1.면역보조제의 독성연구
바이러스성출혈성패혈증 (VHSV)은 국내 넙치 (Paralichthysolivaceus)양식산업에서 경제적으로 큰 손실을 입히고 있는 주요 병원체로 이 질병에 대한 장기적인 대책 마련이 절실히 요구 되고 있다.백신 내 면역보조제는 숙주의 면역 반응을 증진시키는 물질로서 숙주로 하여금 체내로 침입한 병원체를 효과적으로 제거할 수 있게 유도한다.그러나 면역보조제의 어류 체내 부작용과 적절한 사용 농도가 명확하게 규명되어 있지 않다.따라서 본 연구에서는 8.47±0.42 cm의 넙치에,4

1.면역보조제의 독성연구
바이러스성출혈성패혈증 (VHSV)은 국내 넙치 (Paralichthysolivaceus)양식산업에서 경제적으로 큰 손실을 입히고 있는 주요 병원체로 이 질병에 대한 장기적인 대책 마련이 절실히 요구 되고 있다.백신 내 면역보조제는 숙주의 면역 반응을 증진시키는 물질로서 숙주로 하여금 체내로 침입한 병원체를 효과적으로 제거할 수 있게 유도한다.그러나 면역보조제의 어류 체내 부작용과 적절한 사용 농도가 명확하게 규명되어 있지 않다.따라서 본 연구에서는 8.47±0.42 cm의 넙치에,4 종류의 면역보조제 (saponin,aluminum hydroxide,squalene emulsion,FIA)를 복강 내 접종하여 다양한 농도로 독성 실험을 진행하였고,간의 이상 및 조직학적 변화 유무를 관찰하였다.Saponin에 의한 LD50는 105μgfish-1 이었고 조직학적 손상이 일어났으며 16μg fish-1이상에서 AST 수치 증가 160μg fish-1(11U/L)에서 ALT수치 증가 등의 부작용이 나타났고,5μgfish-1의 농도에서는 어떠한 부작용도 관찰되지 않았다.Aluminium hydroxide는 5000㎍ fish-1의 농도에서 독성을 나타냈고,1600㎍fish-1이하의 농도에서는 조직학적 손상 및 AST/ALT 수치에 변화는 없었다.한편,10~20%의 Squaleneemulsion에서는 염증반응의 증가가 보여졌고,5%의 농도에서는 가벼운 염증반응이 유도되었으며,AST/ALT 수치는 대조구와 유사한 양상을 나타냈다.FIA는 어류에 치명적인 손상을 가하진 않으나,주사기로 접종한 부위와 혈관 주변에서 염증이 발생하였다.FIA보다 낮은 독성을 보이는 농도는,5㎍ fish-1의 saponin,1600 ㎍ fish-1 이하의 aluminium hydroxide,5%의 squalene으로 이들 농도를 기준으로 VHSV 백신 제조에 사용하였다.
2.VHSV백신 효능증강 면역보조제 선별
면역보조제가 넙치 (18.5±1.2g)에서 VHSV에 대한 면역 반응 증진을 유도하는지 알아보고자 하였고,각각의 면역보조제는 다양한 형태로 조합하여 백신을 제조하였다.그 결과,불활화된 VHSV 만으로는 면역 후 3주 째의 VHSV 감염에서는 약한 면역형성을 보였으나 (RPS=37%),면역 후 6주 째에는 면역형성을 유도할 수는 없었다.Saponin또한 VHSV 감염방어에 효율적이지 못했다.그러나,squalene과 aluminum hydroxide를 혼합하여 만든 백신은 VHSV 감염에 매우 강한 면역반응을 (RPS=76%)보였다.
