1. 제 1 세부연구과제
미니 돼지에서 대사질환 유전자 ICER Iγ 11β-HSD1가 도입된 형질전환 미니돼지 생산을 위해 ICER Iγ 와 11β-HSD1 유전자를 동정하였다. 동정된 유전자의 과발현을 위해, 돼지 ICER Iγ 유전자는 췌장 특이적이고 최상의 과발현을 위해 돼지와 인간의 인슐린 프로모터 부위를, 11β-HSD1는 지방세포 특이적인 발현을 위해 지방세포 지방산 결합단백질 프로모터 부위를 세분화하고 pγL3 Basic 벡터에 삽입하였다. 돼지와 인간의 인슐린 프로모터의 활성도 평가는 mouse β-cell line (MIN6) 세포에서, 지방산 결합단백질 프로모터의 활성도는 마우스 지방세포주인 3T3-L1 세포에서 활성도 평가를 실시하였다. 프로모터 활성도 평가 결과 세분화된 인간의 인슐린 프로모터에서는 1.5kb의 프로모터에서 유의성 있게 프로모터 활성이 증가하였고, 돼지 인슐린 프로모터와 지방세포 지방산 결합단백질 프로모터는 모든 부위에서 유의성 있게 활성도가 증가하는 것을 확인하였다. 이 중 가장 활성도가 높은 각각의 프로모터를 동정된 유전자에 제한효소를 사용하여 삽입하였다. ICER Iγ 유전자는 테트라싸이클린 처리시기 및 처리양에 따라 발현 정도를 조절할 수 있는 Tet-On 시스템을 적용하였고, 11β-HSD1 유전자는 형광 발현을 통해 발현을 확인할 수 있도록 EGFP 유전자를 삽입하였다. 그리고 두 유전자 운반체 모두 neomycine 저항성 유전자를 삽입해 최종적인 운반체를 제작하였다. 공여핵원세포인 미니돼지의 체세포 제
조를 위해 미니돼지의 귀에서 체세포를 분리하고 운반체를 분리된 체세포에 ICER Iγ와 11β-HSD1 유전자 운반체를 형질주입하고, 250㎍/ml의 G-418을 4주 동안 처리하면서 배양해 neomycin 내성을 통해 형질전환 세포군을 분류하였다. 형질전환 세포군으로부터 지노믹 유전자를 추출해 특정 프라이머를 활용해 PCR을 수행하고, 최종적으로 ICER Iγ 11β-HSD1 유전자 형질전환 세포주를 확립하였다.
2. 제 2 세부연구과제
돼지 미성숙란의 최적의 체외성숙 시스템 확립과 형질전환 복제 배아의 최적의 체외배양 시스템 확립을 위해 돼지 미성숙 난자 및 형질전환 복제 배아에 다양한 물질들을 처리해 그 기전을 연구하였다. GDF9, BMP15가 돼지 난자의 체외성숙과정에서 모계유래 유전자 발현과 유전자 발현의 아데닐산 중합반응에 영향을 주는 것을 확인하였다. 미토콘드리아의 막전위 활동억제제를 사용한 돼지 미성숙난자의 미토콘드리아 수 및 핵 막전위를 감소시킨 결과 미성숙 난자의 MII 단계까지의 성숙률이 크게 감소하는 것을 관찰하였다. 또한 Great wall (Gwl) 유전자가 돼지 난자의 체외성숙 및 생식세포의 분열에 미치는 영향을 살펴본 결과, 세포분열의 MI, MII 단계에서 Gwl 단백질이 현저하게 감소되었다. 형질전환 핵이식 배아를 얻기 위한 미세조작과정에서 시간이 소요되므로 돼지 난자의 체외성숙과정에서부터 난자의 노화를 최소화하는 것이 중요한데, 이에 TSA 처리를 통해 체외성숙 난자의 성숙 동기화 및 노화 지연 기전을 조사하였다. TSA의 처리 농도 증가로 난자의 체외 성숙률은 감소되었지만, MII 단계까지 성숙한 난자의 MAPK 단백질 발현양은 TSA 농도 증가에 따라 증가하였다. 다음으로, 아연이 체외성숙에 미치는 영향을 연구한 결과, 아연이 난자의 감수분열전기에서 감수분열 재개를 촉진하고 MII단계에서의 arrest에 중요한 인자로 작용하는 것을 알 수 있었다.그리고 히스톤 탈아세칠화 억제제인 Scriptaid가 형질전환 복제배아의 리프로그래밍에 미치는 영향을 살펴본 결과, H3K9의 아세칠화는 증가시키고, 메틸화와 ROS는 감소시켰으며, OCT4와 CDX2의 발현은 증가시켰다.
