Ⅳ. 연구개발결과
1. 제1협동과제: 국화의 실용화 가능 유전자 개발 및 기능 검정
애기장대 유래의 화형 및 생장조절 유전자 ARF18 유전자에 대한 상세기능 분석을 위하여 애기장대 형질전환체를 제작하였다. 형질전환체에서 세포 크기 및 수에 관한 해석을 통해 AtARF18이 세포분열 촉진과 기관 크기 조절을 할 수 있다는 사실을 확인하였다. AtARF18 유전자의 식물 기관크기 조절 기작을 상세히 밝히기 위하여 기존에 알려진 세포의 수 및 크기 조절에 관여하는 조절 유전자들, Angustifolia3(AN3)와 Aintegumenta(ANT)의 발현양상 분석과 an3-4 돌연변이체와 ANT 과발현 형질전환체의 유전학적 분석을 통하여 상호 관련성을 조사한 결과, AN3 유전자가 AtARF18 유전자의 하류에서 기관의 크기를 조절하며, AtARF18 유전자와 ANT 유전자와는 서로 독립적으로 기관의 크기조절에 관여하고 있다는 사실을 처음으로 규명하였다. 야생형과 형질전환 국화의 꽃에서 가장 가장자리의 꽃잎을 채취하여 꽃잎의 크기와 모양, 꽃잎 윗면의 세포의 크기와 수를 통계적으로 분석하여 AtARF18 과발현 형질전환 국화의 꽃잎에서 꽃잎의 세포 크기는 120%, 꽃잎 한 장의 전체 면적에서 세포 수가 116% 증가하여 꽃잎의 면적이 135% 증가한 것을 확인하였다. AtARF18 과발현 형질전환 국화 중에서 T-DNA영역이 single copy로 삽입되었다고 확인된 AtARF18 과발현 형질전환 국화에서 RT-PCR을 이용하여 AtARF18 유전자 및 세포 증식 관련 분자 마커인 국화 ANT 유전자 프라이머를 이용하여 발현 양상 분석 결과 발현량이 증가하였음을 확인하였다. 이로 보아 AtARF18 과발현에 의해 세포 증식 조절 인자의 발현량 증대와 세포 증식이 증대되어 국화 꽃잎의 크기가 증대된 것이라고 판단된다.
국화 total RNA를 이용한 RT-PCR 방법과 국화 cDNA library로부터 PCR방법을 사용하여 국화 유래의 개화조절 인자인 CmFT와 CmSOC1을 클로닝하고, 국화의 조기개화 형질전환체 구축을 위해 과발현 벡터를 구축, 애기장대에 도입하였다. 형질전환체에서 CmFT와 CmSOC1 발현량을 RT-PCR로 분석한 결과, 형질전환체는 야생형에 비해 CmFT와 CmSOC1이 과발현 되었음을 확인할 수 있었다. CmFT 과발현 형질전환 애기장대는 단일 조건에서 야생형 보다 파종 후 최대 30일 일찍 개화하였다. CmSOC1 과발현 형질전환 애기장대는 비록 장일에서 꽃대 발생, 개화 시기가 야생형과 큰 차이를 보이지 않았지만, 단일조건에서 10일 이상 빨리, 로제트 잎의 수가 10장 이상 적은 시기에서 꽃대가 발생, 개화하였다. 이로써 국화의 CmFT와 CmSOC1가 개화조절 기능을 가지고 있으며 애기장대의 조기개화를 유도할 수 있음을 확인하였다.
식물은 저/고산소증, 고염분, 가뭄, 빛, 추위, 병원균과 비생체물질 등의 오염원, 노화 등에서 의해 산화스트레스를 겪으며 식물의 산화스트레스는 식물의 생장과 형태형성, 생산성에 영향을 미친다. 애기장대의 NDPK2는 광형태형성, 옥신조절경로, 산화 및 염 스트레스 신호경로에 관여한다. 생장촉진 효과와 산화스트레스를 비롯한 환경스트레스에 대한 저항성을 높일 수 있는 NDPK2 형질전환 국화를 개발하기 위해 애기장대의 NDPK2 유전자를 클로닝하였다. 그리고 국화 최적의 과발현을 위해 CaMV 35S 및 Super 프로모터 외에, 그리고 환경스트레스에 특이적으로 반응하는 유전자인 고구마의 SWPA2 유전자의 프로모터에 각각 삽입한 형질전환 벡터를 제작하였다.
