Ⅳ. 연구개발결과
○ 안토시아닌 고함유 오디 물 추출물을 이용한 고부가가치 제품 개발
- 안토시아닌 고함유 오디 열수 추출물을 이용한 음료는 가용성 고형물, pH, 색도를 측정하여 물 1 L에 오디 30 g을 첨가하여 60분 열수 추출한 추출물을 음료 베이스로 결정하였고, 음료 첨가물은 관능 평가를 통하여 0.63% 매실 농축액, 0.13% 자몽 농축액에 0.08% 오디향(해그린, 오디향592778)을 첨가하여 최종 오디 음료를 개발하였다. 오디음료는 저장성 평가를

Ⅳ. 연구개발결과
○ 안토시아닌 고함유 오디 물 추출물을 이용한 고부가가치 제품 개발
- 안토시아닌 고함유 오디 열수 추출물을 이용한 음료는 가용성 고형물, pH, 색도를 측정하여 물 1 L에 오디 30 g을 첨가하여 60분 열수 추출한 추출물을 음료 베이스로 결정하였고, 음료 첨가물은 관능 평가를 통하여 0.63% 매실 농축액, 0.13% 자몽 농축액에 0.08% 오디향(해그린, 오디향592778)을 첨가하여 최종 오디 음료를 개발하였다. 오디음료는 저장성 평가를 통해 안전성을 확인하였다.
- 최적조건 안토시아닌 함유 물 추출물을 이용한 오디 주스는 가용성 고형물, pH, 색도, 안토시아닌 색소를 측정하여 물 900 mL에 오디 100 g을 첨가하여 15℃에서 8시간 추출한 추출물을 주스 베이스로 결정하였고, 첨가물은 관능평가를 통하여 첨가당은 고과당, 산 비율은 비타민 C : citric acid = 70 : 30으로 하여 최종 오디 주스를 개발하였다. 오디 주스는 저장성과 관능평가를 통하여 안전성과 기호성의 적합성을 확인하였다.
- 비가열처리 오디 주스는 관능평가를 통하여 오디 함량 40%에 여러 가지 과일즙(배, 포도 : 파인애플, 골드키위 = 2 : 1)을 첨가하여 최종 배합비를 완성하였고, 최종 비가열처리 오디 주스는 저장성과 관능평가를 통하여 안전성과 기호성을 검증하였다.
- 비가열처리 오디 주스의 살균 조건은 Ohmic heating을 사용하여 75℃에서 1분간 처리되는 것으로 결정하였으며, 저장성 평가를 통하여 안전성을 검증하였다.
- 오디의 열수 추출물과 여과 추출물을 관능평가를 통하여 비교한 결과 열수 추출물을 푸딩액으로 결정하였으며, 200 g의 오디에 1 L의 물을 넣고 60분간 추출한 추출물 80%에 판젤라틴, 백설탕, 사과농축액, 자몽농축액, 파인애플농축액, 함수결정포도당, 구연산, 오디향을 배합비에 따라 첨가하여 70℃이하에서 모든 재료들을 완전히 용해한 후 오디를 첨가하고 4℃에서 24시간동안 겔화시켜 오디 푸딩을 개발하였고 관능평가를 통하여 기호성을 검증하였다.
○ 오디를 이용한 농축액 개발 및 농축액을 이용한 고부가가치 제품 개발
- 오디 500 g에 물 2 L를 첨가하여 추출한 후 30℃, 40℃, 60℃에서 70 °Bx까지 농축했을 때, 평균 6시간, 4시간, 2시간 30분이 소요되었다. 안토시아닌 색소를 흡광도로 측정한 결과, 낮은 온도에서 고함량 안토시아닌 함유 농축액의 제조가 가능하나, 추출시간과 경제성을 고려하여 30℃를 최적온도로 결정하여 오디의 고함량 안토시아닌 함유 농축액을 개발하였다.
- 오디식혜는 관능평가를 통하여 5% 오디 농축액을 첨가하여 개발하였다. 오디 식혜는 이화학적 특성, 저장성, 관능평가 및 항산화 평가를 통하여 안전성과 기호성 및 기능성을 확인하였다.
- 유효성분 최소 손실 오디 농축액 살균 조건은 Ohmic heating을 사용하여 75℃에서 1분간 처리되는 것으로 결정하였으며, 저장성 평가를 통하여 안전성을 검증하였다.
○ 오디를 이용한 원형유지 병조림 개발
- 오디 병조림 당침액은 관능평가 및 이화학적 특성 측정을 통하여 설탕 : 솔비톨 = 50 : 50, 비타민 C : citric acid = 70 : 30, 젤란검 : 잔탄검 = 70 : 30을 최종 배합비로 결정하였으며, 원형 유지 오디 병조림의 저장성과 관능평가를 통하여 안전성과 기호성을 확인하였다.
○ 여러 건조 조건을 이용한 오디 고부가가치 제품 개발
- 감압건조 기술을 이용하여 개발된 건조 오디는 색도와 조직감 측정을 통하여 3-4% 수분함량과 갈변 현상이 일어나지 않는 75℃, 48시간을 최적조건으로 결정하였고 천연 오디스낵의 저장성과 관능평가를 통하여 안전성과 기호성을 확인하였다.
- 반건조 오디는 관능평가를 통하여 설탕 : 솔비톨 = 70 : 30, 비타민 C : citric acid =50 : 50로 배합비를 결정하였으며, 이화학적 특성 및 관능 평가를 통하여 35℃에서 12시간 열풍건조하는 조건을 확립하였고 저장성과 관능평가를 통하여 안전성과 기호성을 확인하였다.
- 오디 물 추출물을 농축하고 동결건조하여 입자크기에 따라 품질평가를 통하여 식품첨가물 등으로 활용 가능한 오디 건조분말 개발하였다. 오디 건조분말은 수분흡수지수와 수분용해지수 입자크기의 측정을 통하여 입자 크기가 가장 작은 분말로 결정하였다. 입자크기에 따른 오디 건조분말을 이용한 음료를 제조하여 관능평가를 측정한 결과 역시 입자 크기가 가장 작은 분말의 기호도가 가장 높게 나타났고 최종 식품첨가물 등으로 활용가능한 오디 건조분말은 입자크기가 가장 작은 60 mesh로 결정하였다.
- 오디의 동결건조 분말을 비율별로 첨가한 면은 72시간동안 자연건조시켜 건면을 제조하였으며 오디 분말 첨가 반죽의 호화 특성, rheology 특성과 오디 생면의 이화학적 특성과 관능평가를 통하여 오디 동결건조 분말을 1% 첨가하는 것이 가장 좋은 것으로 나타났다.
○ 오디를 이용한 다양한 종류의 소스 개발
- 오디과육을 첨가한 샐러드용 소스는 이화학적 특성과 관능평가를 통하여 오디 과육의 함량을 15% 첨가하는 것으로 최종 결정하였고, 샐러드용 소스는 최종적으로 저장성과 관능평가를 통하여 안전성과 기호성을 검증하였다.
- 오디즙을 첨가한 스테이크용 소스는 이화학적 특성과 관능평가를 통하여 오디즙의 함량을 30% 첨가하는 것으로 최종 결정하였고, 스테이크용 소스는 최종적으로 저장성과 관능평가를 통하여 안전성과 기호성을 검증하였다.
- 오디 콩포트(시럽용) 소스는 이화학적 특성과 관능평가를 통하여 설탕10 : 아가베시럽5: 솔비톨5로 당 배합비를 결정하였으며, 콩포트 소스는 최종적으로 저장성과 관능평가를 통하여 안전성과 기호성을 검증하였다.
○ 오디를 이용한 타블렛(정)
- 오디를 이용한 타블렛(정)은 붕해성, 이화학적 특성, 파손강도, 흡습성 및 관능평가를 측정하여 비타민 C를 10% 첨가한 오디 타블렛을 개발하였다.
○ 오디를 이용한 식품 첨가물 등으로 활용 가능한 천연색소 개발
- 천연색소 개발을 위한 오디의 최적 추출조건은 산첨가와 추출온도에 따른 이화학적 특성과 항산화 특성을 측정하여 25℃에서 citric acid를 0.1% 첨가하여 추출하는 것으로 결정되었다. 천연색소의 소재화를 위하여 오디의 최적 추출조건에 따른 안토시아닌 수율을 측정하였으나 안토시아닌 함량이 너무 극소량 존재하여 오디를 이용한 천연색소의 소재화는 경제성이 없는 것으로 판단되었다.
- 초음파 처리를 통한 천연색소 추출 효율성 증대 기술은 초음파 처리 시간에 따른 안토시아닌 색소 흡광도, 총 안토시아닌 함량, 총 페놀 함량 및 DPPH radical 소거능을 측정하여 5초 간격 초음파로 처리한 조건으로 하였다.
