1. 고기능성 유산균 발효양파음료 개발
양파 착즙액을 이용한 고기능성 유산발효음료를 제조하기 위해서 과채류로부터 702주의 유산균을 분리한 후 발효 시 산도 증가가 우수한 61주의 균주를 1차 선별하였다. 1차 선별된 61주 중에서 앙파 착즙액의 유산 발효능이 우수한 16 균주를 2차 선별하였으며 전자공여능, 아질산염 소거능 등과 같은 항산화활성과 식중독 세균에 대한 항균활성이 우수한 KC-007 균주를 최종 starter 균주로 선정하였다. 균주의 동정을 위해 형태학적, 생리학적 특성을 조사한 결과 KC-0
○ 연구결과
1. 고기능성 유산균 발효양파음료 개발
양파 착즙액을 이용한 고기능성 유산발효음료를 제조하기 위해서 과채류로부터 702주의 유산균을 분리한 후 발효 시 산도 증가가 우수한 61주의 균주를 1차 선별하였다. 1차 선별된 61주 중에서 앙파 착즙액의 유산 발효능이 우수한 16 균주를 2차 선별하였으며 전자공여능, 아질산염 소거능 등과 같은 항산화활성과 식중독 세균에 대한 항균활성이 우수한 KC-007 균주를 최종 starter 균주로 선정하였다. 균주의 동정을 위해 형태학적, 생리학적 특성을 조사한 결과 KC-007은 Pediococcus pentosaceus로 동정되었다(GenBank accession No. EU569832). 양파착즙액을 P. pentosaceus KC-007을 이용하여 발효하였을 경우 생육과 유산발효를 위한 최적조건은 배양온도 40℃, pH 4.0~6.0, 접종량 2%(v/v)로 확인되었다. 양파 착즙액에 2종류의 유산균을 혼합 접종하여 유산발효할 경우에는 P. pentosaceus KC-007과 B. bifidum ATCC 15696을 1:2의 비율로 혼합접종하여 발효하는 것이 가장 높은 산도와 우수한 유산균 생균수를 나타내고 SOD 유사활성도 비교적 높은 것으로 확인되었다. 양파 착즙액에 팽화 홍삼 추출액을 첨가하여 유산발효할 경우에는 팽화 홍삼 추출액의 첨가량에 관계없이 발효 24시간에서 가장 높은 생균수를 나타내었으며 발효액의 전자공여능과 아질산염 소거능은 1%(w/v) 이상의 팽화 홍삼 추출액을 첨가하였을 때 가장 높은 수치를 나타내었다. 양파취를 저감화 하기 위해서는 양파를 2.0%(w/v)의 citric acid 용액으로 24시간 침지하는 것이 가장 효과적인 것으로 확인되었다. 양파 착즙액 발효를 1 L 또는 40 L로 scale-up하였을 경우 유산발효를 위한 최적조건은 양파 착즙액 원액을 pH 5.0으로 조정한 후 P. pentosaceus KC-007과 B. bifidum ATCC 15696을 1:2의 비율로 혼합한 starter를 2%(v/v) 접종하여 37℃에서 24시간 발효하는 것으로 확립하였다.
