○ 연구내용 및 결과(세부과제별) 제1세부과제 C형 간염바이러스의 core와 NS3 유전자, 또 GM-CSF, B7 유전자를 생체 내로 도입시키기 위한 벡터로는 Moloney murine leukinia virus(MoMuLV) 에 기초하여 개발된 MFG 레트로바이러스 벡터가 사용되었다. 이 벡터는 표적세로로의 높은 형질도입 비율, 세포 내에서의 안정적 유지 및 외부 유전자의 지속적 발현 등의 장점을 가지고 있다. MoMuLV의 long terminal repeat (LTR)을 가지고 있어서, 바이러스 particle로 encapsidation 되기 위한 완전한 길이의 subgenomic mRNA를 생성할 수 있다. MFG 벡터에 573bp의 core 유전자 절편과 1767 bp의 NS3 유전자 절편이 삽입되었다. 두 개의 유전자가 함께 발현하게 하기 위하여 murine IRES를 두 유전자 사이에 삽입하였다. 또 granulocyte-macroophage colony stimulating factor (GMCSF) 또는 B7 유전자를 함께 삽입한 벡터도 제조하였다. 제작된 재조합 DNA를 생쥐 간세포주인 BNL CL2에 cationic lipid-mediated gene transfer (lipofectin) 방법에 의해 transfection시켰다. 이렇게 하여 transient expression을 시킨 후 각각의 단백질이 성공적으로 발현되는 것을 확인하였다. Core단백질의 경우 21kDa, NS3 단백질의 경우는 65kDa 의 크기를 가지고 있슴이 단일 항체를 이용한 Western blotting에 의하여 확인되었다. 또 두 개의 유전자를 함께 삽입한 벡터도 모두 각각의 단백질이 합성됨을 알 수 있었다. 제2세부과제 가) Ad/CMV/mB7.1의 제작 생쥐의 mB7.1 유전자가 도입된 MFG.S/B7.puro 플라스미드 (한국과학기술원 최준호 교수 제공)를 XbaI으로 처리하여 얻은 약 1kb의 DNA절편을 pKSII(+)의 Xbal 부위에 도입하여 pKS/mB7.1을 제작하였다. 이 플라스미드를 HindIII/NotI 으로 처리하여 얻은 mB7.1 DNA를 아데노바이러스 transfer 플라스미드의 HindIII/NotI 부위에 도입하여 pX/CMV/mB7.1을 제작하였다. 이 플라스미드를 pJM17플라스미드와 함께 293 세포주에 cotransfection시켜 생성되는 제조합 아데노바이러스를 분리하였다. 이 바이러스의 DNA를 제한 효소로 분석하였을 때 CMV promoter, mB7.1, poly(A)sequence가 아데노바이러스 E1 부위에 도입되었슴을 확인하였다. 나) Ad/CMV/HCV Core1-191의 제작 1차년도에 제조된 아데노바이러스 transfer 플라스미드인 pX/CMV/Core1-191를 pJM17 플라스미드와 함께 293 세포주에 cotransfection 시킨 후 생성되는 바이러스를 라이러스플락검정법을 통하여 순수분리하였다. 이 바이러스의 DNA를 얻은 후 HindIII, XbaI 등의 제한 효소 분석을 통하여 HCV core 유전자가 도입되었음을 확인하였다. 이 바이러스의 타이터를 결정하였고 바이러스로 transduction된 헬라세포에서 Core 단백질의 발현을 검색하였다. 다) Ad/CMV/HCV NS31027-1657 제작 아데노바이러스 transfer 플라스미드 pX/CMV/HCV NS31027-1657를 pJM17 플라스미드와 함께 293 세포주에 cotransfection시켜 상동재조합에 의해 생성되는 바이러스를 얻었다. 이 바이러스의 DNA를 분리한 후 제한 효소 분석을 통하여 아데노바이러스 E1 부위에 CMV promoter, NS3 및 poly (A) sequence가 도입되었음을 확인하였다. 현재 이 바이러스로 감염된 헬라 세포에서 NS3 단백질이 발현되는지를 검색하였다. 제3 세부과제 재조합 FP트로 바이러스 벡터(MFG.Core, MFG.NS3, MFG.Core.NS3, MFG.NS3.Core, MFG.Core.GMCSF, MFG.NS3.GMCSF) 각각을 transfection한 정상간세포주(BNL CL.2) 와 간암세포주 (BNL 1NG A.2) 들을 제조하여, 이들 세포들을 Core와 NS3 단백질에 대한 단일 항체 또는human patient serum을 이용한 Western blotting으로 확인하여 단백질 발현 정도가 높은 clone들을 선별하였다. 단백질의 발현이 확인된 세포주들은 MFG.Core, MFG.NS3, MFG.Core.NS3, MFG.NS3. Core를 도입한 세포주이다. 이와 같이 하여 선별된 정상간세포에서 NS3 단백질을 발현하는 세포주(CL.2-NS3) 와 간암세포주에서 NS3 단백질을 발현하는 세포주(A.2-NS3)를 이용하여 CTL assay를 수행하였다. CTL assay는 JAM test의 방법을 이용하였다. 선별된 세포주를 생쥐에 주사하여 2-4 주 후에 주사한 생쥐로부터 비정세포를 얻었다. 불활성화한 선별된 세포주를 비장세포와 함께 배영하여 in vitro stimulation을 시켰다. 그리고 4-5 일후, 15-18 시간동안 미리 [3H]-thymidine을 incorporation시킨 선별된 세포주와,stimulation된 비장세포를 15-18 시간동안 함께 배양했다. 이렇게하면 target 세포로 이용된, 선별된 세포주가 CTL에 의해 파괴된 정도를 측정할 수 있게 된다. 이와 같이 하여 측정한 결과 NS3 단백질을 발현하는 정싱간세포주와 간암세포주에 의하여 유발되는 CTL은 미약한 것으로 보여진다. 따라서 이 특이적인 CTL을 강화하기 위한 방법을 계속적으로 연구하여야 할 것으로 생각된다. 이를 위하여 HCV의 core 및 NS3와 IL-12를 동시에 발현하는 DNA 벡터를 제조하여 macrophage 또는 여기서 유래된 세포에 도입하여, 이를 생쥐에 주입함으로써 CTL 활성도를 증가시키고자 시도하고 있다.

