1세부과제 : VLCFAs (Very Long Chain Fatty Acids) 생합성의 전사 조절 기작 연구
(1) VLCFAs 생합성을 조절하는 신규 전사 조절인자 발굴: KCR1 promoter:GUS식물체를 이용한 돌연변이체 제작 및 Yeast One Hybrid 시스템을 이용한 KCS2 promoter에상호작용하는 At1g65300 및 At4g31420 유전자를 선별하였다. 그리고 줄기 표피세포에서의transcriptome 분석 및 외부 환경 스트레스에 대한 애기장대 microarray data

1세부과제 : VLCFAs (Very Long Chain Fatty Acids) 생합성의 전사 조절 기작 연구
(1) VLCFAs 생합성을 조절하는 신규 전사 조절인자 발굴: KCR1 promoter:GUS식물체를 이용한 돌연변이체 제작 및 Yeast One Hybrid 시스템을 이용한 KCS2 promoter에상호작용하는 At1g65300 및 At4g31420 유전자를 선별하였다. 그리고 줄기 표피세포에서의transcriptome 분석 및 외부 환경 스트레스에 대한 애기장대 microarray database를 이용하여 확보한 34개의 신규 전사조절인자들 중에서 MYB94와 MYB74 신규 전사조절인자를선별하였다.
(2) 신규 전사 조절인자들의 구조, 발현 및 세포내 위치 분석: MYB94와 MYB74 신규 전사조절인자들이 C-terminal 부위에 전사활성화 도메인을 가지고 있음을 yeast에서 transactivation assay를 통해 확인하였다. 그리고 quantitative real-time RT-PCR 분석 및 선별된 유전자들의 프로모터하에 GUS 유전자가 발현되도록 제작한 형질전환 식물체들을 이용하여 식물 발달학적, 조직학적 및 스트레스별 유전자 발현 분석을 수행하였다. 그 결과,MYB94와 MYB74 유전자 모두 줄기보다 줄기 표피세포에서 4-5배 높게 발현되었으며,전반적으로 지상부 조직에서 고루 발현되고 있었다. 그리고 ABA, 가뭄, NaCl, mannitol을 처리한 조건에서 발현이 유도되고 있음을 확인하였다. 또한 형광단백질을 이용하여MYB94:eYFP와 MYB74:GFP가 핵으로 targeting 됨을 확인하였다.
(3) MYB94와 MYB74 전사조절인자의 식물체에서의 기능 분석: 야생형, 돌연변이,complementation 또는 과다 발현 식물체들을 이용하여 식물 발달학적, 생리학적, 세포학적,또는 생화학적 분석을 통해 식물체에서의 기능 분석을 수행하였다. myb94-ko의 잎에서는 야생형 대비 23% 감소된 cuticular wax 함량이 관찰되었으며, MYB94 OX 잎에서는 wax crystal의 과다축적과 cuticular wax 총 함량이 1.3에서 1.7배 증가되어 있음을 확인하였다.이러한 증가된 큐티클 왁스 함량으로 인하여 가뭄스트레스 조건에서 MYB94 과다발현 식물체잎에서 수분 손실이 야생형 보다 천천히 일어나고 있음을 관찰하였다. 그리고 myb74-ko돌연변이체의 잎에서 cuticular wax 성분 및 함량이 야생형과 큰 차이가 없는 결과와는 다르게, MYB74 OX 잎에서는 14% 증가된 cuticular wax 함량이 검출되었다. 이는 MYB94와 MYB74 모두 cuticular wax 함량을 조절할 수 있는 전사조절인자임을 의미한다.
(4) MYB94 전사 조절 유전자의 target 유전자 규명: 야생형과 MYB94 OX 잎에서 추출한 total RNA를 이용하여 microarray 분석을 수행한 결과 WSD1, KCS2/DAISY, CER1, CER2, AlcFAR3/CER4, ECR/CER10 유전자들의 발현량이 MYB94 OX 식물체에서 증가되어 있음을 확인하였다. 그리고 transcriptional activation assay 및 EMSA를 통해서 MYB94가 WSD1, KCS2, CER2, FAR3, 그리고 ECR 유전자 프로모터에 존재하는 MYB binding consensus sequence 부위에 직접 결합할 수 있음을 확인하였다. 이는 MYB94가 cuticular wax 생합성에 관여하는 유전자들 프로모터의 cis-element에 직접 결합함으로써 유전자의 발현을 조절할 수 있는 활성인자임을 의미한다.