양파 착즙액 유산발효제품의 저장성을 5, 10, 20, 30℃에서 18일 동안 조사한 결과 5℃와 10℃에서는 산도변화가 거의 없었고 유산균 생균수는 20℃에서는 유산균수가 저장기간이 경과하여도 크게 감소하지 않았다. 효모와 곰팡이는 본 발효제품에서 검출되지 않았다. 양파 유산발효음료의 분말 제품을 제조하기 위하여 양파발효음료를 동결건조하여 제품화하였다. 이상의 결과로부터 양파 유산발효제품의 대량생산을 위한 제조공정은 다음과 같이 확립하였다. 즉, 양파를 세척한 후 8등분한 다음 양파취를 제거하기 위하여 2%(%) citric acid 용액에서 24시간 동안 침지하고 착즙한 다음 121℃에서 10분간 가열처리하고 10,000×g에서 30분간 원심분리하여 불용성 물질을 제거한다. 양파 착즙액의 pH를 5.0으로 조정한 후 121℃에서 15분간 가열살균한 다음 대수증식기까지 생육한 P. pentosaceus KC-007과 B. bifidum ATCC 15696을 1:2의 비율로 혼합한 starter를 2%(v/v) 접종하여 37℃에서 24시간 동안 혐기발효시킨다. 발효가 완료되면 상온까지 냉각한 다음 양파 발효액 10%(v/v), 배 농축액 2.38%(v/v), citric acid 0.2%(v/v), 팽화홍삼 농축액 0.02%(v/v), 올리고당 5%(v/v), 고과당 10%(v/v), 아카시아벌꿀 2%(v/v), 비타민 C 0.2%(w/v), 배향 0.2%(v/v), quercetin 추출물 0.29%(w/v), 정제수 69.61%(v/v)의 비율로 배합한 후 병에 충진하여 양파 유산발효음료 제조를 완료한다. 분말제품을 생산하기 위해서는 유산발효음료를 동결건조하여 팩에 충진한다. 추출공정 및 발효 공정을 제어하기 위하여 Governing equation을 이용한 에너지 수지식을 설계하여 MATLAB program의 확장도구인 simulink를 사용하였다.
2. 기능성 양파식초음료 개발
복숭아 낙과로부터 분리한 산생성능이 우수한 초산균은 분리 동정한 결과 한국미생물보존센터에서 Acetobacter tropicalis KFCC 11476P로 균주번호를 부여받았다. GYP 액체배지에서 A. tropicalis 11476P 균주를 이용한 식초제조의 최적 조건을 찾기 위하여 초기 에탄올농도는 GYP 배지에서 30℃에서 10일간 배양하여 균의 생육과 산생산능을 조사한 결과 4%의 에탄올 농도, 초기산도는 초산을 첨가하지 않은 0%, 초기 glucose 농도는 2.5~3% 범위의 농도가 초산균의 생육과 산도 생산이 가장 높은 조건이었다. 초산생산의 최적화를 위해 중심합성계획법에 따라 각 독립변수를 초기 ethanol 함량, 초기 acetic acid 함량, 초기 glucose 농도로 요인변수들을 -2, -1, 0, 1, 2로서 5개로 부호화하여 반응표면분석을 수행한 결과 초기 ethanol 농도 4.67%, 초기 acetic acid 농도 0.03%, 초기 glucose 농도 2.35%이었으며 이때의 산도 값은 3.77%로 나타나 실험결과에 근접함을 알 수 있었다. 식초의 관능특성으로 본 연구에서 제조한 양파식초에 대한 선호도면에서는 시판식초 보다 낮지 않아 양파자체가 갖고 있는 이취는 초산배양에 의해 거의 존재하지 않음을 알 수 있었다. 양파식초의 기능성탐색연구의 일환으로 혈전용해능 및 항혈전기능 정도를 살펴본 결과 혈전용해능은 plasmin 활성의 약 10%에 해당하는 활성을 나타내었으며 항혈전기능은 양파즙은 항혈전기능이 없는 반면 0.1 mL 양파 식초 첨가군에서 APTT, TT, PT 항목이 최대치를 나타내었을 뿐만 아니라 FIB도 대단히 낮아 항 혈전효과가 대단히 큼을 알 수 있었다. 