Ⅳ. 연구개발 결과 및 활용에 대한 건의

ND 바이러스 백신 생산에 적합한 세포주 개발에서는 primary Chick fibroblast와 이들의 transformant 들 그리고 다양한 cell line들에 대하여 ND 백신의 생산성을 비교 검토하였으며, 최적 세포주의 배양 조건을 검토하였다. ND 백신의 생산에 적합한 세포주 확보를 위하여 다양한 동물 세포주를 세포주 기탁기관들로부터 확보하였다. 또한 Chick 수정난으로부터 Primary 세포주를 확보하였으며, 형질 전환을 이용하여 세포주를 개량하였다. 확보된 세포주들의 배양 특성을 비교 조사하였으며 백신 생산성을 검토한 결과 DF-1과 MDBK 세포주가 가장 우수하였다. 또한 Primary fibroblast를 PMA를 이용하여 형질전환 하였을 때 우수한 백신 생산성을 갖는 것을 확인하였으며, Chick fibroblast를 이용한 ND 백신 생산에서 항산화제를 처리할 경우 약 20%의 생산성 증대 효과를 얻을 수 있었다. 본 연구결과는 ND 백신 생산뿐만 아니라 다양한 동물 바이러스 백신 생산에 활용될 수 있을 것으로 판단된다.
동물세포를 이용하여 뉴캐슬병 바이러스를 효과적으로 생산하기 위해서는 고생산성의 고밀도 세포 배양 시스템의 개발이 필수불가결하다. 따라서 본 연구에서는 영양물질 소모 및 노폐물 생산 kinetics의 조사와 함께 영양물질 및 노폐물에 대한 세포성장의 민감성 조사를 통해 배지의 최적화 및 고농도 동물 세포 배양 시스템 구축을 위한 기본 전략을 확립하였다. 생물반응기에서 고정화 세포배양 시스템을 확립하기 위해 최적 미립담체로 Cultispher-G를 선정하였고, 최적 미립담체 농도를 선정하였으며, 최적 고정화 방법을 제시하였고, serial propagation이 가능함을 확인하였다. 또한 세포 성장뿐만 아니라 뉴캐슬병 백신의 생산성도 저해하는 주요 인자인 암모늄 이온의 축적 문제를 해결할 수 있는 암모늄 이온 선택성 제오라이트를 이용한 고정화 흡착법을 도입함으로써 고농도 동물세포 배양 시스템의 기반을 마련하였다. 더구나 세포의 지수적 성장 후 세포의 사멸(apoptosis)이 증가하는 현상은 동물세포의 고농도 배양을 방해하는 결정적 인자이므로 본 연구에서는 세포 사멸인자 3종을 선발하여 동물세포배양시스템에 첨가함으로써 세포 사멸을 방지하는 새로운 고농도 세포배양시스템을 제시하였다.
동물세포를 이용하여 뉴캐슬병 바이러스를 효과적으로 생산하기 위해서는 우선 고생산성의 고밀도 세포 배양 시스템의 개발과 함께 고 생산성 뉴캐슬병 백신 생산 시스템 구축 및 운전 전략의 확립이 필 수 불가결하다. 따라서 확립된 고밀도 세포 배양 시스템을 바탕으로, 백신생산 동안 영양물질 소모 및 노폐물 생산 kinetics 조사, 최적 serum 농도 조사 및 배지 무교환 조건 등을 통해 백신 생산배지를 최적화하였으며, 최적 pH, 최적 MOI, 감염시기, 추수시기 등의 최적 생산 조건을 확립하였고, 또한 생산성 향상을 위해 DEAE-dextran, ascorbic acid, poly-L-lysin, CaCl2 등의 첨가제를 최적화하였다. 한편 3종의 세포사멸억제제 후보물질이 백신 생산에 미치는 영향을 조사함으로써 세포사멸억제제의 백신생산 시스템으로의 응용가능성을 조사하였고, 암모늄 이온의 백신 생산에 대한 영향 조사와 고정화 흡착제의 적용 가능성 조사 등을 통해 미립담체 및 고정화 흡착제를 이용한 바이러스 생산 시스템을 확립하였으며, 확립된 시스템에서의 생산성 향상을 위해 유가식 배양을 도입하였다. 또한 고농도 세포배양과 백신 생산을 결합한 유가식배양 공정 개발을 통하여 백신의 생산성을 100배 향상 시켰다. 이러한 여러 가지 기초 연구를 통해 bioreactor를 기반으로 하는 최적 백신생산 시스템을 구축하고 이러한 시스템을 위한 최적 운전 전략을 확립하였다.