○ 오디의 혈액 순환기계 관련 효과 검증
- 오디 추출물의 ACE 저해 활성 측정 결과, 50% 에탄올 추출물, 산첨가 추출물 및 에틸아세테이트 분획물 중 에틸 아세테이트 분획물 10(133.93%)는 모든 시료 중 가장 높은 ACE 저해 활성을 나타내었다.
- 오디의 HMG-CoA reductase 저해 활성 측정결과, 50% 에탄올 추출물, 산첨가 추출물, 에틸 아세테이트 분획물, 부탄올 분획물 및 에탄올 분획물중 에탄올 분획물 5(133.21%)이 가장 높은 HMG-CoA reductase 저해 활성을 나타냈다.
- F1B Golden Syrian hamster를 이용한 오디의 항동맥경화 효과를 검증하기 위해 실험기간 중 체중변화를 조사한 결과, 오디 추출물을 투여하지 않은 동맥경화유발 사료만을 섭취한 AC군의 체중은 정상대조군에 비해 실험이 진행되는 동안 가장 높게 증가하였으며 1000 mg/kg 오디 추출물 투여군은 체중 감소 효과를 나타냈다.
- 오디 추출물이 동맥경화를 유발시킨 햄스터의 혈중 지질성분에 미치는 영향을 알아보기 위해 혈중 중성지방, 총콜레스테롤, HDL 콜레스테롤 및 non HDL 콜레스테롤 수치를 분석한 결과, 1000 mg/kg 오디 추출물을 투여한 S10은 동맥경화유발 식이를 섭취한 AC군에 비해 낮은 수치를 나타냈다. 오디 추출물의 투여가 동맥경화를 유발한 햄스터의 장기무게에 미치는 영향을 알아보기 위해 간, 심장, 신장 및 비장을 적출하여 무게를 측정한 결과, 간의 무게는 NS10군이 AC군에 비해 1.2배 낮게 나타났다.
제2세부: 오디의 고품질 유지를 위한 수확후 관리기술 개발
Ⅳ. 연구개발 결과
◦수확현장에서의 처리기술개발
- 오디의 유통 중 품질 손상율을 조사하였던바 외형적 손상 발생율은 평균 6.67-20.0%범위로, 대부분 오디 과일 중간부위의 손상이 다른 부위에 비해 압도적으로 높았다. 이러한 결과는 오디의 수확은 수작업으로 완숙과를 위주로 수확하며, 수확용기의 부적합, 작업자의 손에 의한 오디 표면의 손상이 주요인으로 분석되었다. 이를 방지키 위해서는 작업 시 사용하는 장갑, 수확용기의 개선이 필요한 것으로 나타났다. 작업자의 장갑은 가능한 얇고 작업이 편리한 라텍스 계통의 얇은 장갑이, 수확 용기는 기존 4-5리터의 바스켓타입의 용기 보다는 작은 바구니가 보다 효과적인 것으로 조사되었다.
- 수확된 오디는 선별 포장 과정을 거치는데, 이를 위한 수확후 현장 대기시간이 길고, 선별시 용량이 큰 수확용기로부터 소분하는 과정을 거침에 따라 오디의 과육 손상이 크게 발생한다. 이를 개선키 위해서는 수확, 선별 포장 단계를 일원화하여, 이러한 단계를 줄이기 위해 수확 시 유통용 소포장 상자를 직접 사용하는 것이 바람직한 것으로 판단되었다.
- 오디의 과육 손상 정도는 오디의 숙도와 밀접한 관계가 있는데 숙도가 높을수록 손상발생률이 높아, 숙도가 80%정도인 오디는 0.3-1.0%로, 90% 숙도 오디의 2.8-4.7%, 100%숙도 오디의 4.4-11.1%에 비해 손상율이 매우 낮았다. 숙도에 따른 오디의 기호도를 조사였던바 중·장년층은 완숙과를, 젊은 층은 90% 정도 숙도의 오디를 선호하였다. 따라서 100%인 완숙과보다는 90% 정도 숙도의 오디를 수확하는 경우 과피 손상을 상당히 줄일 수 있으며, 포장작업시 조직 손상이 덜 발생하며, 유통기간도 완숙과에 비해 더 연장되는 잇점을 얻을 수 있는 것으로 판단되었다.
- 조사결과, 오디는 수확 후 이송, 선별, 포장 과정을 거치는 시간은 오디의 수확량 대비 선별포장 처리량과 포장 방법에 의해 결정되는데 대부분 수확후 30분-1시간 이내 포장작업이 이루어지며, 포장후 냉장시설까지 이송되는데 소요되는 시간은 최소 1시간 최대 4시간에 걸렸다. 수확된 오디의 품온은 외기의 온도보다 1.2-1.4℃ 정도 낮았고, 과수원에서 선과장으로 이송하기 전 단계에서는 외기보다는 0..5-0.8℃정도 높았다. 또한 수집된 오디가 간이 선과 및 포장장에서 대기하는 동안의 품온은 수확지에서 이송되는 시점의 온도보다 0.8-1.2℃정도 높은 수준을 유지하였다. 또한 선별 작업 전 방치되는 동안 오디 용기 내 품온은 외기보다는 오디의 품온이 1.3-2.0 ℃ 높게 유지되었다. 또한 포장 후 처리장 이동단계에서는 외기보다 2.8℃정도 높았다.
- 오디의 품온 관리를 위해서는, 일정 규모의 이동식 기계적 냉각시설을 오디 수확현지에 갖추는 것이 필요할 것으로 판단된다. 소규모 오디 생산농가의 경우에는 시설비용 및 시설운용이 여의치 않으므로 대형 스티로폼 보온상자의 적용이 적합할 것으로 판단된다. 이 경우 보온상자 내 저온 유지를 위해 PCM (아이스팩)의 사용이 필수적이다. 실증실험으로 현행 사용하고 있는 보냉 상자에 6kg의 오디와 4개의 아이스 팩을 넣은 후 밀봉하여 차량위에 방치하였던 바 3시간 30분간은 외부보다 최대 7℃정도 낮게 유지시킬 수 있었다.
◦생과용 오디의 유통기간 연장기술 개발
- 오디의 적정 저장온도 연구를 위하여 1차적으로 실용적인 측면에서의 저장온도대인 0, 10 및 20℃에서 품질변화를 조사하였고, 아울러 각 저장온도에서 오디 숙도(80, 90, 100%)별 저장성을 비교하였다. 이 실험결과를 바탕으로 저온저장을 위한 최적 조건 설정을 위하여 오디의 숙도별 빙결온도를 조사하였다.
- 오디의 품질 유지를 위한 저장 온도 설정을 위하여 오디를 20℃, 10℃ 및 0℃에서 저장하면서 저장온도에 따른 미생물, 부패율, 관능적 품질의 변화를 조사하였던바 20℃와 10℃에 비해 0℃에서 곰팡이의 성장이 적었으며, 20℃와 10℃에서 2일, 6일째 완전한 부패가 일어난 반면 0℃에선 12일째에 일어났다. 또한 보다 더 적합한 온도 설정을 위하여 0℃근처의 1.5℃, 0℃, -1.5℃로 온도를 재설정 하여 25일간 저장하면서 품질의 변화를 조사하였던 바 총균과 곰팡이, 부패율은 저장기간에 따라 -1.5℃<0℃<1.5℃ 순으로 높은 값을 나타내었다. 관능적 품질평가를 기준으로 1.5℃, 0℃, -1.5℃중 가장 낮은 온도인 -1.5℃가 다른 온도 보다 오디의 품질 유지에 효과적인 것으로 판단되었다.
- 수확 후 오디의 선도연장을 위한 active MA처리효과 조사로 포장 오디 용기 내 탄산가스를 10%, 20% 및 30% 주입 처리하여 밀봉하였던바, 오디의 총 균 및 곰팡이는 통기 포장구의 경우 저장 중 점차 증가하였으나, 밀봉 포장구와 탄산가스 주입 포장구의 경우 그 값의 변화는 미미하였다. 상품성을 관능적으로 평가하였던바 통기 포장구는 10일간, 밀봉 포장구와 탄산가스 포장구는 저장 25일 후까지 상품성을 유지하였다.