산업적으로 유용한 양파식초의 속성생산과 kinetic parameter의 도출을 위하여 회분 및 유가식배양을 심도 있게 시간간격으로 분석하면서 초산균배양 실험을 수행한 결과 초기 에탄올농도 4, 5, 6% 조건에서의 양파식초의 회분배양은 통기효과에 의해 2일이 초과되지 않는 배양 30, 34, 46시간 만에 에탄올이 모두 소비되었으며 이때의 산도는 각각 3.1, 3.8, 4.9%였다. 초기 에탄올의 농도가 5%와 6%로 증가할수록 비증식속도는 0.344, 0.191, 0.114 hr-1로 감소하였고, 초산생산성(P), 비초산생산성(Π)은 증가하는 것을 알 수 있었다. 유가식배양에서는 에탄올만으로는 산도의 증가가 어려웠으며, 단백질영양원인 yeast extract와 corn steep liquor가 추가로 필요함을 알 수 있었다. 유가식배양의 Feeding 방법에서 초기 에탄올농도 4%의 배양조건에서 에탄올이 고갈되는 시점인 배양 28시간에 기질로 양파즙과 에탄올을 7:3의 비율로 하여 peristaltic pump를 이용하여 시간당 8.3 mL의 속도로 15시간까지 continuous feeding 방법으로 유가식배양을 수행한 결과, 잔존 에탄올이 1.15%인 배양 45시간 만에 산도가 4.5%에 도달하였다. 배양 45시간부터 양파즙과 에탄올을 8:2의 비율로 공급한 결과 산도는 5%까지 도달하였고, 배양 72시간만에 산도가 5.72%까지 증가하여(잔존 에탄올 함량은 1.13~1.69%), step feeding 방법의 유가식 배양보다 배양기간이 단축되고 산도가 더욱 높아짐을 볼 수 있었다. 양파식초의 대량생산을 위한 필수적인 전단계로서 120 kg의 양파즙을 제조하여 300 L pilot scale 발효조에서 산소가 충분히 공급되는 조건에서 초기 에탄올 6%의 조건으로 초산균을 배양한 결과 배양 48시간에 에탄올은 고갈되었으며 이때의 산도는 4.6%로 측정되어 실험실규모(2 L)의 발효기 실험결과와 유사하였으며, 산업적인 생산가능기간인 10일의 1/5의 기간이 단축된 2일내에 산도 4%이상의 양파식초의 생산이 가능한 결과를 얻었다.
3. 기능성 양파발효음료 제조공정 개발
다양한 처리를 통한 양파껍질과 과육에서의 유용성분의 추출 수율을 확인하였고, 각 처리의 최적조건을 조합하여 유용성분을 추출한 결과 가장 높은 추출 수율을 얻을 수 있었다. 또한 제1세부과제와 제2세부과제 연구팀과의 공동 연구로 유산균과 초산균이 가장 잘 발효시킬 수 있는 최적 발효기를 제조하였다. 각 단위 공정을 거친 양파 착즙액의 점도, 산도, pH 등의 변화를 확인하였으며, flow rate에 따른 탈취 공정으로 탈취 효과를 확인하였다. 또한 양파 발효음료 및 초산음료에 대한 전체적인 공정도를 설계하였으며, 반응표면분석법을 이용하여 용매, 마이크로웨이브 및 초음파를 이용한 추출공정의 최적화 조건을 수립하였다. 양파 발효음료 및 식초음료를 산업화하기 연구로서, 추출의 scale-up 실험 결과 초기 용매의 온도가 추출 최적온도인 59℃로 투입되고 용매와 시료가 충분히 교반되면 산업화 규모에서도 유용성분의 추출함량이 유지될 수 있음을 확인하였고, 경제성 및 효율성을 고려한 탈취 공정의 선정으로 전체적인 제조공정을 재설계하였다. 또한 추출공정 및 발효공정의 중요한 요인인 온도를 제어하기 위해 에너지 수지식과 MATLAB program을 이용하여 온도 제어의 효과를 확인하였다.
