Ⅳ. 연구개발 결과 및 활용에 대한 건의
시설재배 토양의 질산태 질소 간이검정법을 선발하기 위하여 test strip을 이용한 Nitracheck Reflectometer법, 휴대용 비색계를 이용한 Hanna Ion Specific meter법, 그리고 휴대용 이온분석기 Cardy meter법을 실험실 kjeldahl법 및 이온 전극법과 함께 비교 분석하였다. 평가방법의 신뢰도는 NO3-N 함량이 10 mg/kg에서 340 mg/kg으로 분포되는 20개 시설재배 토양에 대하여 표준편차, 변이계수 및 회수율을 비교하였다.
Ⅳ. 연구개발 결과 및 활용에 대한 건의
시설재배 토양의 질산태 질소 간이검정법을 선발하기 위하여 test strip을 이용한 Nitracheck Reflectometer법, 휴대용 비색계를 이용한 Hanna Ion Specific meter법, 그리고 휴대용 이온분석기 Cardy meter법을 실험실 kjeldahl법 및 이온 전극법과 함께 비교 분석하였다. 평가방법의 신뢰도는 NO3-N 함량이 10 mg/kg에서 340 mg/kg으로 분포되는 20개 시설재배 토양에 대하여 표준편차, 변이계수 및 회수율을 비교하였다.
휴대용 비색계법은 낮은 농도와 높은 농도에서 큰 오차범위를 보였으나 Nitracheck test strip법은 실험실 정밀분석법과 유사한 신뢰도와 회수율을 보였다. 따라서 간편한분석조작과 짧은 분석시간을 구비한 Nitracheck test strip법은 신뢰성 있는 간이 검정법으로 활용 가능한 것으로 평가되었다. 시설재배 토양의 질소공급능력을 예측하는 효율적 지표로 확인된 질산 태질소의 현장 검정법으로 이미 확립된 Test strip 측광기법을 측정기기 종류 및 test strip의 제조번호별로 비교 검토하여 효율적인 기종과 test strip을 선발하였다. 그리고 측정치의 신뢰도를 증진시키기 위하여 질산태 질소표준액에 의한 표준화 과정의 도입여부와 함께 토양시료의 현장 채취방법으로 필름통을 이용한 용적 밀도법에 대한 측정 신뢰도 및 침출액의 여과조건의 차이를 비교하였다. 그 결과 측정기종은 Nitrachek으로 하고 test strip은 질산태 질소 표준액으로 표준화시키는 방법이 측정 신뢰도를 향상시켰다. 그리고 토양시료는 필름통을 이용한 용적밀도법으로 취하고 침출액은 간이법으로 여과시켜 측정하는 토양 질산태 질소의 현장 검정법을 확립하였다. 따라서 토양의 질산태 질소를 영농현장에서 검정하여 질소 시비량을 결정할 수 있는 방법을 확립하였다. 또한 지표면에 염류가 집적되는 특성을 갖는 시설재배 토양의 시료채취 깊이에 따른 화학성의 분포를 조사한 결과 질산태 질소는 다른 화학성에 비하여 가장 크게 영향을 받았다. 따라서 시설재배 토양의 시료채취는 0-20cm 의 일정한 기준을 추천하였다.
