Amphotericin B) 들의 뇌혈관막(blood brain barrier) 투과 효과를 높이는 기술이 개발됨으로써, 가능성 있는 치료 약제들의 치료 효과를 증진시키는 연구 결과를 얻었다. 본 연구실에서는 Amphotericin B의 항체를 성공적으로 개발하였으며, 이들 항체와 transferrin 항체와의 복합체(fusion antiody)를 형성하여 뇌혈관막의 투과 효과를 증가를 가능케 하였다. 그리고 현재 실험동물을 이용한 기초 실험을 통하여 이의 가능성을 증명함으로써 프리온 질환 치료의 가능성을 높이는 성과를 얻었다. 셋째, 프리온 질환의 특징적인 병변의 하나인 퇴행성 신경 변화의 기초 기전을 밝히는 연구를 통하여, 프리온 질환에서는 reactive oxygen species(ROS)의 생성 및 이들과 신경교세포와의 상호연관성이 퇴행성 신경 변화를 유발하는 주된 기초 병리 기전임을 밝혀낼 수 있었다. 이들의 연구 결과는 향후 항산화제를 이용한 퇴행성 신경 질환에 대한 치료 약제 개발의 기초를 제공하게 되었다. 넷재, 부분적으로나마 국내에서 광우병 발병 가능성을 알아보기 위해 전국에 산재한 도살장을 대상으로 도살된 소의 뇌조직을 무작위적으로 선택하여 감염성 프리온 단백의 존재 여부를 판정하였으나, 국내에서는 전혀 발견되지 않음으로써 현재까지 국내는 광우병에 대한 안전지대임을 부분적으로나마 확인할 수 있었다. 그러므로 본 연구실은 국내외적으로 프리온질환의 기초연구 뿐만 아니라 응용연구에서도 국제수준의 기술 경쟁력을 확보하는 계기가 되었다.

As infectious diseases, prion derived chronic neurodegenerative diseases show sporadic incidence in humans and animals such as cow, sheep, or goat. In some case, genetic inheritance of these diseases has been observed in humans. Prion disease results in 100% morbidity, and so it is very fatal. However, its pathogenesis and mechanism are not clearly known, yet. Once prion disease occurs as shown in mad cow disease, its incidence cannot be controlled. Thus, our studies on diagnosis and pathogenic mechanism of prion disease is very important area to be pursued. A main obstacle of these studies is characteristically long period of pathogenic progress. Although the prion researches have been mainly studied in several developed countries, our lab funded by Ministry of Health and Welfare from 1996 established ability to independently perform studies on prion disease in Korea now. The followings are the outcomes of our research funded for last three years. First, a diagnostic technique for prion disease has been established now in our lab. There was no proper way to diagnose any prion infected diseases in humans and animals in Korea until we set up the diagnostic technique by getting fund from Ministry of Health and Welfare. In addition, we have capability to diagnose the inherited familial prion diseases which occur 10 to 15% of total prion diseases. Our lab became only diagnostic center for prion disease in Korea (National Institute of Health Designated Diagnostic Center for Prion). Second, no therapeutics for prion diseases has been developed yet. We developed a technique to enhance the permeability of blood-brain barrier for delivery therapeutics, such as Amphotericin B, to the brain. With one of possible therapeutics, Amphotericin B, we developed monoclonal antibody specific for Amphotericin B. This antibody was genetically fused with transferrin-antibody fusion protein which already proved to cross the blood-brain barrier. These fusion proteins have already demonstrated to enhance the efficacy of Amphotericin B against cryptococcus and the stability in serum. These results indicated the potential treatment of prion diseases with combination of Amphotericin B and antibody fusion proteins. Third, one pathologic characteristics of prion disease is neurologic degeneration. Through studying on the basic mechanism of neurodegeneration in prion disease as well as other similar diseases, the metabolism of NO as a messenger molecule has been changed and the abnormal reactive oxygen species (ROS) have been produced. In addition, interactions of these molecules to other neurons have been the main pathogenic mechanism in neurodegenerative diseases. These findings provide potential of antioxidant agents as preventive and/or therapeutic drugs for neurodegenerative diseases. Last, to investigate the possibility of the incidence of mad cow diseases in Korea, we selectively, but randomly examined prion infection in bovine brains obtained from nationwide butcheries in Korea. As we could not find any infected brain yet, we concluded that Korean cows are still free of mad cow disease. With a three-years support of Ministry of Health and Welfare, we are able to establish worldwide competitive ability on the basic research and the applied research on prion diseases in Korea.
