연구개발사업의 목적 : 암을 효과적으로 치료하기 위해서는 암세포의 발생 및 증식 기작을 이해하는 것이 필수적임. 암은 유전정보 유지에 핵심적인 기작이 망가지면서 돌연변이율이 급격히 증가하여 개시되는데, 특히 세포분열과정을 체크하는 기작 또는 손상된 DNA 복구 기작이 망가지면 극심한 chromosomal instability(CIN)가 초래되어 진행이 빠르고 치료가 어려운 악성종양이 발생함. 난소암과 유방암은 대표적인 CIN-type cancer로 우리나라에서 그 발생률이 급격히 증가하고 있음. 특히 난소암은 표준치료의 한계성으로
연구개발사업의 목적 : 암을 효과적으로 치료하기 위해서는 암세포의 발생 및 증식 기작을 이해하는 것이 필수적임. 암은 유전정보 유지에 핵심적인 기작이 망가지면서 돌연변이율이 급격히 증가하여 개시되는데, 특히 세포분열과정을 체크하는 기작 또는 손상된 DNA 복구 기작이 망가지면 극심한 chromosomal instability(CIN)가 초래되어 진행이 빠르고 치료가 어려운 악성종양이 발생함. 난소암과 유방암은 대표적인 CIN-type cancer로 우리나라에서 그 발생률이 급격히 증가하고 있음. 특히 난소암은 표준치료의 한계성으로 인해 재발률 및 사망률이 높으며, 유방암은 1-2위를 다투는 발생률을 보여, 사회, 경제적으로 매우 중요하고, 모두 BRCA 유전자 변이와 관련성이 높은 암으로 잘 알려져 있음. 본 연구진은 중요 세포분열 체크포인트(Spindle assembly checkpoint, SAC)인 BubR1이 세포분열기에 acetylation 되고, 이러한 BubR1 acetylation 이 암억제인자 BRCA2에 의해 강화되어 유전정보의 안정성을 유지하는데 필수적임을 세포.개체.환자 샘플에서 증명함. 특히 BubR1 acetylation-deficient mouse를 구축하여 SAC의 이상이 tumorigenesis를 시작하는 원인임을 증명하였는데, 이는 SAC 이상으로 인한 암 발생 과정을 연구할 수 있는 최초의 CIN-type cancer model로써 그 가치가 높음. 본 연구는 선행 연구를 바탕으로 BubR1 acetylation-deficient mouse를 p53, p19 유전자 조작 mouse와 교배하여 암 발생의 penetrance를 비교함으로써 개체 수준에서 유전체 불안정성의 시작, 진행 기작을 확립하고, 이를 기반으로 하여 새로운 항암제 개발을 추진하고자 함. 나아가, 난소암 및 유방암을 중심으로 새로운 항암 타깃으로써 BubR1 acetylation의 임상적 응용 가능성을 검증하기 위해, BRCA, BubR1 acetylation 유전자 이상 여부 및 발현을 조사하고, 임상병리학적 인자 및 예후 인자와의 관계를 밝혀 분자생물학적 발암기전을 규명하고, 새로 발굴된 BRCA, BubR1 acetylation 특화된 사항을 활용하여, 표적치료제로서 deacetylase inhibitor들의 유용성을 확인하고자 함.
연구방법 및 연구결과 :
♣ 본 연구팀의 핵심 연구기반
1) 항암 표적인자로써 BubR1 acetylation 규명: BubR1 acetylation은 정확한 염색체 분리를 조절하는 분자적 스위치임 (EMBO J, 2009), BRCA2의 새로운 세포분열조절 기작 규명 (Dev Cell, 2012)
2) SAC 이상과 암 발생 연구를 위한 최초의 CIN-type cancer model 구축: BubR1 acetylation-deficient mouse
3) BubR1 기능 연구에 기반을 둔 임상적 응용연구: 난소암 재발을 예측할 수 있는 prognostic marker로 BubR1 제안 (Br J Cancer, 2009), AcK250-BubR1 항체 제작 및 새로운 스크리닝 기법 제안 (국내. 국제 특허)
♣ SAC 이상으로인한 tumorigenesis 연구: CIN-type cancer mouse modeling
선행 연구에서 구축한 BubR1 acetylation-deficient mouse를 중요 암억제인자인 p53, p19 유전자조작 mouse와 교배하여 인간에게 나타나는 다양한 암의 발병 모델을 새롭게 구축/분석함으로써 CIN-type cancer의 발병 기작을 확립.
♣ 난소암, 유방암 환자에서 BRCA, BubR1 acetylation 이상여부 및 임상적 상관관계 분석
선행 연구에서 제시한 난소암과 유방암에서 BRCA, BubR1의 다양한 발현을 좀 더 심화된 임상적 관점에서 환자군을 계층화하기 위하여, 난소암과 유방암 세포주 및 환자에서 BRCA, BubR1 mutation을 확인하고, 환자조직에서 유래한 일차세포주를 확립. 환자조직을 통해 단백질 발현 여부를 분석하고, 임상병리학적 인자들과 상관관계가 있는지 확인하여, 임상적 예후 인자 및 재발률, 생존율과의 관계를 통계학적으로 분석.
♣ 난소암, 유방암 환자에서 BRCA, BubR1 acetylation 계층화에 따른 targeted therapy
개발된 deacetylase inhibitor들을 이용하여 상기에서 규명한 기전이 증명되는지 세포수준에서 확인하고, 약제를 선정하여 nude mice 등의 동물모델에서 적용하여, 효과적인 deacetylase inhibitor 치료의 가능성 확인.
계층화된 난소암, 유방암 환자군에서 BRCA, BubR1 acetylation status에 따라 deacetylase inhibitor 표적치료를 제시.