Hepatitis C virus (HCV) is the major cause of post-transfusion hepatitis. More than 50% of‘ acutely infected individuals progress to a chronic carrier state that frequentely results in cirrhosis. In addition, HCV infection is an independent risk factor for the development of hepatocellular carcinoma. Currently, INFa is the only treatment‘but its use is limited by low efficacy and high toxicity. Hence, there is an w-gent need for alternative anti-HCV therapy to treat the growing population of patients. The induction of neatralizing antibodies of broad reactivity was hampered by several reasons; heterogeneous nature of HCV genome, high mutation rate, and hypervariable region in envelope proteins. Much information has been accumulated showing that genetic immunization using HCV protein expression plasmids elicit strong cytotoxic T lymphocytes (CTL) responses. Cytotoxic T lymphocyte (CTL), which are believed to be cental in the immunosurveillance of infected and transformed cells, can recognize intracellular' proteins in the form of peptides presented by major histocompatibility complex (MHC) class 1 proteins. Cytokines are important molecules that influence antigen-presentation to (MHC) class I and II molecules on antigen-presenting cells, as well as subsequently stimulate cytokine production by other cells. In some cases, engagement of the CD3/T cell antigen receptor (TCR) complex on small, resting T cells is inefficient to trigger cell-mediated cytotoxicity of' to induce a proliferative response. Recently, the new model was proposed. As this new model, by recognizing antigen on APCs, T helper cell delever a signal that activates the APCs. The licensed APC can then directly stimulate T killer cells. Furthermore, activation of dendritic cells by. CD40 results in increased production of cytokines, especially IL-12. Several studies have shown that the DNA -based immunization technique offers the potential advantage of including cellular immune responses against conserved internal proteins of a virus, as well as the generation of antibodies to viral surface proteins. In order to develop DNA vaccine of HCV, we have constructed various HCV core and/of NS3-expressing vectors and we have demonstrated that cell lines transfected with these DNAs express HCV Core or NS3 proteins. We also studied that cell lines tranfected with recombinant HCV NS3 DNA can induce a specific CTL response in mice, but the specific CTL activity was very low. Now, we are developing a new immunizing agent for a therapeutic and preventive DNA vaccine by introducing IL-12 and HCV Core or NS3 into MHC class I and II expressing cells as APC.
