동충하초의 유전체/전사체 데이터를 기반으로 유용 터페노이드 계열 생산 유전자 집단을 발굴하고 이들의 대사경로와 진화패턴을 파악하는데 중점을 두고 과제를 진행하였다. 터페노이드 생합성 유전자의 모티프 시퀀스를 가지고 있는 후보유전자를 생물정보 분석을 통해 추출하였고, 터페노이드 생산에 관여하는 모든 KEGG pathyway 상의 유전자군을 레퍼런스로 추가적인 탐색을 하여 관련 유전자의 존재 유무를 탐색하였다.터페노이드 백본 물질인 geranyl-PP는 mevalonate 경로로 생산되는 것으로 밝혀 졌으며, MEP/ DOXP 경로의 유전자는 1-deoxy-D-xylulose-5-phosphate synthase (EC 2.2.1.7) 외에는 발견되지 않은 것으로 보아 작동하지 않을 것으로 판단하였다.monoterpene, diterpene, triterpene, sesquiterpene 등의 합성을 위한 전구물질 합성 경로를 통과하는 주요 유전자들이 존재하는 것을 할 수 있었다. Genome mining을 통해 얻은 세가지 터펜계열 유전자 클러스터는 GGPS로서 geranyl-geranyl PP를 형성하거나,Phytoene squalene synthase로서 ergosterol 등의 스테롤 물질을 합성하는데 사용되거나, aristolochene 이라는 균독소 물질을 만드는 구성 물질로 파악되었다. 번데기동충하초의 자실체 발달단계를 5단계로 세분화하고 이들로부터 각각 전사체 데이터를 생산하였으며 이를 곰팡이 특유의 막스테롤인 에르고스테롤 대사경로 관련 유전자들에 맵핑하고 발현양상에 따라 차등발현유전자군을 추출하였다. 이를 KEGG 데이터베이스에서터펜계열 물질의 기초적인 전구물질이 되는 GGPP (geranyl-genanyl pyrophosphate)를 형성하는 terpenoid backbone pathway에 맵핑하였다. 그 결과 터페노이드 계열 유전자의 발현 패턴이 발단단계별로 유사한 것으로 보아 자실체 발단단계가 터펜 물질생산에 미치는 영향은 미미한 것으로 추정하였다. 공통 발현된 유전자는 터펜 백본 합성부터 sesquiterpene, triterpene 및 steroid 물질 합성에 필요한 대부분의 유전자를 포함하고 있었다. 동충하초 건강기능성 지표물질인 cordycepin 생산과 관련된 adenosine 생합성경로 관여 유전자군의 자실체 발달단계별 발현양상을 조사 결과 자실체 발달단계별로 다소 상이한 결과를 얻었다. 첫번째 발단단계에서 과발현된 유전자군은 AMP와 dAMP를 각각 adenosine과 deoxyadenosine으로 전환하는 enzyme이었고, 두번째 발단단계에서의 과발현 유전자는 ATP 생산에 관계한 것이었습니다. 자실체 발단 5단계에서 발현된 유전자는 AMP, adenosine, inosine을 합성하는 유전자군이었습니다. 자실체 전체발달단계에서 전반적으로 높은 발현양상을 보인 유전자군은 AMP의 전구물질인 주로 adenylo succinate를 합성하는 유전자로 나타났다.