4. 기능성 양파발효음료의 산업화 연구
최근 양파를 많이 섭취한 사람들의 경우 암, 심혈관계지환 등 노화관련 질환의 발생률이 현저히 떨어진다는 보고가 잇따르면서 건강기능성 소재로서 양파에 대한 소비자들이 인식이 매우 높아져 있다. 현재 양파의 국내 자급률은 100%에 이르기 때문에 양파를 소재로 한 다양한 기능성식품의 개발은 국내에서 생산되는 농산물을 이용한 건강식품시장의 활성화에 밑거름이 될 것이며, 국내외적으로 우리나라 농산물의 고부가가치화를 추구하는데 크게 기여할 것이다. 따라서 본 연구에서는 우수한 기능성 소재로 입증된 양파를 이용하여 음료의 제조공정 개발 및 최적화시키고자 하였으며, 발효 양파 착즙액에 사과즙, 당, 산, 올리고당 등을 첨가하여 유산 발효 양파음료 시제품을 제조하여 1차, 2차 및 3차 관능평가를 실시한 결과 최종적으로 가장 우수한 배합은 유산발효음료의 경우 양파유산발효액 10%, 고과당 10%, 올리고당 5%, 배 농축액 2.38%, 아카시아 벌꿀 2%, 배향 0.3%, quercetin 추출물 0.29%, 팽화홍삼농축액 0.02% 등이었고, 초산발효음료의 경우에는 양파식초 5%, 올리고당 14.94%, 고과당 4.7%, 레드자몽 농축액 3.3%, 복분자농축액 0.04% 등이었다. 저장시험 결과 개발된 양파발효음료는 냉장온도(4oC)에서 1개월 이상의 유통기한을 가질 수 있음을 확인하였다.
5. 양파발효음료의 기능성 검증
양파의 추출방법과 추출액의 살균·저장에 따른 기능성성분을 분석하였다. 양파 착즙액의 경우 100℃에서 열수 추출한 시료에서 기능성성분의 함량이 가장 높게 검출되었다. 양파 추출액의 살균처리는 기능성 성분의 함량에 영향을 미치지 않았으며, 저장기간 동안 총 phenol, flavonoid, quercetin 함량은 안정적으로 유지되는 것으로 확인되었다. 양파발효음료의 기능성을 검증하기 위하여 유산 및 초산 발효액에 대한 항산화, 항암, 항고혈압 활성을 조사하였다. 유산 발효액은 착즙액에 비하여 높은 전자공여능 활성을 나타내어, 유산 발효에 의해 항산화활성이 상승되는 것으로 확인되었다. 유산 발효에 의하여 폐암세포와 대장암세포에 대한 항암 활성이 증가되었으며, 0.5% 이상의 홍삼 추출액 첨가에 의한 유산 발효는 추가적으로 항암 활성을 증가시키는 것으로 확인되었다. 초산 발효액은 폐암세포와 대장암세포에 대하여 높은 암세포 증식억제 활성을 나타내었으며, 초산발효에 의해 항고혈압 활성이 현저히 증가되는 것으로 확인되었다. 양파발효제품의 기능성을 검증하기 위하여 유산 및 초산발효양파를 투여한 흰쥐에서 혈액과 생화학적 활성변화를 조사하였다. 유산발효양파와 초산발효양파의 섭취는 흰쥐의 혈중 포도당, 중성지방, 총콜레스테롤 농도를 저하시키는 반면, HDL-콜레스테롤 농도 및 항산화 효소의 활성을 증가시키는 것으로 확인되었다. 초산발효양파를 섭취한 흰쥐에서 혈중 포도당 저하와 함께 골격근 내 글리코겐 농도가 낮게 나타나, 인슐린 감수성 증가 효과의 가능성이 제시되었다. 그러나 유산 및 초산발효양파의 섭취와 운동의 상승효과는 관찰되지 않았다.

1. Development of Bio-functional Onion Beverages Fermented with Lactic Acid Bacteria
Lactic-acid bacteria isolated from various fruits and vegetables were screened in order to select appropriate fermentation starters for manufacturing functional fermented onion juice. From the initial screening test comprising 702 isolated LAB, 61 isolates were selected based on their acid production rate. From the second screening test, 16 isolates were selected based on their abilities of lactic acid fermentation and the isolate KC-007 was finally selected as a fermentation starter due to its high antioxidant activity such as electron-donating and nitrite-scavenging activities as well as its high antimicrobial activity against food pathogens. The isolate KC-007 was identified as Pediococcus pentosaceus based on its morphological and physiological characteristics, carbon-utilization pattern, and molecular genetic characteristics (GenBank accession No. EU569832). The optimal temperature, pH, and starter inoculation concentration required for growth of the isolated strain were 40℃, pH 4.0~6.0, and 2%(v/v), respectively. When the onion extracts were fermented with mixed cultures of P. pentosaceus KC-007 and B. bifidum ATCC 15696 with ration of 1:2, the highest acidity, viable cell numbers of starter strains, and SOD-like activity were exhibited.