- 소포장 Active MAP 기술을 오디 수확현장에 적용하기 위하여 관행적인 플라스틱 용기(1kg)에 일정량의 드라이아이스와 오디를 넣거나 이를 진공 포장하여 수확현장 상온에서 2일간 보관하면서 품질변화를 조사한 결과, 드라이아이스를 넣고 진공 포장한 처리구가 관능적으로 가장 우수하였으며, 부패율은 대조구가 50%이었던 비해 드라이아이스 포장구에서는 약 18%, 드라이아이스와 진공 포장구에서는 거의 발생하지 않아 본 실험에서 시도된 드라이아이스와 진공포장구 포장방법이 오디 수확현장에 적용할 수 있는 적합한 active MAP 포장조건으로 여겨졌다
- 오디 포장용기의 구조를 개발하기 위하여 오디의 포장 내 축적 정도에 따른 손상 발생률을 조사하였다. 포장 내 오디의 축적 정도가 바닥으로부터 6cm 정도까지는 손상과 발생과 손상된 조직으로부터의 과즙 누출량이 거의 유사한 수준이었으나, 이 이상 높이에서는 손상과 발생과 손상된 조직으로부터의 과즙 누출량이 유의적으로 차이를 보였다. 따라서 유통량이 가장 많은 1Kg단위로 포장할 경우 포장용기 바닥으로부터 최대 6.5cm정도 까지만 오디를 넣을 수 있도록 구조를 변경하여야 하며, 이를 위하여서는 포장상자의 밑면을 보다 넓게 하여야 할 것으로 판단된다. 아울러 생 오디 포장단위를 500g 또는 250g 정도로 하는 것도 상자 내 오디 자체의 하중으로 인한 조직 손상 발생을 방지할 수 있는 방안이라 판단된다.
- 오디의 운송 중 외포장내로의 외부의 열이 침투하는 것을 방지하키 위하여서는 오디의 운송시 냉장차량을 사용하는 것이 우선되어야 할 것으로 판단된다. 이와 더불어 오디의 외포장용 스티로폼 상자의 열전도를 보다 낮추고 압축강도를 보다 높인 재질을 사용하는 것이 바람직하다. 따라서 현재 오디의 외포장재로 사용되고 있는 스티로폼을 비드법 1종 제품이 주를 이루며 초기 열전도도는 0.037W/m·K 이상으로 이보다 열전도도가 낮은 재질의 제품을 사용하는 것이 바람직하다.
- 예비실험 결과를 기반으로 수확현장에서 적용할 수 있는 신속저온처리기술 개발을 위해 오냉매제와 순환 fan을 장착한 EPS 상자를 고안하였다. 적정 냉매제를 선발하기 위하여 아이스팩(600-700g, 3 packs)과 드라이아이스(EPS 상자부피의 40%)를 단독 또는 병용하여 EPS상자에 담은 후 일정량의(1 kg, 500 g) 오디를 넣고 24시간이 경과한 후 품질을 측정하였다.
관능적으로는 아이스팩과 드라이아이스 병용처리구>드라이아이스 단독처리구>아이스팩 단독처리구>대조구 순으로 향, 맛, 조직감, 외관, 전체적인 기호도 항목에서 우수하였고 부패율도 냉매제 병용처리구에서만 10% 이하로 가장 낮았다.
◦오디의 냉동처리 및 냉동 저장기술개발
- 직접소비용 오디의 경우 과립형태가 중요한 품질인자임에 따라 오디의 빙결온도를 조사하였던바 숙도에 따라 약간의 차이를 보였지만, –1.8~—2.9℃ 범위이었다. 또한, -10, -20, -40℃의 온도에 따른 오디의 빙결온도를 조사하였던바 빙결되는 온도는 거의 유사하였으며 냉동온도가 낮을수록 빙결에 이르는 시간이 짧았다. 이러한 특성을 이용하여 오디를 개체로 냉동시켜 과립을 유지하는 것은 실험실적으로는 가능하였으나, 오디의 조직이 매우 연하여 췩급횟수가 중가하면 할수록 조직의 손상이 심하여 실용화 측면에서는 의미가 적은 것으로 판단되었다.
- -25℃에서 오디를 동결처리 시 -23℃에 도달하는 시간이 개체별로 처리한 경우 1시간 20분 이내에, 250g용기를 사용한 경우 7시간 30분에서 9시간 30분, 1kg 단위의 경우 약 14시간이 각각 소요되었다. 동결 시 포장용기의 뚜껑을 열어 놓은 경우가 밀폐 용기 및 통기구 개설 용기 보다 그 속도가 빨랐으며, 용기의 크기가 클수록 동결 소요시간도 증가하였다. 동결 방법별 외관 및 동결처리 중 오디로 부터 누출된 과즙의 발생정도를 비교하였던바 오디를 낱개로 동결처리 한 경우가 외형이 가장 우수하였고, 과즙의 누출정도는 가장 적었다. 반면에 용기 포장의 경우 용량이 큰 용기일수록 외형변화 및 과즙 누출 정도가 비교적 컸다. 500g 단위의 경우 외형은 250g 단위에 비해 다소 떨어졌으나 과즙의 누출정도는 250g 단위포장과 차이를 보이지 않았다. 한편, 오디를 기존 대형 플라스틱 용기로 수확한 후 포장 단계에서 250g 단위로 소포장한 경우 외관의 매우 불량하였고, 과즙의 누출 정도 역시 매우 심각하였다.
- 가공용 오디의 적정 동결처리기술 연구로 각각 2.5kg 플라스틱 상자, 10kg 스티로폼 상자, 10kg P 박스로 구분하여 포장한 후 –24℃ 냉동보관 하였으며, 추가로 드라이아이스를 포함한 2.5kg 플라스틱 상자와 10kg 스티로폼 상자에 각각 오디를 나눠 담아 드라이아이스 처리에 따른 효과를 조사하였다. 오디를 -24℃에서 동결 처리 시 2.5kg 소포장이 다른 포장에 비해 가장 빠르게 냉동고 외기에 도달하였고, 그 다음은 10kg-P박스, 10kg-스티로폼상자가 뒤를 이었다. 오디의 미생물은 저장기간에 따라 총균과 곰팡이 모두 감소하는 경향을 보였는데 드라이아이스 처리 유무와 포장 방법에 따른 차이는 미미한 반면 시료의 생산 및 취급농가에 따라 보다 큰 차이를 보였다..
- 냉동 오디의 저장 중 품질열화를 억제키 위한 연구결과, 냉동속도 보다는 용기의 크기를 작게 하는 것이 효과적이었으며, 이와 더불어 원료자체가 건전하도록 수확 및 포장 단계에의 처리가 중요한 것으로 판단된다.
- 또한, 냉동오디의 품질 및 온도관리를 위한 포장 기술 확립을 위해 오디를 포장 냉동 후 드라이아이스와 진공 처리하여 품질을 비교하였던 바 대조구와 드라이아이스 처리구와 진공처리구간에는 관능적으로 외관상 차이가 뚜렷하게 나타나지 않았으나, 냉동 전 오디의 상태가 더 큰 영향을 미치는 것으로 조사되었다.

Ⅳ. 연구개발결과
1. 세포독성 및 항산화 활성 효능평가
가. 췌장 베타세포 독성평가 및 농도분석
세포는 mouse insulinoma 인 MIN6N 세포를 선정하였으며 DMEM 배지에 fetal bovine serum을 10% 농도로 첨가하여 배양하였다. 분획별 오디 추출물의 독성 평가에서는 MAE(EtOAc 추출물) 500 μg/ml 에서 세포생존률 84% 이상을 유지하면서 가장 적은 세포독성을 보였으며 MAB (n-BuOH 추출물)는 80.8%, MAH (H2O 추출물)는 75%, MA (Et-OH) 69.9% 의 세포독성을 확인하였다. 모든 분획에서 400 μg/ml의 농도까지는 세포에 큰 영향을 주지 않는다고 판단하였다.
나. 항산화활성 효능평가
이전 실험에 근거하여 세포 독성이 없는 농도내에서 항산화 활성을 DPPH 소거활성을 확인하였다. MA군과 MAH군은 50 μg/ml 농도에서부터 DPPH 소거활성이 크게 증가하여 이후 농도 의존적으로 활성이 증가하였고, MAE와 MAB군은 100 μg/ml 의 농도부터 DPPH 소거 활성이 증가하였다.