토양의 질산태 질소 함량이 46 mg kg-1에서 345 mg kg-1의 분포를 갖는 12개 시설재배 토양에서 시비수준을 무비구, 검정시비구 및 표준시비구로하여 토마토의 생산력을 검정하고 토양 화학성과의 상호관계 및 회귀관계의 평가로부터 질소 적정시비량추천모델을 추정하였다. 무비구 토마토의 건물중 및 양분흡수량과 가장 밀접한 상관을 갖는 화학성은 질산태 질소였다. 토양의 질산태 질소 함량과 무비구 토마토의 건물중, 비료효과 및 시비효율의 회귀관계 분석으로부터 토마토 질소 무시비 재배를 위한 질산태 질소의 상한기준은 280 mg kg-1으로 추정되었다. 토양의 질산태 질소가 46, 96, 126 mg kg-1인 3개 토양에서 무비구 검정시비구 및 표준시비구의 3개 시비수준에 대한 건물중 변화관계로부터 토마토 질소 표준시비를 위한 질산태 질소 하한기준은 100 mg kg-1으로 추정되었다. 토마토에 대한 반복실험을 종합하여 무비구 건물중 및 질소흡수량의 상대량과 질산태 질소 함량의 상호관계를 Cate & Nelson 분할법으로 평가한 질소 시비의 상한기준과 하한기준은 무비구 건물중, 질소 흡수량, 그리고 시비구와 무비구의 건물중과 질소 흡수량으로 평가한 경우와 일치하였다. 그 결과 질소 무시비를 위한 상한기준은 280 mg kg-1으로, 표준시비량이 적용되는 하한기준은 70 mg kg-1으로 평가되었다. 그리고 동일한 평가방법으로 검토한 오이의 질소 무시비를 위한 상한기준은 토마토와 동일한 280 mg kg-1으로, 표준시비량이 적용되는 하한기준은 100 mg kg-1로 평가되었으며 배추의 경우도 상한기준은 200 mg kg-1, 하한기준은 70 mg kg-1로 추정되었다. 따라서 상한기준과 하한기준 사이의 질산태 질소 검정에 의한 토마토, 오이, 배추의 질소 시비량 추천모델은 다음과 같이 개발하였다.
토마토 ; 질소 표준시비량에 대한 비율 Y = -0.476X + 133.33
오이 ; 질소 표준시비량에 대한 비율 Y = -0.5556X + 155.568
배추 ; 질소 표준시비량에 대한 비율 Y = -0.769X + 153.85 (X; NO3-N mg kg-1)
재배기간이 장기간인 작물의 질소 덧거름 시비량 결정방법을 확립하기 위하여 Chlorophyll meter (SPAD502, Minolta)를 활용한 엽록소 검정법을 토마토와 오이에 대하여 검토하였다. 토마토의 평가방법은 NO3-N 함량이 55∼306 mg kg-1인 6개 토양에 대하여 무비구 토마토의 건물중과 질소흡수량, 그리고 재배기간 동안의 경시적 엽위별 엽록소 측정치와의 회귀관계로부터 토마토의 생산능력과 질소공급능력에 대한 엽록소 측정치의 기준을 추정하였다. 토마토 잎의 엽록소 측정치는 단위 엽면적당 질소 흡수량과 유의성 있는 정의 상관을 보여 질소 영양진단을 위한 간이 검정법으로 활용 가능성을 보였다. 토마토 재배기간 동안 엽위별 엽록소 측정치는 정식후 45일째15매 잎에서 최대를 보인 후 감소되었으며 정식후 45일째 이후 토마토의 엽록소 측정치는 토양의 질소 공급능력에 따라 현저한 차이를 나타냈다. 정식후 40일째 14매엽의 엽록소 측정치는 토양의 질산태 질소, 무비구 토마토의 건물중 및 질소 흡수량과 유의성 있는 정 상관의 곡선적 관계를 보였다. 무비구 토마토의 최대 건물중과 최대질소 흡수량에 대한 엽록소 측정치의 기준은 각각 57.1로 동일하게 평가되었다. 2차시험의 정밀검토에서 정식후 15매엽의 엽록소 측정치는 토양의 질소공급능력과 토마토건물중 및 질소 흡수량과 유의성 있는 정의 상관을 보였으며 시비구와 무비구의 건물중 및 질소 흡수량의 차이로 평가한 비료효과와는 유의성 있는 부의 상관을 보였다.