(5) 가뭄스트레스하에서 MYB 전사 조절 유전자들의 상호연관성 규명: 야생형, 단일돌연변이 및 myb96 myb94 이중 돌연변이 식물체에 지속적인 가뭄 스트레스를 처리한 다음,가뭄 스트레스를 처리하지 않은 정상 조건 및 가뭄 스트레스 조건에서 cuticular wax 성분 및함량을 조사하였다. 정상조건과 가뭄스트레스 조건에서 각각 14%와 16% 감소된 cuticular wax 함량 감소가 관찰되었다. 이는 가뭄스트레스 조건에서 MYB94 유전자도 cuticular wax함량 증가에 관여하고 있음을 의미한다. 정상 조건 및 가뭄 스트레스 조건에서, myb96 myb94이중 돌연변이체는 야생형 대비 총 왁스 함량이 각각 44%와 58% 감소되어 있음을관찰하였다. 이는 단일 돌연변이체에서 감소된 비율의 합과 비슷하게 두 유전자가 정상 및가뭄 스트레스 조건에서 cuticular wax 생합성 및 축적을 조절하는데 additive한 기능을가지고 있음을 확인하였다. 이로 인하여 cuticular transpiration 및 chlorophyll이 침출되는속도가 myb94-ko myb96-ko 이중돌연변이체에서 야생형 및 단일 돌연변이체들 보다 빠른속도로 일어나고 있음을 관찰하였다. 그리고 myb96-ko 돌연변이체에서 관찰된 cuticular wax성분 변화 및 함량 감소는 MYB94 유전자의 발현에 의해서 야생형처럼 회복됨을 확인하였다.이는 MYB94와 MYB96이 cuticular wax 생합성을 위하여 combined function을 가지고 있음을 의미한다.
(6) 큐티클 왁스 함량 증가로 인한 가뭄에 저항성을 갖는 유지작물인 카멜리나 형질전환식물체 개발: 애기장대 MYB96 유전자가 과다발현된 카멜리나 형질전환 식물체를개발하였다. 애기장대 MYB96 유전자가 과다발현된 카멜리나 형질전환 식물체에서 cuticular wax crystal 과다축적 및 61%의 cuticular wax 총 함량 증가가 관찰되었다. 이로 인하여 애기장대 MYB96의 카멜리나 과발현 식물체 잎에서 큐티클 투과성의 변화와 함께 비형질전환식물체 대비 향상된 가뭄 저항성능이 관찰되었다. Cuticular wax 함량의 증가는 카멜리나에서발현되고 있는 애기장대 MYB96 유전자가 카멜리나에서 cuticular wax 생합성에 관여할것으로 추정되는 유전자들, CsKCS2, CsKCS6, CsKCR1-1, CsKCR1-2, CsECR 그리고 CsMAH1의 발현량을 증가시킨 결과임을 확인하였다.
2세부과제명 : 사이토키닌 호르몬 수용체 하부 조절 저온 반응 신규 유용 유전자 발굴 및 기능과 신호전달
사이토키닌 신호전달 TCS 구성성분의 저온 반응 기능연구를 위하여 arr1, arr10, arr12 단일및 다중 돌연변이체에서 저온 유도성 type-A ARR 유전자 발현 분석, ARR1 과다발현 형질전환 식물체에서 저온 유도성 type-A ARR 유전자 발현 분석, ARR1 과다발현 형질전환 식물체의 동결 저항성 분석, ahp2, ahp3, ahp5 단일 및 다중 돌연변이체에서 저온 유도성 type-A ARRs 유전자 발현 분석 및 저온처리 하에 AHP-EGFP 단백질의 세포내 분획 분석을 수행하였다. 분석결과 TCS의 type-B ARR 중 ARR1이 AHK2 및 AHK3 를 통하여 인지 전달된 저온 신호에 대응하여 저온 반응성 typ-A ARRs 을 포함하여 하부 표적 유전자들의 발현을 조절하며, AHKs를 통하여 ARRs 로 인 분자를 전달하는 AHPs 를 통하여 저온신호의 전달이이루어진다는 것을 규명하였다. ARR1 의 저온 반응에서의 작동 기전 연구를 위하여 ARR1과 SUMO 단백질과의 상호작용 조사, SUMO 관련 돌연변이체상에서 저온 유도성 type-A ARR유전자의 분자표현형 분석, ARR1의 단백질 안정성과 SUMOylation 자리의 돌연변이와의 상관관계 애기장대 원형질체에서 조사, ARR1의 단백질 안정성을 ARR1 형질전환 식물체에서 조사, ARR1의 SUMOylation을 시험관내 및 애기장대 내에서 확인 및 SUMO E3 ligase의 돌연변이체 상에서 저온 반응성 type A-ARRs 유전자 발현 분석을 수행하였다. 