Ⅳ. 연구개발결과
○ 형광실크누에의 분자생물학적 특성분석 및 표준품 작성(’13~’15)
- 본 연구에서는 기 개발 녹색형광실크 형질전환누에와 국립농업과학원이 보유하고 있는 유색견 누에 유전자원을 이용하여 교차교배를 통하여 실용형질과 녹색형광이 우수한 녹색형광실크를 생산하는 표준품 GM누에 개발하고 이의 특성을 분석함.

Ⅳ. 연구개발결과
○ 형광실크누에의 분자생물학적 특성분석 및 표준품 작성(’13~’15)
- 본 연구에서는 기 개발 녹색형광실크 형질전환누에와 국립농업과학원이 보유하고 있는 유색견 누에 유전자원을 이용하여 교차교배를 통하여 실용형질과 녹색형광이 우수한 녹색형광실크를 생산하는 표준품 GM누에 개발하고 이의 특성을 분석함.
- 녹색형광실크 표준품 GM누에에 도입된 유전자의 연역 아미노산 서열을 바탕으로 기존 알레르겐 데이터와 상동성 분석한 결과(FAO/WHO rule), 녹색형광실크 GM누에로부터 발현하는 단백질의 알레르겐 유발 가능성은 매우 낮은 것으로 확인됨.
- 녹색형광실크 표준품 GM누에 추출물을 일반누에 추출물과 비교하여 기니피그를 대상으로 피부감작성 분석한 결과 GM누에에 의한 피부 알레르겐 및 기타 이상소견을 발견할 수 없었음.
- 표준품 GM누에 추출물을 이용한 토양미생물환경영향 평가의 일환으로 토양미생물의 밀도변화 및 곰팡이와 박테리아 군집상 변화를 분석한 결과, GM누에 실험구는 무처리한 실험구와는 차이가 있었으나 일반누형에처리구와는 유의적 차이가 없이 안전함이 확인됨.
○ 형광실크누에의 안전관리 및 위해성평가 기술 개발(’13~’15)
- 형광실크누에 생물학적으로 집누에의 특성을 가지고 있다. 즉, 이미 가축화 되어 있는 생물체이다. 이동성이 현저하게 둔화되어 외부로 탈출 자체가 불가능에 가깝다. 먹이를 찾는 능력인 포집성도 약하여 사람이 먹이를 주지 아니하면 도태되어 죽게 된다. 다시 말하여 누에나방의 유충단계에서 만에 하나 발생할 수 있는 비의도적으로 환경 노출 또는 유출 시에도 번데기에서 성충으로 탈피할 수 있는 가능성은 거의 없다고 볼 수 있다.
- 밀폐(Contained Use)수준이 아닌 격리(Confined Use)수준에서 사육을 한다고 하여도 LMO에 의한 피해를 예방할 수 있는 안전관리가 충분하다고 할 수 있다.
- 본 연구에서는 형광실크누에의 사육단계에서는 현재 농촌진흥청에서 보급하고 있는 “표준잠실”을 형광실크누에 사육시설로 권장하고 있다. “표준잠실”은 누에유충이 생장할 수 있는 최적의 조건을 만족할만한 시설 수준을 마련하고 있으며, 이 표준잠실에서 형광실크누에를 사육할 경우에는 본 연구에서 제안하고 있는 안전관리 운영규정을 준수하면 농가에서 안전하게 형광실크누에를 사육할 수 있을 것으로 본다.
- 다만, 이를 확인하기 위한 환경방출실험을 추가로 제안한다. 환경방출실험은 유전자변형생물체의 위해성심사 전에 거의 마지막에 수행하는 평가단계로서 실제 “표준잠실”에서 일정기간 형광실크누에를 사육하며 제시된 안전관리지침을 수행하면서 발생할 수 있는 여러 안전사고의 가능성을 확인하는 단계이다.
- 위해성평가 및 심사를 통해 유전자변형생물체인 형광실크누에가 위해성이 없음을 확인함과 동시에 환경방출실험을 통하여 시설 및 운영의 안전관리를 확보하여 바이오안전성에 대한 이중의 대책을 마련할 수 있게 된다.