그리고 토마토 최대 건물중에 대한 부족비율은 15매엽 엽록소 측정치와 유의성 있는 부의 상관을 보였으며 엽록소 기준 측정치 57.1에 대한 부족비율과는 2차함수의 곡선적인 정의 상관을 보였다. 따라서 토마토 최대 건물중에 대한 부족비율을 덧거름 요구량이라고 가정하면 15매엽의 엽록소 측정치에 의하여 덧거름의 시용기준의 결정이 가능하였다. 또한 오이에 대한 엽록소 검정검토에서 정식후 39일째 13매엽의 엽록소 측정치는 토양의 질산태 질소 함량, 무비구 오이 전체 생체중, 무비구 오이의 질소흡수량, 그리고 시비구에서 무비구의 오이 생체중 차이로 평가한 비료효과 등과 유의성 있는 상관관계를 나타냈다. 이러한 결과는 오이에 대한 엽록소 검정방법도 토마토와 유사하게 질소 덧거름을 결정하는 추천방법으로 활용 가능함을 보였다. 따라서 시설재배 작물의 밑거름은 질산태 질소의 토양검정에 의하여 결정하고 덧거름은 토마토의 경우 정식후 40일째 15매엽, 오이의 경우 40일째 13매엽의 엽록소 측정으로 결정할 수 있는 종합적인 질소 시비추천 방안을 확립하였다.
한국토양비료학회에 상기에 검토된 자료들중 상당수를 이미 발표하였으며 추후로 계속 발표 할 계획이다. 이미 확립된 토양의 질산태질소 현장검정법에 대한 시설재배농가의 활용증대를 위하여 청주시 신촌작목반과 청원군 오송작목반 회원들을 협력 농가들로 구성하여 년중 2차례 토양검정을 실시하여 적정시비량을 추천하고 있으며 보다 광범위한 지역으로 확산될수 있도록 현장검정법의 활용성에 대한 홍보와 함께 여러 가지 방안으로 추진하고 있다.

History of salts accumulation in cultivated soils under the plastic film house and relationship between salts accumulation and its impact on environment were investigated to search the cause and solve the salts problems for effective soil management. Especially, effective management of N fertilization was considered as the most important factor for conserving the soil environment. Nitrate problems on vegetables and ground water due to excess application of N fertilizer were discussed with respect to development of an available index for prediction of soil N-supplying capability and optimum application model of N fertilizer for vegetables. The quick test procedure for estimation of soil nitrate nitrogen by means of test strip reflectometer and chlorophyll meter test for nitrogen diagnosis in plant were examined to establish the effective management method of N fertilization through real time test in fields.
To find a quick test procedure for soil nitrate concentration, three methods, i.e. "Nitracheck 404" reflectometer (Eijkelkamp, Netherlands) with nitrate test strip, a portable colorimeter "Hanna Ion Specific meter(USA) with a color development reagent, and a potable ionic meter "Cardy meter(Spectrum, USA) were tested for twenty soils with different nitrate contents ranged between from 10 mg kg-1 to 340 mg kg-1. The standard deviation, coefficient of variability, and recovery from these quick test procedures were compared with those measured by conventional Kjeldahl distillation method and nitrate ion electrode method. The nitrate concentration measured by portable colorimeter method was higher in soils with low concentration and lower in soils with high concentration than those measured by conventional methods. However, concentrations measured by test strip reflectometer method was showed the similar coefficient of variability and recovery as those by conventional methods for all soil samples. From the experimental results in this study, the test strip reflectometer method was thought to be recommendable one revealed the satisfied results for accuracy, quickness, and simplicity for field test of soil nitrate concentration. In order to establish the test strip method as an advanced field test of soil nitrate, a kind of reflectometer (Nitrachek and RQflex) and lot numbers of test strip, quick filtration of extract solution, and soil sampling method by film case were investigated. Consequently field test of soil nitrate was completed by adding the process as follows; selection of Nitrachek as a standard meter, soil sampling by film case and then changing to gravimetric by bulk density, calibration by standard solution of nitrate, and quick filtration of extract solution by filter paper. And sampling depth of 0∼0cm was recommended for soil nitrate test through comparing the effect of sampling depth such as 0~10cm, 0~20cm, 0~30cm depth on concentration of chemical properties.