연구결과는 ARR1의 SUMO 단백질에 의한 변이가 저온에 의한 전사활성에 중요한 기능을 한다는 것을 제시하였다. CRF2 및 CRF3 의 저온 반응에서의 신호전달 및 기능 조사를 위하여 CRF 유전자의in silico 분석 및 qRT-PCR을 통한 저온반응성 확인, CFR2 및 CRF3 promoter를 GFP:GUS보고자 유전자에 융합시킨 형질전환체 분석, crf2 및 crf3 돌연변이체 및 과다 발현 애기장대형질전환체 분석,
CRF2 와 ARR1 간의 생화학적 분자 기전 연구를 수행하였다. 연구결과는CRF2 의 저온에 대한 유전자 발현 조절은 TCS 경로를 통하여 이루어지는 반면, CRF3는 TCS 비의존성 저온 반응 경로를 통하여 유전자 발현이 이루어진다는 것을 보여 주었다. 아울러 CRF2 및 CRF3 가 저온 반응중 측근발달 과정 중 발달 개시 단계를 조절한다는 것을 보여주었다. 저온 및 건조 유도성 type-C ARR22의 기능 규명을 위하여 ARR22의 유도성 과발현에 의한 탈수 및 이온유리 분석시행, ARR22의 유도성 과다발현에 의한 건조 및 이온유리분석시행, 사이토키닌에 의한 arr22 돌연변이체와 ARR22 유도성 과발현체의 건조, 동결저항성 조사, microarray 분석 통한 ARR22 유도성 과다발현의 영향 조사를 수행하였다. 연구결과ARR22가 스트레스 반응 관련 유전자들의 발현을 조절하여 저온 및 건조 저항성을 유도한다는 것을 입증하였다.
3세부과제 : 식물 스트레스 관련 유전자들의 기능 분석 및 전사 조절기작과 신호전달기작 연구
벼, 밀, 유채, 배추, 애기장대 유전체 정보를 활용하여 RNA-결합 단백질 유전자를 비교 분석하여, 밀 RNA-결합 단백질 유전자 3개, 유채 RNA-결합 단백질 유전자 7개, 배추에 존재하는cold shock domain protein (CSDP) 유전자 3개를 분리하였고, 각 유전자의 스트레스 반응 발현양상을 비교 분석하였다. 각 유전자의 RNA 샤페론 기능을 대장균 BX04 및 RL211 균주를 이용한 cold shock assay와 transcription anti-termination assay, nucleic acid-melting assay를 통하여 비교 분석하였다. 특정 family member 만이 RNA 샤페론 활성을 갖고, 특정 도메인이 샤페론 활성에 중요함을 밝혔다.RNA-결합 단백질 유전자 과발현 형질전환체 제작, 돌연변이체 확보 및 스트레스 내성 기능을분석하였다. 애기장대 RNA-결합 단백질 유전자 RBP1, 애기장대 RNA-결합 단백질 유전자PSRP2, 유채 RNA-결합 단백질 유전자 BnGRP1, 밀 RNA-결합 단백질 유전자 TaRZ, 배추CSDP 유전자 과발현 형질전환체 제작 및 분석을 완료하였다. 애기장대 RNA-결합 단백질 유전자 RBP1, 애기장대 RNA-결합 단백질 유전자 PSRP2 KO 돌연변이체 제작 및 분석을 완료하였다. 각 과발현 형질전환체 및 돌연변이체의 저온, 가뭄, 고염분, UV 등 스트레스 반응 분석을 통하여 유전자 기능을 확인하였다.RNA-결합 단백질 유전자가 도입된 작물 형질전환체를 확보하여 분석하였다. 애기장대 AtGRP2및 AtGRP7, 벼 OsCSDP1, OsGRP1, OsGRP6 및 OsRZ2 유전자가 도입된 벼 형질전환체를 확보하여, 가뭄, 저온 등 스트레스 내성 분석을 완료하였다. 애기장대 AtGRP2 및 AtGRP7 발현벼 형질전환체가 가뭄에 강한 내성을 보임을 확인하였다.