2. 제1세부과제: 유용 유전자 도입 국화 형질전환체 대량 생산
국화 유용 유전자 도입 형질전환체를 생산하기 위하여 국내 스탠다드 육성 신품종인 ‘백마’의 기관형성을 통한 식물체 재생 시스템을 확립하였다. 기내 배양된 잎 절편으로부터 신초 기관형성을 유도하기 위한 식물생장조절제와 AgNO3를 포함한 배지조성 및 배양기간의 적정 조건을 확인하여 효율적인 식물체 재생 체계를 확립하였다.
국화의 국내 주요 품종인 ‘신마’의 Agrobacterium 매개 형질전환에서 bar 유전자를 선발마커로 이용하기 위하여, 형질전환된 절편체를 PPT가 포함된 선발배지에서 배양하여 성공적으로 형질전환 식물체를 생산할 수 있는 형질전환 시스템을 구축하였다.
잎 절편체를 Agrobacterium tumefaciens와 공조배양한 후, PPT 1.5mg/L, Cx 500mg/L가 첨가된 선발배지(신초유도배지; TSM)에서 배양하였다. 이후 PPT 2.0mg/L, Cx 500mg/L가 포함된 선발 배지(신초신장배지; TEM1)로 옮겨 배양하였으며, 정상적으로 뿌리가 형성된 형질전환체를 절단하여 PPT 2.0mg/L가 포함된 선발배지(TEM2)로 옮겨 배양하면서 Agrobacterium이 감염되어 있지 않고 신장과 발근이 이루어진 형질전환체를 육성하였다.
화형 및 생장조절 유전자(AtSAP, AtARF), 국화 개화시기 조절(조기개화) 유전자(CmFT, CmSOC1)와 환경스트레스 내성 유전자(AtNDPK2)를 국화 ‘신마’로 도입하여 최종적으로 형질전환 218계통을 선발하였고 형질전환 효율은 1.9%로 나타났다. PCR과 qPCR을 통해 유전자 도입여부와 T-DNA 1-2 copy 형질전환체를 확인하여, 표현형 검정 및 생리학적 기능 분석을 실시하였다. 일부 형질전환 계통에서 잎의 변화, 화형 변화, 개화소요일수 단축, 초장 증가 및 환경스트레스 내성을 갖는 것으로 나타났다. 개화조절 CmFT 유전자 도입 국화 형질전환 계통의 일부는 대조구와 표현형 차이 없이 개화가 5일 빨라진 것으로 나타났다. 환경스트레스 내성 유전자 AtNDPK2 도입 국화 형질전환체의 경우 대조구에 비해 내건성과 내한성 등 다양한 비생물적 스트레스 내성을 가진 형질전환체를 획득하였다.
3. 제2협동과제: 고효율 국화 형질전환용 벡터 개발 및 형질전환체 검증
CaMV 35S, super, sAP1 프로모터의 국화 및 모델식물체에서의 발현양상 연구를 위해서 프로모터 GUS construct를 각각 제작하고 애기장대와 국화 형질전환체를 제작하였다. Genomic PCR을 통해서 DNA 수준에서 형질전환체임을 확인하였다. 애기장대 형질전환체에서는 GUS 염색을 통해서 sAP1 프로모터가 CaMV 35S 프로모터와 달리 잎에서의 발현은 우수하나 뿌리에서의 발현은 매우 약한 발현 특징을 확인하였다. 이러한 결과를 통해 애기장대 유래의 sAP1 프로모터가 국화 잎에서의 외래 유전자의 발현에 이용될 수 있는 새로운 프로모터임을 알 수 있었고 향후 제작된 다수의 형질전환체의 뿌리, 줄기, 꽃에서의 발현량을 조사함으로써 그 이용 범위를 명확히 할 수 있을 것으로 생각된다.
