메밀의 품종개발을 위하여 재배적 특성과 수량과의 상관관계를 분석한 결과 보통종은 초장이 길수록 주경절수와 화방수가 많았으며 분지수, 소화경수가 많을수록 그리고 개화기가 빠를수록 주당수량이 증가하였다. 고루틴, 메밀쌀 수율이 높은 쓴메밀 개발을 목표로 2013년에 교배친 93조합을 작성하여 후대 F1에 대한 생육 및 수량특성을 조사하였고, 종피열개 특성을 지닌 rice type 3계통(RTB1∼3)과 루틴함량이 높고 재배특성이 우수한 TB7 등 12계통에 대해 인공교배(2012년)를 실시하여 얻은 26조합에 대해 후대 수량특성을 조사하였다. 교배계통에 대한 선발을 통해 고루틴 계통 12조합 224개체, rice type 계통 6조합 84개체를 선발하였다.
보통메밀 품종육성을 위해 선발된 중간생태형 계통 ‘대관3-6호’, ‘대관3-7호’ 그리고 동주화계통 ‘대관3-8호’, ‘대관3-9호’, ‘대관3-10호’에 대해서 제주, 고창, 평창, 3지역에서 봄과 가을재배를 실시하여 지역적응시험과 생산력 검정시험을 실시하였다. 그 결과 ‘양절메밀’ 대비 생산력검정시험에서 4%, 지역적응시험에서 3% 수량이 높았으며 메밀쌀 수량은 지역적응시험에서 6% 높은 우수한 특성을 보였다. 품질특성으로는 표준품종인 양절메밀 대비 과피비율이 낮고 메밀 쌀비율은 73.5%로 높으며 천립중이 30.5g으로 대립종의 특성을 보였다. 대관3-6호의 종실 루틴함량은 15.4mg/100g으로 표준품종인 양절메밀과 비슷한 수준을 보였다.
메밀 연작피해 규명 및 경감 대책 개발을 위하여 메밀 특화단지(제주, 고창, 평창) 농가의 재배관리법과 수확량 등을 조사한 내용을 보면 농가의 65%가 재래종을 심고 있었으며, 97%가 2~5년 이상 연작을 하고 있었다. 수확량이 많은 농가는 1년 2작 윤작 또는 2년 이하 연작이 대부분이었으며 수확량이 적은농가는 봄, 가을 2기작 메밀재배를 포함하여 5년 이상 연작을 하는 경우가 대부분이었다. 10a당 120kg 이상의 높은 종실수량을 보인 농가는 토양관리(심토파쇄) 및 작부체계에 깊은 관심을 보인 농가였다. 농가의 수확량 증대와 연작피해를 줄이기 위해 윤작과 신품종을 재배 한 결과 메밀을 연작하고 재래종을 재배한 것보다 수량이 21% 증수되었으며 천립중도 무겁게 나타났다. 그러나 윤작의 효과인지에 대한 검토가 필요할 것으로 생각된다. 또한 연작장해를 해결하기 위한 녹비작물 재배효과를 보기위해 겨울동안 호밀을 재배하고 메밀을 재배하였으나 2015년 봄 동안 가뭄으로 초기생육이 전반적으로 부진하였으며 녹비재배에 따른 생육 및 수량이 크게 차이가 없었다. 지속적인 검토가 필요할 것으로 생각된다.메밀의 질소시비량에 따른 수량반응을 보기위해 쓴메밀 1품종(대관3-3호), 보통메밀 1품종(양절)을 이용하여 재배하였다. 질소 시비를 위해 대조로 콩 복합비료 20kg/10a, 무시비, 1.5배구에 콩복합비료 30kg/10a, 2배구에 콩복합비료 40kg/10a, 2.5배구에 콩복합비료 50kg/10a을 시용하고 생육 및 수량, 토양화학성 등을 조사하였다. 보통메밀과 쓴메밀의 시비반응이 달라 다른 관리가 필요할 것을 생각되었다. 보통메밀은 표준시비량의 1.5배 이하로 시용하는 것이 좋을 것으로 판단되었고 쓴메밀의 경우 시비량이 많을수록 수량이 증가하였으나 염류집적의 우려가 있으므로 표준시비량의 2배 이하로 시용하는 것이 좋을 것으로 판단된다.
