1. 버섯파리류 해충의 유인살충 트랩 개발
가. 국내 버섯파리류 해충의 분자생태적 특성
국내에 느타리버섯 재배사에서 대발생하는 버섯파리 L. mali와 C.fuscipes의 유전적 다양성과 지역적인 변이를 조사하기 위하여 미토콘드리아 COI 유전자를 분석하였다. 채집 개체중 L. mali 55개체, C.fuscipes 41개체를 선발하여 mtDNA COI 유전자의 일부(406 bp)의 염기서열을 결정한 결과, L.mali의 경우 매우 낮은 염기서열변이 (0.2%)를 나타내었

1. 버섯파리류 해충의 유인살충 트랩 개발
가. 국내 버섯파리류 해충의 분자생태적 특성
국내에 느타리버섯 재배사에서 대발생하는 버섯파리 L. mali와 C.fuscipes의 유전적 다양성과 지역적인 변이를 조사하기 위하여 미토콘드리아 COI 유전자를 분석하였다. 채집 개체중 L. mali 55개체, C.fuscipes 41개체를 선발하여 mtDNA COI 유전자의 일부(406 bp)의 염기서열을 결정한 결과, L.mali의 경우 매우 낮은 염기서열변이 (0.2%)를 나타내었다. 반면 C. fuscipes는 41개체로부터 총 10개의 haplotype(CF1~CF10)으로 최대 1.2%의 유전적 다양성을 보였다. C. fuscipes에서 집단 사이의 유전적 거리(FST)는 0.345--0.094, 세대당 이동 암컷 이동율(Nm)은 0.947로 나타났다.
나. 버섯파리 유인살충트랩 제작
유인 트랩 제작을 위한 색상 선발은 최종적으로 실재 느타리버섯 재배사 시험 결과를 토대로 결정하였으며, 버섯파리 유인을 위한 끈끈이 트랩의 색깔은 보라색이 효과적이었다.
버섯파리 유인을 위한 유인물질은 느타리버섯 재배시 발생되는 휘발성 향기 성분 중 약 50% 이상을 차지하고 있는 1-octen-3-ol 과 3-octanone 선발하여, n-hexane에 희석하여 1.5% agar 배지에 넣어 formulationtlzu 유인효과를 검정하였다. L. mali 암컷 성충에 대한 유인효과는 0.5 mg의 1-octen-3-ol에서 났으며, C. fuscipes에서는 1 mg의 1-octen-3-ol에서 암컷성충에 대한 유인효과가 높았다. 또한 3-octnone의 유인력 시험 결과와 종합하여 분석할 때, 1-octen-3-ol과 3-octnone의 비율이 2:1로 조성된 1 mg의 혼합물을 유인제로 사용하였을 때 버섯파리에 대한 유인효과가 가장 높았다. 이를 기초로 원통형, 삼각형, 고깔형 및 막대형의 여러 형태로 제작하여 실재 유인효과를 검정한 결과, 막대형 트랩에서 유인효과가 높은 것으로 나타났으며, 그 유인 효과는 약 33%정도로 나타났다.
다. 활용 건의
유인살충 트랩은 느타리버섯 재배사 내에서 버섯파리의 밀도 감소에 효과가 인정되어,특허를 출원할 예정이다. 국내에 서식하는 버섯파리류 해충의 분자 생태적 특성은 국제 SCI 학술잡지(Appl. Entomol. Zool.)에 게재하였다. 따라서 본 연구의 유인살충 트랩은 계속적인 효율성 제고 연구와 함께 개발 기술은 산업체 이전에 따른 실용화가 가능할 것으로 기대된다.
2. 버섯파리류 해충방제용 미생물상충제 개발
가. 버섯파리류 해충 방제용 새로운 Bacillus thuringiensis 656-3균주의 분리
국내 느타리버섯 재배사의 토양으로부터 버섯파리류 해충(Lycoriella mali 및 Coboldia fuscipes)에 강독성을 보이는 Bacillus thuringiensis 656-3 균주를 분리하였다. B. Thuringiensis 656-3(Bt 656-3) 균주는 이중피라이드형 독소를 생산하며, B. thuringiensis subsp.morrisoni(H8a8b)의 H antiserum과 반응하였다. 또한 Bt 656-3의 플라스미드와 단백질 양상은 B. thuringiensis subsp. morrisoni PG-14과 유사하였다. 그러나독소 유전자 특이 프라이머를 이용한 PCR 분석에 의한 유전자 타입 결정에서 Bt 656-3은 reference 균주인 B. thuringiensis subsp. morrisoni PG-14와 달리 cry4A, cry4B, cry10A, cry11A 및 cry1Ac 유전자를 함유하고 있었다. 이러한 유전자 타입 결정 결과로 볼 때, Bt 656-3은 새로운 균주로 판단된다. 아울러 Bt 656-3의 버섯파리류 해충에 대한 독성 검정 결과, Bt 656-3의 LC50치는 reference 균주인 B. thuringiensis subsp. morrisoni PG-14에 비해 C. fuscipes(73.6 ngㆍmL-1, 24시간 처리 후)에서 약 1.68배, L. mali(257.4 ngㆍmL-1, 24시간 처리 후)에서 약 1.95배정도 높게 나타났다.
