연구의 목적 및 내용:
본 연구의 최종 목표는 혈관신생 및 림프관신생과 관련된 질환 중 특히 망막혈관질환,조직재생, 암전이, 림프부종, 염증, 면역질환에서 VEGF-angiopoietin system의 상호관계 이상에 대한 병태생리학적 작용을 총체적이고 통합적으로 분석하고 이해하고자 함. 이를 위하여 VEGF-angiopoietin system을 target으로 제작된 유전자 변형동물들을 이용하여 상기질환 모델을 제작하고 현상기전을 규명하며 이에 상응하는 치료단백질을 질환 모델에서 시험하여 새로운 치료방법을 설정하고자 하였음.
연구의 목적 및 내용:

망막혈관질환에서 혈관 신생의 작용 기전과 역할 규명
1. 망막혈관질환 모델 확립 (ROP, LICN 모델)
2. ROP 또는 LICN 모델에서 관찰되는 vascular tufts 및 맥락막신생혈관의 생성과정을 시각화

림프부종, 염증 및 면역 질환의 진행과 해소에 작용하는 림프관신생의 작용 기전과 역할 규명
1. 림프관 림프절 형성에 있어 T 임파구와 인터페론의 역할 규명
2. 복막염 내 림프관 신생 및 기능에 있어 대식세포의 역할 규명
3. 조건부 림프관 사멸 동물 모델을 이용한 림프관의 기관특이적인 역할 및 기능 규명
연구결과의 활용계획:
1. 차세대 재조합 Ang1 단백질의 합성
- 기존의 COMP-Ang1 단백질의 약동학 및 효능을 강화시킨 새로운 Ang1 단백질의 제작
2. 혈관내피세포와 주위세포와의 관계 규명
- Ang1, Ang2, VEGF-A, PDGF-b의 다양한 상호작용에 기인한 혈관신생과정과 주위세포 결집의 기전 규명 규명
3. Tie2 신호를 활성화 시키는 암혈관신생 억제제의 개발
4. RhoJ 표적단백질 저해인자의 생성

The goal of this study is to understand interactions between VEGF system and angiopoietin system in angiogenesis and lymphangiogenesis, which are closely related to pathophysiology of retinal vascular diseases, tissue regeneration, cancer metastasis, lymphedema, and inflammati
Purpose & contents:
The goal of this study is to understand interactions between VEGF system and angiopoietin system in angiogenesis and lymphangiogenesis, which are closely related to pathophysiology of retinal vascular diseases, tissue regeneration, cancer metastasis, lymphedema, and inflammation/immune diseases. To elucidate these underlying mechanism, our research team will use genetically modified mice, which are targeted VEGF-angiopoietin system. This study will provide important mechanical insights for how to generate novel therapeutic methods for the angiogenesis- and lymphangiogenesis-dependent diseases.
Result:
Investigation of fundamental mechanism in angiogenesis and lymphangiogenesis
1. Generation of the genetically modified mice
2. Synthesis of recombinant proteins including VEGF-Trap, DAAP, VEGF-C, soluble VEGFR-3, sTie2 and COMP-Ang1
Role of angiogenesis in retinal vascular disease
1. Establishment of retinal vascular disease model including ROP and LICN
2. Visualization of vascular tuft formation and choroidal neovascularization in ROP and LICN model