2. 세포사멸 및 증식 효능평가
가. 세포 보호능 및 회복능 효능평가
(1) 세포 보호능 효능평가
세포의 산화적 스트레스를 유발하기 위한 H2O2의 농도는 0.7mM의 농도에서 50.7%를 보였으므로 최적 농도는 0.7 mM의 농도를 선정하였으며 분획별 오디 추출물의 전 처리 후 0.7 mM의 과산화수소를 처리하였을 때 MAE군은 50 μg/ml에서 과산화수소 처리군에 비해 약 11% 증가한 세포생존률을 보였고 100 μg/ml의 농도에서 약 20% 증가한 세포생존률을 보이며 가장 높은 세포 보호능을 보였고, MA 군은 50 μg/ml에서 13%, 최대 17% 생존률 증가를 확인하였다. MAB 군과 MAH군은 각각 11%, 14% 증가한 세포 생존률을 보였으나 MA와 MAE에 비해 적은 세포 생존률을 보였다. 이 중 가장 활성이 우수했던 MAE 분획을 용매별 극성과 분자량에 따라 세분화한 14가지 분획으로 나누어 세포 보호능을 진행한 결과 14가지 분획중에 MAE1과 MAE2 군에서 각각 최대 21%, 20%의 세포 생존률을 보이며 가장 우수한 세포 보호능을 보임을 확인하였다. 위 시료군과 별도로 추출용매인 Et-OH 조성을 50%로 변경해 추출한 시료군과 가장 상위분획인 MA군에 각각 유기산 (Citric, Malic, Fumaric acid)을 첨가한 시료군을 추가로 실험하였다.
실험결과 세포 보호능은 보이나 시료군 A와 시료군 B 보다는 미약함을 확인하여 차 후 실험은 시료군 A와 시료군 B를 선별하여 진행하였다.
(2) 세포 회복능 효능평가
효능 비교를 위하여 분획별 오디추출물을 전 처리한 보호능외에 후 처리한 회복능 평가를 진행하였다. MA군은 과산화수소 (H2O2) 처리군에 비해 13% 증가한 세포 생존률을 보였으며 최대 15% 증가율을 보였다. MAE 군은 100 μg/ml에서 과산화수소 (H2O2) 처리군에 비해 11% 증가한 65%의 세포 생존률을 확인하였고, 최대 13%의 세포생존률을 보였다. MAB와 MAH는 이전과 마찬가지로 세포생존률 증가 경향을 보였으나 MA와 MAE에 비하여 낮은 세포 생존률을 보였다. 이러한 결과를 통해 분획별 오디추출물은 세포 회복능 보다는 보호능에서 비교적으로 우수한 효능을 보였다.
나. 세포 내 활성산소 소거능 평가
(1) 과산화물 (ROS) 소거능 평가
과산화수소 처리군에서 ROS의 양이 급격하게 증가함을 확인하였고, MA군에서 농도 의존적으로 과산화물이 소거됨을 확인하였고, 400 μg/ml 에서 약 44%의 감소를 보였다.
MAE군에서도 마찬가지로 농도 의존적으로 과산화물이 소거되었고, 최대 47%의 감소능을 보였다. MAB와 MAH 군에서도 과산화물 소거능이 확인되었으나 MA와 MAE에 비하여 낮은 과산화물 소거능을 보였다. 분획 중 MAE를 세분화한 시료군 B에서는 세포보호능과 마찬가지로 MAE1과 MAE2에서 가장 높은 과산화물 소거능을 보였다. 시료군 C에서도 마찬가지고 소거능을 보였으나 시료군 A와 시료군 B 보다는 미약한 소거능을 보임으로써 차 후 실험에는 가장높은 활성을 보인 시료군 A의 MA, MAE 와 시료군 B의 MAE1, MAE2를 사용하였다.
(2) 과산화 지질 억제능 효능평가
MA군에서는 100 μg/ml에서부터 유의적으로 과산화지질 억제능이 증가하기 시작하여 400 μg/ml 농도에서 26%의 억제능을 보였다. MAE군에서는 최대 30%의 과산화 지질 억제능을 보임으로써 가장 높은 활성을 보였고, MAE1과 MAE2는 각각 30%와 28%의 과산화 지질 생성 억제능을 확인함으로써 결과적으로 이전 실험들을 종합하였을 때 MAE군이 가장 우수한 효능을 보임을 확인하였다. 차 후 실험은 MAE군으로 진행하였다.
다. 세포사멸 저해 효능평가
(1) Apoptosis 저해 효능 평가
Control (18%) 에 비하여 과산화수소 처리 군에서 Apoptosis가 58%까지 증가하였으며 MAE 100 μg/ml 군에서는 과산화수소 처리 군에 비하여 23% 감소한 35%의 apoptotic cell rate를 보임으로써 산화적 스트레스로 인한 췌장베타세포의 apoptosis 진행을 억제하였다.
(2) DNA fragmentation
Control 군에서의 파란색 형광은 약하게 관찰되며 morphology가 정상적인 것을 확인하였다. 과산화수소 처리시에는 DNA fragmentation이 일어나면서 파란색형광이 밀집되어 강하게 관찰되는 부분이 증가하였고, morphology 또한 변형된 모양이 관찰되었다. MAE 100 μg/ml 처리시에는 과산화수소 처리 군에 비하여 형광이 감소되고 Control과 유사하함을 확인하였다. 다른 분획인 MA, MAB, MAH를 처리한 결과도 마찬가지로 산화적 스트레스로 인한 DNA fragmentation이 감소됨을 확인하였다.
3. 메커니즘 규명 및 In vitro 항당뇨 효능평가
가. Western blot
(1) Apoptosis 관련인자 확인
Apoptosis 저해 효능이 어떠한 내부 신호를 경유하는지 확인하기 위하여 Apoptosis 인자인 Caspase-3와 AIF (Apoptosis Inducing Factor)를 단백질 수준에서 확인하였다. 과산화 수소 처리시에 Caspase-3 활성화 form인 Cleaved caspase-3 의 활성이 증가하였고, AIF 또한 발현이 증가함을 확인하였다. MAE 처리 시에 50 μg/ml에서 70 μg/ml 로 농도가 증가 할수록 활성이 감소함을 확인함으로써 Caspase-3의 활성과 AIF 발현에 영향을 주는 것을 확인하였다.
(2) 항산화 효소 활성 (Catalase, SOD) 확인
항산화 효소의 발현을 확인한 결과, 과산화수소 처리시에 SOD (Superoxide Dismutase)와 CAT (Catalase)의 활성이 증가함을 확인하였고, MAE 처리시에 과산화물 소거에 의하여 SOD와 CAT의 발현이 감소함을 확인하였다.
나. In vitro 항당뇨 효능평가
(1) 인슐린 분비능 확인
Insulin secretion에서 Control 에 비하여 과산화수소 처리군에서 Insulin 분비가 약50%까지 유의적으로 감소한 것을 확인하였고, 과산화 수소 처리군에 비하여 MAE 처리시에 농도 의존적으로 Insulin 분비능이 증가하였다.
(2) 포도당 흡수능 평가
Glucose uptake 실험에서는 Control군에 비하여 과산화수소를 처리하였을 때 형광이 감소하면서 포도당 흡수가 감소하였고, MAE를 처리하였을 때 농도 의존적으로 포도당 흡수가 증가하면서 산화적 스트레스로 인하여 손상된 glucose uptake가 MAE에 의해서 회복됨을 확인하였다.
4. In vivo 항당뇨 효능평가
가. 체중 및 장기 무게 측정
(1) 체중 측정
베타세포에 손상을 주는 Streptozotocin (STZ)을 이용하여 ICR mouse에 당뇨를 유발하였고, 유발 후 4 주간 오디추출물 (MAE)을 이틀 간격으로 경구 투여 하였다. Normal군에 비하여 STZ 투여 군에서 체중 감소를 나타내었고, 4주간 MAE를 처리한 실험군에서는 체중감소를 완화시키는 결과를 보였다.
(2) 장기무게 측정
장기 무게의 경우 다른 장기에는 큰 변화가 일어나지 않았지만 STZ를 처리한 실험군에서 비장의 크기가 비대해 지는 경향을 확인하였고, MAE를 4주간 경구 투여한 실험군에서 완화됨을 확인하였다.
나. 혈중 인슐린 및 혈당 측정
(1) 혈중 인슐린 측정
Normal 군에 비하여 STZ를 복강 투여한 군에서 베타세포 손상에 의한 Insulin의 분비가 감소됨을 확인하였고, 4주간 MAE를 경구투여한 실험군에서 회복됨을 확인하였다.
(2) 혈당 측정
Insulin 분비에 따른 혈당의 증감을 확인한 결과, STZ 처리 군에서는 4주차까지 지속적으로 혈당이 증가하였으나 MAE를 4주간 경구투여한 군에서는 STZ 단독처리 군에 비하여 혈당이 감소한 경향을 확인하였다.
다. 혈중 콜레스테롤 및 중성지방 측정
(1) 혈중 콜레스테롤 측정
Normal군에 비하여 STZ를 처리한 군에서 혈중 콜레스테롤의 양이 증가함을 확인하였고, STZ만을 투여한 실험군과 비교하였을 때 MAE를 4주간 경구 투여한 실험군에서 유의적으로 콜레스테롤의 수치가 낮아짐을 확인할 수 있었다.
