○ 연구결과
xanthophyll을 다량으로 함유하고 있는 소재를 찾기 위해 국내산 작물과 미세조류로부터 소재 탐색을 수행하였다. 특히 루테인과 함께 황반 내에 다량 분포하는 것으로 알려진 제아잔틴 분석 결과 국내산 고추, 파프리카, 구기자 등에 많은 양의 제아잔틴이 있었음을 알 수 있었다. 해수클로렐라의 한 종인 Chlorella ellipsoidea는 고추나 파프리카에 비해 10-100배 이상 많은 제아잔틴을 함유하고 있었다. 또한, 고추의 제아잔틴이 소화흡수력이 떨어지는 것으로 알려진 zeaxanthin diesters 위주로 구성되어있는 반면 해수클로렐라는 흡수가 잘 되는 것으로 알려진 lipid free 형태의 제아잔틴이 주성분으로 존재하기 때문에 품질 면에서 우수할 것으로 기대된다.
루테인이 풍부한 미세조류 종균을 얻기 위하여 루테인 및 아스타잔틴 고 함량 균주를 선별하는 실험을 수행하였다. 25 종 이상의 미세조류 중 총 5 종의 클로렐라를 선별하였으나 기존 제품 생산에 이용하는 클로렐라 종균에 비해 생장속도와 바이오매스 함량이 낮아 실용화가 어려웠다. 이에 xanthophyll 함량 증대를 위해 인위적인 돌연변이원 처리를 통한 우수 종자 선별 실험을 수행하였고, 클로렐라로부터 카로티노이드 생산성을 증대시키기 위해 최적배양방법을 개발하는 연구를 수행하였다. 반응요인설계를 통한 클로렐라 배양배지조건 설정 실험에서 EDTA (g), MgSO4 (g), A5(mL)이 중요한 영양인자로 확인되었으며 최적배지 내에 1 : 8 : 12로 구성되었을 때 가장 높은 카로티노이드 생산성을 나타내었다.
클로렐라의 생이용성을 증진시키기 위해 다양한 물리적가공처리를 수행하였고 고전압 펄스 처리, 초고압 처리, 습식분쇄를 이용하였다. 클로렐라의 추출 및 생이용성 증대에는 습식분쇄가 가장 효과적인 것으로 판단되어 습식분쇄에 대한 최적화 실험을 수행하였다. 습식 분쇄는 고압균질기인 microfluidizer를 사용하여 이루어졌고 클로렐라에 적용 시 압력, 반복 횟수를 증가시킴에 따라 클로렐라 입자 크기가 작아져 분쇄효율이 늘어남을 확인할 수 있었다. 공정 조건을 최적화하기 위한 실험에서 클로렐라 용액의 희석 배수와 처리 압력, 통과 횟수가 중요한 영향요인이었다. 클로렐라의 분쇄 시 농축 배양액을 5-10배 희석한 후 80 MPa 이상의 압력으로 5회 이상 처리하는 조건이 클로렐라 입자 저감화에 효과적이었다.
클로렐라 루테인의 생이용성을 연구하기 위해 위-장-관 모사 시스템을 구축하여 분석하였다. microfluidization을 거친 시료의 경우 아무런 가공처리를 하지 않은 시료에 비해 체내 흡수가능하다고 알려진 micelles이 소화 과정 시 보다 효율적으로 생성됨을 확인할 수 있었다. Caco-2 세포수송 분석 시스템을 이용한 연구에서 분쇄처리를 한 클로렐라가 실제로 장내 세포에 더 높은 효율로 수송되었다. 위-장-관 모사 시스템의 효율성, 재현성을 검증하기 위하여 동물실험을 수행하였다. 쥐에게 생이용성이 증가된 시료를 투여했을 때 일반 시료를 투여했을 때보다 3배 이상 높은 농도의 루테인이 쥐의 혈장에서 분석되었다. 또한 쥐의 간, 눈 조직으로부터 루테인 분석을 실시했을 때 높은 농도의 분쇄 클로렐라를 투여한 그룹일수록 조직 내 루테인 함량도 높은 것으로 측정되었다. 특히 동물실험 결과와 위-장-관 모사 시스템 결과를 비교하였을 때 일치하는 부분이 많아 본 과제에서 구축한 위-장-관 모사 시스템이 클로렐라로부터 루테인 생이용성을 분석하는데 적합한 모델임을 확인할수 있었다.
