Ⅳ. 연구개발결과
1. 제1세부과제
활성추적분리법을 통한 천식 효능성분 탐색 및 구조 동정
○ 머위 부위별 용매를 달리하여 추출한 추출물의 6종의 암세포에 대한 MTT assay를 한 결과 항암 등에 관하여 조사한 결과 부위에 추출용매에 따라서 세포의 생존율이 달랐는데, 극성이 낮은 용매로 추출할수록 세포의 생존율이 감소하였다.
○ 머위로부터 항아러지에 효능을 나타내는 성분을 activity-guided purification방법으로 분리한 결과 항알레르기 효과가 높은 bakkenolide B와 신물질인 pet
Ⅳ. 연구개발결과
1. 제1세부과제
활성추적분리법을 통한 천식 효능성분 탐색 및 구조 동정

머위의 부위별 극성용매 추출물이 6종 암세포의 생존율에 미치는 영향
○ 머위의 잎, 엽병 및 근경을 각각 hexane, EtOAc, BuOH 및 물추출물에 대하여 6종의 암세포인 AGS (human gastric cancer), HepG2 (human hepato cellular cancer), HCT-116 (human colorectal cancer), and MCF-7 (human breast cancer) cell lines에 대한 세포생존율을 조사하였다.
○ 세포의 생존율은 극성이 낮은 hexane, EtOAc 추출물에서 생존율이 낮아 항암 효과가 있는 것으로 조사되었다.
○ 식물체의 부위별 효과을 조사한 결과 HepG2 cell line의 경우에는 IC50이 4.31 μg/ml으로서 근경보다는 2.5배, 엽병보다는 6.3배 효과가 좋았다.
○ 이러한 결과를 종합하면, 머위는 암에 효과가 있는 식의약품 또는 의약품으로서 개발 가능성이 있는 것으로 판단되었다.
머위로부터 Bakkenolide B의 순수분리, HPLC분석 방법 및 채취 시기별 함량 분석
○ 머위(Peatasites japonicus)의 잎으로부터 생리활성물질을 탐색하여 항천식 효과가 있는 물질로 알려진 bakenolide B를 순수분리하였으며, 1D-NMR, 2D-NMR 및 GC-MS spectrum 데이터를 이용하여 구조를 동정하였다.
○ 머위의 잎, 엽병 및 근경 등의 채취시기별 bakkenolide B의 함량을 평가하기 위하여 HPLC 분석 방법을 확립하였다.
○ Bakkenolide B의 민감도는 210 nm와 215 nm의 파장에서 254 nm, 235 nm 및 265 nm보다 높았으며, 분석을 위한 회귀 직선식은 y=7.841-6.262(파장 215 nm)로서 상관값(r2)이 0.999이상으로서 유의성이 매우 높았다.
○ 검출한계(LOD)는 1.05, 정량한계 (LOQ)는 3.38이었다. 회수율은 3종류의 농도값에 대하여 98.6에서 103.1%로서 매우 높았다.
○ 이러한 결과들로 미루어 볼 때 머위에서 bakkenolide B의 함량은 HPLC 분석방법으로서 가능한 것으로 증명되었다.
○ 식물체의 부위 즉 잎, 엽병 및 근경에서의 bakkenolide B의 함량을 측정한 결과, 잎에서 가장 높았고, 다음은 엽병이었으며, 근경에서 함량이 가장 낮았다.
○ 3월 초부터 8월 초까지 1개월 간격으로 식물체의 부위별 bakkenolide B의 함량을 조사한 결과, 3월부터 8월까지 채취시기가 지연될수록 감소하는 경향이었다.
○ HPLC를 이용한 Petasites species의 bakkenolides 분석은 단순하고, 급속하게 분석할 수 있는 방법으로 확인되었다.
○ 신물질인 petatewalide B는 다양한 효능을 검증하여, 머위의 새로운 효과 제시가 가능할 것으로 기대된다.