(2) 혈중 중성지방 측정
STZ만을 투여한 실험군과 비교하였을 때 MAE를 4주간 경구 투여한 실험군에서 유의적으로 중성지방의 수치가 감소하였으므로, 당뇨 유발시에 나타날 수 있는 혈중 중성지방의 증가를 회복함을 알 수 있었다.
5. 추가 연구내용
가. 오디 유래 안토시아닌 분리정제
(1) 에탄올 농도에 따른 추출물의 효능평가
에탄올 용매 비율 100%, 70%, 50%, 30% 따라, 각각 300 μg/ml 의 농도에서 80%, 84%, 79%의 세포 생존율을 보였으며 70% 에탄올 오디 추출물 군은 200 μg/ml의 농도에서 91.5%의 높은 생존율을 확인할 수 있었다. 100%, 70 %, 50%, 30% 에탄올 오디 추출물 군은 70 μg/ml 의 농도부터 DPPH 소거활성이 증가하여 이후 농도 의존적으로 소거활성능이 증가하였으나 에탄올 용매 50%, 30% 의 오디 추출물은 소거활성능 증가폭이 70% 보다 작았다. 70%에탄올 오디 추출물 군은 300 μg/ml 농도에서 68.9% 의 높은 radical 소거능을 보였으며 세포독성이 없는 농도내에서 오디 추출물의 농도 의존적으로 항산화 활성이 증가함을 확인하였다. 과산화물 소거능 평가와 과산화 지질 억제능 평가에서도 마찬가지로 70% 에탄올 오디 추출물이 다른 군에 비해 우수한 효능을 확인하였으므로 차 후 실험은 70% 에탄올 추출물로 진행하였다.
(2) 안토시아닌 분리정제 및 함량 확인
에탄올 용매 비율에 따른 각 분획에서 70% 에탄올 오디 추출물이 391 mg/g 으로 가장 많은 양의 안토시아닌을 가지고 있음을 확인하였다. 이 물질을 정제하여 standard와 시료의 피크를 확인하였을 때 Retention time 이 같음을 확인하여 분리된 정제물질이 Cyanidine-3-glucoside 임을 확인하였다.
나. 오디로부터 분리된 C3G의 항당뇨 효능평가
(1) 세포사멸 및 항산화활성 효능평가
췌장 베타 세포에서 고혈당 유도를 하였을 때 (25 mM glucose, 18 h) 72.9% Cell viability를 보였으며 삼투압 컨트롤으로는 mannitol 25 mM 이 사용되었다. 고혈당에 의해서 감소한 Cell viability를 C3G 70 μg/ml 처리시에 83.8%를 보이며 시료가 세포를 보호하였다. 항산화활성 실험에서는 High glucose 처리군에서 7.6배의 과산화물이 생성 되었고, C3G 70 μg/ml군에서는 6.5배로 과산화물 생성의 감소가 보였으며, 100 μg/ml 에서는 5.4배로 감소하였다. 또한 형광 현미경을 통해 ROS 소거를 가시적으로 관찰하였다. Control의 apoptotic cell rate는 약 4.9%에 비하여 high glucose 처리 시 apoptotic cell rate가 약 33.9% 증가함을 확인하였다. C3G 70 μg/ml 처리 시 High glucose 처리군에 비해 약 9% 줄어든 24.8% 의 apoptotic cell rate를 보이며 High glucose 유도 산화적 스트레스로 인한 췌장 베타세포의 apoptosis 진행을 억제함을 확인하였다.
(2) 세포보호 작용 메커니즘 확인
Apoptosis 와 관련있는 인자인 p-ERK, p-JNK, p-38의 발현이 High glucose 처리된 군이 비해 C3G를 처리한 군에서 발현량이 저해되어 세포를 보호 하는 것을 확인하였다. 또한 anti-apoptotic protein인 Bcl-2이 C3G 처리 군에서 발현이 많이 되어 apoptosis를 저해하였고, pro-apoptotic protein인 Bax의 발현량은 감소하였다. C3G 처리 군에서 cytochrome C의 방출은 적게 일어남을 확인 하였다. 그리고, Cleaved caspase-3의 발현량은 C3G 농도 의존적으로 감소 하여 apoptosis를 억제하였으며, NF-κB의 핵내로의 translocation이 감소함을 확인하였다.
(3) 인슐린 분비능 확인
Control 에 비하여 High glucose 처리군에서 Insulin 분비가 약 74.8%까지 유의적으로 감소하였고, High glucose 처리군에 비하여 C3G 70 μg/ml과 50 μg/ml처리 시에 Insulin 분비능이 각각 78.1%, 84.2%로 증가하였으므로 고혈당으로 저해된 Insulin 분비능이 회복됨을 확인하였다.
제1위탁 : 오디로 부터 색소 및 기능성 소재의 분리 동정 및 표준화
Ⅳ. 연구개발결과
오디로부터 70% EtOH 을 이용하여 기능성성분 추출 조건을 확립하였다. 또한 상기 추출용매에 4종 유기산을 처리하여 안토시아닌을 높은 수율로 추출하는 방법을 확립하였다. 추출물에 대하여 column chromatography를 이용하여 6종의 플라보노이드를 분리, 정제하였다. 각 정제된 화합물에 대해서는 NMR, IR, MS 등의 스펙트럼 데이터를 이용하여 각각 astragalin, quercetin, isoquercetin, rutin, morachalcone A 및 isobavachalcone으로 구조 동정하였다. 제조한 추출물과 분리한 화합물에 대해선 공동연구팀에 활성시료로 제공하였다. 안토시아닌의 저장 효과를 높이기 위하여, 처리하는 유기산의 종류와 적절한 농도를 결정하였다. 오디를 이용한 소재의 표준화를 위하여 주요 활성 화합물 rutin, morachalcone A, artoindonesianin O 및 isobavachalcone에 대하여 HPLC를 이용하여 정량 분석법을 확립하였다.

- Mulberries hydrothermal extracts containing high anthocyanin content were determined by soluble solids, pH and color. The extracts were used as a drink base. Thirty grams of mulberries in 1 L of water were extracted for 60 min. Plum extracts (0.63%), grapefruit concentrates (0.13%), and mulberry scent (0.08%; Haegrin, mulberry scent 592778) were added as drink additives based on sensory evaluations. Safety and suitability of the drink were assessed by storability and sensory evaluations.
- Soluble solids, pH, color, and anthocyanin pigments were measured to determine the optimal conditions for the production of mulberry juice by using the extracts as the juice base. Mulberries were extracted with hot water for 8 h at 15 ℃ by adding 100 g of mulberries to 900 mL of water. High-fructose corn syrup and citric acid with vitamin C (vitamin C: citric acid = 70:30) were added to develop the mulberry juice. Safety and suitability of the juice were assessed by storability and sensory evaluations.
- The optimal proportions of the non-heat-processed mulberry juice were determined by adding pear and grapes: pineapple and gold kiwi in a 2:1 ratio to mulberry (40%) using sensory evaluations. Safety and suitability of the final non-heat-processed mulberry juice were assessed by storability and sensory evaluations.
- The optimal sterilization conditions of the non-heat-processed mulberry juice were determined to be 1 min at 75 ℃ using Ohmic heating. Safety and suitability were assessed by storability and sensory evaluations.
- Sensory evaluations of the hot water and filter extracts determined that hot water extract was optinal for pudding juice. Plate gelatin, sugar, apple concentrate, grapefruit concentrate, pineapple concentrate, hydrocrystalline glucose, citric acid, and mulberry scent were added according to the mix proportions into 80% extract produced by adding 200 g of mulberries to 1 L of water and subjecting the mixture to extraction for 60 min. After all the ingredients were completely dissolved at a temperature below 70℃, the mulberries were added and gelled for 24 h at 4℃. The suitability of the mulberry pudding was assessed by sensory evaluations.
○ The development of mulberry concentrates and high value-added products from concentrates
- When concentrating extracts by adding 500 g of mulberry to 2 L of water at 30℃, 40℃, 60℃, the average time required for concentration was 6 h, 4 h, and 2.5 h, respectively. For the concentrates containing high levels of anthocyanins, 30 ℃ was determined to be the optimal concentrate temperature, and the anthocyanin pigments were measured using absorption.
- The mulberry Sikhye was developed by adding 5% mulberry concentrate, as judged by sensory evaluations. Safety and suitability of the mulberry Sikhye were determined by tests evaluating the physiochemical characteristics, storability, antioxidant activity, and sensory evaluations.