클로렐라 내 xanthophyll 이 안구세포에서 갖는 기능성을 분석하기 위해 xanthophyll이 다량 함유되어있는 클로렐라 추출물을 망막신경절세포에 처리하여 생리활성을 탐색했다. 클로렐라 추출물은 산화스트레스를 받은 세포를 다시 회복시키고 활성산소 생성을 억제하는 효과가 있었으며 apoptotic 단백질의 발현을 농도 의존적으로 감소시켰다. 클로렐라 추출물이 안구 세포에 대해 갖는 기능성으로 유추해볼 때 생이용성이 증가된 클로렐라를 섭취 시에도 황반변성 억제 효과가 나타날 가능성이 높을 것으로 보인다.
나노화 클로렐라를 음료 제형으로 적용하기 위한 가공 적성 평가를 수행하였다. Suspension의 안정성은 10 배 희석과 80 MPa 이상의 압력에서 5 cycle 이상일 때 가장 안정하게 유지되었고 20일 이후에도 안정성이 유지됨을 확인하였다. 이러한 결과를 바탕으로 포스파티딜세린을 첨가한 기능성 클로렐라 시제품을 생산하였다.

Recently, the needs for the pharmaceuticals preventing eye disease are growing because of worldwide growing elderly population, ozone depletion caused by environmental pollution, and the frequent use of electronic equipments which continued to stress for the eye. It was reported that cataract and age-related macular degeneration, two typical eye diseases, were caused by a reduction of xanthophyll (lutein and zeaxanthin). As the content of xanthophyll present in the macula was decreased with aging, many people must intake lutein or zeaxanthin as food for the health of eye. According to these demands, lutein market as pharmaceuticals is growing significantly.
Chlorella is a rich source of lutein and there are many reports related in the extraction of lutein from chlorella. Though chlorella has abundant lutein, it is considered that the extraction efficiency of lutein and its bioavailability is very low due to the thick cell wall of chlorella. In this reason, physical processing improving the lutein bioaccessibility from chlorella is important for industrialization.
This research aims to develop functional food suppressing macular degeneration from plant resources composed of xanthophyll. To this edd, we are focused on developing technology which enables to increase to increase the lutein bioavailability from chlorella containing high levels of lutein.
To select the xanthophyll-rich materials, the extent of xanthophyll was analyzed using LC-MS from various plant and microalgae. Zeaxanthin was abundant in paprika and korean red pepper. Especially, Chlorella ellipsoidea, a kind of seawater microalgae, has 10 times higher level of zeaxanthin than paprika. In order to improve the lutein productivity from chlorella, culture method for lutein-rich chlorella was optimized with response surface design.
Physical processing using pulsed electric field generator, high hydrostatic press, and microfluidizer was carried out to overcome the limitation of chlorella in nutrition absorption and to increase lutein bioavailability of chlorella. Microfluidization was most effective technique among three processing technologies. The key factors of microfluidization to be determined were dilution level, treatment presure, and recycle number.
In order to evaluate the effect of physical processing on the lutein bioavailability of chlorella, in vitro digestion system was established and applied to the micro fluidized chlorella. The lutein micelles that are actually available to human body were more efficiently formed comparing with no treatment group. The result of Caco-2 cell transmembrane analysis showed that the celluar transport of lutein was approximately three times greater from microfluidezed chlorella than from untreated chlorella. Also, in vivo rat model was performed to assess the reliavbility of in vitro digestion system developed from this project. The level of lutein in the plasma, liver, and eye from rats to which the microfluidized chlorella was administrated had the conecntration-dependent manner, while no organs from rats to which the general chlorella was injected were analyzed to increase the lutein significantly. Inaddition, the protective activity of chlorella extracts mainly composed to lutein on retinal ganglion cells was carried out in order to evaluate the actual function of lutein originated from chlorella on the eye. Chlorella extracts had very effective in the inhibition of reactive oxygen and the decrease of the expression of apoptotic proteins on the eye. In these results, it was concluded that physical processing using microfluidization enabled to improve the bioaccessibility of lutein from chlorella.
For the industrialization of nano chlorella into beverage market, the physicochemical properties of microfluidized chlorella and its processing properties were studied. Finally, xanthophyll product, which has the bioavailable lutein, was developed with the addition of phosphatidyl serine.
Through our research, the application of physical processing on the chlorella was effectively increased the lutein bioavailability from chlorella and we developed the novel functional food containing the high level of xanthophyll.