부원료인 어성초와 수세미로부터 생리활성 성분의 순수분리 및 구조동정
○ 어성초(Houttuynia cordata Thunb)로부터 3 – 5종, 수세미(Lufa cylindrica)로부터 2종의 물질이 순수분리 되었다.
○ 순수분리된 성분들은 향 후 항알레르기, 항염증 등의 효능을 검증할 계획임
○ 어성초와 수세미로부터 순수분리한 성분의 효능 검증 후 지표성분으로 활용이 가능할 것으로 기대된다.
2. 제2세부과제
머위에서 분리한 bakkenolide B의 항알레르기와 항염증 효과
머위 속 (Petasites genus)의 항알레르기 효과와 항염증 효과를 밝히기 위해서, Petasites japonicas 잎에서 추출한 몇 가지 물질들의 효과를 연구하였다. Bakkenolide B는 Petasites japonicas 잎에서 분리한 물질이다. 항알레르기의 측정지표인 β-hexosaminidase 활성을 측정하기 위해서 RBL-2H3 비만세포주에서 항원으로 유도된 탈과립 반응 억제효과를 측정하였다. 항염증 효과를 확인하기 위해서 복강 대식세포에서 western blotting 방법을 이용해 inducible nitric oxide synthase (iNOS)와 cyclooxygenase2 (COX-2)의 발현을 측정하였다. Bakkanolide B의 in vivo상에서 효능 검사를 하기 위해 ovalbumin으로 유발된 마우스 천식 모델을 사용하였다. Petasites japonicas 잎의 주요 성분인 bakkenolide B가 RBL-2H3 비만 세포주의 탈과립을 농도의존적으로 억제시키는 것을 확인하였다. 또한, 마우스 복강 대식세포에서 bakkenolide B가 iNOS와 COX-2 발현 유도를 억제시켰다. 그리고 ovalbumin으로 유발된 천식 모델에서 얻은 기관지 세척액에서 bakkenolide B가 호산구, 대식세포, 림프구의 축적을 유의성있게 억제시켰다. Bakkenolide B는 알레르기 반응과 염증 반응을 억제하는 효과를 가지고, 천식 치료제로써 이용될 수 있다.
머위에서 분리한 새로운 petatewalide B의 다중 효과
머위 속 (Petasites genus)의 효과를 밝히기 위해서, Petasites japonicas 잎에서 새로운 구조의 물질인 petatewaldie B를 발견하였다. Petatewaldie B는 bakkenolide 구조를 가지고 있으며, 항알레르기의 측정지표인 β -hexosaminidase 활성을 측정하기 위해서 RBL-2H3 비만세포주에서 항원으로 유도된 탈과립반응 억제효과를 측정하였을 때, 억제효과를 나타내었다. 하지만 세포내 칼슘농도에 대해서는 억제효과를 보이지 않았다. Petatewaldie B는 항염증 효과를 확인하기 위해서 복강 대식세포에서 western blotting 방법을 이용해 inducible nitric oxide synthase (iNOS)와 cyclooxygenase2 (COX-2)의 발현을 측정하였을 때, iNOS를 억제하였으나, COX-2를 억제하지는 않았다. Petatewaldie B는 NO의 생성을 억제하였다. 또한 petatewalide B는 C6 glioma세포에서 세포막전위를 농도의존적으로 증가시켰다. 본 연구를 통해서 머위에서 분리한 petatewlide B가 항알레르기효과와 항염증효과를 가질 뿐만아니라 C6 glioma 세포의 막전위를 증가시키는 것을 보였다.