선발된 국화 형질전환체들의 잎 형태, 화형 및 개화시간 등의 화훼학적 중요 표현형의 변이정도를 야생형 신마와 비교 조사하고 표현형의 다양한 변화와 삽입된 transgene의 발현량과의 상관성을 조사하기 위해서 RT-PCR과 qRT-PCR를 수행하여 도입된 유전자들의 발현 정도를 정량적으로 조사하였다. 특히 중요 표현형 조사대상이 잎과 꽃이므로 RT-PCR에 사용되는 RNA는 장일조건에서 배양되는 영양생장기의 국화 잎과 단일 조건에서 개화가 유도된 생식생장기의 국화 꽃봉오리에서 각각 분리하여 사용하였다. 또한 각각의 도입 유전자 특이적 Forward primer, 사용한 벡터의 3‘UTR 특이적 reverse primer set와 국화의 모든 조직에서 항상 일정하게 발현하는 actin 유전자 특이적인 primer set를 control로 함께 사용하여 새롭게 도입된 유전자들의 발현 정도를 정량적으로 조사하여 발현양상을 관찰하였다.
AtSAP1, AtARF18, AtNDPK2와 CmFT 유전자가 발현하는 국화 형질전환체들의 삽입된 T-DNA 수를 southern blot 분석을 통해서 조사하였으며, Event화를 하기 위해서 선발된 single T-DNA가삽입된 국화 형질전환체들은 향후 GM 안전성 평가에 필요한 T-DNA가 삽입된 국화 chromosome에 대한 정보를 얻기 위해 삽입된 T-DNA의 flanking sequence를 확보하고자 하였다. 이를 위해서 이미 확립한 TAIL-PCR 방법으로 증폭하고 염기서열을 규명하였다. 확인된 T-DNA 염기서열에서 벡터부분과 left 또는 right border sequence를 확인함으로써 삽입된 T-DNA상태를 조사하였고 확인된 T-DNA 염기서열 중에서 border sequence 다음의 염기서열과 유사성이 있는 염기서열을 NCBI Blast search를 통해서 조사하여 T-DNA 삽입부위가 gene coding 영역인지 또는 intergenic 영역인지 구분하였다.
4. 제3협동과제: 고부가가치 장미 형질전환
형질전환 재료의 획득 및 선발을 위해 장미 ‘Sweet Yellow' 1품종 및 KR056002 등 2계통 유래 배발생캘러스를 3-6주 간격으로 품종 또는 계통별로 총 2,180 paltes를 계대배양 하였으며, 증식과정에서 분화된 배와 배 주변의 배발생캘러스를 선발하여 아그로박테리움 공동배양법을 이용한 유전자 도입을 위한 시료로 사용하였다.
장미 형질전환을 실시하시 위해 내환경성 유전자 SOD2삽입 고유 벡터인 pPZP200-bar를 제작 완료한 후 Agrobacterium tumefaciens LBA4404 competent cell에 형질전환시켜 SOD2 유전자의 삽입을 확인하였고, GM작물실용화사업단으로부터 pPZP200-Bar 벡터내 삽입 상태로 분양받은 GPD 유전자와 함께 2011년 및 2012년도에 아그로박테리움과의 공동배양을 통해 'Sweet Yellow' 1품종과 KR056002 등 2계통 유래의 배(배발생 캘러스 포함)로 도입하였으며, 신초를 재분화 시키고자 배발생, 배발아, 신초유도, 신초생장, 다신초유도 등 생육단계에 따라 선발배지를 달리하여 총 1,441 plate를 교체하였다. SOD2 유전자 도입 후 재분화 선발배지에서 획득된 식물체 41개체(KR056006유래 9개체 및 KR056002유래 32개체)를 대상으로 PCR분석을 이용하여 SOD2 유전자가 도입되었음을 확인하였다. 서던분석에 의해 SOD2 형질전환체 19개체에 목표유전자 SOD2가 1-4 copy 도입되었음을 확인하였고, RT-PCR 분석에 의해 형질전환체 4개체 모두 도입 목표유전자 SOD2의 전사체가 정상적으로 발현함을 확인하였으며, western 분석에의해 SOD2 형질전환체 6개체 모두 목표유전자 SOD2의 단백질이 정상적으로 발현함을 확인하였다. Nested PCR을 이용한 도입유전자 주변 염기서열 분석에 의해 SOD2 형질전환체의 도입 유전자 주변에 장미 genome 염기서열로 추정되는 376-1,064bp의 염기서열을 확인하였으며, 이들 염기서열의 FASTVAL 검색 결과 현재까지 유사한 sequence 정보가 없음을 확인하였다. 150mM NaCl 첨가 장미 증식배지에 내환경성 증진 유전자 SOD2 형질전환체 8개체를 처리한 결과 형질전환체가 비형질전환체에 비하여 피해가 30% 덜 나타나 형질전환체의 내염성이 증진되었음을 확인하였다. 또한 내건성 및 내한성 검정 형질전환체가 비형질전환체에 비하여 내건성과 내한성이 증진되었음을 확인하였다.