When the onion extracts was supplemented with puffed red ginseng extract, viable cell number of lactic acid bacteria was the highest at 24 hr of fermentation in all examined concentrations of puffed red ginseng extract. The electron donating ability and nitrite scavenging ability were highest when red ginseng extract was added at the concentration of more than 1%(w/v). It was found that soaking the onion into 2.0%(w/v) citric acid solution for 24 hr was the best method for the reduction of off-flavor of onion, When the onion extract was fermented in large scale with 1 L or 40 L, the optimal fermentation conditions were obtained as follows; pH of the onion extract was adjusted to 5.0, and mixed cultures of P. pentosaceus KC-007 and B. bifidum ATCC 15696 with ration of 1:2 were inoculated in the concentration of 2%(v/v) and then fermented at 37℃ for 24 hr. When the shelf-life of lactic acid fermented onion extract was examined at 5, 10, 20, or 30℃ for 18 days, there was no change in acidity at 5 and 10℃, and viable cell number of lactic acid bacteria was not noticeably decreased. Yeasts and fungi did not detected in the stored products.
Fermented onion extract was freeze dried to manufacture the powder product. From the above results the manufacturing process for the mass production of lactic acid fermented onion extract was set up as follows; onions are washed, cut into 8 pieces, and soak into 2%(%) citric acid solution for 24 hr followed by extraction. Extracts are autoclave at 121℃ for 10 min and centrifuge at 10,000×g for 30 min to remove insoluble materials. The pH of onion extract is adjusted to 5.0, autoclave at 121℃ for 15 min, and mixed cultures of P. pentosaceus KC-007 and B. bifidum ATCC 15696 with ration of 1:2 are inoculated in the concentration of 2%(v/v). Fermentation is carried out at 37℃ for 24 hr in anaerobic condition. Fermented broth is cooled down and formulate as follows with indicated concentrations; 10%(v/v) fermented broth, 2.38%(v/v) pear extract, 0.2%(v/v) citric acid, 0.02%(v/v) puffed red ginseng extract, 5%(v/v) oligosaccharide, 10%(v/v) high fructose syrup, 2%(v/v) acacia honey, 0.2%(w/v) vitamin C, 0.2%(w/v) pear flavor, 0.29%(w/v) quercetin extract, 69.61%(v/v) water. The formulated product is finally packed into glass bottle. The software system for the temperature control in the fermentation process was developed using Simulink in MATLAB program.
2. Development of Bio-functional Onion Vinegar Beverage
As a part of basic research for enhancing the availability of onions having short shelf-life and for the development of a commercial onion vinegar having functionalities of both of onions and vinegar, we isolated acetic acid bacteria having excellent acetic acid productivity from fallen peaches, and have found the optimum culture condition by using the properties of the response surface method (RSM) and conducted batch and fed-batch culture and compared with organoleptic properties through organic acid compositions of onion vinegar and investigated antithrombotic activity and kinetic parameter and scale-up culture of onion vinegar. In order to produce vinegar using onion, 12 acetic acid bacterium were selected and isolated from the juice of fallen peaches, among which a strain showing the highest productivity of acetic acid was identified as Acetobacter tropicalis 11476P. The cultural characteristics of A. tropicalis 11476P for maxmium acetic acid production were investigated in GYP medium and onion juice. The GYP medium containing 0% initial acidity, 4% ethanol and 2.5% glucose was the most suitable acetic acid production with A. tropicalis 11476P, and the acidity of culture medium was 4.02% after 10 days shaking cultivation of 30℃. Also optimum conditions for acetic acid production by RSM using the fractional factorial design with 3 variables and 5 levels were involved with initial ethanol content 4.67%, initial acidity 0.03% and initial glucose concentration 2.35% and predicted level of acetic acid production at these conditions was 3.77%. Onion vinegar with A. tropicalis 11476P after yeast and onion vinegar with only A. tropicalis 11476P have revealed higher sensory scores in overall tasted preference than the commercial vinegar in proportion to the ratio of the acetic acid to the total organic acid. The in vitro antithrombotic activities of the onion vinegar of 0.1% maximally prolonged the activated partial thromboplastin time (APTT) and prothrombin time (PT) compared with onion juice having no antithrombotic activity. On the results of fermentation properties of A. tropicalis 11476P varing the initial ethanol concentration to 4, 5, 6% on the fermentor with aeration (DO 80%), specific growth rate (μ) was decreased to 0.344, 0.191, 0.114 hr-1, respectively according to increase of initial ethanol concentration, while the acidity was increased to 3.1, 3.8. 