시중에서 판매되는 식육가공품(소시지, 혼합소시지, 햄, 프레스햄)은 돼지고기, 닭고기, 쇠고기, 연육 등 다양한 원료육을 이용하고 있고, 증량제인 옥수수전분 및 대두단백을 대체적으로 사용하고 있었다. 축산물 가공기준 및 성분규격(식약처 고시)에 제시된 축산물가공품의 유형을 잘못 표시한 사례도 있었고, 유형이 표시되어 있지 않는 제품도 있었다. 원료육(돼지고기, 닭고기) 및 증량제(옥수수전분, 농축대두단백) 혼합비율에 따른 혼합소시지 품질은 닭고기 및 대두단백 첨가가 단백질 함량을 증가시켰고, 옥수수전분 첨가는 제품 내 수분함량을 감소시켰다. 또한 경도 등 조직감과 지방산화 증가는 옥수수전분을 첨가하였을 때 나타났다. 혼합육제품(소시지) 제조 원료육이 돼지고기 단독만 사용하는 것이 여러 가지 원료육을 혼합하는 것보다 제품의 단백질 함량은 낮고, 지방함량은 높았다. 조직감과 색도(명도, 적색도), 저장 중 지방산패도는 원료육 종류보다 옥수수전분 첨가가 더 영향이 컸다. 혼합육제품(소시지)에 증량제인 옥수수전분 첨가한다면 최소 기준이 5% 이내로 첨가하는 것이 바람직한데, 이는 수분이 옥수수전분 5% 첨가, 단백질이 5%, 육색(명도, 적색도)이 1%, 조직감(경도)이 3%, 관능평가(전체만족도)가 5% 첨가하였을 때 가장 적합하였기 때문이다. 혼합육제품(소시지)에 증량제를 첨가하였을 때 제품 특성은 수분이 대두단백 1% 첨가, 단백질이 1%, 육색(명도, 적색도)이 5%, 조직감(경도)이 5%를 첨가하는 것이 적합하여 혼합육제품(소시지)에 증량제인 대두단백을 첨가한다면 최소 기준이 5% 이내로 첨가하는 것이 바람직한 것으로 사료된다. 닭고기 가슴살 및 기계발골 계육을 첨가한 소시지의 제품 특성에서 유화 안정성은 닭고기 가슴육은 5% 이내로 첨가해야 하고, 기계발골계육은 35%까지 첨가해도 적정하고, 무기물 함량은 인, 칼슘, 철, 구리 함량에서는 닭고기 가슴살이 15% 첨가가 적절한 것으로 나타났다. 관능평가에서는 닭고기 가슴살은 15% 이내, 기계발골 계육은 25% 이내로 첨가하는 것이 바람직하며, 육색은 닭고기 가슴육 5% 첨가하는 것이 적합한 것으로 사료된다. 조직특성은 닭고기 가슴육과 기계발골 계육 첨가는 5%가 적합한 것으로 나타났다. 따라서 닭고기 가슴살과 기계발골 계육의 첨가량은 5% 이내로 설정하는 것이 바람직할 것으로 사료된다. 시중 판매 식육가공품의 분석결과를 종합하면, 지방함량이 가장 높은 프레스햄(식육통조림, 원료육: 돼지고기, 증량제: 대두단백, 베지 첨가)이 육색 명도도 가장 높았고, 지방함량이 가장 낮고, 수분 및 단백질 함량이 가장 높은 햄(원료육: 돼지고기, 비분쇄)이 조직특성도 높게 나타났다. 식육가공품의 유형에 따라 프레스햄이 소시지와 유사한 조직 특성을 보였고, 오히려 프레스햄 식육통조림이 가장 부드러운 것으로 나타났다. 혼합소시지 품질규격(안)은 전분 및 대두단백 5%이하로 설정하였다. 이러한 사유는 축산물 가공기준 및 성분규격 고시에는 전분만 10% 이하로 제시되어 있어 대두단백에 대한 첨가기준과 설정이 필요하기 때문이다. 주관과제에서 제공받은 혼합육제품(소시지)의 구성원료에 대한 정성, 정량분석을 위해, 원료종의 핵 DNA 서열 정보를 이용한 분석법을 개발하여 적용하였다. NEUROD1과 RHO 유전자서열에서 개발한 종 특이적인 프라이머들이 PCR 과정에서 종 특이적인 증폭에서의 효율이 높았으며, MC3R 유전자 범용 프라이머를 이용한 PCR 증폭은 돈육, 계육과 선별적으로 PCR 산물의 길이가 다른 어육 특이적인 PCR 산물을 형성하였다. RHO 유전자의 원료 특이적 프라이머들은 재현성이 매우 높아, 정량분석용 유전자 진단체계에 적합한 것으로 판단되었다. 농도별 시험에서도 원료종의 성분비에 따른 증폭양상이 높은 상관을 나타내었다(R2 최소값 0.99872 ∼ 최대값 1.0000). 혼합육제품(소시지)의 주원료인 육 단백질과 증량제로 이용된 대두단백을 구분을 위해 대두단백 특이적인 정량분석을 Sandwich ELISA 방법으로 검출하여 정량하였다. 육 성분으로만 구성된 혼합육 시료에서는 대두단백이 전혀 검출되지 않았고, 대두단백을 참가한 경우는 2.5∼20ppm 수준에서 검출되었다. SLS 용액과 proteinase K로 분해한 혼합육제품(소시지) 시료에서 전분은 불용성으로 검출되었고, 정성분석을 통해 전분의 함유를 확인하였다. 황산-페놀 비색법을 활용한 시험에서 원료육 처리구에서는 녹말이 검출되지 않았으며, 옥수수 전분을 처리한 시료에서만 전분이 검출되었다. 시중판매 제품에 대한 현장접목시험을 위해, 원료육 성분의 검출은 mtDNA 유전자에 대한 OS-PCR과 실시간 PCR, PCR-direct sequencing 시험, 정량분석을 위해서는 RHO 유전자에 대한 실시간 PCR을 수행하였다. 식물성 증량제 중 대두단백은 Sandwich ELISA 분석, 전분 검출과 정량은 황산-페놀비색법을 적용하였다. 시중제품 40건에 대한 원료육 정성분석 결과에서 시료 중 2건에서 label에는 표기되지 않은 어육이 사용된 사례들을 확인하였다. 시중 소시지 제품 80건과 17건의 햄류에 대한 정성-정량분석에서 원료육과 증량제를 선별적으로 정량하였고, 몇몇 시료에서 표기와 매우 다른 원료육과 증량제가 검출되었다.