○ 형광실크누에의 인체위해성평가 기술 개발(’14~’15)
- DsRed2 및 EGFP 단백질의 대량정제 : 형광실크누에의 인체위해성평가 항목 중 단회투여독성 및 물리화학적 안정성 시험 등을 수행하기 위하여 대체산물로서 대장균 발현계를 이용하여 대량정제 시스템을 확립하였다. 대량 정제된 단백질은 SDS-PAGE 및 Western blot 분석방법으로 순수한 DsRed2 및 EGFP 단백질을 확인할 수 있었다
- 숙주와 대체단백질간의 구조적·기능적 동질성 평가 : 형광실크누에의 도입단백질과 대장균을 이용해 발현시킨 도입단백질간의 구조적 기능적 동질성을 평가하기 위해 형광실크 누에의 발현단백질 및 대량 정제 단백질의 아미노산 서열의 번역 후 변이 유무를 관찰하였으며, N-말단 아미노산 서열 분석 및 펩타이드 질량분석을 수행하였다. 그 결과 형광실크누에 및 대장균에서 만들어진 EGFP 단백질의 경우 단백질 생성 후 구조적인 변이(glycosylation)가 일어나지 않았음을 확인할 수 있었으며, 대장균에서 발현시킨 DsRed2 및 EGFP 단백질의 N-말단 아미노산 서열 분석결과 DsRed2의 경우 두 번째 아미노산부터 EGFP의 경우 첫 번째 아미노산 서열부터 10개의 잔기가 동일하게 분석되었다. 또한 DsRed2 및 EGFP 단백질의 펩타이드 질량 분석 결과 대장균에서 발현시킨 DsRed2의 경우 약 29824 m/z 이온이 확인되었으며 EGFP의 경우 26464 m/s 이온 값이 검출되었으나 형광실크누에에 도입된 EGFP의 경우 펩타이드 질량 분석을 통해 질량값을 확인할수 없었다.
- DsRed2 및 EGFP 단백질의 물리화학적 안정성 검정 : 형광실크누에에 도입된 DsRed2 및 EGFP 단백질이 소화능과 열에 대하여 안정성을 가지는지 확인하기 위하여 인공위액,인공장액 및 열처리를 한 결과 인공위액에서는 두 단백질 모두 5초 이내에 완전히 분해됨을 확인할 수 있었고, 인공장액에서는 EGFP의 단백질은 5초 이내에 완전히 분해되었으나, DsRed2의 경우 분해되지 않는 양상을 보였다. 하지만 DsRed2의 경우 인공위액효소에서 5초 이내에 완전히 분해되어 소화경로상에서 쉽게 분해가 되기 때문에 소화과정후 독성을 나타낼 가능성은 낮은 것으로 판단되었다. DsRed2 및 EGFP 단백질의 열처리 후 형광강도를 측정한 결과 열을 가한지 10분 안에 급속한 감소를 보이며 95% 이상의 단백질이 분해되어 두 단백질 모두 열에 대한 안정성이 낮은 것으로 확인하였다.
- 형광실크누에 EGFP 단백질의 발현시기, 발현량 평가 : ELISA 분석을 통해 성장시기별 형광실크누에의 도입단백질 발현량을 측정한 결과, 형광실크누에 1령, 2령, 3령, 4령, 5령,번데기에서는 도입단백질이 확인되었으며, 알과 인편에서는 도입단백질이 확인되지 않았다. 또한 형광실크누에 도입 4령 애벌레에서는 도입단백질의 양이 1령, 2령, 3령, 5령,번데기에 비하여 낮게 발현되는 것을 확인하였다.
- EGFP 단백질의 단회투여 독성 평가 : 암 수 ICR 마우스에 대한 단회 경구 투여 독성평가를 실시한 결과 마우스에 대한 최소 치사량은 2 g/kg 이상의 고용량이며, 2 g/kg투여 용량에서도 임상증상 및 부검소견 등에서 특별한 이상소견은 전혀발견되지 않아 EGFP 단백질은 급성독성의 측면에서 매우 안전한 물질인 것으로 판단되었다.
- 형광실크누에의 유전독성 평가 : Ames test에 따라 형광누에고치 분말과 5령의 형광누에 동결건조가루에서 복귀돌연변이시험을 진행한 결과 복귀돌연변이 독성이 없는 것으로 확인되었다.
○ 형광실크누에의 환경위해성평가 기술 개발(’14~’15)
- 숙주생물체 (이하 집누에나방 혹은 집누에) 유충 사육과정에서 환경위해성 평가
․ 집누에 사육 시작은 신석기시대 (기원전 1,600~1,700년경)로 알려져 있다
․ 집누에의 학명은 Bombyx mori이며, 곤충강, 인시목, 집누에나방과 집누에나방속에 속한다
․ 집누에는 자발적인 먹이탐식능력이 대부분 상실되었다.
․ 집누에 유충은 자연환경 조건에서는 이동성이 극히 낮고 생존능력이 거의 없다.