- Ohmic heating for 1 min at 75 ℃ was determined to be the optimal sterilizing condition for the mulberry concentrate with minimal loss of active contents. Safety was determined by storability evaluations.
○ The development of bottled mulberry friuts that preserve the original form of mulberries
- The optimal ratios for bottled sugared solutions were determined to be sugar : sorbitol = 50 : 50, vitamin C : citric acid = 70 : 30, and gellan gum : xanthan gum = 70 : 30 as judged by sensory evaluations and physiochemical property measurements. The bottled mulberry fruits and their safety and suitability were identified through storability and sensory evaluations.
○ The development of high value-added mulberry products using various drying conditions
- The optimal conditions for the vacuum drying of mulberries (for which the end-product had a 3–4% moisture content and did not brown) were 48 h at 75 ℃. Safety and suitability of the natural mulberry snack were identified by storability and sensory evaluations.
- The optimal semi-dry mulberry mix contained sugar : sorbitol in a 70 : 30 ratio and vitamin C : citric acid in a 50 : 50 ratio, as determined by sensory evaluations. The optimal hot air drying conditions were 12 h at 35 ℃, based on the physiochemical properties and sensory evaluations. Safety and suitability of the semi-desiccated mulberry snack were identified by storability and sensory evaluations.
- Dried mulberry powder for use as a food additive was developed using quality evaluations, according to particle size, by concentrating and freeze-drying mulberries.
The powder with the smallest particle size was determined by measuring the absorptive and solubility index particle size. Sensory evaluations of drinks containing the dried mulberry powder revealed that the drink made using the smallest particles was preferable. The particle size of the end product to be used as a food additive was determined to be 60 mesh.
- Dried noodles were produced by adding various amounts of freeze-dried mulberry powder to noodles and drying for 72 h. Evaluation of the pasting and rheological properties of the mulberry powder-added dough and physiochemical and sensory evaluations of the wet mulberry noodles indicated that the 1% mulberry freeze-dried powder-added noodle was optimal.
○ The development of various sauces using mulberries
- The addition of 15% mulberry pulp was preferred for mulberry-supplemented salad dressings, based on physiochemical and sensory evaluations. Safety and suitability of the end product were determined by storability and sensory evaluations.
- The optimal amount of mulberry juice added to steak sauce was 30%, as determined by physiochemical and sensory evaluations. Safety and suitability of the end product were determined by storability and sensory evaluations.
- The optimal sugar mixture for the mulberry compote (syrup) sauce was determined to be sugar 10: Agave syrup 5: Sorbitol 5, based on the physicochemical properties and sensory evaluations. Safety and suitability of the final compote were determined by storability and sensory evaluations.
○ The development of tablets using mulberries
- Mulberry tablets containing vitamin C were determined to be optimal products based on measurements of disintegrability, physicochemical properties, breaking strength, and hygroscopic properties and sensory evaluations.
○ The development of natural pigments to be used as food additives by using mulberries
- The optimal extraction conditions for the development of natural pigments were 0.1% citric acid at 25 ℃, as determined by the physiochemical and antioxidant properties as a function of the amount of acid added and extraction temperature. The yield of anthocyanin at the optimal mulberry extraction conditions was measured. The anthocyanin content was very low; hence, the materialization of natural pigments using mulberries will be expensive.
- Ultrasonication at 5-s intervals improved the natural pigment extraction efficiency, as determined by the anthocyanin absorbance, total anthocyanin content, total phenol content, and antioxidative ability (determined by a DPPH radical scavenging assay).
○ Identify the positive effects of mulberry on the blood circulatory system - Ethyl acetate fraction 10 (133.93%) had higher ACE inhibition activity than did 50% ethanol extract, acid-added extract, or ethyl acetate fraction.
- Ethanol fraction 5 (133.21%) had higher HMG-CoA reductase inhibition activity than did 50% ethanol extract, acid-added extract, ethyl acetate fraction, butanol fraction, and ethanol fraction.
- The anti-arteriosclerotic effects of mulberries were validated; the weight of the F1B Golden Syrian hamsters increased in the AC group provided only arteriosclerosis-inducing feed as compared with that in the normal control group. The group injected with 1,000 mg/kg mulberry extract showed a reduction in weight.
- Neutral fat in blood, total cholesterol, HDL cholesterol, and non-HDL cholesterol were evaluated to assess the effect of mulberry extracts on blood lipid components. S1 injected with 1000 mg/kg mulberry extract had lower values than the AC group fed with the arteriosclerosis-inducing feed.
- To assess the effects of mulberry extract injections on the organ weights of arteriosclerosis-induced hamsters, the weights of the liver, heart, kidney, and spleen were measured. The results showed that the liver weight of hamsters in the NS10 group was 1.2 times lower than that of hamsters in the AC group.

Ⅳ. R&D Results
◦Treatment technology development at harvesting site
- As this study investigated quality damage rate of mulberries during handling, appearance damage rate was 6.67-20.0% on average. Mostly, the damage in the middle part of the mulberry was overwhelmingly higher than other parts. Such a result shows that mulberry's surface damage is caused by worker's hands, since mulberry harvest is carried out manually, and mulberries are harvested, centered on fully matured fruit. To prevent this, the improvement of gloves and harvesting containers used during the work is necessary. It was investigated that latex thin gloves offering convenient work is effective and that small basket is more effective than existing 4-5 liter basket type container regarding harvesting container.
- Harvested mulberries go through sorting & packing process. In this process, onsite waiting time is long, and thus, mulberry's flesh damage occurs greatly, according to subdivision process from large containers. To improve this, using small size packaging is judged desirable in harvesting by unifying harvesting and sorting & packing stages.
- The degree of mulberry's flesh damage is closely related with mulberry's maturity. As maturity is higher, damage rate is higher. If mulberry's maturity was 80%, the damage rate was 0.3-1.0%, which was very low, compared to 2.8-4.7% of 90% matured mulberry, and 4.4-11.1% of 100% matured mulberry. According to mulberry preference survey via mulberry's maturity, the middle aged prefer fully matured fruit, and young generation prefers 90% matured mulberry. From this, if 90% matured mulberry is harvested, rather than the fully matured fruit, the damage can be reduced quite a lot, and less tissue damage will take place in the case of packing. And, keeping period of the 90% matured mulberry can be extended more than that of the fully matured fruit.
- Harvested mulberries undergo transportation, sorting, and packing processes, and the time for these processes is decided by sorting & packing quantity and packing method.
Mostly, packing is conducted within 30 minutes to an hour after harvest. The time spent to transport to cooling facility after packing is from an hour to four hours. The harvested mulberry's internal temperature of fruit was lower than air temperature by 1.2-1.4℃. In the stage before mulberry is transported from an orchard to sorting site, it was higher than air temperature by 0.5-0.8℃. The collected mulberries' internal temperature of fruit during waiting in the sorting & packing site maintained 0.8-1.2℃ higher level than the temperature at the time of transporting from the harvesting site.
While the collected mulberries are left alone before sorting & packing, the mulberry's internal temperature of fruit was higher than air temperature by 1.3-2.0℃. In the stage to move to treatment site after packing, it was 2.8℃ higher than air temperature.
- To manage mulberry's internal temperature of fruit, mobile mechanical cooling facility with a certain size is judged to be required. For small-sized mulberry production farm, the application of large styrofoam insulation boxes is considered suitable from facility cost and operation aspect. In this case, the use of PCM (ice pack) is essential to maintain low temperature within the insulation box. Through the field test, after putting and sealing 6kg mulberries and four ice packs in the currently used insulation box, and leaving the box on a car, maximum 7℃ lower temperature was maintained than air temperature for three hours and 30 minutes.
◦Development of keeping period extension technology of fresh mulberries
- To research mulberry's proper preservation temperature, this study examined quality change by mulberry's preservation temperature from a practical aspect. The temperature applied to the experiment was 0℃, 10℃ and 20℃, respectively, and this study compared preservation attributes by mulberry's maturity (80%, 90% and 100%) at each preservation temperature. Based on the experiment results, mulberry's freezing point was investigated by mulberry's maturity to set optimum conditions for low temperature preservation.
- For preservation temperature setting to maintain mulberry's quality, this study examined the changes of microorganisms, deterioration rate and organoleptic quality, according to preservation temperature, while preserving the mulberries at 20℃, 10℃ and 0℃. The growth of fungi was smaller at 0℃ than at 20℃ and 10℃. Also, complete deterioration occurred at 20℃ and 10℃ on the second and sixth days; however, it occurred at 0℃ on the 12th day. For more proper temperature setting, this study examined quality change, while preserving mulberries for 25 days by re-setting temperature at 1.5℃, 0℃, -1.5℃, which are 0℃ or around 0℃. As a result, the values of total colony, fungi and deterioration rate showed higher values in the order of 1.5℃, 0℃ and -1.5℃, according to preservation period. Based on sensual quality evaluation, the lowest temperature, -1.5℃, was judged to be more effective in mulberry quality retention than 1.5℃ and 0℃.
