Avian influenza virus (AIV) is an enveloped virus from the Orthomyxoviridae family and can be classified as either low pathogenicity (LP) or high pathogenicity (HP). It has an eight-segmented, single-stranded, negative-sense RNA genome. Because of its continuing evolution, AIV is able to modify its genome by any of the several types of mutations, including antigenic variation, natural selection, and reassortment. Because of this, AIV outbreaks are the cause of serious concern in asian countries. Especially the economic losses incurred in the poultry industry because of H9N2 low-pathogenic avian influenza (LPAI) are extreme, and this infection has been endemic in domestic poultry farms in Korea since 1996.
AIV virus targets the epithelial cells of the upper respiratory tract. The early steps in infection, as well as the pathogenicity of AIV, depend on the cleavage of the haemagglutinin (HA) glycoprotein of AIV. A major route of infection of AIV in chickens is through cells of the respiratory epithelium. Although animal systems (in vivo) are the best systems for studying disease response, we use an in vitro systems (cell lines and organ cultures) that offer the opportunity to study mechanisms of infection and the immediate host responses under highly controlled conditions. Tracheal epithelial cells (TECs) are an important tool for studies of viral respiratory diseases. Primary TECs have been cultured from human, mouse and hamster. It is also necessary to diagnose viral respiratory disease and reveal infection mechanisms in chicken. We isolated tracheal epithelial layers from tracheal of 20-day-old chicks and cultured primary TECs from the isolated layers. Ciliated cells which were a typical morphology of TECs were observed in cultured primary TECs and maintained until cell passage 5 (15 to 20 days). When we analyzed expression patterns of epithelial marker genes (retinoic acid responder, FGF-binding protein, virus activating protease) in TECs compared to immortalized chicken embryonic fibroblast cell line (DF-1), all the marker genes are highly expressed in TECs than in DF-1. After H1N1 virus infection, we found major 20 antiviral genes of differential expression. The relationship between transfected-TEC and H1N1 virus infection was investigated by microarray. We found transfected-TEC had high expression gene related resistance for cell death and immune sensitivity.
As a innate immune protein, the Mx protein is one of the antiviral GTPases, induced by interferon. The Mx system in the chicken has a genetically RNA-virus resistant or sensitive character that provides diversity in response to experimental infections such as with influenza viruses. The innate immune response nonspecifically induces antiviral activities by recognition pathogen-associated molecular patterns. Those are recognized by variable pathogen recognition receptors, and following signal transduction pathway transmits its signal to induce type I IFNs by auto- or paracrine effects. The type I IFNs effect on not only sensitized cell, but also neighbor cells to maintain antiviral states. Finally, IFN-inducing antiviral effectors such as Mx GTPase, PKR, RNase L or OAS are produced and inhibit virus replication.
Host immunity, particularly innate immunity, is important in the pathogenesis of AIV. Since current knowledge on the avian immune response to AIV infection is limited, we used microarray analysis to examine global changes in immune-related gene expression induced by H9N2 virus infection in chickens. We optimized challenge dose and condition of H9N2 and H5N1 viruses in this study. H9N2 virus infection induced strong innate immune responses in brown layer chickens than white layer chicken. The increased transcription of 2′ -5′ -oligoadenylate synthetase, C-type lectin, and myxovirus resistance genes was confirmed by real-time RT-PCR.
In our project, antiviral genes including chicken IFN-α/β/γ, Mx, c-type lectin gene were cloned into mammalian expression vector. Virus replication was inhibited in transiently antiviral gene expressing chicken cells infected with avian influenza. Additionally, we evaluated the resistance of transgenic chicken expressing a 3D8 scFv antibody against H9N2 and H5N1 viruses. Transgenic chickens showed partial resistance against H9N2 infection in this study.