천식과 복막염 동물모델에서 S-Petasin의 치료효과
Petasites genus의 항알레르기 및 항염증 효과를 확인을 위해, Petasites formosanus(머위의종)으로 부터 핵심 sesquiterpene인 s-petasin의 효과를 천식과 복막염모델에서 연구하였다. 오브알부민으로 유도하는 마우스 천식모델에서는, s-petasin은 호산구, 대식세포, 림프구의 기관지세척액으로 축적되는 것을 명확히 억제하였다. s-petasin은 RBL-2H3 비만세포주에서 항원으로 유도하는 β-hexosamindase의 탈과립을 억제하였으나, 세포내 Ca2+ 증가를 억제하지는 못하였다. S-petasin은 마우스 복강 대식세포에서 LPS로 유도되는 iNOS를 RNA와 단백질레벨에서 억제 하였다. 또한, s-petasin은 NO (iNOS의 생성물)의 생성을 대식세포에서 농도의존적으로 억제하였다. 거기에 더해, s-petasin은 복막강에 다형 및 단핵백혈구 축적을 억제를 명확히 보였다. 이 연구는 Petasites genus의 s-petasin이 천식과 복막염과 같은 알레르기성 및 염증성 질병에서 비만세포에서의 탈과립의 억제, 대식세포에서의 iNOS유도와 NO생성의 감소와 염증성세포의 축적의 억제를 통한 치료적 효과를 가지고 있음을 보여 준다.
오미자에서 분리한 새로운 항염증물질 α-cubebenoate의 동정
Schisandra chinensis 의 추출물은 예로부터 항피로제, 강장제로 사용되어왔다. 만성피로 증후군은 염증과 항산화 스트레스와 연관되어있기 때문에 Schisandra chinensis 구성물질로 항 염증 평가하였고, Schisandra chinensis 로부터 분리한 α-cubebenoate의 효과를 연구하였다. α-cubebenoate는 Schisandra chinensis 열매의 추출물로부터 분리 되었다. in vitro 실험에서는 mouse peritoneal macrophage에 LPS로 COX-2와 iNOS를 유발시켜 RT-PCR과 Western blotting으로 정량하였다. 세포 배양액 내의 NO와 PGE2는 Griess reagen와 EIA 방법으로 측정하였다. in vivo 실험에서는 LPS로 복막염을 유도한 마우스 모델을 사용하여 α-cubebenoate의 효능을 테스트 하였다.
결과 : α-cubebenoate (5-10μg/ml)는 mouse peritoneal macrophage 의 iNOS, COX-2의 발현을 mRNA와 protein level에서 저해하였고. LPS로 유도된 NO와 PGE2 생성 또한 α-cubebenoate (5-10μg/ml)에 의해 저해 되었다. 게다가, α-cubebenoate는 mouse peritoneal macrophage 내에 LPS로 유도된 JNK의 활성을 저해시켰다. 하지만 ERK, p38 MAPK는 억제하지 못 했다. 또한 LPS에의한 복막염 모델에서 α-cubebenoate는 복막 내 다형핵 백혈구의 축적을 상당하게 감소 시켰다. α-cubebenoate는 LPS에 의해 증가된 iNOS, COX-2를 농도 의존적으로 저해시켰고 그 때문에 peritoneal macropage의 NO, PGE2의 생성을 억제한 것이다. α-cubebenoate는 또한 LPS로 유도 된 복막염 모델에서 다형핵 백혈구 축적을 저해시켰다. α-cubebenoate는 항염증을 통해 Schisandra chinensis의 항피로 성분의 역할을 하며 염증성 질환 치료로 사용 될 수 있을 것이다.
3. 제3세부과제
○ 머위 잎에서 알레르기성 천식에 효과적 치료제로 알려져 있는 petasin을 효율적으로 추출하기 위해 반응표면분석법을 사용하여 petasin 추출공정 최적화를 실시하였다. 추출물의 petasin 함량은 에탄올 농도, 추출 rpm, 추출 시간의 유의적인 영향을 받으며 특히, 에탄올 농도의 영향이 크며 이의 수준이 높아짐에 따라 증가하는 것으로 나타났다. 추출조건에 따른 petasin의 거동은 유의적인 회귀식으로 설명이 가능한 것으로 확인되었다. 반응표면분석 결과, 머위 잎으로부터 petasin의 추출에 최적인 조건으로 에탄올 농도는 79.92%, 추출 rpm은 178.10, 추출시간은 2.06 hr으로 각각 결정되었다.
