상기 목적 및 내용에 따라 연구한 결과, 연구 1차년도 세포배양, 기저막침윤억제 실험, 전자현미경연구, 유세포 분석기연구 및 분자생물학적 연구결과 세포살살효과는 흔합파장광선이 apoptosis를 유도하므로서 초래되며, 세포주기의 G2M기의 억제 기전이 있음을 확인했으며, 2차년도 연구결과 252, 312, 갱4 및 760nm 파장이 세포살상효과가 강함을 확인했으며, 혼합광선조사에 의한 피부의 병변은 2(2/33)례에서 stratum corneum의 비후가 관찰되었으나, 비특이 소견이며, 24례에서 stratum corneum이 straum spinosum으로부터 분리되어 있었음. 3차년도 연구비 지원은 평가단으로부터 중단되었으나, 일부 2차년도에 이미 시행된 결과에 의하면, 실험동물의 표재성암 model 생산이 성공하였으며, 현재 이를 이용한 2차원 배양을 실험 중이며, 암세포의 seeding model 인 Ehrlich-Lettre Ascities carcinoma 세포주와 C57BL 생쥐를 이용한 일부실험에서, 실험군에서는 seeding 이 발생하지 않았음을 의미하는 매우 희망적인 결과를 성취하였다. 그러나, 3차년도인 1999년도 연구비의 지원중단을 평가단에서 결정하였으므로 자비로 연구를 지속하고 있으나, 추후 언제 이 연구가 중단될지 언급할 수 없다.

Unexpected appearance of the metastatic cancer, suspicious for caused by the seeding, after cancer surgery which was performed under the accurate staging and proper procedure is critical and agonizing problem for both patient and surgeon. For the reduction of the seeding in cancer surgery, various meticulous technique has been developed and resulted the reduction of them. The pleural effusion and ascites in metastatic terminal cancer is one of the condition troubling patient and doctor. In this research, I studied the effect of the light of mixed wave length which discovered to have cytocidal effect after exposure to treat superficial tumor and reduce the seeding and internal metastasis.
For this study, four kinds of prostate cancer cells, PPC-l and DU-145, and one bladder transtional cell carcinoma T-24 and J-82 were used. The mixed light was exposed to the cells in culture dish at the suspension and attatched state on culture dish. The effectg of the light, invasion study on reconstituted basement membrane using modified Boyden chamber, transmission / scanning electron microscope(TEM/SEM), flow cytometry, and electrophoresis were used for this study.
There was little difference in the result of the study regardless of the cancer cell line.
The growth of the cancer cell was reduced markedly and the further growth was not observed after the exposure of the light. The ligjt exposed cells at the suspension state just after the subculture lost the capability to adhere on the bottom of the culture dish.
The light exposed cells at post subculture 24 hour in attatched state on bottom of the culture dish become rounder and lost their adhesion capability with time passing being floating in culture medium. Those floating cells lost their capability to adhere to the culture dish and no more proliferation was observed. The transmission electron microscopic examination showed condensation of the chromatin, fragmentation of the chromatin, appearance of the vacuoles within cytoplasm, focal breakdown of the nuclear membrane and cytoplasmic membrane blebbing suggesting apoptosis. At the invasion assay, the invasion was markedly reduced compared to control group and at the scanning electron microscopic study, the appearance of the pseudopod for the penetration to the reconstituted basement membrane was not seen. The cell cycle analysis by flow cytometry for cells in apoptotic state showed G2/M phase arrest. The fraction of the cancer cells in apoptosis was increased with the time passage. The electrophoresis of the DNA extracted from the light exposed cells from floating and attatched ones showed typical laddering sign in all cell lines. The electrophoretic finding suggest that the DNA from the floating cells were more fragmented into small molecular weight than attatched cells.
The mice skin after 10 months from the exposure of mixed light showed exfoliation without evidence of the cancerous change in light microscopic and electron microscopic examination. The 252, 312, 434 and 760 nm wave length showed strong cytocidal effects. The animal model with seeding and internal metastasis with EhrIich-Lettre Ascities carcinoma cell showed mixed light exposed makred reduction of them.
With this research, we conclude that the mixed wave length light induce the apoptosis and it has the effects of anti-proliferation and anti-invasiveness which are irreversible. And with the passage of time, the induction of the apoptosis is increased at single exposure. The results of this study show that the mixed wave length light has the potential to prevent cancer cell seeding, internal metastasis and treat superficial cancers.