The electrical conductivity in soils under the plastic film house showed the best correlation with soil nitrate, suggesting that salts accumulation is closely affected by application rate of nitrogen fertilizer. Consequently salts accumulation in plastic film house soils was closely related with nitrate contamination in ground water and abnormal accumulation of nitrate in vegetables, which can affect health concerns of human and livestock and ecological impact on water resources. Soil nitrate was identified as the most available index for evaluation of vegetables productivity, showing the best correlation coefficient and standardized partial regression coefficient between soil nitrate and productivity of chinese cabbage and tomato.
In order to establish N fertilizer recommended scheme for tomato. the pot experiment was conducted with and without fertilization in different twelve soils which have NO3-N concentrations ranging from 46 to 344 mg kg-1. Total dry weight (DW) of tomato plant grown without fertilization ranged from 28.9 to 112.5 g plant-1 showing a sharp difference in soil fertility. Increases in DW of tomato plants by application of fertilizer N were significantly appeared in the soils which have NO3-N concentration ranging from 46 to 164 mg kg-1. However, DW of tomato plants was not significantly affected by nitrogen application to soils containing more than 208 mg kg-1. The soil NO3-N concentration was significantly and positively correlated with dry weight(r=0.83) and N uptake (r=0.78), and negatively correlated with N fertilizer efficiency and total fertilizer effect estimated from DW difference between fertilization and no fertilization plot.
The upper critical level of soil NO3-N concentration for growing without fertilizer N was estimated 280 mg kg-1 by examining (ⅰ) relationships between soil NO3-N concentration and DW, N, P, and K uptake by the upper part of tomato plant grown without fertilization; (ⅱ) a relationship between soil NO3-N concentration and deficit index for maximum dry weight(110 g plant-1) of tomato plant grown without fertilization; (ⅲ) a relationship between soil NO3-N concentration and total fertilizer effect; (ⅳ) a relationship between soil NO3-N concentration and fertilizer N efficiency. The lower critical level of soil NO3-N concentration for recommended application rate of fertilizer N was estimated 100 mg kg-1 by regression between dry weight of tomato applied with different rate of fertilizer N for three soils contained with NO3-N concentration of 46, 96 and 126 mg kg-1, respectively.
To establish N fertilizer recommended scheme for the chinese cabbage cultivation in plastic film house based on the soil test of nitrate nitrogen, relationship among the content of soil nitrate and fertilizer effects and fertilizer N use efficiency were investigated from nine soils which differed amount of nitrate
nitrogen from 14 mg kg-1 to 226 mg kg-1. The amount of nitrate nitrogen in soil showed a positive correlation with the dry weight of chinese cabbage in the plot of no fertilization. When the fertilizer effects were calculated by difference between the plots of fertilization and no fertilization in the dry weight and the amount of N uptake, a negative correlation was obtained between the amount of nitrate nitrogen in soils and the fertilizer effects. There was also a negative correlation between the amount of nitrate nitrogen in soils and fertilizer use efficiency.
Recommendation of application rate of nitrogen fertilizer based on content of NO3-N in soils was evaluated by the regression equation among the content of soil nitrate, fertilizer effects and fertilizer N use efficiency. In case the content of NO3-N nitrogen in soil is more than 200 mg kg-1, No N fertilization is recommended; However, The standard N fertilization(320 kg ha-1) is recommended for the soils with less than 50 mg kg-1. For the soils ranged from 50 mg kg-1 to 200 mg kg-1 in the amount of nitrate nitrogen, an equation has been developed in order to calculate the recommended amount of fertilizer N.