(1) Identification of novel genes, which are involved in transcriptional regulation of VLCFA biosynthesis: The transgenic Arabidopsis plants expressing the GUS gene under the control of the KCR1 promoter were generated (T3 generation). Two novel genes(At1g65300 and At4g31420) that bind to the KCS2 promoter (1.5 kb) were selected via yeast one hybrid system. In addition, MYB94 and MYB74 transcription factors were selected using transcriptome data from Arabidopsis stem and stem epidermal peels and from ABA and drought-treated Arabidopsis seedlings.
(2) Analysis of structure, expression pattern, and subcellular localization of novel transcription factors: MYB94 and MYB74 contain transcriptional activation domain in their C-terminal regions. The spatial and temporal expression patterns of MYB94 and MYB74 were examined in various Arabidopsis organs and in seedlings after treatments with ABA, drought, salt, and osmotic stresses by using qRT-PCR and GUS reporter system. The MYB94 and MYB74 transcripts were ubiquitously expressed in aerial organs. The expression levels of the MYB94 and MYB74 were approximately 4- to 5-fold higher in stem epidermal peels than in the stems. In addition, the levels of MYB94 and MYB74 transcripts increased in response to drought, ABA, NaCl, and mannitol treatments. The fluorescent signals from the MYB94:eYFP and MYB74:GFP constructs were observed in the nucleus of tobacco leaf epidermal cells or in transgenic Arabidopsis roots.
(3) Functional analysis of MYB94 and MYB74 in planta: To understand the roles of MYB94 and MYB74 genes in planta, T-DNA-inserted knock-out mutants were identified. In addition, complementation lines and transgenic Arabidopsis plants overexpressing MYB94 or MYB74 under the control of the cauliflower mosaic virus (CaMV) 35S promoter were generated. The total wax loads decreased by 23% in the leaves of the myb94-ko, but increased by approximately 1.3- to 1.7-fold in the leaves of the transgenic Arabidopsis plants overexpressing MYB94 (MYB94 OX) lines relative to the WT. Deposition of epicuticuar wax crystals was also detected on the leaves of MYB94 OX lines, but not shown in WT leaves. Cuticular transpiration occurs more slowly in the leaves of MYB94 OX lines than in the leaves of WT under drought stress conditions. The total wax loads increased by 14% in the leaves of the MYB74 OX line, but was not significantly altered in the leaves of myb74-ko relative to the WT. These results indicate that MYB94 and MYB74 play as positive regulators in cuticular wax accumulation.
(4) Identification of target genes of the MYB94: To investigate the target genes of the MYB94, microarray analysis was analyzed using total RNAs, which were extracted from the leaves of WT and MYB94 OX. The expression of WSD1, KCS2/DAISY, CER1, CER2, AlcFAR3/CER4, and ECR/CER10 genes were up-regulated in MYB94 OX relative to WT. MYB94 was identified to bind to the MYB binding consensus motifs of the WSD1, KCS2, CER2, FAR3, and ECR gene promoters. Microarray analysis, transcriptional activation assay, and EMSA revealed that MYB94 activates the expression of WSD1, KCS2, CER2, FAR3, and ECR genes via direct binding to their promoters.
(5) Functional relationship between MYB94 and MYB96 under the drought conditions: Cuticular wax composition and amounts were examined in the leaves of WT, myb94-ko, myb96-ko, and myb96 myb94 double mutants under normal and drought stress conditions. Under normal and drought stress conditions, the total wax load was decreased by 14% and 16% in the leaves of myb94-ko relative to the WT, respectively. In addition, the total wax load was more reduced in the leaves of myb96 myb94 than in single mutants(myb94-ko; 31-44%, myb96-ko; 20-25%). This result indicates that MYB96 and MYB94 have an additive function in the biosynthesis and accumulation of cuticular wax under drought stress. The cuticular transpiration occurred more rapidly in myb96 myb94 leaves than that in single mutants. The reduced total wax load in the myb96-ko leaves was restored to WT levels in transgenic myb96-ko plants expressing MYB94 under the control of the CaMV 35S promoter, indicating that MYB96 and MYB94 have combined function in cuticular wax biosynthesis and accumulation.
(6) Development of drought-resistant transgenic Camelina sativa plants via cuticular wax accumulation: Transgenic Camelina plants overexpressing Arabidopsis MYB96 were developed. Deposition of epicuticular wax crystals and total wax loads increased by 22–61% on the surfaces of transgenic leaves compared with non-transgenic plants. Cuticular transpiration occurred more slowly in transgenic leaves thannon-transgenic plants. Transgenic Camelina plants overexpressing Arabidopsis MYB96 exhibited drought-stress resistance by altering cuticle permeability. The expression levels of CsKCS2, CsKCS6, CsKCR1-1, CsKCR1-2, CsECR, and CsMAH1 were up-regulated in transgenic Camelina leaves than those in non-transgenic leaves. These results indicate that heterologous overexpression of Arabidopsis MYB96 activated the expression of Camelina wax biosynthetic genes that consequently caused an increase of cuticular wax biosynthesis and accumulation in transgenic Camelina plants.