- 집누에나방 누에씨 생산 및 관련된 환경위해성 평가
․ 집누에나방 성충의 생존일수는 10일 내외이었다.
․ 성충은 암수 모두 자발적으로 날아오르지 못하며, 비행능력도 상실되었다.
․ 성충은 자발적으로는 날 수 없으므로 생태계 및 환경에 미치는 영향은 무시할 수 있다.
- 근연종과의 생물 관계성 평가
․ 집누에의 가장 근연종은 동일한 속(屬)에 속해있는 멧누에나방, Bombyx mandarina(이하 멧누에) 이다.
․ 멧누에는 극동아시아지역에 서식하는 완전 야생성 곤충이며, 뽕잎을 기주식물로 하는 등 생리적으로 집누에와 유사하다.
․ 생리적으로 멧누에나방은 집누에나방과 교차생식이 가능하다. 즉 종간잡종을 만들 수 있으며 잡종 제1대는 생식력을 갖는다.
․ 자연상태에서 집누에나방은 이동성이 거의 없으므로 특별한 경우가 아니면 두 종 사이의 교차생식은 거의 이루어지지 않는다.
- 집누에나방과 형질전환누에의 생물학적 차이
․ 형질전환누에는 집누에와 마찬가지로 야외환경에서 생존이 거의 불가능하다
․ 형질전환누에는 몇 가지 실용형질에서 근소한 차이를 나타내지만 근본적으로 집누에와 동일하다.
- 집누에나방과 형질전환누에와의 환경 영향 평가
․ 형질전환누에 유충을 고압멸균 및 소각처리 한 경우에 표지유전자는 불활화 되었다.
․ 잠사류 (사육폐기물)의 토양 매립 1개월 이후부터는 표지유전자가 불활화되었다.
․ 잠사류 (사육폐기물)가 매립지 토양에 미치는 화학적 및 생물학적 영향은 없는 것으로 나타났다.

The risk assessment and safety management of living modified organisms(hereafter referred to as the “LMO”) must be conducted in accordance with the Act on Transboundary Movement of Living Modified Organisms (hereunder referred to as the“LMO Act”), the domestic law set in place to implement the Cartagena Protocol on Biosafety, which is an act supplementing the Convention on Biological Diversity.Fluorescent silkworms are living modified organisms and therefore relevant risk assessment and safety management work must be conducted in adherence to the LMO Act. According to the LMO Act, initial risk assessment must be conducted by the developer, and based on that assessment data, the government conducts the risk review to confirm safety of the pertaining materials. To produce fluorescent silkworm cocoon, those entities that wish to rear fluorescent silkworms can raise only those of which safety has been confirmed by a risk review.
In recently, many kinds of GM silkworm were developed in Korea that can produced a different kind of hybrid cocoons fused fluorescent proteins. But, the proper risk assessment and management of the GM(Genetically Modified) silkworm have not been executed yet in Korea. According to the LMO Act of Korea, the risk assessment is essential to the commercialization of these GM silkworms. Therefore, it is necessary for mass production of the GM silkworm to demonstrate whether it could effect the risk on human health or environment in compared with the non-GM silkworm. This study therefore was performed with four subjects, a standardization of GM silkworm producing the green fluorescent cocoon, a analysis of human health and environmental risk assessments, and a development of safety management of the GM silkworm.
1. A standardization of GM silkworm producing the green fluorescent cocoon.
We developed a GM silkworm producing a chimeric cocoon fused a green fluorescent protein(EGFP) in previous study, but the GM silkworm has decreased in the convenience of breeding and the productivity of silk. Commercial feasible traits of the GM silkworm is significantly low. Therefore, to improve the practical traits of the GM silkworm, we have performed a cross-inbreeds between the green fluorescent GM silkworm and some strains of colored-cocoon’s silkworms. In the result, we founded the most valuable cross fertilization manner is in the GM silkworm female(T59B) and non-GM silkworm male(Jam126), and then its offspring(F1) was named as the standard GM silkworm producing the green fluorescent cocoon. We analyzed the genetical and optical characteristics of the GM silkworm as well as hybrid cocoon protein. A human health risk assessment of the standard GM silkworm was conducted by skin sensitization test with guinea pig. The results of skin reactions and common symptoms observation were not differ between GM silkworm and non-GM silkworm. An environmental effect of the GM silkworm on the soil was performed as environmental risk assessment. in the result, there was no significant difference in microbe population between the GM silkworm and non-GM silkworm.
