Ⅳ. 연구개발 결과 및 활용에 대한 건의 1. 닭 장내 미생물의 분리 및 체계화 SPF 10수와 야외 성계 20수로부터 677종의 장내미생물의 분리하여 형태학적, 배양학적 특성을 분석하고 자료를 체계화하여 향후 관련 분야의 연구에 유용한 자료로 활용될 수 있도록 하였다. 2. 경쟁적 배제(CE) 미생물군의 선발 총 30군의 defined 미생물군과 1군의 undefined 미생물군을 포함한 총 31군의 장내미생물군을 확보하였고, 살모넬라 배제 활성을 검증하여 6종의 미생물군을 1차 선발하고 재검증하여 가장 좋은 활성을 보인 undefined 미생물군을 경쟁적 배제(CE) 미생물군으로 최종 확정하였다. 3. 유용 미생물군의 구성 균주 분석 생체내 살모넬라 배제 활성 검증 결과 1차 선발된 6종의 미생물군의 구성균주를 당이용성을 바탕으로 속까지 동정하여 결과를 체계적으로 정리하였다. 구성 균주는 주로 Lactobacillus가 많았고 Bifidobacterium, Streptococcus, Enterococcus, E.coli, Propionibacterium, Leuconostoc, Corynebacterium 등으로 구성되어 있었다. 4. 경쟁적배제(CE) 미생물군의 유용 균주 생육특성 Bifidobacterium, Bacteroides, E. coli, Lactobacillus, Streptococcus의 선택배지를 개발하고, Bifidobacterium, E. coli, Lactobacillus의 단독(개별)배양시 최적 배양 온도, 배양시간 및 배지에 따른 생육양상 등을 조사하였다. 장내 유익균인 Bifidobacterium, Lactobacillus의 생육을 촉진하는 인자를 탐색한 결과 proteose peptone No.3, trypticase peptone 및 galactooligo당을 선발하였다. 5. 경쟁적 배제(CE) 미생물군의 혐기 혼합 배양 기술 확립 CE 미생물의 대량생산용 배지로 glucose, yeast extract를 선정하고 10g/L의 탄소원, 질소원을 3.5시간 간격으로 3회 fed-batch 배양하였다. 용존산소를 제거하기 위하여 0.25g/L의 L-cysteine을 첨가하여 대량 배양을 시행한 결과 1010cfu/ml의 고농도혐기배양이 가능하였다. 6. 경쟁적 배제(CE) 미생물 제제의 제조 및 효능검증 50L 발효기에서 fed batch 방법으로 배양한후 이를 냉동건조하여 CE 제제를 대량으로 제조하였다. 이를 이용하여 생체내 살모넬라 배제 효능 검증을 한 결과 대량 배양물이 살모넬라를 97%이상 배제함을 알 수 있었다. 7. 경쟁적 배제(CE) 미생물군 배양물의 분말화 기술 확립 대량배양물 분말화 조건으로 냉동건조가 적합하였고, 냉동보호제로는 15% skimmilk를 선발하였다. 냉동보호제에 CE미생물군 구성 균주 중 Bifidobacteria와 Lactobacillus에 대한 보호 효과가 뛰어난 galactooligosaccharide를 5% 첨가하여 유용균의 생존율을 증가시킬 수 있었다. 8. 경쟁적 배제(CE) 미생물 제제의 적정처리기술 확립 개발된 CE 미생물 제제의 적정처리 방법은 음용수에 희석하여 투여하는 방법이 부화란에 직접 분무하는 방법보다 살모넬라 배제 효능이 우수하였고, 처리 농도는 최소 1수당 108cfu이상은 투여해야 효능이 있음을 알 수 있었다. 따라서 CE 미생물 제제는 음용수에 희석하여 직접 투여하는 방식으로 1수당 최소 108cfu이상이 투여되도록 하였다. 9. 경쟁적 배제(CE) 미생물 제제의 농장시험(field test) CE제제 투여시 무투여군에 비해 약 4.4배의 증체율 효과를 보여 주었고 사료요구율도 유의미하게 감소하여 생산성의 증대 효과가 확인되었다. 장내환경의 변화효능을 검증한 결과 CE제제가 투여 초기(1주령 이하)에 Bifidobacterium의 신속한 정착과 Clostridium과 같은 장내 유해균을 억제하는데 기여하는 것으로 판단되었다. 또한 장내 휘발성 지방산(VFA) 중 propionic acid와 butyric acid 함량이 CE제제 투여시 무투여군에 비해 현저히 높아 propionic acid의 경우 사육 3주후 CE제제 투여군에서 최대 9배이상 높은 농도로 검출 되었다.상기의 결과는 본 연구에 의해 개발된 경쟁적 배제 미생물제제는 닭의 장내환경을 크게 변화시켜 살모넬라의 장내 정착을 효과적으로 억제함을 알 수 있었다. 본 연구결과는 항생제 사용의 규제 강화, HACCP 실시에 대한 필요 증가등 양계 산업 환경의 변화에 적극 대처 할 수 있는 진보된 위생관리 기술의 실제적인 대안으로 활용될 수 있다.

Result and Suggestions 1) Screening and systematization of intestinal microbes of chicken Six hundred sixty seven intestinal microbes were isolated from 10 SPFs and 20 adult chicken and systematized in cultural and biological characteristics. 2) Selection of CE microflora Thirty defined microflora and one undefined micro flora were obtained. Six microflora were selected among 31 microflora for their excellent CE activities in the intestine of chicken in the first stage. Finally the undefined microflora showing the best CE activity were selected as the CE microflora. 