2세부과제 : 사이토키닌 호르몬 수용체 하부 조절 저온 반응 신규 유용 유전자 발굴 및 기능과 신호전달
Two-component signal transduction is commonly used as a stimulus-response coupling mechanism to allow organisms such as eubacteria, archea, and a few eukaryotes to sense and respond to changes in many different environmental conditions. Typically, the histidine protein kinase senses extracellular stimuli by autophosphorylation and transfers a phosphoryl group to the response regulator, resulting in activation of downstream proteins that elicit a specific response. In Arabidopsis plants, a multi-step two-component system is well established as a key element of plant hormone cytokinin signaling. However, recent accumulating evidence demonstrates that the two-component signaling system (TCS) is also involved in plant adaptation to environmental stresses such as abscisic acid (ABA), drought, cold, and osmotic stress in addition to cytokinin signaling. In this study, we focused on the role of TCS during cold signaling in Arabidopsis. Arabidopsis has three sensor histidine-kinase cytokinin receptors, CYTOKININ RESPONSE1/ARABIDOPSIS HISTIDINE KINASE4 (AHK4)/WOODEN LEG1, AHK2, and AHK3. ARABIDOPSIS HISTIDINE PHOSPHOTRANSFER PROTEINs (AHPs) mediate the transfer of a phosphoryl group from the AHK proteins to the ARABIDOPSIS RESPONSE REGULATORs (ARRs).ARRs are classified into type-A, type-B, and type-C. The type-B ARRs (ARR1, 2, 10–14, and 18–21) contain the receiver domain and a large C-terminal region harboring a Myb-like DNA-binding domain and a glutamine-rich region for transcriptional activation, and are not inducible by cytokinins, and function as positive regulators of cytokinin signaling. Type-A ARRs (ARR3–9 and 15–17) are composed of the receiver domain and a C-terminal extension and are inducible by cytokinins, and act mainly as redundant negative regulators of cytokinin signaling. Type-C ARRs (ARR22 and ARR24) have a domain structure similar to the type-A ARRs, but their expression is not induced by cytokinins, and their role in cytokinin signaling is unclear. We have previously shown that AHK2, AHK3, and cold-inducible type-A ARRs play roles in cold signaling.However, the roles of type-B ARRs and AHP s have not been investigated in cold signaling. Here we showed that ARR1 and AHP2, AHP3, and AHP5 play a positive role in cold-inducible expression of type-A ARRs. arr1 mutants showed greatly reduced cold-responsive expression of type-A ARRs compared with that of wild type, whereas ARR1-overexpressing Arabidopsis exhibited hypersensitive cold-response of type-A ARRs as well as enhanced freezing tolerance with cytokinin, suggesting that ARR1 functions as a positive factor of cold signaling.Transgenic Arabidopsis expressing ARR1ΔDDK:GR lacking the N-terminal receiver domain showed wild-type expression levels of type-A ARRs in response to cold, indicating thatthe signal receiver domain of ARR1 might be important for cold-responsive expression of type-A ARRs. ahp2 ahp3 ahp5 triple mutations greatly reduced type-A ARR expression in response to cold, whereas the single or double ahp mutants displayed wild-type levels of ARR expression, suggesting that AHP2, AHP3, and AHP5 are redundantly involved in cold signaling. Taken together, these results suggest that ARR1 mediates cold signal via AHP2, AHP3 or AHP5 from AHK2 and AHK3 to express type-A ARRs. We further identified a cold transcriptome affected by ahk2 ahk3 mutations by microarray analysis, revealing a new cold-responsive gene network regulated downstream of AHK2 and AHK3. We found that 57 cold-responsive genes were down-regulated by the ahk2 ahk3 mutation compared withthat of the wild-type under cold treatment at 1 °C for 2 h with a ratio of ≥1.5. Among them, 10 genes, including APRR5,ARR15, CRF2, ARR5, were found to be targets of ARR1. While AHK2 and AHK3 function independently of CBFs, they modulate several CBF3 target genes via negative regulation of MYB15, which acts as a negative regulator of CBF3, indicating that a link might exist between the CBF3 pathway via MYB15 and the AHK2/AHK3-responsive cold signaling pathway. However, most of the cold-regulated genes downstream of AHK2 and AHK3 were not regulated by CBF3 or ARR1, indicating that those cold-responsive genes are regulated by unidentified transcription factors acting downstream of AHK2 and AHK3.Functional characterization of CRF2 and CRF3 were carried out, showing that CRF2 and CRF3 are involved in regulating lateral