This study was carried out to develop some methods for detection and quantification of meat origins (species) and extenders, simultaneously to establish the quality standards in the processed meat products. The processed meat products available on the markets (e.g., sausage, mixed sausage, ham, press ham) were usually processed with various types of raw meat materials such as pork, chicken and beef etc, and extenders such as corn starch and soy protein. The mixed sausages were usually added with chicken and soy protein to increase the protein content, while the corn starch was added in order to reduce the moisture content in the product. When compared to the products made with a variety of raw meat materials, the mixed meat products (sausages) manufactured with only raw pork material generally had lower protein content but higher fat content. The addition of corn starch generally had greater effect on the texture and color (redness and lightness) traits, as well as fat rancidity during storage. In case, if the extenders such as; corn starch and starch are added into the mixed meat products (sausages), the minimum added level within 5% is recommended. From the quality characteristics point of view of sausages made with chicken breast and mechanically deboned chicken meat, the added level of chicken brisket and mechanically deboned chicken meat should be within 5%. The analytic results revealed that among the products available on the markets, press ham (canned meat, meat materials: pork, extender: soy protein, vegetable) had the highest fat content and brightness. Depending on the natural characteristics of each type of processed meat products such as press ham and sausage, however, the canned meat was found the most tender compared to the others. Finally, in order to ensure the quality standard of the mixed sausages, the added level of starch and soy protein within 5% is recommended. For detecting meat sources, pork-, chicken-, and fish-meat, universal primers for the mitochondrial DNA (mtDNA) genes were designed and applied to screen the use of meat sources using polymerase chain reaction (PCR) technique. Three different PCR-based assays were tested for origin-specific detection of each meat source including pork, chicken and fish: PCR-RFLP for PCR products amplified using universal primers, TaqMan real-time PCR using universal primers and origin-specific probes, and conventional PCR assay using pork-, chicken- and fish-specific primers. Using the detection methods developed, meat sources (pork, chicken, and fish) were deferentially detected by producing the origin-specific fragments in PCR-RFLP and/or origin-specific PCR products in conventional PCR and real-time PCR in the processed meat products, sausages and hams. In addition, the relative amounts of each PCR product could be calculated for quantification by real-time PCR. Moreover, several methods were tested and established to select the suitable DNA extraction method and pre-treatment condition for sample lysis. From the results of real-time PCR each meat source were detected over 1×10-14g/ul levels. For the each single meat source, these molecular methods yielded possibly to discriminate the sources even from different conditions for production conditions such as mixing proportion, particle size and from different storage conditions. Among the plant-derived extenders, detection and quantification methods were also established for soy protein and corn starch. The soy protein was detected and quantified using the sandwich ELISA assay and the concentrations of corn starch was calculated using the sulfuric acid-phenol spectrophotometric assay after lysis using sausage lysis solution designed in this study. Assays for soy protein and cor starch recommended the appropriate dialysis procedure for the biochemical analyses. For detecting and quantifying the processed meat products supplied by the supervising research party, molecular methods had been developed using information of nDNA genes and applied to examine. Species-specific PCR primers designated from NEUROD1 and RHO genes showed highly efficient results in origin-specific PCR. Universal primers designed from MC3R gene served the specific PCR products in the product length discriminating the fish-meats from pork- and chicken-meat sources. From the results of preliminary studies, origin-specific primers designed from RHO gene showed the most efficient in the analyses of detection and quantification of meat sources by real-time PCR as well as conventional PCR. The high levels of significant correlation was observed in the amplification patterns to concentrations of template DNA (R2 value: min. 0.99872-max. 1.0000). Soy protein-specific sandwich ELISA assay was applied to detect and quantify deferentially from animal proteins derived from pork, chicken, and fish meats. This assay showed no results from the samples produced using only meat sources but showed the concentration levels from 2.5-20ppm of soy proteins from the cases of use of soy protein as one of extenders. Corn starch did not dissolve during the lysis step of the SLS and proteinase K and detected as insoluble phase after sample lysis, and that was verified by biochemical identification assay. Sulfuric acid-phenol spetrophotometric analysis showed that non-starch products have not significant signal for the starch contents but starch-added products have significant levels of absorbance score in the assay. From the test of commercial products, OS-PCR, real-time PCR, and PCR-direct sequencing were applied to detect the meat sources using mtDNA genes and real-time PCR was carried out to calculate the relative concentrations of each source using RHO gene. The results from the source meat identification showed that the labels of two sausage samples had not the records for use of fish meat in production. The results showed that some of product labels had not quitely different contents of meat sources and plant-derived extenders from the analyses for source identification and relative quantification of 80 commercial sausages and 17 industrial hams.