구축된 bioreactor를 기반으로 하는 세포배양 및 백신 생산 시스템과 자동화 시스템의 통합 및 scale-up을 위하여, 우선 MOI, infection time, harvest time 등의 백신생산 관련 운전변수를 최적화하였으며, 제2세부에서 확립된 자동화 시스템을 이용하여 고농도 동물세포 배양 공정과 백신 생산 공정이 병합된 유가식 생물반응기 시스템을 구축하였다. 이들을 바탕으로 협동과제에서 선정된 세포주를 이용하여 구축된 생물반응기 시스템에서 생산함으로써 확립된 시스템에서의 ND백신의 생산성을 조사한 결과 9×1010 pfu/ml 정도의 획기적인 백신 생산성을 얻을 수 있었다.
생물반응기에서의 배양을 위한 기초실험으로 T-flask와 spinner flask에서 기초배양 조건을 조사했으며, 이 결과를 토대로 생물반응기의 운전 전략을 확립하였다. T-flask와 spinner flask에서의 배양 결과, 배양 중 용존산소의 부족 현상과 pH의 감소가 세포 성장에 심각한 부정적인 요인임이 관찰되었다. 따라서 이러한 저해 요인들을 극복하기 위하여 pH와 용존산소를 정밀 조절할 수 있는 동물세포 배양기를 자체 설계ㆍ제작하여 사용하였다.
생물반응기 내의 용존산소량이 세포성장에 미치는 효과와 효율적인 산소 전달을 위해 배양액 내에 정밀한 microsparger를 설치하여, 적은 양의 직접 공급된 공기에 의해 배양액내의 용존산소를 정밀 조절하는 전략을 개발하였다. 한편 microsparging aeration의 경우, 생성된 미세한 공기방울이 터짐으로써 세포성장에 부정적인 영향을 미칠 수 있으므로, 이러한 현상을 극복하기 위해 동물세포배양기 내에 다공성의 실리콘튜브 또는 PTFE polymer 튜브를 설치하여 용존산소를 공기방울 없이 공급하는 방법을 설계하였다. 또한 세포배양기의 scale-up시 가장 중요한 산소전달계수 (kLa) 측정을 위해 용존산소의 물질 수지식을 이용하는 dynamic method에 대해 실험방법을 확립하였으며, 상기에서 언급한 다양한 공기전달 방법에 대해 실제로 kLa 값를 온라인 상에서 측정하여 용존산소 공급의 효율성을 비교 검토하는 방법을 완성하였다. 이와 함께 배양공정 데이터 수집 및 분석을 위해 컴퓨터 프로그램을 완성하였으며, 이를 기초로 배양공정 자동제어시스템을 설계하여 동물세포의 대사특성을 조사하였다. 특히 동물세포 성장과 관련된 변수 (즉 specific oxygen uptake rate, oxygen uptake rate, cell concentration 등)에 대한 온라인 측정을 통해 세포농도를 컴퓨터상에서 자동 추정하였으며, 이를 위해 가장 적합한 제어방법을 확립하였다.
본 실험 결과 microsparging aeration system과 항거품제를 함께 이용함으로써, 소량으로 기체산소를 지속적으로 공급하여도 공정상에서 문제점이 발생하지 않는 결과를 확인하였으므로, direct microsparging에 의한 산소공급 시 사용할 수 있는 기체산소의 물질수지식과 가스분석장치에 의해 세포배양공정 변수를 자동으로 측정하는 방법을 확립하였다. 즉 중요 배양공정 변수의 실시간 측정 및 모니터링을 위한 이론식을 확립해서, 이들 주요 공정변수들을 컴퓨터상에서 분석하고, 적시에 배양공정을 조정해 주기 위한 배양공정 계측 및 제어시스템을 확립하였다. 또한 세포배양 및 바이러스 생산을 위한 유가식배양 공정 개발 연구를 동시에 수행하였다. 유가식배양에 의한 숙주세포의 배양생리적 특성을 규명함으로써, 고농도 세포배양이 가능해졌으며 또한 ND 바이러스의 생산성이 큰 폭으로 증가됨을 확인 할 수 있었다.