제2협동과제는 현미동충하초 유래 유용물질 생산 유전자 형질전환체 제조 연구를 다루었다. 예비실험으로 노란다발동충하초를 재료로 PKS-NRPS hybrid 유전자의 knockout실험을 agrobacterium을 이용한 일반적인 상동유전자재조합 방법을 통해 수행하였지만 성공 효율이 매우 낮아 실질적인 기능유전체 분석을 위해서는 새로운 방법고안이 필요했다. 원핵생물의 외래유전자 방어면역체계로 알려져 있는 CRISPR 시스템을 형질전환을 2013년도에 유전체 편집으로의 적용에 성공하면서, 이후 다양한 생물군에 대한 형질전환 기술로서 도입되고 있는데, 이 기술을 동충하초 연구에 적용하고자 먼저 선행 연구가 잘 되어 있는 human cell 형질전환 재현실험을 수행하였다. 4개의 타겟 영역을 지정하고 human cell에 도입시켜 Cas9 유전자의 정상발현을 확인하였고,최종적으로 해당 영역의 PCR product를 NGS 시컨싱하여 목적한 부분의 deletion을 통한 gene disruption이 성공적으로 수행되었음을 확인하게 되었다. 예비실험 목적으로 수행되었지만 형질전환 효율은 12% 정도로 매우 높게 나타났습니다. 예비실험 결과를 바탕으로 동충하초 전용 CRISPR/Cas9 벡터를 디자인하였다. 동충하초 시스템에서 CRISPR 유전자가 정상 발현할 수 있도록 codon optimization을 진행하고 강력한 동충하초 특이적 promoter 유전자를 선발 도입하였다.
동충하초의 phenylpropanoid 대사 경로에 관계하는 phenylalanine ammonia lyase(PAL) 유전자의 발현조절 연구를 수행하였다. PAL은 phenylalanine 기질을 전구물질인 trans-cinnamic acid로 전환하는 핵심적인 역할을 수행하고 있는데 이는 flavones,isoflavaone 등 기능성을 나타내는 색소 물질 생산에 관여하는 유전자로서 식물에서는 활발히 연구되었으나 곰팡이의 경우에는 잘 알려지지 않았다. 동충하초 유전체 내에서 PAL로 추정되는 768 amino acid 염기서열을 추출하였고 여기에는 PAL 발현 조절인자로 알려진 calmodulin biniding domain이 포함되어 있음을 확인하였다. calmodulin biniding site은 두 개의 칼슘이온과 결합하여 PAL 유전자를 감싸안고 비활성화 시키는 것으로 알려져 있다. 100개의 아미노산이 대략적으로 1KDa을 나타내는 바 약75KDa에 위치한 PAL 단백질을 전기영동을 통해 확인하였고 PAL 유전자의 activity를 다양한 pH와 온도조건에서 확인함으로써 최적 발현 조건을 설정하였다. CaM 기질 첨가와, W-7 저해제 첨가를 통해 Camodulin binding이 PAL 활성을 억제하는 것을 확인하였고, CaM이 binding 할 수 있는 영역을 지닌 peptide를 인공합성하여 첨가해준 결과 저해도가 줄고 PAL의 활성이 증가하는 것을 다시 확인하였다. CaM 저해제인 W-7첨가전후의 중간대사산물 정량 비교결과 이 역시 PAL의 저해로 생산되는 cinnamic acid의 양이 크게 차이나는 것을 확인할 수 있었다. 다만, 최종적인 쿠마릭산의 정량이 크게 차이나지 않는 것으로 보아 중간산물인 cinnamic acid를 이용한 또 다른 대사경로가 존재하는 것을 추정할 수 있었다. W-7이 색소에 관계한 phenylpropanoid의 대사의 저해 인자를 억제함으로써 동충하초 배양액의 색깔에도 차이를 나타내었다. Heat,cold, dark에 의한 PAL의 activity 변화와 CaM 억제에 의한 activity 회복을 확인하였고 이를 통해 PAL은 CaM에 의한 환경인자에 영향을 통해 조절되며 특히 열, 냉, 암처리에 의한 CaM 활성화 및 빛 처리에 의한 CaM 억제를 확인할 수 있었다. 이는 명배양을 통해 동충하초 자실체의 색소 생산이 증진되는 기존의 데이터와 일치한다. 이렇게 만들어진 전구물질은 phenyl-propanoid 경로와 알려지지 않은 또 다른 경로를 통해 소비되는 것으로 밝혀졌다.
