정상세포에 대한 부작용을 줄이기 위하여 고선량 방사선대신 저선량 방사선을 반복 처리하여 암을 치료하는 분할방사선 치료법의 큰 문제점은 암세포가 방사선에 대한 저항성을 획득한다는 것임. 이러한 적응방사선저항성(adaptive radioresistance)을 획득한 종양세포는 결국 재성장하면서 더욱 악성화되는 것으로 알려져 있으나 이에 대한 체계적인 연구는 미흡함.
본 연구에서는 분할방사선에 의해 유도되는 적응 방사선저항성의 획득 기전을 “방사선 처리 후 세포 생존 활성화 기전” 및 “방사선에 의한 암줄기세포 활성화 기전”으로 세

본 연구에서는 분할방사선에 의해 유도되는 적응 방사선저항성의 획득 기전을 “방사선 처리 후 세포 생존 활성화 기전” 및 “방사선에 의한 암줄기세포 활성화 기전”으로 세분하여 연구하였으며, 이와 아울러 이러한 경로와 관련된 인자들을 제어할 수 있는 펩타이드 발굴을 통한 “적응방사선저항성 제어 원천기술” 개발연구를 수행하였음.
분할방사선에 의하여 암줄기세포의 집적과 상피성-중간엽성 세포형질전이와 함께 방사선과 항암제에 대한 내성 획득 암악성화 기전에는 K-Ras, PKCδ, JNK, Rac1, LCK, PI3K/AKT, MAPK (ERK, JNK, p38), Nitric Oxide 등이 관련되어있음을 밝혔음. 또한, 2-pyrone 의 새로운 유도체인 BHP 그리고 천연물에서 얻은 Phlorotannin 성분인 Eckol과 Phloroglucinol이 분할방사선 조사후 획득된 암세포의 악성화를 타겟할 수 있으며, 그 관련 신호기전 연구를 밝혔음. 이러한 분할방사선 유도 암악성화 관련신호전달기전들의 연구를 최종적으로 취합하여, 암세포의 악성화 관련 신호인자들의 신호전달 네트워크를 구축하고자 하였으며, 이에 대한 연구성과로 17편의 국외논문 게재와 10개의 국내특허 등록 및 출원을 하였음.

Radiation is widely used for cancer treatment. For clinical application of ionizing radiation (IR), high dose of radiation is often fractionated to reduce the side-effects that may be caused by damages of neighbored normal cells around tumor.
However, recent studies have reported that IR paradoxically promotes the malignant cancer cell phenotype, allowing relapse after treatment. Notably, this effect is frequently observed after exposure to doses of fractionated radiation that are not sufficient to eradicate the primary tumor. Although the mechanisms underlying this effect are not well understood, it may reflect the selective expansion of intrinsically IR-resistant cancer cells or acquisition of radio-resistance after exposure to IR.
In this context, considerable emerging evidence suggests that a subpopulation of cancer cells is highly tumorigenic and self-renewing, properties reminiscent of normal stem cells. Because these cells constitute a small percentage of cells in tumor and their gene expression profiles resemble those of normal stem cells, they have been called tumor-initiating cells or cancer stem-like cells (CSCs). These CSCs are thought to be responsible for tumor relapse after treatment and, as such, are regarded as good potential therapeutic targets. Moreover, the resistance of CSCs to anticancer treatments such as radio- and chemotherapy implicates these cells in IR-induced malignant progression in cancer. In this study, we aimed to understand the cellular and molecular signaling mechanisms underlying the maintenance of CSCs and acquisition of resistance to anticancer treatment. With promising progression, we found that the involved-mechanisms of K-Ras, PKCδ, JNK, Rac1, LCK, PI3K/AKT, MAPK (ERK, JNK, p38), Nitric Oxide etc. in the maintenance of CSCs and acquisition of resistance to anticancer treatment. Also, we proved the anticancer effects of natural compound of Phlorotannin (Eckol, Phloroglucinol) that target IR-induced malignant phenotype in cancer. In addition, we attempted to develop peptide-based CSC targeting cancer therapy. Finally, converging our studies, we suggest the signaling networks that are responsible for IR-induced cancer malignancy.
