Ⅳ. 연구개발결과

2. E3-ligase의 비알콜성 지방간질환 목표인자에 대한 조절기작 규명 및 질환 조절 천연소재 개발
비알콜성 지방간 질환 조절에 있어 중요한 인자로 알려진 PPARγ단백질을 조절하는 인자로 MKRN1, CHIP, MKRN3를 찾았고, 그들의 PPARγ 단백질 조절기작을 밝혀냈다. MKRN1과 CHIP 단백질의 직접적인 상호결합에 의해 각각 PPARγ 단백질의 감소능을 가지고 있음을 확인했다. 그리고 이러한 PPARγ 단백질의 감소능이 proteasome 의존적인 ubiquitylation의 증가에 기인한 기작임을 밝혔다. 지방아세포와 마우스 배아세포를 이용한 유전자 knock-in, -out 실험을 통해서 이러한 기작이 지방세포로의 분화를 억제시키는데 역할을 한다는 것을 확인하였다. 또 다른 e3-ligase인 MKRN3도 PPARγ와 결합은 하지만, 단백질을 증가시키는 기작을 확인하였다. 그리고 공동연구를 통해 camptothecin이라는 기능성 성분이 PPARγ 단백질의 발현을 억제하고, 지방세포 분화를 막는 기능이 있음을 확인하였다.
3. 비알콜성 지방간 질환 유발 동물 모델 확립 및 천연소재 적용연구
비알콜성 지방간 질환 동물모델을 확립하기 위해 정제된 adenovirus PPARγ를 마우스 혈관에 투여하여 PPARγ 과발현 동물모델을 형성하였고, 이후 지방간병변이 발생하는 것을 확인하였다. 이후 제 1세부과제 연구팀에서 제공한 천연물 소재를 투여하였을 때 간 조직내에서 PPARγ의 발현이 감소하는 것을 확인하였다. 또한, PPARγ 발현 조절 유전자(Galectin3) KO마우스를 이용하여 새로운 비알콜성 지방간 질환 동물모델을 확립하였다. Galectin-3 KO에 의해 마우스 지방조직 감소와 간 내 중성지방 축적 억제를 확인하였고, 지방 축적과 합성 관여 유전자들의 발현이 감소하는 것을 확인하였다.

Liver is the central organ for coordinating the regulation of energy metabolism and lipid trafficking. The excess intake of dietary lipid is leading to fatty liver, a liver lesion which can later progress to NAFLD, NASH. Besides, an imbalance between the synthesis and oxidation of fatty acids in the liver are also involved in the pathophysiology of NAFLD. Several medicines are being tested for the treatment of NAFLD, and recently the use of plants emerged as a possible means to alleviated NAFLD. However, no medication has been approved for treating NAFLD. In present study, we examined the beneficial effects of natural materials, candidates for the treatment of NALFD, on hepatic fat accumulation and production of oxidative stress. In addition, to radical treatment of NAFLD, we identify a novel biomarker which regulated PPARγ as fatty liver inducer. And also, we confirmed the improvement of NAFLD by transgenic mouse model and natural materials.
First, we investigated regulatory mechanism in NAFLD and investigation of natural materials. This study was performed to investigate the effect of natural materials on lipid metabolism in OA-treated HepG2 cells. To verify the inhibition of natural materials of OA-induced lipid accumulation, HepG2 cell were treated with natural materials in the presence of OA. Then cells were stained with various staining solution such as Oil-Red O and Nile Red. In the results, Aralia elata(Miq.) Seemann, Pinus densiflora Sieb. & Zucc., Aster yomena treated group effectively reduced the lipid accumulation compared to OA group regardless of staining solution. In addition, the mRNA expression of lipogenic genes such as SREBP1c, ACC1 and FAS was significantly decreased and PPARα, CPT1 and ACC2 which are lipolysis genes, expression was increased in Aralia elata(Miq.) Seemann, Pinus densiflora Sieb. & Zucc., Aster yomena group compared to OA group. Furthermore, we also confirmed that the inhibitory activity of Aralia elata(Miq.) Seemann, Pinus densiflora Sieb. & Zucc., Aster yomena on NAFLD in vivo. Finally, we suggested that the reason of inhibiting lipid accumulation in the liver was due to recovery of insulin resistance by natural materials.
Second, we identified the regulatory effects of E3-ligase on NAFLD. We observed that several e3-ligases controling the PPARγ, could be linked to NAFLD. MKRN1 and CHIP (STUB1) degrade and induce ubiquitylation of PPARγ. Corroborating these data we confirmed that these two e3-ligases could suppress the adipocyte differentiation. Another e3-ligase MKRN3 (isoform of MKRN1) also induces ubiquitiylation of PPARγ protein without its degradation suggesting ubiquitin dependent modulation of protein function. Lastly, our group found that Camptothecin, a chemotherapeutic agent, suppresses expression of PPARγ protein with inhibition of adipogenesis. In conclusion, we provided many candidate proteins and chemical regulating NAFLD.
Third, we established the mouse model of NAFLD and studies of natural material. To establish the NAFLD-mouse model, we injected purified adenoviral PPARγ in mouse tail vein. After injection, we confirmed the lipid accumulation in liver. And then, we treated the natural materials to confirm the regulating PPARγin liver. We identified inhibition of PPARγexpression. In addition, we established the novel NAFLD mouse model "Galectin-3 KO" which regulating PPARγ. Galectin-3 was decreased the lipid accumulation in fat and liver. And also, mRNA expression of lipogenic genes was significantly decreased by galectin-3.
Taken together, we identified the natural materials and novel biomarker for improvement of NAFLD and confirmed the molecular mechanism.