4.9% within the fermentation time of 2 days. Fed batch fermentation at the time of exhaustion of residual ethanol was started and the effective feeding substrate to increase of acidity was additively need to protein source such as yeast extract, corn steep liquor except ethanol. Also, fed batch culture by carrying out the continuous substrate feeding showed the increased acidity than it by step substrate feeding. A acidity of onion vinegar by a continuous fed-batch culture can be produced 5.72% at fermentation period 72 hr when the ethanol content of onion juice is reduced to a range of 1.13 to 1.69%. The pilot scale fermentation in order to the industrial onion vinegar production was performed under the conditions that initial ethanol concentration of 6%. The acidity of pilot scale (120 L) onion vinegar at fermentation period 48 hr exhausted the residual ethanol reached to about 4.6%. We come to the conclusion that a industrial brewing vinegar of total acid 4~29% can be produced within 2 days by inoculating and fermentation our identified acetic acid bacteria at onion juice having the initial 6% ethanol.
3. Development of Manufacturing Process of Bio-functional Fermented Onion Beverages
The yield of quercetin and quercetin glycosides from onion edible part and skin part was investigated under various conditions which included solvent types (methanol, ethanol, water and combination of the solvents), time (15-60 min), temperature (30-60℃), ratio of sample over solvent, grinding methods, sample amount, drying methods (freeze dry and hot air dry), microwave, ultrasound and enzyme (cellulase and viscozyme) treatments. For solvent extraction, the maximum yield of quercetin was achieved with 30 mL/g of 80% methanol at 60℃ for 15 min. The methanol concentration and the extraction time showed no effect on the extraction yield. As extraction temperature increased, the extraction yield increased. Enzyme treatment increased the extraction yield about 1.5 times more than the control. 60 sec of microwave treatment improved quercetin extraction above twice more than the control. 30 min of ultrasound treatment showed 7% of improvement on quercetin extraction. The maximum yield was 17.76±1.29 mg/g from onion skin using combination of optimum conditions in various treatment. Also optimum medium for microbes was developed. Onion fermentation process was designed and optimized as measurement of viable cell count, pH, acidity, reducing sugar, nitrite scavenging ability and superoxide dismutase. Viable cell count increased to 5.3×108 from 3.1×104, pH decreased to 3.80±0.04 from 5.76±0.03, acidity increased to 2.31±0.01% from 0.52±0.03%, reducing sugar decreased to 81.41±3.97 from 101.19±8.23 μmol, electron donating ability increased to 77.56% from 25.53%, nitrite scavenging ability in pH 3 increased to 76.75±4.94% from 27.38±3.22%, that in pH 4.2 increased to 74.68±1.13 from 25.47±0.65, in pH 6 increased to 73.60±3.24% from 25.10±0.58 and superoxide dismutase was similar to control after 48 hr fermentation. Deodorization process was designed through flow rate. 30 mL/min of flow rate was highly deodorized from sensory evaluation. The microwave assisted extraction (MAE) and ultrasonic assisted extraction (UAE) of quercetin from onion skin were optimized and compared to conventional solvent extraction (CSE) by response surface methodology (RSM). A central composite design was employed and the response variable was yield of quercetin. The obtained models produced reasonable fittings of the experimental data on yield of quercetin (R2=0.937, 0.988 and 0.809 for CSE, MAE and UAE, respectively).