During the last decade, various transgenic plants of chrysanthemum and rose has been produced by using an Agrobacterium-mediated transformation technique. However, the fairly low efficiency of transformation and lack of the valuable genes that can be used for molecular breeding of chrysanthemum and rose still remain to be solved.
As there is a significant commercial market for chrysanthemum and rose, commercial growers have been trying to produce flowers consistently throughout the year and to reduce the production costs. However, in terms of year-round production, unexpected challenges, such as premature flower budding, occasionally arise due to deviations from normal environmental conditions in greenhouses. Despite the considerable research efforts that have focused on gaining more complete understanding of the effects of the environment on growth and flowering in chrysanthemum and rose, very little is still known about the molecular and genetic mechanisms that control flower development.
Therefore we tried to develop the novel promoters and genes for securing patents. And then we produced the transgenic plants with the value for commercialization using plant genetic transformation technology of major floricultural crops. Eventually, on the basis of core technology such as the construction system for genetically modified plants, we will develop new varieties of major floricultural crops, chrysanthemums and rose with international competitive power.
1. We isolated Arabidopsis thaliana Auxin Response Factor 18(AtARF18) and Sterile Apetala(AtSAP) involved in regulation of plant growth and development and generated expression vector system with the CaMV 35S or Super promoter using pCAMBIA3300. We analyzed the alterations in flower development of AtARF18 overexpressing transgenic Arabidospis and chrysanthemum. In histological analyses of AtARF18 overexpressing Arabidopsis and chrysanthemum, transgenic plants showed increase in leaves and petals caused by increase in cell number and cell size and expression level of Aintegumenta(ANT), positively regulate cell proliferation in the ARF18-mediated cell proliferation pathway. These results indicate that AtARF18 enables to modify the size of shape of flowers by regulating cell proliferation in chrysanthemum flower.
Secondly, we isolated flowering timing regulators, Flowering Locus T(FT) and Suppressor of Overexpression of Constans 1(SOC1) from chrysanthemum ‘Jinba’, generated expression vector system for CmFT and CmSOC1 overexpression under the CaMV 35S or Super promoter using pCAMBIA3300, and introduced into Arabidopsis. In various organs and tissues of chrysanthemum, CmFT is strongly expressed in leaves and weakly in flower bud, but not detected in stem and petals. CmSOC1 is strongly expressed in leaves, stem, and flower bud and weakly in petals. CmFT or CmSOC1 overexpressing transgenic Arabidopsis showed the early flowering and increase in the expression level of Apetala 1(AP1). Especially, transgenic Arabidopsis showed striking early flowering in short-day condition, flowering at about 30 days after sowing and 10 rosette leaf number compared to wild type flowering at about 62 days after sowing and 20 rosette leaf number. CmFT overexpressing transgenic chrysanthemum also showed early flowering, forming flower buds and increase in the expression level of Leafy(LFY). These results indicate that chrysanthemum FT and SOC1 function as positive regulator of flowering. In addition, these results may suggest the differences of flowering regulation mechanism between long-day plants(represented with Arabidopsis) and short-day plants(represented with chrysanthemum).
Finally, we isolated Arabidopsis Nucleoside Diphosphate Kinase 2(NDPK2) to respond to abiotic/biotic stresses and generated expression vector system for AtNDPK2 overexpression under the CaMV 35S or Super promoter using pCAMBIA3300.
2. Through Agrobacterium tumefaciens-mediated transformation of chrysanthemum ‘Jinba’, plant development and growth gene(AtSAP, AtARF), flowering regulation gene(CmFT, CmSOC1) and abiotic stress tolerance gene(AtNDPK2) under the control of the CaMV 35S promoter, Super promoter or IbSWPA2 promoter were introduced into leaf explants of chrysanthemum via Agrobacterium tumefaciens LBA4404 mediated transformation. 218 independent transgenic plants were generated from the transformed explants via shoot organogenesis and rooting on selection medium supplemented with 1.5-2.0 mg·L-1 phosphinothricin(PPT) at a transformation efficiency of 1.9%. From transgenics, the integration of transgenes, AtSAP, AtARF, CmFT, CmSOC1, AtNDPK2 and bar, were confirmed using PCR and qPCR. Following acclimatization, the transgenic plants were cultivated in pots under long-day conditions. The transgenic plants reached a height of 30 cm after 5 weeks, following which they were subjected to short-day conditions to induce floral initiation and flowering.