나. Bacillus thuringiensis 656-3 균주의 대량배양 및 제제화
버섯파리 해충 방제용으로 분리 선발된 Bt 656-3의 대량을 위한 배지는 다낵질원으로 대두박을 탄소원으로 밀기울을 사용하였다. 대두박과 밀기울의 혼합 비율은 최종 농도가 4%의 범위에서 대두박과 밀기울을 20:20의 비율로 조제하여 대량배양기에서 배양하였을 때, 세포 성장도나 포자 형성 면에서 가장 양호하였다.
제제화는 Bt 656-3의 독소, 포자및 배징서 유래된 밀기울과 대두박의 입자를 포함하고 있는 건조 분말화된 배양침전물 40g 과 metamorphic starch 전분 60 g을 혼합하여 제제화하였다. 이러한 조건의 제제에서 2 g의 분말제제를 물 750 ml에 섞어 10 m2에 살포할 수 있는 양으로, 이때 Bt 656-3의 최종농도는 5 x 107 cfu가 함유되며, 1회 살포시 실재 느타리버섯 재배지에서 최소 90% 이상의 살충력을 가진다. 제제는 살포 후 5일째까지 약 90%의 살충율을 보였다.
다. Bacillus. thuringiensis 656-3 제제의 느타리버섯 재배사 실증시험
느타리버섯 재배사 포장 실증 시험에서 Bt 656-3 제제 처리구와 무처리구 간의 느타리 버섯 수확량을 비교한 결과, 제제 처리구에서 1차 수확시 약 7%, 2주기에서는 약 17%, 3주기는 약 83%, 4주기에서는 약 36%의 수확량이 증대되었다. Bt 656-3 제제 살포와 유인살충트랩 설치를 느타리버섯 재배나 내에 동시에 처리함으로서 버섯파리 살충율 증대에 따른 느타리버섯 수확량의 조사에서는 제제와 트랩을 동시에 처리하였을 때 Bt 656-3 제제만을 살포하였을 때보다 1주기에 약 1%, 2주기에는 약 4%, 3주기에서는 약 14%, 4주기에서는 약 32%정도 수확량이 증대되었다.
라. 활용 건의
버섯파리류 해충방제용 미생물살충제 개발에 있어서 가장 중요한 관건은 속효성 강독성의 새로운 Bt 균주이다. 따라서 본 연구에서 분리된 Bt 균주는 현재 특허 출원된 상태이다(명칭 : 버섯재배지 토양으로부터 분리된 바실러스투린지엔시스 656-3 균주 및 이를 함유하는 해충방제용 조성물; 특허 출원번호 10-2003-0003628). 아울러 국제 SCI 학술잡지(Current Microbiology)에 게재하였다. 따라서 본 연구의 기초적인 대량배양과 제제화에 이은 재배사내 실증 시험 결과, 본 연구에서 분리한 Bt656-3 균주를 이용한 버섯파리류 해충방제용 미생물살충제 개발 기술은 산업체 이전에 따른 실용화가 가능함을 입증하였다.

1. Molecular ecological characterization of mushroom flies in Korea
Mitochondrial DNA sequence variation of the mushroom pest flies, Lycoriella mali(Diptera : Sciaridae) and Coboldia fuscipes(Diptera : Scatopsidae), in Korea was studied.
We analyzed a portion of Mitochondrial COI gene sequences(406 bp) to investigate the genetic diversity and geographic variation of the mushroom flies. L. mali showed minimal sequence divergence(0.2%) in two mtDNA haplotypes, whereas C. fuscipes showed an intermediate level of sequence divergence(1.2% at maximum) compared with other relevant studies. While L. mali was fixed with one haplotype except for one population, C. fuscipes possessed a total of ten mtDNA haplotypes, and six of these occurred commonly in multiple populations. We ascribed the difference in the level of genetic variation between the two species to a difference in the degree to which they are dependent on cultured mushroom, which is fluctuating food source. Im C. fuscipes, as in other cosmopolitan insect species, a high rate of female migration(Nm=0.947-infinite) and little genetic differentiation(FST=0.345--0.094)between population were estimated.