The optimum conditions were achieved with 55.6% ethanol at 59.3℃ for 13 min for CSE; 64.5% ethanol for 148 sec of microwave treatment for MAE and 45.7% ethanol for 20.7 min of ultrasonic treatment with power of 721.8 W for UAE. Under the optimum conditions, the highest yields of quercetin were 7.13, 8.99, and 7.41 mg/g for CSE, MAE and UAE, respectively. These results implied that MAE was more efficient and faster extraction method than CSE and UAE. Based on the first and the second detail project results, overall process of fermented onion beverage and fermented onion vinegar were redesigned. Considering economics efficiency, 2%(v/v) citric acid was suitable as deodorization process compared to using activated carbon.
To control temperature at 59℃ and 37℃ for extraction and fermentation, the heat transfer model for steam jacket was developed. Then the control system was simulated using MATLAB program. And the simulation showed a proportional control was appropriate for the control purpose. The specifications for the hardware system for the control system were determined according to the production size. The yields of quercetin and its glycosides from onion skin in lab-scale and pilot-scale under the optimal condition acquired for lab-scale (59℃, 13 min, 56% ethanol) were compared to each other. The yields were 8.35±0.07 mg/g and 7.94±8.68 mg/g for lab-scale and pilot-scale, respectively. It showed that the optimum condition can be applied to industrial scale to extract the maximum quercetin contents from onion skin if there were good agitation and proper temperature control.
4. Industrial Application of Bio-functional Fermented Onion Beverages
Fermented functional onion juice was developed using lactic acid bacteria and acetic acid bacteria as fermentation starters of onion. From the preliminary studies, we selected Pediococcus pentosaceus and Acetobater tropiclis as fermentation starters. The optimum recipe of functional lactic acid fermented onion juice based on manufacturing process and sensory evaluation were determined as 10% of fermentation liquer, 10% of fructose, 5% of oligosaccharide, 2.38% of pear concentrate, 2% of honey, 0.3% of pear flavor, 0.29% of quercetin extract, 0.02% of ginseng extract, etc. In the case of acetic acid fermented onion juice, the optimum recipe were determined as 5% of onion vinegar, 14.94% of oligosaccharide, 4.7% of fructose, 3.3% of grapefruit concentrate, 0.04% of Korean bramble concentrate, etc. Preservation tests showed that the shelf life of fermented onion juices could be greater than 1 month at 4℃ of storage temperature.
5. Verification of functionality of Fermented Onion Beverages
An investigation was carried out to study the effects of extraction, heat treatment, and storage temperature on functional ingredients of onion. The highest levels of functional ingredients were observed in samples extracted using water at 100℃. Heat treatment of onion extracts did not effect on the levels of all functional ingredients tested. During the storage, the levels of total phenol, flavonoid, and quercetin were not significantly changed regardless of storage temperature. In order to evaluate the biofunctional effects of lactic acid or acetic acid fermented onions, antioxidant, anticancer, and antihypertensive activities were determined. DPPH radical scavenging activity of lactic acid fermented onion juice was greater than that of control onion juice. The fermented onion juice showed strong anti-proliferative effects against all cancer cells tested. The anticancer activity of the fermented onion juice was further increased when it was supplemented with puffed red ginseng extracts at the concentration of over 0.5%(w/v). Onion vinegar was found to have stronger growth inhibition against all cancer cells tested as well as greater ACE inhibitory activities, compared to control onion juice. In order to evaluate the physiological functionalities of the fermented onions, changes of blood components and biochemical activities were determined in Sprague Dawley rats. The fermented onion diets resulted in decreasing the levels of glucose, triglyceride, and total cholesterol in blood, but increasing HDL-cholesterol levels and superoxide dismutase activities. Especially, lactic acid fermented onion diet reduced the serum glucose levels with significant decrease in glycogen levels in muscle, suggesting that the fermented onion may have a capacity to increase insulin sensitivity. However, the enhancement of exercise performance was not observed in rats supplemented with the fermented onions.