○ 머위(Petasites japonicus) 잎에서 알레르기성 천식 억제 효능이 있는 bakkenolide B의 최적추출공정을 반응표면분석법을 이용하여 결정하였다. Bakkenolide B의 추출함량은 추출온도, 시간, pH의 유의적인 영향을 받았으며, 추출온도 127.1℃, 시간 46.6분, 추출용매의 pH 7.7에서 최대치 121.6 μg/g이 예측되었다. 비만세포의 탈과립반응 억제효과의 회귀분석 결과는 유의성이 인정되지 않았고 낮은 상관계수를 보였다.
○ 머위(Petasites japonicus) 잎의 유독성분인 pyrrolizidine alkaloids 성분의 함량감소를 위한 방법을 조사한 결과, pyrrolizidine alkaloids 의 한 종류인 senecionine은 머위 속에 상당한 양이 함유되어 있음을 알 수 있었고 이 성분은 열과 알칼리에 약한 성분임을 알 수 있었다. 그러나 senecionine 성분을 감소시키는 가장 효율적인 방법은 화학적 처리보다 열풍 건조방법이 효과적이며 적용 가능한 방법이었다.
○ 머위 30~70%, 수세미 10~30% 및 어성초 10~30%의 비율로 혼합하여 중앙점 반복을 포함하여 총 12개의 실험점을 설계하였고, 이 설계를 바탕으로 혼합물은 121℃에서 45분간 중탕 추출을 하여 항알레르기 및 항균효과를 측정한 결과, 항알레르기 효과는 Response surface와 trace plot을 보면 머위의 함량이 많을수록 좋은 효과를 나타내었고 수세미는 다소 약한 영향을 미친다는 것을 알 수 있었다. 다중회귀분석 결과 상관계수 R2=82.10%이며 유의적인 회귀식으로 설명이 가능한 것으로 확인되었다. 따라서 최적비율은 머위 0.75%, 수세미 0.14% 그리고 어성초 0.11%의 비율로 결정되었다. 항균효과는 혼합비율에 따른 각 추출물들이 그람양성균인 Staphylococcus aureus (KCCM 40881), Staphylococcus epidermidis (KCCM 35494)에 대해서는 유효하나 그람음성균인 Escherichia coli (KCCM 11234), Pseudomonas aeruginosa (KCCM 11328)에 대해서는 미미하거나 효과가 나타나지 않았다.
4. 협동과제
○ 알레르기성 천식 완화에 효능을 가지는 기능성식품의 원료배합비와 제조공정을 제3세부과제와 협동하여 최적의 공정을 완성하였으며, 이에 따라서 1, 2 및 3차의 시작품을 제작하고 각 시작품의 관능적 품질 검사결과를 토대로 원료배합과 제조공정을 개선하여 최종 제품을 제조하였다.
○ 머위의 기능성분을 알코올로 추출하고 추출물의 건조분말을 껌의 기능성 원료로 첨가한 껌 1종을 개발하였으며, 최근에 유행하고 있는 알레르기성 비염에 효능이 있는 음료제품을 개발하고자 머위에 항균성이 우수한 어성초를 부원료로 첨가한 음료제품을 개발하였다.

○ In Vitro Anticancer Activity of Hexane and Ethyl acetate Extracts from Leaf, Petiole and Rhizome of Petasites japonicus

○ In Vitro Anticancer Activity of Hexane and Ethyl acetate Extracts from Leaf, Petiole and Rhizome of Petasites japonicus
Wild Petasites japonicus, which is widely distributed in Korea, Japan and China, is used as a culinary vegetable and a traditional medicine for treatment of several human diseases, including asthma, migraines and tension headaches, and allergic rhinitis. In this study, twelve extracts from the leaves, petioles and rhizomes of P. japonicus were evaluated in vitro for their antiproliferative activity against AGS (human gastric cancer), HepG2 (human hepato cellular cancer), HCT-116 (human colorectal cancer), and MCF-7 (human breast cancer) cell lines. Plant extracts were fractionated using hexane, EtOAc, BuOH and distilled water, after which the antiproliferative activity was measured by MTT assay. Cell viability decreased in a dose dependent manner in response to the hexane and ethyl acetate extracts for all cell lines, with a significant decrease in cell viability being observed for 100 μg/ml and 75 μg/ml in the AGS and HepG2 cancer cell lines. The leaf extract appeared to be more potent against HepG2, as indicated by IC50 values of 4.31 μg/ml for approximately 2.5- and 6.3-fold compared with rhizome and petiole extracts, respectively. The IC50 values of hexane extracts from leaves and rhizomes against AGS, HepG2, and MCF-7 human cell lines were greater than those of other extracts, while ethyl acetate extracts from petioles showed higher significant effect for all cell lines. Hexane and EtOAc extracts exerted their antiproliferative activity in four cancer cell lines. Because of the rich natural supply, simple extraction procedure and high yield, further studies of the LH of wild P. japonicus are warranted. Overall, the results indicate that P. japonicus has the potential to be developed into selective anticancer nutraceutical and/or pharmaceutical treatments with low cost.