3.최적 스쿠알렌농도와 aluminum hydroxide의 영향
VHSV백신에 가장 유효하게 사용할 수 있는 squalene농도를 알아보고자 하였다.Squalene 은 다양한 농도 (1%,5%,10%)에서 VHSV에 대한 면역증강 반응을 유도하였다.RPS 는 5% Sq+IV,5% Sq+AI+IV,10% Sq+IV,10% Sq+AI+IV 에서 각각 31%,58%,46%,33% 로나타났다.한편 백신 접종 후 10주가 경과한 시점에서,면역보조제의 농도에 따라 폐사가 결정지어졌고,squalene과 aluminum hydroxide에 의한 면역 상승 효과 또한 확인할 수 있었으며 각 RPS는 1% Sq+IV,1% Sq+Al+IV,5% Sq+IV,5% Sq+Al+IV,10% Sq+IV,10%Sq+Al+IV에서 각각 53%,71%,65%,83%,77%,91%로 나타났다.면역 10주째의 공격시험에서는 squalene의 농도가 높을수록,같은 농도의 squalene에서는 aluminium hydroxide가 첨가된 것일수록 높은 효능을 보였다.한편 ,FIA는 백신 접종 후 4주가 경과한 시점에는 면역형성을 유도할 수 없었으나,10주 후 71%의 RPS를 나타내면서 비교적 VHSV에 대한 면역형성이 늦어지는 것으로 보여졌다.
본 연구의 결과로부터 squalene을 5%나 10% 사용한 것이 모두 비슷한 정도로 효능이 좋으며,squalene에 aluminum hydroxide(0.5%)를 첨가하는 것이 시너지효과가 있음을 알 수 있었다.이후의 실험에서는 넙치에 독성이 약하도록 5%의 squalene을 사용하였으며 aluminum hydroxide0.5%를 첨가하여 백신의 지속기간을 1년 이상 확인하였다.
4.최적의 면역보조제 첨가 VHSV백신의 방어력 지속기간
최적의 면역보조제인 Squaleneemulsion (5%)과 aluminum hydroxide(0.5%)가 포함된 불활화 VHSV 백신을 약 15.2±0.4 g의 넙치에 접종하고 일정시간 경과 후 15℃로 옮겨 ,107.1TCID50/fish의 바이러스로 공격감염하였다.그 결과 RPS가 60% (3주),64% (6주),71%(12주),55% (18주),52% (24주),50% (40주),45% (57주)로 나타나 면역보조제가 포함된 VHSV백신은 넙치를 VHSfh보터 방어할 수 있음을 알 수 있었다.또한 백신 처리 후 18주가 경과한 실험어들은 6°C의 저수온 조건에서도 56%의 RPS를 나타냈다.이들 실험어의 항체가(OD;1.08± 0.13)는 면역 후 3주에 확인되었고,이는 점진적으로 감소하여 24주가 경과한 시점에서는 검출되지 않았다.면역보조제가 혼합된 백신에 의한 눈에 띄는 부작용은 없었으며,6주와 12주에 비장과 장에서 약간의 염증성 반응이 유도되었고,이는 24주 후에 정상적으로 회복되었다.
5.최적의 면역보조제 첨가 VHSV백신의 작용기전
백신에 의해 면역이 형성된 어류는 백신 처리 후 8주까지 VHSV (107.1TCID50/fish)감염에 효율적으로 반응하였고,백신 처리된 그룹은 60%의 생존율,대조구 그룹은 25%의 생존율을 나타냈다.백신 처리 된 그룹에 바이러스 감염 후 3시간,6시간,12시간,1일,2일,4일,7일에 걸쳐서 시료를 채취하여,바이러스 증식 수치와 면역 유전자 발현을 비교 분석하였다.백신처리하지 않은 그룹은 감염 후 4일이 경과한 시점에서 백신 처리구에 비해 유의적으로 높은 바이러스 증식을 확인할 수 있었으며 백신을 접종한 구에서 항원제시와 세포자멸사에 관여하는 인자인 MHC classII,CD8perforin과 caspase3가 높은 발현을,백신을 접종하지 않은 구에서는 염증반응과 인터페론의 생성에 관련하는 유전자인 interleukin 1 and 6,Mx, ISG15,Type1interferon의 발현이 높았다.이는 바이러스감염 초기의 항원제시와 세포자멸사가 바이러스의 증식을 효율적으로 막아 백신이 방어력을 제공할 수 있도록 도우며,염증반응과 인터페론의 유도는 바이러스의 양이 증가함에 따라 유도되므로 백신을 접종하지 않은 구에서 높게 발현되는 것으로 생각되었다.
