연구의 목적 및 내용
신경조율(Neuromodulation)은 손상된 뇌신경 기능이 다시 활성화되도록 외부 자극을 가하여 조율하는 신경재활 기술이다. 최근 신경재활의 효용이 전기적 치료법인 경두개 전기자극법(Transcranial Direct Current Stimulation), 자기적 치료법인 경두개 자기자극법(Transcranial Magnetic Stimulation), 침습적 전극을 통하여 직접 전기자극을 가하는 심뇌자극법(Deep Brain Stimulation) 등의 다양한 방식으로 입증되고 있다. 하지만 이러한
연구의 목적 및 내용
신경조율(Neuromodulation)은 손상된 뇌신경 기능이 다시 활성화되도록 외부 자극을 가하여 조율하는 신경재활 기술이다. 최근 신경재활의 효용이 전기적 치료법인 경두개 전기자극법(Transcranial Direct Current Stimulation), 자기적 치료법인 경두개 자기자극법(Transcranial Magnetic Stimulation), 침습적 전극을 통하여 직접 전기자극을 가하는 심뇌자극법(Deep Brain Stimulation) 등의 다양한 방식으로 입증되고 있다. 하지만 이러한 방법들은 직접적인 전류주입, 자극의 낮은 집중도, 침습적 방식으로 인해 사용의 제약이 따르는 한계가 있다. 초음파를 이용한 신경자극은 이러한 단점을 극복하면서 동시에 고집중도의 신경자극이 가능하므로, 본 과제에서는 초음파를 이용한 신경조율 기술을 개발하고, 효과의 기전에 대해 연구하고자 한다. 이를 위해 초음파 자극에 의해 유발되는 신경신호를 전기·광학적으로 동시 측정할 수 있는 다채널 미세전극 어레이(Microelectrode Array, MEA)를 개발하고, in vivo 신경신호 검출에 응용한다.
연구결과
[1세부] 저주파/저강도 초음파 자극 뇌 신경 조율 기술 개발
• 신경세포(neuronal cell) 조율용 저주파/저강도 초음파 자극 시스템 개발: 신경 세포 자극용 단채널 초음파 자극 시스템을 구현하고, MEA 시스템에서 배양된 쥐 해마 신경 세포에 저강도/저주파 초음파 자극 시의 신경 세포의 활동성이 증가하는 연구 결과를 도출하여, 초음파 신경 자극시스템의 신경조율 성능의 유효성을 확인하였음.
• 신경조직(brain tissue) 조율용 초음파 자극 시스템 개발: 뇌 해마 조직에 저강도/저주파 초음파 자극 시, 해마 조직의 다른 뇌 영역에서 (DG, CA3, CA2, CA1) 서로 다른 신경 조율 활동성을 검출하여, 구현한 초음파 뇌 조직 자극 시스템의 신경 조율 성능의 유효성을 확인하였음.
• Realistic 유한요소 뇌 머리 모델을 생성하고, 유한요소법을 이용하여 초음파 자극 현상의 시뮬레이션 및 자극 해석 기술을 개발하여, 효율적인 초음파 신경 자극 시스템 개발의 기초 기술 확보.
[2세부] 초음파 신경조율의 전기ㆍ광학적 동시측정을 위한 MEA 기반기술 개발
• 초음파신경조율을 위한 다채널 전극어레이 시스템(multi-electrode array, MEA)을 구축하였으며, 전기적, 광학적 동시측정을 위한 기반기술을 개발하였음.
• 초음파가 신경조율을 나타내는 기전을 밝히기 위해 온도변화에 따른 신경세포의 변화를 측정하고 온도감지 수용체가 온도변화에 의해 신경신호에 미치는 영향을 분석하였음.
• 광학적 신경신호의 측정을 위해 칼슘인디케이터와 표면플라즈몬 현상에 의한 반응을 in vitro, in vivo에서 측정하여 초음파에 의한 신경조율 연구를 수행하였음.
[3세부] 고감도 플라즈모닉 센서를 이용한 신경신호의 광학적 측정 기술 개발
• 유전체 나노격자 구조를 사용하여 고감도 광학 센서를 제작하고 이를 바탕으로 구현된 플라즈모닉 파장분리기를 기반으로 한 실시간 광학 이미징 원천 기술을 개발하였음.
• 기존 미세전극 어레이 구조를 변형하여 전기광학적 동시측정이 가능한 다채널 전극 기판을 개발하고, 고감도 특성의 비표지 광학 측정기법을 신경조율에 적용하였음.
• 금속 나노입자와 적외선 광을 이용하여 쥐의 좌골 신경세포를 광학적으로 자극하고 신경신호를 검출하는 연구를 통하여 in vivo 환경에서의 신경조율 연구를 수행하였음.
연구결과의 활용계획
• 본 연구의 새로운 MEA 시스템을 활용하여 초음파, 광학, 복합자극, 전기자극 등의 다양한 방법을 활용한 신경조절 현상을 중/장기적으로 실험을 수행하고 특성 및 기전을 분석하는데 활용함
• 개발한 초음파 신경조율 방법과 다양한 신경반응의 측정 방법은, 신경신호전달 경로를 분석하여 다양한 뇌신경과학 및 뇌공학 분야 연구에서 활용할 수 있음.
• 기존 전기적 신경조율 방법의 한계를 극복할 수 있는 비침습적 초음파 신경조율 시스템 개발을 통하여 각종 신경치료 및 신경재생 분야의 연구 및 개발의 활성화에 기여함.
• 본 연구에서 확보한 기초/원천 기술을 통하여 초음파, 광, 자기장 등 다양한 파동에너지가 결합된 다중모드 신경조율 시스템을 개발할 수 있음. 또한 본 연구에서 확보한 기초 기술로 고가의 기존 형광 현미경을 대체할 수 있는 새로운 개념의 생체 이미징 시스템 개발함.
• 세포배양기술, 표면공학기술, 나노/마이크로 스케일 공정기술, 생체신호 측정기술, 전자회로 설계 기술을 접목함으로써 전자공학, 신경생리학, 생체의공학 분야의 학제 간 연구 활성화함.

Neuromodulation techniques via electrical (i.e., tDCS (transcranial direct current stimulation) and DBS (deep brain stimulation)) magnetic (i.e., TMS (transcranial magnetic stimulation)) stimulation have gained widespread attention. However, ultrasound (US) neuromodulation has been sparse, although it offers several advantages over the conventional techniques (i.e., non-invasiveness, higher penetration and focus.) We propose to develop US neuromodulation techniques and validate them using cells, brain slices, and small animal brains via electrical and optical measurements of neural activities using newly developed multi-electrode arrays (MEAs) which are highly sensitive and US compatible.
