▣ 국내 주요 젖소 유방염 원인균 및 항생제 내성패턴에 대한 분석
지난 2003년 이후 국내 젖소 유방염 원인균의 분포 및 항생제 내성양상에 관한 문헌보고 분석을 통한 국내 주요 유방염 원인균 및 항생제 내성패턴을 확인하였으며, 2011년 1월-12월 전국 31개 목장, 711건의 유방염 관련 우유검사의뢰 샘플로부터 국내 주요 유방염 원인균 분포 및 항생제 내성양상을 분석완료 하였음

Ⅳ. 연구개발결과
▣ 국내 주요 젖소 유방염 원인균 및 항생제 내성패턴에 대한 분석
지난 2003년 이후 국내 젖소 유방염 원인균의 분포 및 항생제 내성양상에 관한 문헌보고 분석을 통한 국내 주요 유방염 원인균 및 항생제 내성패턴을 확인하였으며, 2011년 1월-12월 전국 31개 목장, 711건의 유방염 관련 우유검사의뢰 샘플로부터 국내 주요 유방염 원인균 분포 및 항생제 내성양상을 분석완료 하였음
▣ 자성나노복합체를 이용한 유방염 원인균의 신속 고효율 진단 기술 개발
국내 젖소 유방염의 주요 원인균종으로 규명된 총 7종에 대한 종 특이적 다클론항체 제작을 완료하였으며, 개발한 항체에 대한 자성 나노복합체를 제작하였음. 또한 다양한 희석배수에 대한 반응성을 비교 분석함으로써, 혼합 및 조성액을 최적화하였으며, 주요 유방염 원인균 표준균주를 활용하여 우유에 인공 접종한 뒤, 시료 전처리 및 자성나노 복합체 적용을 위한 프로토콜을 확립함은 물론 시제품 제작을 완료하였음.
▣ 젖소 유방염 및 항생제 내성 제어를 위한 신규 안티센스 올리고머 발굴
Duffield et al (2010)에 의하여 제시된 바 있는 후보유전자의 down-selection 기법을 토대로 표적인자 후보유전자를 선별한 뒤, 최종 17종의 신규 후보유전자에 대한 선별을 완료하였고, 페니실린(blaZ) 및 메티실린(mecA) 내성유전자 총 2종에 대한 안티센스 올리고머 탐색 및 선발하였음. 또한 후보 안티센스 올리고머의 기능성 검증을 위하여 spot assay 기법을 확립하고, 검사 조건을 최적화하였음.
▣ 세균침습성 펩타이드(bacterium-penetrating peptides) 선별
변성 프라이머와 Invert fusion PCR cloning (IFPC)기법을 이용하여 녹색형광단백질 (GFP)과 translational fusion 시켜 세균침습성 펩타이드 리이브러리 구축시스템을 완성하였고, 총 5,000여개의 세균침습성 펩타이드 라이브러리로부터 유효한 후보펩타이드를 선별하였음. 더불어 이들 후보펩타이드의 기능성 검증을 위하여, HTS (High-throughput screening)을 위하여 다양한 직경의 bead를 활용하여 효율적인 단백질 추출 기법을 확립하였음.
▣ 주요 유방염 원인균에 대한 세균침습성 안티센스 PNA 복합체의 기능성 평가
표준 세균침습성 펩타이드와 PNA올리고머 중합체 (Peptide-PNA)를 제작하였고, 기능성 검증을 위하여 표적유전자 발현침묵효과의 정성분석을 위해 colony counting analysis 및 spot test를 실시하였고, 최소유효농도가 10~20uM 수준임을 확인하였음. 또한 transcriptional level에서의 정성 정량적 분석을 위해 RNA isolation 과 Real time-quantitative PCR을 실시하였으나, 전사 수준에서 별 다른 차이를 확인하지 못하였음. Translation level에서의 발현을 모니터링하기 위하여, cmK::lacZ & deoD::lacZ translational fusion 시스템을 구축하였고, 검증 PNA 중합체가 표적유전자 발현을 용량 의존적인 방식으로 억제함을 확인하였음.