세포 고정화 배양의 경우, 담체 내 균일한 균체분포와 담체내로의 기질 (글루코스 및 글루타민) 공급이 효율적으로 이루어지기 위해서는 수학적인 모델링과 제어기법, 아울러 불균일 시스템의 특성을 나타내는 효율인자와 담체 중심에서의 기질농도 등에 대한 정밀한 계산이 필수적이다. 이를 위해 고정화 Vero 세포배양의 kinetics data를 바탕으로, 물질수지에 의거한 미분방정식을 도입하여 효율인자, Thiele modulus 및 담체내의 기질구배 등을 구할 수 있었다. 특히 확산계수, 담체의 균체부하량 (cell loading)등이 효율인자 및 담체 중심의 기질농도에 미치는 영향을 예측하여, 다양한 동물세포 배양 모델에 적용하였으며, modelling 및 simulation 연구에 의해 회분식배양, 반복회분식배양, 유가식배양 및 perfusion 배양공정의 최적화에 필요한 중요한 요인들을 분석할 수 있었다.
결론적으로 본 연구에서 확립된 자동화 고정화 생물반응기에서 고농도 동물세포배양을 통한 최적 ND 백신 생산 시스템은 기존의 수정란에 의한 뉴캐슬병 백신 생산 방법 보다 효율적이고 경제적이며 환경문제가 적은 새로운 뉴캐슬병 백신의 생산 방법으로, 이 시스템의 확립을 위한 핵심 요소 기술 및 scale-up 기술은 ND 백신뿐만 아니라 다양한 동물 또는 인간 바이러스 백신의 생산에 활용될 수 있다.
또한 본 연구에 의해 축적된 자동화 고농도 동물세포배양 공정의 know-how는 동물백신 이외에도 기존 동물세포 배양에 의해 생산되고 있는 인간 백신 및 단일클론항체, 치료용 단백질 등의 고부가가치 유용 생물의약품들의 국내 개발에도 적용이 되어서 가격 경쟁력 확보가 가능하며 이들 제품군들의 국내 생산을 앞당길 수 있다. 본 연구에서 확립한 기술은 참여 기업체에 기술이전 한 후에 안정성 검사 및 현장 실험을 거쳐서GMP 시설을 갖춘 제3의 산업체와 공동으로 산업화 시켜나갈 예정이다.

The conventional production method has several problems like labour-intensive process, impossibility of automation, and the disposal of used eggs. A new production method using animal cell culture could be one of the promising alternatives to overcome these problems, so the development of an industrial ND vaccine production system employing animal cell bioreactor is keenly required. The development of highly productive host cell lines, the establishment of an optimized ND production strategy through high density animal cell culture, and the development of automatized and scaled-up bioreactor system are necessary in order to accomplish substantial reduction in production cost through development of a novel ND vaccine production system based on animal cell bioreactor.
In this work, cell lines were developed to be used for the production of ND vaccine. Primary chick fibroblasts and their transformants, and various mammaline cell lines were tested for their ND vaccine productivity. The optimization of the cell culture process was also investigated. Mammalian cell lines were obtained from various type culture collections. Primary fibroblasts were prepared from chick embryos. Transformation of the primary cells were carried out by mutagens. Finally, DF-1 and MDBK cells were selected as the most useful cell lines. Transformants from the primary fibroblasts were also a recommendable cells for the production of ND vaccine. Anti-oxidants increased the vaccine productivity 20%. The cell lines and results obtained in this work could be used in the production of other various vaccines.