○ Isolation, Quality Evaluation, and Seasonal Changes of Bakkenolide B in Petasites japonicus by HPLC
The leaves of Peatasites japonicus are a traditional oriental medicine with diverse biological activities. A simple and specific analytical method for the quantitative determination of bakkenolide B constituents from methanolic extract of the leaves of P. japonicus was developed. Bakkenolide B was isolated from the leaves of P. japonicus, and its structure was elucidated based on 1D, 2D NMR, and GC-MS spectral data. A liquid chromatographic method was developed to evaluate the quality of P. japonicus through determination of major active compound, bakkenolide B. The wavelengths at 254 and 215 nm were chosen to determine bakkenolide B. The recovery of the method was in the range of 98.6 to 103.1%, and bakkenolide B showed good linearity (r2=0.999) within test ranges. The developed method was applied to the determination of bakkenolide B in the plant part and seasonal changes. The results showed that the content of bakkenolide B in the leaf was higher than in the petiole and rhizome. In this study, a simple, rapid, and reliable high-performance liquid chromatography method was used to determine the percentage and composition of bakkenolide B in P. japonicus procured from different Petasites species plants in South Korea. The method can be employed in routine quantitative analysis and quality control of different products in the market.
○ Isolation and structure elucidation of pure compounds from adjunct Houttuynia cordata Thunb and Lufa cylindrica
Three – five pure compounds from Houttuynia cordata Thunb and 2 pure compounds from were obtained from Lufa cylindrica These compounds screen the biological activity including antiallergic and antiinflammatory activity etc.
2. 2nd Reseach Section
○ Anti-allergic and anti-inflammatory effects of bakkenolide B isolated from Petasites japonicus leaves
To elucidate the anti-allergic and anti-inflammatory effects of Petasites genus, we studied the effects of several compounds isolated from Petasites japonicas leaves. Bakkenolide B was isolated from Petasites japonicus leaves. Antigen-induced degranulation was measured in RBL-2H3 mast cells by measuring b-hexosaminidase activity. Induction of inducible nitric oxide synthase and cyclooxygenase 2 was measured by Western blotting in peritoneal macrophages. Ovalbumin-induced asthma model was used for in vivo efficacy test of bakkanolide B. We found that bakkenolide B, a major component of the leaves, concentration-dependently inhibited RBL-2H3 mast cell degranulation. Bakkenolide B also inhibited the gene inductions of inducible nitric oxide synthase and cyclooxygenase 2 in mouse peritoneal macrophages. Furthermore, in an ovalbumin-induced asthma model, bakkenolide B strongly inhibited the accumulation of eosinophils, macrophages, and lymphocytes to bronchoalveolar lavage fluid. Bakkenolide B has suppressive properties for allergic and inflammatory responses and may be utilized as a potent agent for the treatment of asthma.