▣ 항생제 내성균에 대한 세균침습성 안티센스 PNA 복합체의 내성제어능 평가
메티실린 내성 유전자(mecA)에 대한 표준 세균침습성 펩타이드와 PNA올리고머 중합체 (Peptide-PNA)를 제작하였고, transcription & post-transcription levels에서의 정성 정량적 분석을 위해 real time-quantitative PCR과 Western 분석을 실시하였음. 병원 내 감염 MRSA의 대표 균주인 N315와 사회감염 MRSA의 대표 균주인 USA300 strain을 이용하여 분석을 실시한 결과, 전사수준보다는 단백질 합성 단계에서 발현 억제를 확인하였음.
▣ 실험동물 감염모델을 이용하여 세균침습성 안티센스 PNA 복합체의 in vivo 방어효능 분석
마우스 복강접종 모델 및 마우스 유방염 감염모델을 확립하였고, 이들 모델에 개발한 세균침습성 PNA 후보물질을 다양한 농도의 유방염 원인균 공격접종에 대한 in vivo 방어효능을 검증하였음.
▣ 세균침습성 안티센스 PNA 복합체의 제형화 및 in vivo 안전성 분석
제형화된 세균침습성 PNA 복합체에 대한 생체내 안전성 분석을 위하여, 마우스 유방염 감염모델을 활용하여, PPNA 과용량 처치군 과 비처치군을 10일간 관찰한 뒤, 일부 조직에 대한 조직병리학적 분석을 수행하였으며, 별 다른 독성을 확인하지 못하였음.
▣ 목적동물을 활용한 임상평가
현장 목적동물에 대한 치료제의 효능을 평가하기 위하여, 5개의 목장으로부터 체세포 50만이상의 급성유방염 감염우의 분방에 개발된 PPNA 제제를 직접 적용한 결과, 체세포의 변화 및 분방 내 세균수의 감소 경향을 확인할 수 있었음.

The project was performed to develop a novel technology for control of bovine mastitis and the related antibiotic resistance using bacterium-penetrating antisense PNAs. To the end, we examined the prevalence and antibiotic resistance profiles of bovine mastitis-causing bacterial agents in Korea during the year 2011 and compared the data to those reported since the year 2003. The seven major causative bacterial agents associated with bovine mastitis in Korea were selected and used to develop the rapid and reliable immuno-separation technique with both the microorganism-specific polyclonal antibodies constructed in this project and the magnetic beads. Staphylococcus aureus is an important zoonotic bacterial pathogen that has been frequently associated with food poisoning and nosocomial infections in humans as well as mastitis in dairy cows. Since the microorganism can easily acquire bacterial resistance to several antimicrobial drugs, novel antibiotics and/or non-antibiotic based anti-infective strategies need to be developed for the intervention of notorious methicillin- or multidrug-resistant S. aureus. Here we report a novel bactericidal peptide nucleic acid (PNA) that can antisense the cytidine monophosphate (CMP) kinase of S. aureus, a putative essential component for bacterial species. Based on the genome sequence of S. aureus N315, a set of PNA conjugates with a bacterial penetration peptide [(KFF)3K-L-] were synthesized to antisense the seven potentially essential genes (cmk, deoD, ligA, smpB, glmU, pyrH, and ftsA) and further evaluated for their antibacterial properties in vitro as well as in vivo. Our experimental analyses demonstrated that two peptide conjugated-PNAs (P-PNAs) targeting either cmk or deoD genes, Pjyh-cmk1 and Pjyh-deoD1, had the strongest growth inhibitory effects against S. aureus ATCC 29740 (a bovine mastitic milk isolate) in a dose-dependent manner. Using a translational fusion system with the lacZ reporter gene, such an inhibitory effect was due to the antisense effect against the target genes rather than the antigene effect. In vivo application of Pjyh-cmk1 resulted in the significant reduction of bacterial loads in the intra-peritoneally infected mice with a sublethal dose of S. aureus at 20 hrs post-infection. Moreover, Pjyh-cmk1 could dramatically increase the survival rate of the mice lactated after intra-mammary infection as well as seemed to be effective when applied to the mastitic cows with severe intramammary infection. Our characterization of Pjyh-cmk1 demonstrated the bactericidal activity against S. aureus, an important zoonotic bacterial pathogen, as well as in vivo effectiveness. This is another demonstration implying possible application of the antisense P-PNA as an alternative anti-infective agent. Taken together, we developed a novel technology to control bovine mastitis and its related antibiotic resistance using bacterium-penetrating antisense PNAs as well as the rapid immuno-magnetic separation techniques against the major bacterial pathogens associated with bovine matitis in Korea. The developed anti-infective and diagnositic tools can be applicable for the clinical practice.