봄메밀은 수확기에도 잎이 마르지 않아 기계수확시 지장을 초래하는 경우가 있어 이를 해결하기 위해 메밀 잎 고조제를 처리 시험을 추진하였다. 고조제는 다이쾃 디브로마이드를 사용하였고 처리시기는 수확전 3일, 7일, 10일에 처리하였고, 처리농도는 다이쾄디브로마이드 800배희석, 400배 희석, 300배 희석, 소금물(3.5%)을 처리하고 잎 마름 정도와 수량손실률을 조사하였다. 메밀은 잎이 얇고 작아서 3일처리구에서도 잎이 거의 말랐다. 고조제 처리농도가 높을수록 경엽이 빨리 말랐으며 800배로 7일전에 처리하며 잎이 충분히 마르고 수량의 손실도 없는것으로 나타났다.
메밀재배 중 비가 많이 오면 배수가 원활하지 않아 습해의 피해가 많다고 한다. 이는 파종시세조파(골뿌림)을 권장하고 있으나 농가에서는 대부분 산파(흩어뿌림)하고 있어 배수가 원활하지않기 때문인 것으로 파악이 되었다. 따라서 배수를 원활하게 하기위해 개선형산파(90cm 두둑을 만들고 산파)를 제안하였다. 2015년 봄 가뭄으로 과습이 발생하지 않아 처리구별 생육 및 수량에 큰 차이 없었다. 파종방법 개선을 통해 습해를 줄일 수 있다면 좋은 방안이라 생각된다.
본 연구는 보통메밀과 쓴메밀의 transcriptome NGS data를 이용하여 SSR 표지인자를 발굴하고 두 종의 SSR 특성을 비교 후 개발된 SSR 표지인자의 유전자원 평가 및 교배후대세대 선발 이용가능성에 대하여 조사하기 위하여 수행되었다. 또한 쓴메밀의 엽록체 염기서열을 완성하고 이를 보통메밀과 비교 분석하여 쓴메밀 구분 특이 마커를 개발하고 활용하고자 수행되었다. 쓴메밀과 보통메밀의 엽록체 유전체를 비교분석하여 약 100bp 크기의 InDel 마커를 개발하였다. 개발한 InDdel 마커와 PCR을 이용하여 분석한 결과 2번 마커를 제외한 5개의 마커는 비교유전체 분석을 통하여 얻어진 결과 일치하였다. 분석을 위해 사용된 각 8종의 쓴메밀과 보통메밀은 원산지와 관계없이 모두 구분이 가능하였다. 이는 본 연구결과에서 개발된 InDel 마커가 두 종을 구분하기 위한 마커로서의 유용성이 있음을 알 수 있었다. InDel 마커를 이용하여 보통메밀과 쓴메밀 두 종의 DNA를 이용하여 두 가지 종에 특이적 밴드 검출을 확인하였다. 보통메밀과 쓴메밀의 각 DNA 농도를 20ng/ul, 10ng/ul, 5ng/ul, 1ng/ul, 500pg/ul, 250pg/ul,100pg/ul, 10pg/ul의 조절하여 In/del-6 마커 증폭용 프라이머 세트를 이용하여 PCR을 수행하였다. PCR후 전기영동 결과 본 연구결과의 InDel 마커를 이용할 경우 10pg/ul까지도 검출이 가능함을 알 수 있어 미량이 포함된 시료에서도 활용이 가능함을 알 수 있었다. 또한 본 연구결과인 InDel 마커를 이용하여 메밀 가공품에 대한 활용성을 검토하고자 국내에서 시판중인 쓴메밀 가공품인 메밀국수 2종과 쓴메밀차 6종을 구매한 후 Macherey-Nagel(MN) 회사의 NucleoSpin Food kit를 이용하여 DNA를 추출하였다. 추출 후 20ng/ul로 정량 후 상기의 PCR조건과 InDel # 6 마커를 이용하여 PCR 수행 한 결과 보통메밀로 만든 국수(A)와 쓴메밀 100%로 만든 국수(B)를 확인 할 수 있었다. 또한 쓴메밀차 6종 중 1종(차 F)은 보통메밀과 쓴 메밀이 혼합되어 있음을 알 수 있었다.