○ Multiple effects of a novel compound, petatewalide B from Petasites japonicus
To explore the effects of Petasites genus, we isolated a novel compound from Petasites japonicus leaves (a butterbur species). It was found as a new bakkenolide-type sesquiterpene and named petatewalide B. Petatewalide B inhibited the antigen-induced degranulation of b-hexosaminidase in RBL-2H3 mast cells, but did not affect antigen-induced Ca2+ increase in the cells. Petatewalide B also showed inhibition of the LPS-induced induction of iNOS, but not COX-2 in mouse peritoneal macrophages. Nitric oxide production was also inhibited by petatewalide B in macrophages. Petatewalide B increased the membrane potential of C6 glioma cells in a concentration-dependent manner. The current study shows that petatewalide B from Petasites genus not only has anti-allergic and anti-inflammatory effects but also induces a transient increase of membrane potential in C6 glioma cells.
○ Therapeutic effects of s-petasin on disease models of asthma and peritonitis
To explore the anti-allergic and anti-inflammatory effects of extracts of Petasites genus,we studied the effects of s-petasin, a major sesquiterpene from Petasites formosanus (a butterbur species) on asthma and peritonitis models. In an ovalbumin-induced mouse asthma model, s-petasin significantly inhibited the accumulations of eosinophils, macrophages, and lymphocytes in bronchoalveolar fluids. S-petasin inhibited the antigen-induced degranulation of b-hexosaminidase but did not inhibit intracellular Ca2+ increase in RBL-2H3 mast cells. S-petasin inhibited the LPS induction of iNOS at the RNA and protein levels in mouse peritoneal macrophages. Furthermore, s-petasin inhibited the production of NO (the product of iNOS) in a concentration-dependent manner in the macrophages. Furthermore, in an LPS-induced mouse model of peritonitis, s-petasin significantly inhibited the accumulation of polymorphonuclear and mononuclear leukocytes in peritoneal cavity. This study shows that s-petasin in Petasites genus has therapeutic effects on allergic and inflammatory diseases, such as, asthma and peritonitis through degranulation inhibition in mast cells, suppression of iNOS induction and production of NO in macrophages, and suppression of inflammatory cell accumulation.
○ Identification of a novel anti-inflammatory compound, a-cubebenoate from Schisandra chinensis
Extracts of Schisandra chinensis have been used as an anti-fatigue and tonic agent. Because chronic fatigue syndrome is related to inflammatory and oxidativestress, we assessed whether Schisandra chinensis has anti-inflammatory constituents and studied the effect of a novel a-cubebenoate isolated from S.chinensis. a-Cubebenoate was isolated from an extract of Schisandra chinensis fruits. The inductions of inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX-2) by lipopolysaccharide (LPS) were quantified by RT-PCR and Western blotting in mouse peritoneal macrophages. Nitric oxide (NO) and prostaglandin E2 (PGE2) were also measured in the media by Griess reagent and EIA method. A mouse model of LPS-induced peritonitis was used to test the in vivo efficacy of a-cubebenoate. a-Cubebenoate (5 - 10 mg/ml) inhibited the inductions of iNOS and COX-2 in mouse peritoneal macrophages at the mRNA and protein levels. LPS-induced productions of NO and PGE2 were inhibited by a-cubebenoate (5 - 10 mg/ml). In addition, a-cubebenoate inhibited the LPS-induced activation of JNK, but not those of ERK and p38 MAPK in mouse peritoneal macrophages. Furthermore, in the LPS-induced in vivo peritonitis model, a-cubebenoate (1 mg/kg) strongly inhibited the accumulation of polymorph nuclear lymphocytes in the peritoneal cavity. a-Cubebenoate inhibited LPS-induced expression of iNOS and COX-2 in a concentration-dependent manner, thereby suppressing productions of NO and PGE2 in vitro in peritoneal macrophages. a-Cubebenoate also inhibited LPS-induced accumulation of polymorph nuclear lymphocytes in LPS-induced peritonitis model in vivo. a-Cubebenoate may act as an anti-fatigue constituent of Schisandra chinensis through anti-inflammation and could be of therapeutic use as a treatment for inflammatory diseases.