6.개발된 면역보조제첨가 백신의 현장적용시험
완도 1개소,제주 2개소의 넙치양식장에서 현장적용실험을 실시했으며 자연적인 발병이 없어 완도와 제주 1개소의 백신접종 넙치를 VHSV로 공격감염을 실시하였다.완도의 넙치에서 방어력을 확인할 수 있었으며 제주실험어는 에드워드병이 원인으로 폐사가 발생하여 VHSV효능을 검증하기는 어려웠다.

Viral hemorrhagic septicemia (VHS) in olive flounder (Paralichthys olivaceus) remains an unsolved health problem in Korean aquaculture. Vaccination plays a significant role in modern aquaculture, and the duration of protection provided is of vital importance. Adjuvants play a major role in enhancing the immune response to a given antigen. Most of the adjuvants are toxic at certain doses, and toxicity varies in different species. Moreover, there are no standard dosage limits set for adjuvant use in fish vaccines. To know safe ranges of several adjuvant, acute mortality, serum enzymes (AST/ALT) indicating hepatic injury and histopathological changes due to intra-peritoneal administration of different concentrations of a panel of adjuvants were conducted. Adjuvant used were quillaja saponin, aluminum hydroxide, squalene emulsion and Freund’s incomplete adjuvant (FIA) with a dose ranging study of saponin (500, 160, 50, 16 and 5 μg fish-1), aluminium hydroxide (5000, 1600, 500,160 and 50 μg fish-1), squalene emulsion (20, 10 and 5%), and FIA to determine the acceptable dosage for vaccination in olive flounder fingerlings measuring 4.66 ± 0.41 g, 8.47±0.42 cm. Saponin was highly toxic with a LD50 of approximately 105 μg fish-1 (22.4 mg kg-1) causing severe histological damage and AST level was high at dose above 16 μg fish-1 and ALT, specific for liver damage was high only at 160 μg fish-1 (11 U/L) and was safe at 5 μg fish-1. Aluminium hydroxide was toxic at 5000 μg fish-1 and was acceptable at dose below 1600 μg fish-1 with moderate histology and AST/ALT levels similar with control. Squalene emulsion showed increased inflammation at 20% and 10% emulsions and the inflammatory response was mild at a concentration of 5% oil emulsion and AST/ALT levels being similar to control in 10 and 5% emulsions and elevated in 20% on both sampling days. FIA was not lethal, but induced severe inflammation at injection site and around blood vessels. In comparison to FIA, saponin found to be safe at dose of 5 μg fish-1, aluminium hydroxide below 1600 μg fish-1, and squalene at 5% emulsion and could be accepted for vaccination studies. These results provide an insight for the selection of safer dose of adjuvants for intra-peritoneal vaccination of olive flounder.
The selected adjuvants at their acceptable levels were evaluated with viral haemorrhagic septecemia virus (VHSV) antigen to enhance the immune response of olive flounder (18.5±1.2g).
The vaccine was formulated with different combination of adjuvants. Vaccine group comprised of formalin inactivated vaccine (IV), squalene (5%) emulsion with inactivated vaccine (IV+Sq), squalene (5%) emulsion with aluminum hydroxide and inactivated vaccine (IV+Sq+Al) and saponin (50μg/fish) with inactivated vaccine (IV+Sp). We found that the inactivated vaccine alone provided moderate protection at 3 weeks post vaccination (RPS=37%) and it could not protect fish at 6 weeks post vaccination to virus challenge. However, the magnitude of protection provided by squalene and aluminum hydroxide adjuvanted vaccine was much high (RPS=76%) indicating the need for adjuvants to enhance immune response of olive flounder to VHSV antigen. Saponin adjuvant failed to enhance any response.