연구결과에 따른 기대성과 : 현대 항암 치료의 키워드는 ‘표적 치료’로 이는 정확한 분자적 수준의 기작을 이해할 때 가능함. 선행연구를 통해 밝힌 BubR1 acetylation의 세포분열 조절 기작은 유전체 안정성 유지 기작에 대한 새로운 패러다임을 제시함으로써, 이를 타깃으로 하는 항암 전략을 제안하는 기반이 될 것임. 특히 SAC 이상으로 인한 악성 종양 연구에 최적화된 동물 모델을 확보하였고, 이를 활용하여 구축될 새로운 CIN-type cancer 모델은 인간에게 나타나는 암의 발병 기작 규명 및 표적치료를 위한 신약의 검증등에 응용 가능성이 높음. 또한 연구 결과 새로이 규명될 분자적 기작과 그에 기반을 둔 임상적 연구결과가 효과적인 신약 스크리닝 기법의 개발 및 암의 진단과 예후측정 등에 직접적으로 응용될 수 있을 것으로 기대함. 특히 기개발된 deacetylase inhibitor의 활용 가능성이 높은 바, BRCA, BubR1 acetylation이 중요한 작용을 하는 난소암과 유방암에서 획기적인 암 치료 표적으로 향후 환자의 맞춤치료가 가능해질 것으로 기대함.

Purpose of the research: In order to approach effective cancer therapy, it is necessary to understand the basic mechanisms behind cancer cell development and proliferation. Cancer is triggered by impaired mechanism in genetic information maintenance leading to escalation of genetic mutations. Especially, impaired mechanisms in cellular division or DNA repair cause significant chromosomal instability (CIN) of which could generate malignant tumors that are hard to cure due to their rapid proliferating nature. Namely, breast and ovarian cancers are typical CIN-type cancers and incidence of those two cancers are markedly increasing in Korea. In fact, ovarian cancer bears high recurrence and death rate due to ineffective standard therapy and breast cancer is currently one of the most common cancers in Korea and thus there’s a popular demand in development of more effective treatment against those two cancers. Indeed, ovarian and breast cancers are both known to be highly related with mutations in BRCA genes. Using human patient sample, we previously showed that a key regulator in spindle assembly checkpoint (SAC), BubR1, is acetylated during mitosis and this acetylation is mediated by BRCA2 that collectively work to maintain genetic stability in cell.
Most importantly, we generated BubR1 acetylation-deficient mouse to prove that defects in SAC leads to spontaneous tumorigenesis; in fact, this mouse model is the first in vivo CIN-type cancer model that could be used to study correlation between defective SAC and tumorigenesis. Our research focuses on the establishment of mechanisms underlying initiation and progression of genetic instability and possible anticancer drug development by using mouse models that harbor p53 or p19 mutation along with BubR1 acetylation deficiency and compare cancer penetrance among those mouse models. Furthermore, to verify the potential of BubR1 acetylation as a novel anticancer target for clinical application, we are going to investigate mutations and expression level of BRCA and acetylated BubR1 and reveal relationship between pathological and prognostic factors of breast and ovarian cancers, eventually unveiling the molecular biological mechanism behind cancer development. Moreover, our research is to confirm deacetylase inhibitors as valid candidate for novel anticancer drugs, expanding on the new findings regarding BRCA and BubR1 acetylation.
Research methods and results:

1) Verification of BubR1 acetylation as an anticancer target: BubR1 acetylation is a molecular switch regulating precise chromosome segregation (EMBO J, 2009), investigation of BRCA’s new role in mitosis (Dev Cell, 2012)
2) Construction of first CIN-type cancer model to study correlation between defective SAC and carcinogenesis: BubR1 acetylation deficient mouse
3) Translational research based on the study of BubR1 function: suggestion of BubR1 as a prognostic marker to prevent recurrence of ovarian cancer (BRJ Cancer,2009), AcK250-BubR1 antibody generation and novel antibody screening method (domestic and international patent)
♣ Research in tumorigenesis by SAC abrogation: CIN-type cancer mouse modeling
Cross BubR1 acetylation-deficient mouse with mutated p53 and p19 tumor suppressors to construct mouse models that could recapitulate human carcinogenesis and thus establish mechanism behind CIN-type cancer.
♣ Exploring the correlation between abnormal BRCA and BubR1 acetylation in breast and ovarian cancer patients.
To clinically stratify patients according to variation in BRCA and BubR1 expression pattern in breast and ovarian cancer, confirm BRCA and BubR1 mutation in cancer cells from human patients. Furthermore, from primary cells originated from patient cancer tissue, analyze protein expression level and whether there is a correlation with pathological factors. Moreover, from cancer tissue analysis data, statistically define prognostic factors and recurrence and survival rate.
♣ Target therapy based on the stratified ovarian and breast cancer patients according to variations in BRCA and BubR1 acetylation
Using deacetylase inhibitors currently under development, confirm that above investigated mechanism is valid in cells and select drug candidates and test them in vivo using animal models such as nude mouse model. This will open room for target therapy against breast and ovarian cancers.
♣ Expected research achievement:
The main issue regarding anticancer therapy nowadays is ‘target therapy’ and it requires understanding of precise molecular mechanism behind cancer development. Previous work from our lab regarding BubR1’s regulatory roles in mitosis via acetylation suggested a novel mechanism in maintenance of genetic stability and is expected to guide effective anticancer therapy in near future. Especially, we already have mouse models harboring SAC abrogation and generation of new CIN-type mouse models recapitulating human cancer are expected to have wide range of applications, from investigation of molecular mechanism behind carcinogenesis to potential development of target therapeutic approaches. Moreover, we could test whether deacetylase inhibitors are workable in ovarian and breast cancers, of which would lead to breakthroughs in cancer therapy.