3. 제 1 협동연구과제
미니돼지 ICER Iγ와 11β-HSD1 유전자의 과발현 운반체가 형질주입된 체세포를 이용해 형질전환 복제배아를 생산하였고, 이러한 복제배아들의 체외발달율 확인하였다. 준비된 대리모에 외과적 수술을 통한 ICER Iγ와 11β-HSD1 유전자 형질전환복제배아들의 수정란이식을 실시하였다. ICER Iγ 형질전환 세포주를 이용해 체세포핵이식을 수행해 총 440개의 복제 배아를 생산하였다. 그 중 55개의 복제 배아를 체외배양해 체외 발달능을 평가하였다. 복제 배아 생산 후 48시간뒤에 분할률은 90.0%, 7일후의 배반포 형성률은 15.0%로 통상적인 체세포 핵이식 배아의 체외 발달률을 보였다. 11β-HSD1의 경우, 체세포 핵이식 방법을 통해 6마리 산자를 얻었고, 이중 한 마리가 11β-HSD1 유전자 부위와 선별유전자 부위를 모두 지니고 있었다. 이 11β-HSD1 형질전환 돼지의 체세포로부터 형질전환 세포주를 수립하고, 이를 체세포 핵이식에 이용하는 일명 re-cloning 기법으로 형질전환 복제 돼지를 생산하였다. 이 기법을 통해 생산된 2641개의 11β-HSD1 형질 전환 복제 배아를 총 28두의 대리모에 이식해 그 중 5두(17.8%)의 대리모에서 임신이 확인되었다. 기존 방식과 비교할 때, 임신률이나 분만효율에서는 유의적인 차이가 없었다. 그러나, re-cloning을 통해 생산된 산자 중 14마리에서 Target 유전자와 Reporter 유전자의 도입을 확인한 결과, 14마리 모두에서 Reporter 유전자와 Target 유전자의 도입이 확인되었다. 이는 기존의 체세포 핵이식 기법을 통해 22마리의 산자 중 9마리(40.9%)에서만 Reporter 유전자의 도입이 확인되고, 그 중 3마리(13.6%)에서만 Target 유전자의 도입이 확인된 것과 비교할 때, 형질전환 돼지 생산 효율이 유의적으로 향상되었다.

Metabolic syndrome is defined as the simultaneous presence of abdominal obesity, hyperglycemia, hypertension, hyperphagia, hyperleptinemia, and dyslipidemia with the insulin resistance as a common pathophysiology. Diabetes mellitus is a metabolic disease caused by impaired insulin secretion from the pancreatic β cells and increased insulin resistance in peripheral tissues. Type 1 diabetes mellitus (also referred to as insulin dependent diabetes mellitus or T1DM) is characterized by the failure to produce sufficient quantities of insulin. Individuals with this form of the disease require exogenous insulin for survival. Type 2 diabetes mellitus (also referred to as non-insulin-dependent diabetes mellitus or T2DM) is caused by defects in insulin secretion along with insulin resistance.
First, inducible cyclic AMP (cAMP) early repressor (ICER) Iγ acts as an endogenous inhibitor and disrupts the transcriptional regulation of cAMP responsive element binding protein (CREBP) responsive genes. Since the overexpression of ICER Iγ causes severe diabetes in a transgenic mouse model similar to human diabetes mellitus, we used an ICER Iγ construct containing a pancreas tissue specific and adjustable promoter in the current study. We created a doxycycline (dox)-inducible ICER Iγ expression system. We also developed a unitary tet-on system that combined a tet-on activator cassette controlled by the human/pig insulin promoter with a responder cassette containing genes encoding ICER Iγ controlled by the TRE promoter. To determine whether dox-enhanced ICER Iγ expression influences insulin production, we first introduced the unitary tet-on ICER Iγ vector into mouse pancreatic β cell line, and then treated the cells with 0.1-1 mg/mL dox. We observed a robust increase in ICER Iγ expression and decreased insulin production. In addition, we generated porcine transgenic fibroblasts containing dox-inducible ICER Iγ.
Second, overexpression of 11β ‑hydroxysteroid dehydrogenase type 1 (11β‑HSD1) induced obesity and the insulin resistance in rodent adipose tissue. Based on these, overexpression of 11β‑HSD1 may be suitable for the generation of a porcine model of metabolic syndrome. It was evaluated that promoter activities of the porcine adipose fatty acid‑binding protein (aP2) gene generates adipose tissue‑specific 11β‑HSD1 expres-sion. In adipose cell line, the maximum promoter activity (‑2,826 to +51 nt) of aP2 was 200‑fold higher than that of a promoter-less construct. In addition, 11β‑HSD1 transcriptional levels were significantly increased following the introduction of the aP2 promoter into 3T3‑L1 adipocytes. Transgenic fibroblasts were generated containing 11β‑HSD1 cDNA controlled by the aP2 promoter with two screening markers, green fluorescence protein and a neomycin‑resistance gene. Based on 11β-HSD1-transgenic (TG) fibroblasts, we produced TG piglets overexpressing 11β-HSD1 without obesity through SCNT and re-cloning method. Integration of target gene into their genomic DNA was confirmed from all of them. However, all live piglets died within one month, and showed hypoglycemia. Overexpression of 11β-HSD1 induced the over-production of cortisol, and up-regulation of gluconeogenesis/lipogenesis-related genes in liver and muscle. Also, it stimulated AMPK/SIRT signaling, which controls energy balance. To compensate for energy loss by anabolism, expression of mitochondrial biogenic genes was increased.