After screening steps, squalene containing aluminum hydroxide adjuvanted vaccine was selected. Next, concentration of squalene required to enhance the immune response of olive flounder (15.2±0.4 g) was evaluated by immunizing with different formulation of vaccines containing different percentages of squalene (1%, 5% and 10%). The response was moderate at 4 weeks post vaccination in 5% and 10% groups with RPS of 31, 58, 46 and 33% in squalene (5%) emulsion and inactivated vaccine (5Sq+IV), squalene (5%) emulsion with aluminum hydroxide and inactivated vaccine (5Sq+Al+IV), squalene (10%) emulsion and inactivated vaccine (10Sq+IV) and squalene (10%) emulsion with aluminum hydroxide and inactivated vaccine (10Sq+Al+IV) respectively. At 10 weeks post vaccination, a concentration dependent response was observed and also the synergistic effect of squalene and aluminum hydroxide was evident.
The RPS obtained was 53, 71, 65, 83, 77 and 91% in 1Sq+IV, 1Sq+Al+IV, 5Sq+IV, 5Sq+Al+IV, 10Sq+IV and 10Sq+Al+IV respectively. FIA provided protection only at 10 weeks post vaccination with RPS of 71% but not at 4 weeks post vaccination, probably due to the depot formation and slow release of antigen. Adhesion of internal organs and peritonitis in fish vaccinated with FIA as adjuvant at 4 weeks post vaccination, where the other groups were not having any side effects. Furthermore, the group administered with squalene and aluminum hydroxide adjuvant alone without antigen failed to provide protection against VHSV challenge indicating the protection was antigen specific. With these results, it was confirmed that squalene at 5 % and 10% can enhance the immune response synergistically with aluminum hydroxide against VHSV antigen.
The efficacy, safety, and duration of protection of an inactivated vaccine formulated with a squalene emulsion (5%) and aluminum hydroxide (0.5%) was studied. The vaccinated fish (15.2±0.4 g) showed significant protection at 3, 6, 12, 18, 24, and 40 weeks post vaccination (wpv) (except week 57) than non-vaccinated fish to an intraperitoneal challenge of 107.1 TCID50/fish at 15°C, with relative percent survival (RPS) of 60%, 64%, 71%, 55%, 52% and 50% (45% at 57 week) respectively, covering the duration of natural infection. Fish challenged at 18 wpv at 6℃ showed 56% RPS and protection at a lower temperature. The antibody titer was high at 3 wpv with an OD of 1.08 ± 0.13, but decreased gradually and was undetectable by 24 wpv. The vaccine formulation was safe without injection site reactions, adhesions, or pigmentation observed at 6, 12, 18, or 24 wpv. Inflammatory reactions were observed in the spleen and intestine at 6 and 12 wpv but were normal by 24 wpv.
The vaccinated fish provided a significant protection against VHSV (107.1TCID50/fish) challenge at 8 wpv. The percentage survival observed in vaccinated group was 60% and it was 25% in non-vaccinated group with an RPS of 47%. The challenged fish were sampled at 3h, 6h, 12h, 1d, 2d, 4d and 7d post challenge and the viral copy number showed an increased replication in non-vaccinated group at 4d compared to vaccinated group, which might lead to the disease and mortality. The study of immune gene expression suggests that the genes responsible for specific immune response are not significantly up-regulated. The absence of up-regulation of these molecules might indicate a defective specific immune response in fish and might have a different mechanism to provide protection.
This work reveals the toxicity levels of the selected adjuvants in olive flounder and the potential of a formalin-killed VHSV vaccine formulated with squalene and aluminum hydroxide adjuvant to enhance and prolong the efficiency in olive flounder fingerlings. The vaccine provided good protection against VHSV and offers an effective strategy to control VHS by vaccinating with a single dose before winter. Furthermore, the duration of protection, single dose, and the minimum side-effects will make this formulation a better suited vaccine for field conditions.