○ In research to optimize the extraction process of petasin from P. japonicus leaves by response surface methodology (RSM), the petasin content was significantly affected by ethanol concentration, extraction rpm, and extraction time, tending to more increase with increasing ethanol concentration. The optimum condition for petasin extraction from Petasites japonicus leaves was 79.92% in ethanol concentration, 178.10 in extraction rpm, 2.06 hours in extraction time, respectively.
○ Petasin extracted from Petasites japonicus leaves has been well known to be effective in the treatment of allergic asthma. This study was carried out to optimize the extraction process of petasin from P. japonicus leaves by response surface methodology (RSM). The dried powder of P. japonicus leaves was extracted at ethanol concentrations ranging from 40% to 80%, extraction rpm ranging from 125 rpm to 225 rpm, and extraction time ranging from 1 to 3 hours. The effects of the extraction conditions on the dry yield and petasin content of the extracts were investigated using a second-order Box-Behnken design. The petasin content was significantly affected by ethanol concentration, extraction rpm, and extraction time, tending to increase more with increasing ethanol concentration. The optimum condition for petasin extraction from Petasites japonicus leaves was 79.92% in ethanol concentration, 178.10 rpm in extraction rpm, and 2.06 hours in extraction time, respectively.
○ Optimal conditions for extraction of bakkenolide B from Petasites japonicus leaves were determined by using response surface methodology. The response surface plot described for bakkenolide B content showed that the maximum bakkenolide B content was predicted as 121.6 μg/g at the extraction conditions of 127.1℃, 46.6 min, and pH 7.7. The extraction temperature and time were important factors in case of bakkenolide B content. The correlation coefficient between the inhibition effect of mast cell degranulation and bakkenolide B content analyzed by the regression equation showed low correlation coefficient.
○ In research to develope the process for reduction of toxic pyrrolizidine alkaloids from Petasites japonicus, heating by using warm air and alkali treatment were effective methods, however, heating by using warm air was more effective and adaptable method in industrial field.
○ To find the optimal ratio of Petasites japonicus, Luffa cylindrica and Houttuynia cordata for development of a functional drink, all of which are supposed to have anti-respiratory diseases effects such as rhinitis, the experiment was designed by mixture design and included 12 experimental points with center replicate for three different independent variables (Petasites japonicus 30~70 %; Luffa cylindrica 10~30 %; and Houttuynia cordata 10~30 %.). Based upon this design, the mixture was extracted in hot water at 121℃ for 45 minutes and observed anti-allergy activity and anti-microbial activity. The response surface and trace plot described for the anti-allergy activity showed that Petasites japonicas was relatively important factor. The correlation coefficient (R2) value 82.10% for the inhibition effect of degranulation analyzed by the regression equation. Analysis of variance showed that the model fit was as statistically significant (p<0.05). The optimal ratio of mixture was Petasites japonicus 0.75%, Luffa cylindrica 0.11% and Houttuynia cordata 0.14 %. Anti-microbial activity for each extraction of mixture was valid on gram-positive such as Staphylococcus aureus ( KCCM 40881 ), Staphylococcus epidermidis ( KCCM 35494 ) while was less effective on gram-negative such as Escherichia coli ( KCCM 11234 ), Pseudomonas aeruginosa ( KCCM 11328).
4. 4th Reseach Section
○ To optimize the optimal process of functional drink easing the allergy asthma, the study was carried out in collaboration with the 3rd Reseach Section. The optimal process was determined by the research result on the material mixture ratio and manufacturing process for the functional drink. Trial products (1st, 2nd and 3rd) were made and carried out the sensory test. As a result of this test, The final product was finally produced by the revised material mixture ratio and manufacturing process.
○ The gum which contained extraction of Petasites japonicus was developed. Functional compound of Petasites japonicus was extracted at ethanol concentrations ranging from 30% to 50% and made in dried powder for the gum.
○ Allergic rhinitis is one of the most rapidly increasing respiratory diseases in modern society. The functional drink with anti-allergy rhinitis effect was also developed by using Petasites japonicus, Luffa cylindrica and Houttuynia cordata, all of which has been known to be effective in anti-microbial activity.