root development at the initiation step during cold response. To explore a potential function of the cytokinin TCS in the Arabidopsis dehydration stress response, we investigated the expression of all type-A ARR genes and a type-C ARR, ARR22, in both wild type and ahk single, double, and triple mutantsin response to dehydration compared to cytokininas well as dehydration tolerance of ahkmutants. We found that drought significantly induced the expression of a subset of ARR genes, ARR5, ARR7, ARR15, and ARR22. The results of expression analyses in ahk single, double, and triple mutants demonstrated that the cytokinin receptors AHK2 and AHK3 are redundantly involved in dehydration-inducible expression of ARR7, but not that of ARR5, ARR15, or ARR22. Dehydration tolerance assays showed that ahk2 and ahk3 single mutants exhibited enhanced dehydration tolerance compared with that of wild-type plants and ahk4 mutants, and that ahk2 ahk3 double mutants exhibited stronger drought tolerance than that of ahk3 ahk4, which exhibited more enhanced drought tolerance than that of wild-type plants and ahk single mutants. Taken together, these results demonstrate that while the cytokinin receptors AHK2 and AHK3 are critically involved in the dehydration tolerance response, both cytokinin receptor-dependent pathway and receptor-independent pathway occur in the dehydration response regulating ARR gene expression. In addition, preincubating ahk2, ahk3, ahk4, and the wild-type plants with cytokinin induced enhanced dehydration stress tolerance in these plants, demonstrating that cytokinins are involved in regulating plant response to dehydrationstress.Although the roles of type-A and -B ARRs are well established in Arabidopsis plant signaling, roles of type-C ARRs, ARR22 and ARR24, remain elusive. We showed that inducible overexpression of ARR22 in Arabidopsis enhanced dehydration, drought, and cold tolerance in a dexamethasone-dependent manner, whereas mutation of the putative phospho-accepting Asp to Asn in ARR22 (ARR22D74N) abolished these tolerance phenotypes. Overexpression of ARR22 decreased electrolyte leakage in dehydration-, drought-, or cold-stressed transgenic Arabidopsis plants compared with that of ARR22D74Nor compared with wild-type plants.Transpiration rates and stomatal apertures were not affected by ARR22 overexpression. No significant difference in both dehydration and freezing tolerance was observed between wild-type and arr22 mutants with or without cytokinin preincubation, consistent with the lack of phenotypes of arr22 mutants in their vegetative development. Meta-profile analyses of the microarray data on ARR22-responsive genes indicate that ARR22 modulates expression of a variety of abiotic stress-responsive genes which might contribute to increasing drought and freezing tolerance. Taken together, these results suggest that ARR22 plays a positive role in the stress tolerance response in part via enhancing cell membrane integrity and that phospho-histidine phosphatase activity of ARR22 may be required for this function.

Title; Isolation and functional analysis of RNA transcript-regulating genes involved in stress tolerance
This research aimed to isolate RNA transcript-regulating genes and to analyze the functional roles of these genes in stress tolerance. In particular, special focus was given to determine the roles of RNA-binding proteins (RBPs) in Arabidopsis (Arabidopsis thaliana), rice (Oryza sativa), wheat (Triticum aestivum), and rapeseed (Brassica napus) under various stress conditions, including cold, salt, drought, and heat stresses. Many genes, including glycine-rich RBPs (GRPs), zinc finger GRPs (RZs), and cold shock domain proteins (CSDPs), were isolated from Arabidopsis, rice, wheat, and rapeseed, and their roles in stress response were determined using loss-of-function mutants and overexpression transgenic plants. In addition, the mechanistic roles of RBPs as RNA chaperones were analyzed. Our results showed that several GRPs, RZs, and CSDPs play important roles in plant response to stress conditions. Moreover, certain family members of GRPs, RZs, and CSDPs displayed RNA chaperone functions during stress tolerance. Collectively, the current research demonstrated important roles of GRPs, RZs, and CSDPs in stress tolerance in diverse plants, suggesting that these genes can be utilized to develop transgenic plants with a higher productivity under stress conditions.