2. Development of attraction trap
For attraction trap to mushroom flies, L. mali and C. fuscipes, color of trap was selected violet in mushroom house. The 1-octen-3-ol and 3-octanone were tested as attraction materials. The 1-octen-3-ol and 3-octanone were diluted with n-hexane and formulated with 1.5% agar at a rate of 2:1(1-octen-3-ol : 3-octanone). One 1 mg of formulated attraction materials were released at the concentric box of the bottom of traps. The sticky trap was designed as plate type. In oyster mushroom house test, the attraction trap was approximately 36%(C. fuscipes)-38%(L. mali) more effective than nontreatment in capturing mushroom flies.
3. Suggestion for the application
The molecular ecological result of mushroom flies in Korea is previously published in SCI Journal(Appl. Entomol. Zool. 2001). In addition, the attraction trap in this study could be more effective for control of mushroom flies. We expect that the attraction trap may be useful in the development of environmentally-friendly biological for control of mushroom flies.
Part 2. Development of microbial agent for control of mushroom flies
1. Isolation and characterization of a novel starin of Bacillus thuringiensis 656-3 for control of mushroom flies
Bacillus thuringiensis 656-3, isolated from a soil sample collected at mushroom houses, showed high toxicity to mushroom flies, Lycoriella mali and Coboldia fuscipes. B. thuringiensis 656-3 produced bipyramidal inclusions and reacted with the H antiserum of B. thuringiensis subsp. morrisoni(H8a8b). The plasmid and protein profiles of B. thuringiensis 656-3 were similar to those of its reference strain, subsp. morrisoni PG-14. However, PCR analysis using cry gene primers showed that B. thuringiensis 656-3, unlike its reference strain, had cry4A, cry4B, cry10A, cry11A and cry1Ac genes, suggesting that B. thuringiensis 656-3 was a unique strain with respect to gene type.
In addition, B. thuringiensis 656-3 showed a high level of toxicity against mushroom flies, L. mali and C. fuscipes. The LC50 calues of B. thuringiensis 656-3 were higher-approximately 1.68-fold to C. fuscipes(73.6 ngㆍmL-1 after 24 h) and 1.95-fold to L. mali(257.4 ngㆍmL-1 after 24 h)-than those of B. thuringiensis subsp. morrisoni PG-14, respectively.
2. Mass-culture and Formulation of B. thuringiensis 656-3
For mass-culture of B. thuringiensis 656-3, soybean cake and wheat bran were used as medium source. B. thuringiensis 656-3 was cultured in the fermenter containing soybean cake(2%) and wheat bran(2%) mixed with 4% of final concentraton. In this condition, the growth and spore forming of B. thuringiensis 656-3 were most effective.
For formulation of B. thuringiensis 656-3, the cultured B. thuringiensis 656-3, including toxin, spore, soybean cake and wheat bran particles, was freeze-dried and mixed with metamorphic starchat a rate of 40:60(dried B. thuringiensis 656-3 culture : metamorphic starch). Two grams of B. thuringiensis 656-3 formulation, developed as powder formulation, was added to 750 ml of water. Seven hundred milliters of the formulation suspension containing 5 x 107 cfu was sprayed at 10 m2 of mushroom house, showing that the insecticidal effect of B. thuringiensis 656-3 formulation to mushroom flies, L. mali and C.fuscipes, was maintained over 90% by the fifth day after spraying.
3. Insecticidal effect of B. thuringiensis 656-3 formulation to mushroom flies in mushroom house
Insecticidal effect of B. thuringiensis 656-3 formulation to mushroom flies in mushroom house was surveryed as the yield of mushroom. The yield of mushroom house with B. thuringiensis 656-3 formulation was increased approximately 7% for the first harvest, 17% for the second, 83% for the third, and 36% for the final harvest compared with control. In addition, the yield of mushroom house with B. thuringiensis 656-3 formulation and attraction was increased approximately 1% for the first harvest, 4% for the second, 14% for the third, and 32% for the final harvest compared with treatment of formulation only.
4. Suggestion for the application
The result of Bacillus thuringiensis 656-3 is previously submitted for patent(Patent tilte : Bacillus thuringiensis 656-3 isolated from the soil of mushroom houses and a composition for the prevention of insects containing the same; Patent submission number 10-2003-0003628) and published in SCI Journal(Current Microbiology, 2003).
Accordingly, the novel B. thuringiensis 656-3 strain could be more effective for control of mushroom flies. We expect that the novel B. thuringiensis 656-3 strain may be useful in the development of environmentally-friendly biological agent for control of mushroom flies.