- As an investigation of active MA treatment effect for the extension of mulberry's freshness after harvest of them, carbon dioxide was injected into packing mulberry container by 10%, 20% and 30%, and the container was sealed. As a result, the total colony and fungi of mulberry gradually increased while mulberries are preserved in the case of packaging treatment with vent holes. However, the change was minimal in the cases of sealed packaging treatment and packaging treatment with carbon dioxide injection. In the sensual evaluation of merchantable quality, the packaging treatment with vent holes maintained merchantable quality for 10 days, and the sealed packaging treatment and packaging treatment with carbon dioxide injection maintained more than 25 days.
- To apply the small size packaging active MAP technology to mulberry harvesting site, this study put a certain amount of dry ice and mulberries in a usual plastic container (1kg), or conducted vacuum packing, and then, examined quality change, while preserving them in the harvesting site at room temperature for two days. As a result, the vacuum packing treatment injected with dry ice was the most excellent in sensual evaluation, and the control group's deterioration rate was 50%, while packaging treatment with dry ice showed about 18% in deterioration rate. And the dry ice and vacuum packaging treatment showed almost no deteriorate rate. Therefore, the packing method using dry ice and vacuum packing was regarded as proper active MAP packing condition that can be applied to mulberry harvesting site.
- To develop mulberry packing container's structure, this study examined damage rate, according to accumulation degree of mulberry within packaging. The dipped quantity of juice was almost similar level from damaged fruit and damaged tissue up to 6cm from the bottom. Above the 6cm level, the dripped quantity of juice from damaged fruit and damaged tissue showed significant difference. Therefore, if packing is conducted in the unit of 1kg, whose handling quantity is the most, the structure needs to be changed to put mulberries up to 6.5cm maximum from the container bottom. To do so, the bottom of the container needs to be widen more. Also, making the mulberry's packing unit 500g or 250g can be a measure to prevent mulberry's tissue damage, arising from mulberry's load within the container.
- To prevent the penetration of heat into external packaging during the transportation of mulberries, the use of cooling vehicle should be preferred. Also, it is desirable to lower heat conduction of styrofoam box for mulberry's outer packing, and to use the material enhancing compression strength more. Therefore, it is desirable to use number 1 type product by bid method instead of the currently used styrofoam as outer packing material, and the material with lower heat conduction than 0.037W/m․K in terms of initial stage heat conduction.
- To develop a quick low temperature treatment technology that can be applied at harvesting site, based on the preliminary experiment results, this study devised an EPS box that installed refrigerant and circulation fan. To select proper refrigerant, after putting three ice packs (600-700g) and dry ice (EPS: 40% of box volume), respectively, or together, and then a ceratin quantity of mulberries (1kg, 500g), this study measured quality after 24 hours. Sensually, scent, flavor, tissue, appearance and overall taste items were excellent in the order of ice pack and dry ice composite treatment, dry ice single treatment, ice pack single treatment and control group. Deterioration rate was the lowest at 10% or less in the refrigerant composite treatment.
◦Development of mulberry refrigeration treatment and refrigeration preservation technologies
- Concerning mulberry for direct consumption, as granule shape is an important quality factor, this study examined freezing point of mulberry. Actually, slight difference was shown, according to mulberry's maturity, but, the temperature difference was between –1.8℃ and —2.9℃. According to the examination of mulberry's freezing point, according to -10℃, -20℃ and -40℃, the freezing points were almost similar, and the time to reach freezing point was shorter, as refrigeration temperature was lower. Using this feature, it was possible to maintain mulberry's granule by freezing mulberry, however, mulberry's tissue damage became more severe, as mulberry touching frequency increased, due to very weak mulberry's tissue. And thus, it was not much meaningful in terms of commercialization.
- When freezing mulberries at -25℃, it was within an hour and 20 minutes in the case of treating a mulberry to reach -23℃, between seven hours and 30 minutes and nine hours and 30 minutes in the case of using 250g container, and about 14 hours in the case of 1kg container, respectively. When the packing container lid was open in the case of freezing, the speed was faster than the sealed container and packaging treatment. As container size was bigger, the freezing time increased. As a result of comparing the appearance and dripped quantity of juice from mulberry by freezing treatment by freezing method, the appearance was shown the most excellent and dipped quantity of juice was the smallest in the case of freezing individual mulberry. Meanwhile, as packaging container's volume was bigger, appearance change and dipped quantity of juice were relatively larger. In the case of 500g unit of mulberries, the appearance was slightly inferior to 250g unit, however, it did not show difference in terms of the dipped quantity of juice, compared to 250 packing unit. In the case of packing mulberries in the unit of 250g of in small size in the packing stage, after mulberries were harvested with existing large plastic containers, the appearance was very inferior, and the dipped quantity of juice was very serious.
- This study classified packing containers into 2.5kg plastic box, 10kg styrofoam box and 10kg P box, respectively, packed mulberries with them and preserved them at–24℃ using the research result on proper freezing treatment technology of mulberries for processing. In addition, mulberries were put into a 2.5kg plastic box including dry ice and a 10kg styrofoam box, respectively, and then, this study examined the effect according to dry ice treatment. When mulberries were frozen at -24℃, 2.5kg of small packing was frozen the fastest, compared to other packaging, followed by 10kg P box and 10kg styrofoam box. The microorganisms of mulberry showed a trend of diminishing total colony and fungi, according to preservation period. While the difference was minimal, according to the treatment with or without dry ice, and packing method, bigger difference was demonstrated, according to production and handling farm.
- As a result of research on controlling of quality deterioration during the preservation of frozen mulberries, it was more effective to make smaller containers than reducing freezing speed. Also, treatment at the harvesting and packing stages is judged important to make raw material itself sound.
- To build packing technology for quality and temperature management of frozen mulberries, this study froze mulberries after packing them in small packaging, and then, treated with dry ice and vacuum packing to compare quality. As a result, no clear difference was demonstrated in appearance sensually among the control group, treatment with dry ice and treatment with vacuum packing. However, the state of mulberry before refrigeration was investigated to affect more.

Ⅳ. Results
A. Cytotoxicity and efficacy evaluation of anti-oxidative activity
(1) Evaluation of Cytotoxicity in MIN6N beta-cells
The MIN6N pancreatic beta-cells were derived from a mouse pancreatic islet cell line. MIN6N beta-cells were cultured in DMEM (11 mmol glucose, GIBCO, USA) supplemented with 10% inactivated FBS and 1% penicillin/streptomycin and maintained at 37oC in a humidified 5% CO2 incubator. Cell viability was greater than 84% in the presence of MAE (EtOAc extracts), up to a concentration of 500 μg/ml and MAB extract (n-BuOH extracts), MAH (H2O extracts), MA ( Et-OH extracts) viability was value 80.8%, 75% and 69.9%, respectively.
(2) Efficacy evaluation of anti-oxidative activity
To determine their radical scavenging activities, this study measured scavenged DPPH radical in the nontoxic concentration of cell viability. Fractionated extracts (MA, MAH) scavenged the DPPH radicals in a dose-dependent manner, with a 50 μg/mL and DPPH radical scavenger activity of fractionated extracts (MAE, MAB) increased in a dose-dependent manner, with a 100 μg/mL B. Efficacy evaluation of Apoptosis and proliferation
(1) Protective effect and recovery ability
The cyto-protective effect of MAE on the H2O2-treated MIN6N beta-cells was measured using the MTT assay. In this study, H2O2 was used to induce oxidative stress in MIN6N beta-cells. At the concentration of 0.7 mM H2O2 cell viability was reduced to 50.7% compared with control. Therefore, 0.7 mM H2O2 concentration was selected for experiments. Treatment of MAE 50 μg/ml increased cell viability to 11% in comparison with H2O2-treated MIN6N beta-cells and increased to 20%, the highest protective effects, in the presence of MAE 100 μg/ml. Fractionated extracts MA was confirmed an increase in the cell viability value of 13% at the 50 μg/ml. MAH and MAB, Respectively, showed a cell viability was increased by 11% and 14% ,showed a cell viability of less than the MAE and MA. Among of 14 types fractions, sample of MAE1 and MAE2 showed the highest values of cell viability at 21%, 20% respectively. To analyze after-treatment efficiency as a recovery effect, each samples was treated under H2O2 condition. MA and MAE produced an increase in the cell viability value. This cell viability was showed that more than the results of treat-MAB and MAH in the recovery ability experimentation.