SSR markers are only reported in common buckwheat with lower numbers based on enriched repeat libraries and Next-generation sequencing (NGS) is a cost-effective alternative. Here we report the development of new cDNA SSR (cSSR) markers characterized from transcriptome sequence data of F. esculentum and F. tatricum using NGS and these cSSR markers were applied for the validation of buckwheat germplasm. From the transcriptome sequences, a total of 8,360 and 6,518 putative cSSR markers were identified in common and Tartary buckwheat, respectively using SSR Locator. Among the cSSR markers, tetra-mer repeats are most abundant and occurred in rates of 54.9 and 37.3% in common and tartary buckwheat,respectively. To test the usefulness of newly developed cSSR markers with previously developed SSR markers based on the SSR motif enrichment library, in analysis of genetic relationship in tartary buckwheat, we selected 5 representative cSSR markers and applied to investigate the polymorphism of tartary buckwheat germplasm and found these markers can be applied in genetic diversity of germplasm with high polymorphism. Also, we report the chloroplast (cp) genome sequence of tartary buckwheat (Fagopyrum tataricum) obtained by next-generation sequencing technology and compare this with the previously reported common buckwheat (F. esculentum ssp. ancestrale) cp genome. The cp genome of F. tataricum has a total sequence length of 159,272 bp, which is 327 bp shorter than the common buckwheat cp genome. The cp gene content, order, and orientation are similar to those of common buckwheat, but with some structural variation at tandem and palindromic repeat frequencies and junction areas. A total of seven InDels (around 100 bp) were found within the intergenic sequences and the ycf1 gene. Copy number variation of the 21-bp tandem repeat varied in F. tataricum (four repeats) and F. esculentum (one repeat), and the InDel of the ycf1 gene was 63 bp long. Nucleotide and amino acid have highly conserved coding sequence with about 98% homology and four genes—rpoC2, ycf3, accD, and clpP—have high synonymous (Ks) value. PCR based InDel markers were applied to diverse genetic resources of F. tataricum and F. esculentum, and the amplicon size was identical to that expected in silico. Therefore,these InDel markers are informative biomarkers to practically distinguish raw or processed buckwheat products derived from F. tataricum and F. esculentum. We also verified these InDel markers in processed buckwheat products such as noodle and tea to identify the amount of bitter and common buckwheat.
This study was carried out for develop some of the cultivation techniques about reducing continuous cropping injury, nitrogen application settings, benefits of defoliant and sowing method for preventing moisture injury. First, we investigated status of buckwheat cultivation and yield of farms. 65% of farmers have planted native variety, 97% of them have done continuous cropping cultivation more than 2 to 5 years. It turned out farmers done continuous cropping less than two years produce greater yields and more than five years produce low yields. Farmers that harvest more than 120kg per 10a have interested in soil management (subsurface fracturing) and cropping systems. To increase yields and reduce the continuous cropping injury, we grew new variety and crop rotation planting. As a result, the yields increased 21% than continuous cultivation and growing native species. But, the result should be considered whether this is effect of the crop rotation. In addition, we cultivated a Rye to reduce the injury by continuous cropping during the winter. At the beginning of the cultivation, Buckwheat's growth was generally not good due to drought during the spring of 2015. There was no effects of a green manure crop cultivation. To investigate the quantity reaction according to the amount of applied fertilizer of buckwheat, we cultivated tatary buckwheat, common buckwheat. Nitrogenous fertilizer was processed 0, 1.5(30kg/10a), 2(40kg/10a) and 2.5(50kg/10a) times based on soy compound fertilizer 20kg/10a. The amount of fertilizer nitrogen reaction between Common buckwheat and tatary buckwheat differed from each other. Common buckwheat had more yield in 1.5 times of standard amount of fertilizer, in the case of tatary buckwheat, the more nitrogen is provided, the more yield is increased. Spring buckwheat has a problem that don't go dry the leaves at harvest time. This defoliant processing test was done to dry its leaves because it caused problems when mechanical harvesting. Diquat dibromide was used for defoliant and it was processed 800, 400, 300 times as much as diluted, salt water(3.5%) before harvest for three, seven, ten days ago.Buckwheat's leaves almost dried at three days treatment plot because it was thin and small. The more defoliant concentration is high, the more the leaves dry quickly. It turned out that the leaves dry enough and there is no loss of water when the defoliant is processed the solution 800 times seven days ago. During the cultivation, heavy rain cause excess-moisture injury because drainage is not smooth. To prevent it, drill seeding was encouraged but farmers mostly have done broadcast planting Improved broadcast planting was suggested and tested for smooth water drainage. But there was no excess moisture, difference of growth and quantity because of drought at 2015 spring.
