines/chemokines, 신생혈관생성과 신생림프관생성을 감소시킴으로써 항종양 및 항전이 활성을 나타냄.
본 연구의 결과는 고지방 섭취와 기능성 식품성분에 의해 조절되는 고형암의 성장과 암전이 기전을 규명하는데 크게 기여하였음.
연구결과의 활용계획
(1) 본 연구의 결과를 암예방 및 치료 연구를 위한 기초 자료로 이용함.
(2) 본 연구에서 확립한 in vitro co-culture model과 암세포와 M2-대식세포의 in vivo co-injection model은 종양미소환경을 구성하는 세포들의 상호작용 규명을 위한 이상적인 model 로서, 앞으로 미소환경에 대한 연구에서 뿐만 아니라 종양미소환경을 타겟으로 하는 암예방 기능성 성분의 개발을 위한 연구에 활용함.
(3) 본 연구에서 탐색된 다양한 표적분자 (MCP-1, M-CSF, CXCL13 등)들은 종양미소환경에서의 cytokines/chemokines network 규명에 도움을 주며, 이를 타겟으로 하는 후속연구에 기초로 활용함.
(4) 항종양/항전이 효능이 관찰된 기능성 식품성분 (BITC, piceatannol)은 기능성 식품 개발에 선도물질로 활용함.

In order to examine the mechanisms by which a high-fat diet and/or obesity stimulates tumor growth and progression, we tested the following hypothesis. Macrophages expedite tumor growth and metastasis. Adipose tissue, primary tumor, and metastasized organs recruit macrophage population into their midst, and subsequent interactions among the adipocytes, macrophages, cancer cells, and host cells induce changes in cytokine/chemokine profiles leading to the stimulation of inflammation-related cancer growth and metastasis. The ultimate goal of the proposed study was to search various bioactive food components with the ability to inhibit this high-fat diet-stimulated tumor growth and metastasis and to elucidate the underlying mechanisms.
Result
To achieve this goal we used several mouse tumor models and in vitro co-culture models.
Our results demonstrated that:
(1) High-fat diet (HFD) increases tumor angiogenesis and tumor lymphangiogenesis through increased expression of HIF-1α, VEGF-A, VEGF-C, VEGF-D, and increased secretion of various cytokines/chemokines, thereby increasing the solid tumor growth and metastasis, and decreasing the survival rate in mouse tumor models.
(2) HFD increases M2-macrophages in tumor tissues, and the crosstalk between M2-macrophages and tumor cells stimulates macrophage infiltration into tumor tissues through the release of various cytokines (MCP-1, M-CSF etc.). Moreover, the crosstalk between M2-macrophages and tumor cells increases tumor angiogenesis and tumor lymphangiogenesis, thereby increasing the solid tumor growth and metastasis.
(3) HFD increases the levels of various cytokines in adipose tissues, and adipose tissue-conditioned medium (ATCM) stimulates proliferation and migration of cancer cells and increases angiogenesis. Furthermore ATCM-induced cancer cell migration and angiogenesis were stimulated by co-culture with M2-macrophages or cancer cells.
(4) Knockdown of MCP-1 and M-CSF by shRNA decrease macrophage infiltration and macrophage differentiation into the M2 phenotype, which in turn, inhibit the solid tumor growth and metastasis.
(5) We selected bioactive food components with the ability to inhibits inflammation, metastasis, and adipogenesis. Among these, BITC and piceatannol suppressed tumor angiogenesis and tumor lymphangiogenesis in mouse tumor models.
Expected Contribution
(1) The information regarding a role of target molecules will help develop the cancer-preventive agents.
(2) Established mouse tumor models and co-culture systems will be used in cancer prevention research, and the information regarding the crosstalk between cancer cells and macrophages will help develop the cancer-preventive agents that target the tumor microenvironment.
(3) The selected target molecules (MCP-1, M-CSF, CXCL13 etc.) will help to explain the cytokines/chemokines network, and can contribute to the development of cancer-preventive agents.
(4) The selected bioactive food components (BITC and piceatannol) can be developed as cancer preventive agents.