2. Development of safety management guidelines of GM silkworm.
The purpose of this study is to present techniques and methodology for the preparation of risk assessment data for fluorescent silkworms, and to provide standard safety management guidelines that can prevent damage to humans and the environment during the stage of farm rearing after the risk review. The evaluation of the impact of fluorescent silkworms on humans and the environment was prioritized as the scope of this study. The ultimate aim of developing fluorescent silkworms is to distribute them to farms, which is why this study is focused on the development of assessment guidelines for the production of data that enables one to judge whether the fluorescent silkworm is safe enough to be grown in farms and what level of safety management must be conducted in the process. In particular, a differentiation was made, according to the life cycle of the silkworm moth,between the stage at which it can be grown at farms, and at which it cannot. The former is the rearing stage, while the latter is the silkworm’s egg stage, and this study proposes safety management guidelines at the rearing stage. Biologically, the fluorescent silkworm has the characteristics of the mori silkworm. In other words, it is an organism that has already become domesticated. As its mobility is conspicuously slowed, escape is almost impossible. The ability of the silkworm to find food is weak, and it cannot prevail and survive without being fed by humans. Even in the case of unintentional exposure to or leakage into the environment, which can only occur at the larva stage of the silkworm moth, there is almost no likelihood that the larva can become a pupa or emerge into an imago. In other words, even if the silkworm is grown at the “confined use” level, rather than the “contained use” level, there are sufficient safety management mechanisms that prevent damage from the LMOs. In this study the standard silkworm-rearing room that is currently being supplied by the Rural Development Administration is recommended as the breeding facilities of the fluorescent silkworm at its rearing stage. The standard silkworm-rearing room provides facilities at levels that satisfy optimum conditions enabling the growth of silkworms, and we believe farms shall be able to grow the fluorescent silkworm safely if they adhere to the safety management and operation provisions proposed by this study. The results of this study shall be used for the establishment of safety management guidelines for the breeding of the fluorescent silkworm in farms, and can be applied not only to the fluorescent silkworm, but for farm breeding of silkworms that produce protein that are beneficial to humans. When considering the fact that the fluorescent silkworm is an insect that has already been domesticated, these guidelines can be used for provision of safety management guidelines for the breeding of genetically modified animals.
3. Human health risk assessments of GM silkworm producing florescent silk
Silk has had a reputation as a luxurious and sensuous fabric but it is not popular due to the expensive price and poor durability. To develop the silk materials that apply the various industries, the artificially synthesized gene can be introduced into the silkworm and expressed in the silk gland. Transgenic silkworms for the mass production of green fluorescent silks are generated using a fibroin H-chain expression system. For commercial use, safety assessment of the transgenic silkworms is essential. Based on the regulations of safety assessment, we have evaluated physical and chemical stability and functional equivalence. First, we tried to develop the separation and large-scale purification of EGFP and DsRed2 proteins. To evaluate their toxicity, we showed that EGFP and DsRed2 could be degraded in intestine and stomach from the test using artificial intestinal and gastric juices, so that EGFP and DsRed2 may be easily digested in 5 seconds through stomach not intestine. Also, in vitro heat stability tests have shown that introduced proteins exposed to high temperatures lose biochemical function. From the studies, we concluded EGFP and DsRed2 expressed in transgenic silkworm dose not induce allergen when proteins made by transgenic silkworms was taken in.
4. Environmental risk assessments of GM silkworm producing florescent silk
Breeding the begin with the silkworm is known as the Neolithic era (about BC 1600-1700 years), scientific name is Bombyx mori, Insecta : Lepidoptera : Bombysidae : Bombyx mori L. Silkworm is lost almost spontaneous phagocytic capacity, the larvae in the natural environment conditions, mobility is very low, there is almost no viability. Silkworm adult survival days of is about 10, male and female both flight capacity is degenerate, it can not fly. Since the adults can not spontaneously fly, it is possible to ignore the impact on the ecosystem and the environment. The most Allied species of silkworm is a belong to the same genus wild-silkworm (Bombyx mandarina). Wild-silkworm is a wild-type insects that live in the Far East Asia region, and the mulberry leaves a host plant, similar to the physiologically silkworm. Physiologically wild-silkworm make interspecific hybrids are cross reproductive silkworm, with a hybrid first generation fertility. Since silkworm has little mobility in a natural state, cross reproductive between the two species is little effected. Transgenic silkworm is almost impossible to survive in the outdoor environment as silkworm, although there is a slight difference in some of the practical traits, which is fundamentally the same. When the larvae transformed silkworm autoclaved and incineration, the marker gene is inactivated. The soil from fille-in of breeding waste after a month, marker gene has been inactivated. Breeding waste did not have chemical and biological effects on the reclaimed land of the soil.