Phenotypic characteristics of the AtSAP and AtARF transgenic plants were investigated following flowering. Transgenic plants showed an almost normal flowering time (similar to that of the WT), except for plant height, flower diameter, leaf and flower morphology which were increased in the transgenics. The transgenic lines harboring CmFT, like the WT plants, grew vegetatively under long-day conditions, but they flowered 5 days earlier than the WT plant under short-day conditions, which flowered 8 weeks after the short-day treatment. The AtNDPK2 transgenic lines generated were selected for evaluation of tolerance to various abiotic stresses. Transgenic plants demonstrated enhanced tolerance to low temperature and drought stresses at both the leaf and whole-plant levels.
Phenotypic examination of WT and transgenic plants that flowered under the short-day conditions revealed the increase of plant height and leaf and flower morphology in AtSAP and AtARF transgenics, early and perpetual flowering in CmFT transgenics, and the increase of abiotic stress tolerance in AtNDPK2 transgenics, compared to the WT plant.
3. We examined the phenotypic and molecular characteristics of the transgenic lines introduced four genes(AtSAP1, AtARF18, AtNDPK2 and CmFT) that previously identified as key regulatory genes relevant to flowering time and environmental stress. All transgenic plants were characterized by genomic PCR and RT-PCR analysis carried each report genes in their genome. RT-PCR analysis showed that introduced gene was normally expressed in transgenic lines. To investigated the number of the inserted T-DNA of chrysanthemum transgenic plants was studied through the Southern blot analysis. Among all the transgenic plants, we selected the transgenic lines with one copy of the T-DNA introduced into AtSAP1, AtARF18, AtNDPK2 and CmFT through a Southern blot analysis. Also, we used thermal asymmetric interlaced polymerase chain reaction (TAIL-PCR) to amplify unknown sequences adjacent to known insertion sites in transgenic plants. A flanking T-DNA sequencing method was used to determine that left or right border sequences flanked four genes in the transgenic plants. We expected that there are no similar sequence information from NCBI data base and T-DNA inserted into the intergenic region or the non-coding region.
In conclusion, we introduced the four genes into chrysanthemum and characterized transgenic plants. Through a fast and efficient analysis we selected 12 transgenic lines with a single copy T-DNA 38and obtained flanking sequences for each individual in the transgenic plants. 4. Somatic embryos (including embryogenic calluses) derived from in vitro roots of ‘Sweet Yellow' and two breeding lines (KR056002 and KR056006) bred by crossing between Rosa hybrida ‘Tineke’ and ‘Mirinae Gold’ were subcultured on fresh medium at 3- to 6-week intervals and used as materials(explants) to obtain transgenic plants resistant to abiotic stresses. Totally, we subcultured 2,180 times from May 1, 2011 to October 27, 2014.
pPZP200-bar vectors containing SOD2 or GPD gene were constructed. The GPD gene was obtained from The National Center GM Crops(NCGC). The SOD2 and GPD genes were introduced into the somatic embryos (including embryogenic calluses) derived from three genotypes, respectively, using Agrobacterium-mediated transformation method. And then we subcultured the explants into new fresh medium supplemented with both 250 mg·L-1 cefotaxime and 2 mg·L-1 PPT appropriate for each stage. There are stages such as embryo maturation, embryo germination, shoot primodia induction, shoot growth, multi-shooting, and rooting. Totally, we replaced 1,441 plates to obtain 41 putatively transgenic plants. Nine of 41 plants were from KR056006 and 32 KR056002. We confirmed the introduction of SOD2 gene into transgenic plants by PCR analysis, and reconfirmed 19 plants having 1 to 4 copies of SOD2 gene by Southern analysis. Also, we identified the normal transcript expression of the SOD2 gene in 4 plants by RT-PCR analysis and normal protein expression the SOD2 gene in 6 plants confirmed by Western analysis. We determined the sequences of 376 to 1,064 flanked the SOD2 gene in the transgenic lines using Nested PCR method. These sequences did not match any gene known by FASTVAL. Furthermore we identified the resistance to high salt, low water content, and low temperature in the SOD2-transgenic lines.