제2협동과제는 현미동충하초 유해 물질을 구조동정하고 인공 합성하여 기본적인 생리활성 평가 및 제3, 4협동과제의 약리효능 연구 재료를 생산하는 역할을 수행하였다.동충하초의 에탄올, 부탄올 추출물을 조건별로 분획화하고, 각각의 분획에서 발견된 생리활성이 높은 특이 피크를 추출하여 구조동정을 수행하였다. 이를 통해 기등록된 8종과 신규 12종을 포함 총 20종의 천연물질을 구조동정하고 합성하였다. 구조동정 결과를 기초로 다양한 유도체를 합성하여 동충하초 유래 생물소재의 폭을 넓히고자 하였으며 그 결과 55종의 유도체를 추가로 생산하였다. 특히, 세포실험에서 높은 항암 활성을 보인 KTH-13물질의 유도체 합성을 통해 약 9배와 2.5배의 항암효과를 보이는 새로운 유도체를 생산할 수 있었다. 노랑다발동충하초 외에도 붉은자루동충하초와 풍뎅이동충하초에서 유래한 다양한 물질의 분리를 수행하였고, 이들에 대한 항암활성 데이터를 또한 축적하였다.
제3협동과제에서는 현미동충하초 유래 생물소재의 약리효능 평가 결과를 다루었다.먼저 번데기동충하초의 부탄올 조추출물에 대한 in vivo 처리에 의한 위염억제능을 육안으로 확인하였다. 특히 염증을 유발하는 Lipo-polysaccharide(LPS)를 처리한 후 번데기동충하초 부탄올 추출물 처리에 의해 점차 세포면역에 관계한 Phospho-I-kappa-B-alpha 단백질이 활성화됨을 확인하였습니다. 이는 붉은자루동충하초 부탄올 추출물 처리에서도 유사하게 나타났다. 풍뎅이동충하초 조추출물 처리시 부탄올 프랙션에서 높은 염증감소 효과가 확인되었는데 이는 효과는 농도 투입농도 증가와 정비례하게 증가하는 것을 확인할 수 있었다. 노랑다발동충하초의 에탄올 부탄올 추출물에 의한 염증감소효과를 확인하였고 이들의 기전을 구명하기 위한 관련 유전자의 발현양상 변화를 관찰하였다. 노랑다발동충하초에서 유래한 유망 생물소재 물질인 KTH-13의 세포사멸 효능 평가 결과 도스가 증가함에 따라 다양한 tumor cell의 apoptosis가 증가함을 확인할 수 있었다. 동충하초의 주정 추출물에 대한 아토피 치료효능을 쥐실험을 통해 형태적으로 평가했다. 인위적 염증유발에 의해 피부의 각질층에 의해 피부두께가 증가하지만 주정추출물 처리에 의해 점차 두께가 감소함을 확인할 수 있었고 IL-13, IL-4, il-6 같은 피부염 관련 전사인자가 추출물 처리에 의해 모두 감소함을 확인하였다. 또한 동충하초 유래 균독소 물질인 Beauverin 처리가 암유발 단백질이 결합하는 3차구조를 파괴하는 효과를 confocal 촬영에 의해 확인할 수 있었다. 이외에도 KTH-13의 구조를 기초로 한 다양한 analog의 암세포 사멸 효능을 스크리닝하였고,KTH-13-aniline, methylation 등 다양한 유도체의 항암 효과를 구명하고 관련 표적 단백질 구명 연구를 수행하였다.