Recommendation scheme of N fertilizer for the cucumber in plastic film house based on the soil test of nitrate nitrogen as the same estimation method as tomato and chinese cabbage was developed from eight soils which differed amount of nitrate nitrogen from 67 mg kg-1 to 343 mg kg-1. The soil NO3-N concentration was significantly and positively correlated with fresh weight of cucumber and N uptake by upper part of cucumber applied with no fertilizer and negatively correlated with N fertilizer efficiency and total fertilizer effect estimated from fresh weight difference between fertilization and no fertilization plot. These results obtained from biological assay for cucumber production in condition of no fertilizer showed the same tendency as those of tomato and chinese cabbage.
And another recommendation model of optimum application rate of N fertilizer for tomato, chinese cabbage, and cucumber were analyzed by non linear regression of Mitscherlich equation and partitioning method by Cate and Nelson through the scatter diagram of combination experiments. Consequently, the upper critical level of soil NO3-N concentration for growing without fertilizer N was estimated by 280 mg kg-1 equally for tomato and cucumber and 200 mg kg-1 for chinese cabbage.
And the lower critical level of soil NO3-N concentration for recommended application rate of fertilizer N was estimated by 70 mg kg-1 equally for tomato and chinese cabbage and 100 mg kg-1 for cucumber. Consequently, optimum application rate of N fertilizer on the soil containing NO3-N concentration between upper critical level and lower critical level was recommended as the linear equation as follows;
Tomato ; Y = -0.476X + 133.33
Cucumber ; Y = -0.5556X + 155.568
Chinese cabbage ; Y = -0.769X + 153.85

To find diagnosing method of nitrogen status in tomato plant for determining optimum application rate of side dress fertilizer N, chlorophyll reading values were measured by portable chlorophyll meter(SPAD 502, Minolta), and compared with nitrogen supplying capability of soils. Regression between dry weight, amount of nitrogen uptake, and chlorophyll reading at stalk positions of tomato applied with no fertilizer were evaluated for 6 green house soils with different nitrate concentrations ranged from 55 mg kg-1 to 306 mg kg-1. The chlorophyll reading of tomato leave was significantly correlated with amount of nitrogen per unit area of leave suggesting that chlorophyll content is useful for nitrogen diagnosis of tomato plant. The chlorophyll reading showed peak at the 15th leaf of stalk position on the 45th days after transplanting and this suggested that below or near the 15th leaf and before or near the 45th days after transplanting is the critical stalk position and time for diagnosing nitrogen status of tomato by chlorophyll test. The chlorophyll reading at the 14th leaf on the 40th days after transplanting was significantly correlated with soil nitrate status, dry weight and amount of nitrogen uptake by tomato grown with no fertilization. From the above correlation, the chlorophyll reading value of 57.1 at the 14th leaf of tomato was estimated as the critical level for maximum dry weight and amount of nitrogen uptake by tomato grown with no fertilization. And deficit ratio for maximum dry weight of tomato applied with no fertilizer was negatively correlated with chlorophyll reading of 15th leaf at the 40th day after planting. From the result, correlation between deficit ratio for maximum dry weight and dificit ratio for maximum chlorophyll reading, 57.1 was positively significant showing the regression of quadratic curve. If the deficit ratio for maximum dry weight of tomato were considered as required amount of side dress fertilizer N, the chlorophyll test on the 15th leaf at the 40th day after planting could use for determination of application rate. The similar estimation method by chlorophyll meter was conducted for cucumber. Chlorophyll reading on the 13th leaf at the 40th day after planting as a critical stalk position and season was closely correlated with soil nitrate, total fresh weight and N-uptake of cucumber applied with no fertilizer, and fertilizer total effect estimated by fresh weight difference between no fertilization and fertilization plot. From the result, it was possible for cucumber to use the chlorophyll test on the 13th leaf at the 40th day after planting for optimum application rate of side dress fertilizer N showing the same tendency as that of tomato. Consequently, chlorophyll reading of tomato and cucumber leaves measured by potable chlorophyll meter were thought to be available as a rapid plant test for predicting the nitrogen supplying capability of green house soils.
