Korean research group for the development of artificial retina for retinal stimulation, silicon retinal tack for epiretinal fixation of the electrode array, and the stimulation chip for animal experiment.
Both subretinal and epiretinal approaches are investigated in our group.
Polyimide liquid crystal polymer was chosen as the base material for the retinal stimulator to meet these requirements. It is cheap, flexible, and well known by its biocompatibility and resistance to acid or solvent. It also recovers its original shape even after rolling or folding. Most of all, it can be used in silicon microfabrication process, which enables bulk production of the polyimide stimulator in various design with high microelectrode density. Gold metal is widely used as an electrode material for neural stimulation but shows weak adhesion to polyimide base. To overcome this problem, thin titanium layer was pre-sputtered between polyimide base and gold layer.
Various shapes of polyimide microelectrode arrays (MEA) are designed to reduce the tissue damages and to take more intimate contact to the retina. To prevent tearing of edge.
microelectrode array is designed to have rounded corners and circular holes for retinal tack.
Manufactured MEAs showed suitable impedance for electrical stimulation, between 800 to 1.5 ㏀.
In vitro and in vivo biocompatibility of a polyimide MEA was tested by co-culture with human retinal pigment epithelial cells (RPE) and in the rabbit eyes, respe ctively before in vivo electrical stimulation experiment. RPE cells showed good affinity to MEA and there seemed no abnormal morphological changes or no piled-up growth. Also there was no histological difference between control and operated rabbit eyes except damaged photoreceptors in subretinally implanted group.
ERG revealed no difference between the transplanted and the healthy eye.
The prototype of the implantable retinal stimulation system was proposed for preclinical and clinical experiments. Current stimulation pattern was adopted as the electrical signal for neural stimulation be ca use the current stimula tion can pro vide mo re sta ble stimulation current lev el to the electrode-retina interface than the voltage stimulation regardless of the impedance change at the interface. Charge-balanced biphasic electrical stimulation was used as an electrical signal pattern.
Pulse amplitude, duration, frequency, and selective activation of the specific electrode can be modified by wireless communication between external unit and implanted circuit.
Using microelectromechanical systems (MEMS) technology, micro-scaled silicon retinal tacks can be easily produced in mass and their design can be easily modified or customized. To evaluate the usability of the silicon retinal tack, various types of tacks are designed.
To understand neuronal circuitry in the retina, extracellular recording in the isolated rabbit retina was performed using 8x8 multi-electrode array (MEA, Multi Channel Systems GmBH, Germany).
Followed by spike sorting, the patterns of interaction between pairs of neurons in the retinal ganglion cells were studied by examining the cross-correlation function, which represented the firing rate of one neuron as a function of time before or after a spike from the other neuron.