제4협동과제는 현미동충하초 생물소재의 항바이러스 효능 평가에 관련한 연구를 진행하였다. 동충하초 조추출물의 in vivo 위암예방효눙에 대한 효능을 분석하였다. 인간위암세포주를 쥐에 이식한 xenograft 모델을 확립하고 물과 동충하초 추출물한 섭취한후 종양의 크기를 관측한 결과 동충하초 섭취시 종양 크기 생장에 어느 정도 저해를 일으키는 것을 확인할 수 있었다. 바이러스에 의한 위암 발생은 EBV 바이러스의 감염 및 증식에 의해 일어나는데 동충하초 추출물 처리 후 EBV 바이러스의 대부분의 유전자 발현이 증가하는 양상을 보여주었다. 특히 잠복감염상태의 바이러스를 활성화 시켜 더 이상의 감염을 일으키지 않도록 하는데 관여하는 BZLF1 유전자의 발현을 근소하게 증가시킴이 발견되었고 이에 대한 심도있는 연구가 필요했다.동충하초 유래의 다양한 천연화합물과 KTH-13을 비롯한 유도체 화합물을 확보하였고 기본적인 세포독성에 따른 스크리닝을 수행하여 물질 투입 도스를 결정하였다. 동충하초의 지표물질인 cordycepin 처리시 EBV바이러스 유전자의 발현 변화를 관찰한 결과, 이 단일 컴파운드는 추출물에서의 결과와는 달리 바이러스의 잠복/증식에 관련한 유전인자의 발현을 억제하는 것으로 밝혀졌다. 동충하초 추출물 중 EBV 바이러스 증식에 영향을 주는 단일물질을 찾기 위해 4개의 피크를 찾아냈는데 각각은 adenosine, uridine, guaonosine,cordycepin으로 동정되었고, 각각의 세포독성을 측정하였다. 그리고 EBV 바이러스의 주요 유전자 발현에 이들의 처리가 미치는 영향을 관찰하였다. 그 결과 uridine, guanosine, cordycepin이 유사한 패턴으로 관련 발현을 억제하는 반면, adenosine의 경우 BNRF1, BZLF 등의 유전자를 크게 활성화 시켜 추출물 중 실질적인 EBV바이러스의 암 유발 활동을 저지하는 것으로 밝혀졌다. 동충하초에서 추출물 중 adenosine이 EBV의 증식감염으로의 활성화를 통해 정상세포가 종양으로 전환시키는 기작을 저해하는 중요인자라는 것을 구명해낼 수 있었다. 본 과제 수행을 통해 22편의 SCI 논문, 12건의 산업재산권 출원, 19건의 기능성 소재 개발 등의 정량적 성과를 채웠으며 비SCI와 유전자분석 개발 실적은 추가 달성하였다.

The genus Cordyceps, which includes Cordyceps sinensis, Cordyceps militaris, Cordyceps pruinosa, andCordyceps bassiana, comprisese ethnopharmacologically valuable mushrooms in Korea, China, and Japan. The genus Cordyceps has been traditionally used for the treatment of various inflammatory and infectious diseases, such as eczema, skin diseases, chronic bronchitis, asthma, and tuberculosis, or used as a tonic for longevity, endurance, and vitality. However, in spite of their excellent pharmaceutical and nutraceutical potential, it is difficult to mass produce these mushrooms. To overcome the limitations of mass production of Cordyceps species, our group has developed an artificial culturing condition with a grain rice-enriched medium that is able to achieve mass production of fruit bodies of Cordyceps militaris, Cordyceps pruinosa, and Cordyceps bassiana under defined culture conditions.
To find useful bio-materials from Cordyceps spp., this project conducted a convergence research on genomics, bioinformatics, molecular biology, chemical analysis, pharmacology, and clinical virology. A number of genes synthesizing useful secondary metabolite including cordycepin were predicted by genome mining. Transcriptome analysis of C. militaris suggested the metabolic pathways for producing useful terpenes.
Natural products have pharmacological or biological activity that can be of therapeutic benefit in treating diseases. Cordyceps spp., Dong-Chung-Ha-Cho in Korea (winter worm and summer grass), are well known as a traditional medicinal mushroom and have been widely used for the treatment of various diseases. They contain many kinds of active components (such as cordycepin, polysaccharides, ergosterol and mannitol, etc.), and have been used due to its various physiological activities. But, among many Cordyceps species, C. bassiana has not yet been evaluated for their effective compounds. The present study aimed to identify and purify the bioactive compounds on the attributes of anti-inflammation and anti-cancer activities from artificially cultivated C. bassiana.
First, the ethanol extract and precipitates were obtained from the fruit bodies of C. bassiana and the ethanol extract was sequentially partitioned into n-hexane, n-butanol, ethyl acetate and H2O fraction. Ethanol extract, hexane and butanol fractions were effective on the anti-cancere ffects. But the anti-inflammation effect was only shown on butanol fraction. In the comparison of anti-inflammation and anti-cancer effects of C. bassiana, anti-cancer effect was much higher than anti-inflammation effect in the extracts from C. bassiana.