3) Analysis of CE microflora Six microflora selected in the first stage were identified in their ability of sugar utilization. The main microbe was Lactobacillus, and Bifidobacterium, Streptococcus, Enterococcus, Propionibacterium, Leuconostoc and Corynebacterium were also found. 4) Growth characteristics of CE microflora Selective media and the growth patterns for Bifidobacterium, E. coli, Lactobacillus and Streptococcus were investigated. Proteose peptone No.3, trypticase peptone and galactooligosaccharide were chosen as the growth factors for Bifidobacterium and Lactobacillus. 5) Establishment of the mixed culture technique for CE microflora CE microflora were cultivated by fed-batch method. Glucose and yeast extract of 10g/L were fed by 3 times, every 3.5 hour, respectively. L-cysteine of 0.25g/L was used to remove dissolved oxygen and the maximum cell number was over 1010 cfu/mL. 6) Verification of the effects of CE products CE microflora were fermented in 50L fermentor and lyophilized to CE products. CE products eliminated Salmonella by 97% in the in vivo test. 7) Development of drying technique for CE products Lyophilization method was selected for drying CE culture, and 15% Skim milk was chosen as a cryoprotectant. The viability of useful microbe in CE product was increased by addition of 5% galactooligosaccharide, which has a excellent preservation function for Bifidobacterium and Lactobacillus, 8) Establishment of the proper treatment technique CE product was more effective when it was diluted in water and taken to chicken than when sprayed to eggs. CE product was effective at the treatment concentration of 108 cfu per chicken. So CE product was diluted in water and taken to chicken directly at the concentration of 108 cfu per chicken. 9) Field test of CE products CE products enhanced weight gain by 4.4 times and reduced feed conversion rate significantly. And they also contributed to rapid stabilization of Bifidobacterium and to suppress the harmful microflora such as Clostridium in intestinal environment. The propionic acid and butyric acid contents in intestine of CE treated chicken were higher than those of control. After 3 weeks of CE treatment, propionic acid contents of CE treated group was 9 times as much as those of control. This results suggest that CE products change the intestinal environment of chicken to suppress the intestinal colonization of Salmonella effectively. The products can be an alternative for use of antibiotics to cope with the regulation of antibiotics and the introduction of HACCP.