The development of a high density cell culture system with high productivity is prerequisite to the efficient production of New Castle's Disease Vaccine (NDV) using animal cells. Therefore this study set up a basic strategy for the development of optimized media and a high density cell culture system through the investigation of the kinetics and cell growth sensitivities of nutrients and waste byproducts. In order to establish immobilized cell culture in a bioreactor system, Cultispher-G was selected as a optimum microcarrier, optimum microcarrier concentration was determined, optimal cell immobilization method was suggested, and the feasibility of serial propagation was verified. Ammonia was identified as a key component which inhibited both cell growth and NDV production. Thus, a high density cell culture system was devised by introducing a immobilized adsorbent system using ammonium ion selective zeolite which reduced ammonia accumulation problem. Furthermore, three apoptosis inhibitors were screened out because the increase of apoptosis after exponential growth of cells is a key factor preventing from high density cell culture, and a novel high density cell culture system employing these apoptosis inhibitors was suggested.
The efficient production of New Castle's Disease Vaccine (NDV) also requires the development of a highly productive NDV production system and its optimum operating strategy as well as the development of high density cell culture system. Therefore the production medium was optimized through the studies of nutrient consumption kinetics and waste production kinetics, optimum serum concentration, and media-replacement-free condition. The optimum operation conditions including pH, MOI, infection time, and harvest time were also determined, and the additives such as DEAE-dextran, ascorbic acid, poly-L-lysin, CaCl2 were optimized for the enhancement of productivity. The applicability of apoptosis inhibitors to vaccine production system was examined through the studies of the effects of three apoptosis inhibitors on virus propagation. Besides, a novel virus production system employing immobilized adsorbents and microcarriers was devised based on the study of the effect of ammonium ion on virus production and applicability of immobilized adsorbents, and fed-batch operation mode was introduced to the developed system for the productivity improvement. In addition to that, the ND vaccine productivity was increased by 100 times through the development of a novel system combining a fed-batch high density cell culture system and a fed-batch vaccine production system. An optimum vaccine production system based on bioreactor and its optimum operation strategies were established through the various basic studies mentioned above.
Prior to the integration and scale up of cell culture system, vaccine production system, and automation system, operation variables of vaccine production such as MOI, infection time, harvest time were optimized based on bioreactor system. And a scaled-up bioreactor system incorporating not only high density culture system and vaccine production system but also fed-batch and automation was developed. ND vaccine was produced in a developed system employing cell lines selected by cooperative team(Korea University), and it was possible to achieve the considerably high titer of 9×1010 pfu/ml.
Efficient aeration systems using porous silicone tubing or PTFE polymer tubing were designed for supplying dissolved oxygen into the cell culture medium without formation of excess forms.
Experimental method for measuring oxygen mass transfer rate (kLa), the most important factor in the scale-up process was established by applying mass balance for the dissolved oxygen in the bioreactor (i.e. dynamic method), and then various aeration systems were compared for their oxygen-supplying efficiency through on-line measurement of the intrinsic value of kLa. For data acquisition and analysis in the Vero cell cultures, automatic computer-controlled cell culture process was developed, thus enabling on-line estimation of the important cultivation parameters such as oxygen uptake rate and cell concentration. Notably a special antifoam agent was found to efficiently eliminate viscous forms generated when dissolved oxygen was continuously supplied with direct microsparging aeration system. This results made it possible to directly estimate valuable cells' physiological features during the Vero cell culture by applying a theory of gaseous oxygen mass balance and installing vent gas analyzer. In parallel, an efficient fed-batch culture process was developed for mass production of Vero cells, leading to highly enhanced titre of ND virus. Finally, based on kinetic growth data of the immobilized Vero cells, effectiveness factor, Thiele modulus and concentration profiles of carbon and energy sources (glucose and glutamine) inside a microcarrier were estimated. These process parameters were then successfully utilized for modelling and simulation studies for the optimization of various cell culture modes such as batch, repeated fed-batch, fed-batch and perfusion operation processes.
In conclusion, the ND production system developed in this study based on high density animal cell culture in a bioreactor is a novel production method which is more efficient and environmentally friendly than conventional methods. And key technologies and know-hows accumulated during the development of the ND production system could be applied to the production of other animal or human vaccines.