Next, the butanol fraction of C. bassiana (CBBF) was purified to get bioactive compounds through the purification steps using silica gel, sephadex LH-20 column chromatography and HPLC. Finally, 13 compounds were identified in the first(1st) year, 5 compounds were identified in the second (2nd) year, and 2 compounds were identified in the third (3rd)t year identified from C. bassiana. through GC-MS,LC-MS/MS, 1H-NMR and 13C-NMR.
And, after identification of compounds from C. bassiana, synthesis of each compound was carried out for the further functional tests. 13 compounds were synthesized in the first(1st) year, 22 compounds were synthesized in the second(2nd) year, and 36 compounds were synthesized in the third (3rd)t year for the extensive studies for the functional tests.
Thus, Cordyceps bassiana or compounds from Cordyceps bassiana can be used as a useful medicinal food or drug in the near future. More extensive identification of bioactive compounds from C. bassiana will be further needed for potent anti-inflammatory and anti-cancer remedies.
The inhibitory activities of the Cordyceps pruinosa butanol fraction (Cp-BF) and the Cordyceps militaris butanol fraction (Cm-BF) were investigated by determining inflammatory responses of lipopolysaccharide (LPS)-treated RAW264.7 macrophage cells and by evaluating HCl/ethanol (EtOH)-triggered gastric ulcers in mice. The molecular mechanisms of the inhibitory effects of Cp-BF and Cm-BF were investigated by identifying target enzymes using biochemical and molecular biological approaches. Cp-BF and Cm-BF strongly inhibited the production of NO and TNF-α, release of reactive oxygen species (ROS), phagocytic uptake of FITC-dextran, and mRNA expression levels of interleukin (IL)-6, inducible NO synthase (iNOS), and tumour necrosis factor-alpha (TNF)-α in activated RAW264.7 cells. Finally, orally administered Cp-BF and Cm-BF was found to attenuate gastric ulcer and block the phosphorylation of IκBα induced by HCl/EtOH. Therefore, these results suggest that the anti-inflammatory activity of Cp-BF and Cm-BF may be mediated by suppression of IKKα and its downstream NF-κB activation. Since our group has established the mass cultivation conditions by developing culture conditions for Cordyceps pruinosa and Cordyceps militaris the information presented in this study may be useful for developing new anti-inflammatory agents.Above all., we examined the anticancer activity of 5 isolated compounds derived from the butanol fraction (Cb-BF) of Cordyceps bassiana. For this purpose, several cancer cell lines such as C6 glioma, MDA-MB-231, and A549 cells were employed and details of anticancer mechanism were further investigated. Of 5 compounds isolated by activity-guided fractionation from BF of Cb-EE, KTH-13, and 4-isopropyl-2,6-bis(1-phenylethyl)phenol, Cb-BF was found to be the most potent antiproliferative inhibitor of C6 glioma and MDA-MB-231 cell growth. KTH-13 treatment increased DNA laddering, upregulated the level of Annexin V positive cells, and altered morphological changes of C6 glioma and MDA-MB-231 cells.
Cordyceps species are known to produce numerous active components and are used for diverse medicinal purposes because of their varied physiological activities, including their ability to protect the liver from damage as well as their anticancer, antidepressant, anti-inflammatory, hypoglycemic, antimicrobial effects. Cordycepin, an adenosine derivative, differs from adenosine in that its ribose lacks an oxygen atom at the 3' position. Several research groups have reported that cordycepin has antiviral activity against several viruses including influenza virus, plant viruses, human immunodeficiency virus(HIV), murine leukemia virus, and Epstein-Barr virus(EBV). In this study, we identified the epigenetic mechanisms by which cordycepin and adenosine exerted their anti-gammaherpesvirus (Epstein-Barr virus (EBV) and Kaposi’s sarcoma associated herpesvirus (KSHV)) effects. We then showed that cordycepin and adenosine possess antitumor activities against gastric carcinoma cells and endothelial cells, respectively. These results suggest that cordycepin and adenosine have antiviral and antitumor activities against gammaherpesviruses and their host carcinoma cells.