(2) Intracelluar ROS scavenging activity
Intracellular ROS levels in H2O2-treated cells were determined using the ROS-sensitive fluorescent probe H2DCF-DA. In experiments, H2O2-treated cells dramatically increases amounts of intracellular ROS and treated MA was able to scavenge intracellular ROS in dose-dependent manner. The ROS scavenging activity decreased level of ROS up to 47% at MA 400 μg/ml compared to cells treated only with H2O2. Also, ROS scavenging activity was confirmed in MAB, MAH group but this activity was less than MA and MAE. Sample group B divided in MAE group was the highest ROS scavenging activity in the presence of MAE1 and MAE2.
Whereas, sample group C showed result of low ROS scavenging activity. Therefore, samples of MA, MAE, MAE1, and MAE2 were used for next experiments.
(3) Anti-apoptotic efficacy
Treated with 0.7 mM H2O2 cells group caused 58% of the cells to undergo apoptosis. However, pre-treatment with 100 μg/ml MAE significantly inhibited H2O2-induced apoptotic cell death (35%). In this study, MAE was shown to decrease undergoing apoptotic cell rate in H2O2-treated MIN6N beta-cells.
C. Mechanism analysis and I n vitro anti-diabetes efficacy evaluation
(1) Western blot analysis
To further analyze the effects of MAE on inhibition of apoptosis, the effects of MAE on apoptosis signaling pathways such as Caspase-3 and AIF (Apoptosis Inducing Factor), plays an essential role as executioner in apoptosis, were studied by western blot analysis. MIN6N beta-cells were cultured in H2O2 condition, with or without MAE and then the phosphorylation levels for cleaved caspase-3 and AIF were examined. The cleaved form of caspase-3 was appeared in H2O2-treated group.
However, MAE (from 50 to 70 μg/ml) dose dependently inhibited the H2O2-dependent phosphorylation of caspase-3 and AIF proteins. Furthermore, it is well known that anti-oxidative enzymes such as catalase (CAT) and superoxide dismutase (SOD) have major effects on the apoptosis signaling pathway. The two anti-oxidative enzymes were distinctly appeared in H2O2-treatedgroup. However, pre-treatment with MAE reduced the SOD and CAT expression compared with H2O2-treated group.
(2) In vitro anti-diabetes efficacy evaluation
The evaluation of anti-diabetic efficacy, effect of MAE on insulin releas and glucose uptake in MIN6N beta-cells, pre-treatment with MAE produced an increase in insulin secretion and glucose uptake in a dose-dependent manner compared with the H2O2-treated group.
D. I n vivo anti-diabetes efficacy evaluation
(1) Measurement of body and organs weight
Changes body and organs weight in normal and experimental groups are measured for 4 weeks. Loss of body weight was observed in streptozotocin (STZ)-induced diabetic mice compared to normal mice. But, treatment with MAE mice improved the body weight as compared to diabetic mice. Also, the weight of organs such as liver, heart, and kidney was not change in the MAE treated group compared to the diabetic mice except the spleen, treatment with MAE mice decreased the spleen weight as compared with increased spleen weight of diabetic mice. Thus, it was seemed that MAE may can be protection against weight changes.
(2) Measurement of insulin secretion and blood glucose level
The level of serum insulin significantly decreased in the STZ-induced diabetic mice caused by the destruction of pancreatic beta-cells as compared with normal mice. The administration of the MAE mice increased the levels of serum insulin as compared to diabetic mice. These results suggest MAE improved the insulin release of STZ-induced diabetic mice. The level of blood glucose related with insulin release increased in the STZ-induced diabetic mice. But, the administration of the MAE mice decreased the levels of blood glucose as compared to diabetic mice.
(3) Measurement of cholesterol and neutral fat
The level of cholesterol and neutral fat significantly increased in the STZ-induced diabetic mice as compared with normal mice. The administration of the MAE decreased the levels of cholesterol and neutral fat as compared to diabetic mice. These results suggest MAE inhibits the level of cholesterol and neutral fat induced diabetic mice.
E. Supplement research
(1) Separate purification of anthocyanin isolated from Mulberry
Cell viability was greater than 85% in the presence of H2O and EtOH extracts, up to a concentration of 200 μg/ml and in the case of EtOH extract, viability was still above this value (91.5%) at a 200 μg/ml dose. DPPH scavenging activity of 100%, 70%, 50% and 30% ethanol mulberry extract increased in the concentration of 70 μg/ml, but the best DPPH scavenging activity was 70% ethanol mulberry extract. It was confirmed that the antioxidant activity is increased in a manner dependent on the concentration of the mulberry extract. This DPPH scavenging activity showed a high radical scavenging effects of 68.9% at a concentration of 300 μg/ml. All fractionated extracts were able to scavenge intracellular ROS in dose-dependent manner.
Consistent with its protective effects on cell viability, 70% EtOH extract was a much more effective suppressor of intracellular ROS compared with other fractions. The ROS scavenging activity increased up to about 34.1% at 70% EtOH extract 70 μg/ml compared to cells treated only with H2O2. All fractionated extracts were able to lipid peroxidation inhibitory activity in dose-dependent manner. The inhibitory effect of 30%, 50%, 70%, and 100% EtOH extracts were 21.6%, 32.5%, 32.5% and 22.6% at 70 μg/ml compared to cells treated only with H2O2, respectively. These results indicated that the ability of fractionated EtOH extracts to inhibit lipid peroxidation on H2O2 condition might be due to their intracellular ROS scavenging activity. The 70% EtOH extract was found to possess the highest total monomeric anthocyanin content among various concentrations EtOH extracts, which is 25% higher than other EtOH extracts. The pure cyanidin-3-glucoside standard (kuromanin chloride) and purified anthocyanin isolated from mulberry were identified by high performance liquid chromatography (HPLC) retention time.
(2) Anti-diabetic efficacy of Cyanidin-3-glucoside isolated from Mulberry Since high-glucose (HG) condition increase osmolarity in cells, the MIN6N beta-cells were also cultured with 25 mM mannitol, osmotic control, to distinguish the effects of glucose and osmotic pressure. The HG-dependent increase into 7.6 times of intracellular ROS level and green fluorescence intensity was dramatically decreased after pretreatment C3G 70 μg/ml compared with HG group. Also intracellular ROS was visually observed through a fluorescence microscope. HG, but not mannitol, caused 33.9% of the cells to undergo apoptosis. However, pretreatment with 70 μg/ml C3G significantly inhibited HG-induced apoptotic cell death (24.8%). To further analyze the effects of C3G on inhibition of apoptosis, the effects of C3G on mitogen-activated protein kinase (MAPK) signaling pathways were studied by western blot analysis. The phosphorylation levels for ERK, JNK, p38 were examined. C3G (50 and 70 μg/ml) dose dependently inhibited the HG-dependent phosphorylation of MAPK proteins, mitochondrial permeability-mediated activation of apoptotic proteins, such as Bcl-2 family proteins, and cytochrome c were examined. This study confirmed the release of cytochrome c and expression of Bcl-2 family proteins that protein expression of Bcl-2 was decreased and Bax was increased in HG-treated cells. In contrast, expression of the Bcl-2 family proteins was regulated similar to the control in C3G 70 μg/ml pretreatment group. Caspase-3 plays an essential role as executioner in apoptosis. The cleaved form of caspase-3 was appeared in HG-treated group. However, pre-treatment with C3G 70 μg/ml reduced the cleaved caspase-3 compared with HG-treated group. Nuclear factor-kappa B (NF-κB) has been known to be involved in oxidative stress-induced cell death in different cell type. HG-induced nuclear translocation of NF-κB p65 was inhibited by pre-treatment with C3G 70 μg/ml. As a final step for the evaluation of anti-diabetic efficacy, effect of C3G on insulin release in MIN6N beta-cells, Pre-treatment with 70 μg/ml C3G produced an increase in insulin secretion (84.2%) compared with the HG-treated group (74.8%).

70% EtOH was used for effective extraction of active components from the mulberries. And three organic acids citric acid, malic acid, and fumaric acid were used to extract anthocyanins with high yield. From the extracts six flavonoids were isolated through repeated SiO2, and ODS column chromatography methods. The purified flavonoids were identified to be rutin, quercetin, isoquercetin, astragalin, artoindonesianin O and isobavachalcone on the basis of spectroscopic data. The extracts and compounds were evaluated for several pharmacological activities. To increase the stability of anthocyanins the kind and concentration of treated organic acids were determined. And for standardization of active materials and products, the quantitative analyses for the principal components rutin, morachalcone A, artoindonesianin O, and isobavachalcone were established using HPLC method.