Porcine oocyte aging resulting from asynchronised IVM impairs embryo developmental competence. We investigated whether trichostatin A (TSA; an inhibitor of histone deacetylation) prolongs the maturation time and prevents the aging of oocytes. Porcine oocytes were cultured in medium containing increasing concentrations of TSA (300 nM) for 24, 44 or 64 h. The percentage of oocytes that underwent germinal vesicle breakdown was significantly lower in the TSA-treated group (300 nM) than in the control group. TSA did not affect oocyte quality at MII based on levels of maturation-promoting factor, the phosphorylation status of mitogenactivated protein kinase or histone H3K9 acetylation analysis. We also compared the preimplantation developmental competence and the viability of pathenogenetic embryos treated with 100nM TSA for 24 h and then continuously cultured for another 24 h in TSA free condition. No significant differences were observed for either parameter between the TSA treated and control groups. This study investigated the role of zinc in the metaphase (M) II arrest of porcine oocytes. N,N,N',N'-tetrakis (2-pyridylmethyl) ethylenediamine (TPEN), a Zn(2+) chelator, was used to deplete free zinc from porcine MII stage oocytes. TPEN treatment significantly (P<0.01) reduced the zinc content in the cytoplasm. The percentages of oocytes in which second polar body emission and pronuclear formation occurred increased as the concentration of TPEN increased (P<0.01), and reached 93.64 ± 5.53% and 90.61 ± 9.10%, respectively, following treatment with 10 μM TPEN. Zinc depletion also resulted in cortical granule release and spindle depolarization. Maturation-promoting factor activity, as assessed by examining p34(cdc2) activity, decreased (P<0.05) following zinc depletion. Following TPEN treatment, embryos developed to the 4-cell stage but failed to reach the blastocyst stage. Next, we examined the impact of zinc depletion on phosphorylation of PKC substrates. Phosphorylation of PKC substrates was reduced (P<0.05) in zinc-depleted oocytes, and this was rescued by phorbol 12-myristate 13-acetate (PMA) treatment. However, treatment of oocytes with both PMA and TPEN did not affect pronuclear formation or second polar body emission. Also, we investigated the role of zinc during meiotic resumption in in vitro matured porcine oocytes. The level of phosphorylated mitogen activated protein kinase (MAPK) and p34cdc2 kinase activity were reduced when zinc was depleted, and reduced the levels of phosphorylated PKC substrates in a dose dependent manner. Real-time PCR analysis showed that expression of the MAPK and maturation promoting factor related genes C-mos, CyclinB1, and Cdc2 was down-regulated following zinc depletion. Treatment with the PKC agonist PMA increased phosphorylation of PKC substrates and MAPK and increased p34cdc2 kinase activity. This rescued the meiotic arrest, even in the presence of TPEN. Activation of PKC by PMA increased the level of zinc in the cytoplasm. These data demonstrate that zinc is required for meiotic resumption in porcine oocytes, and this appears to be regulated via a PKC related pathway.

The basic mechanisms of metabolic syndrome are not completely known yet. Therefore, animal disease models are required for the study of metabolic syndrome. So, we tried to produce the pigs with overexpression of 11ß-HSD1 gene by somatic cell nuclear transfer (SCNT). However, the low transgenic (TG) efficiency has been an obstacle to the production of TG pigs. And, a SCNT method in which somatic cells derived from TG pig are used as the nuclear donor (re-cloning method) is an effective technique for TG pig production. In this study, we attempted to increase TG efficiency by re-cloning method. Pregnancy efficiency, production efficiency and TG efficiency were compared with sources of donor cells (transfected TG fetal fibroblast vs. TG fibroblast derived from newborn TG cloned pig). A total of 1382 and 881 TG SCNT embryos were produced from fetal fibroblast vs. cloned fibroblast, and then transferred to 13 and 10 recipients. The pregnancy rate was not significantly different (30.8% vs. 20.0%). Seventeen live piglets and five stillborn piglets were born from four recipients in the fetal fibroblast used group and eight live piglets, two stillborn piglets and three mummies were born from two recipients in the cloned fibroblast used group. There were no significant difference in the production efficiency (3.7% vs. 5.0%). All of the thirteen re-cloned piglets showed reporter and target gene integration. But, of twenty two fetal fibroblast-cloned piglets, reporter gene integration was confirmed in nine, only three clone piglets showed reporter gene integration. TG efficiency was significantly increased in re-cloning group (13.6% vs. 100.0%). In this study, TG efficiency of 11β-HSD1 overexpressed pigs was improved by re-cloning method.
